Effect of Temperature on Reproduction and Development of Rice Water Weevil, Lissorhoptrus oryzophilus (Coleoptera: Curculionidae) (벼물바구미의 생식과 발육에 미치는 온도의 영향)

  • This study was carried out to test the effects of temperatures between 2$0^{\circ}C$ and 3$0^{\circ}C$ on the reproduction and development of the rice water weevil, Lissorhoptrus oryzophilus. Preoviposition periods were much longer (17.2 to 51.0 days) in the overwintering adult females collected in March than those collected in May, regardless of temperature. Oviposition periods, however, were longer (16.9 to 22.0 days) in the adult females collected in May than those collected in March at the same temperatures. The longer oviposition period observed in the females collected in May were directly associated with higher fecundity. Egg periods were shortened from as temperature increased, but the hatching rate was highest (100%) at 27$^{\circ}C$. The developmental periods from egg to adult were shortened as temperature increased : from 77.9 days at 2$0^{\circ}C$ to 38.3 days at 3$0^{\circ}C$. The developmental zero point temperature (T) and the total effect temperature (K) for egg were 16.3$^{\circ}C$ and 62.1 dgree days, respectively. The T and K from egg to adult emergence were 13.9$^{\circ}C$ and 577.6 dgree days, respectively. The adult females of the first generation did not oviposit at 2$0^{\circ}C$, but did at $25^{\circ}C$ and 3$0^{\circ}C$. The intrinsic rate of natural increase (r$_{m}$) increased as temperature augmented. Net reproductive rate (Ro) per generation was highest (75.3) at $25^{\circ}C$.>.

Conservation status assessment of archaeological bone from Fourier Transform Infrared Spectroscopy and histological Analysis (적외선 분광 분석과 조직 분석을 통한 출토 인골의 보존 상태 평가)

  • Fourier transform infrared spectroscopy(FTIR-ATR) was applied to chemical analysis for conservation status of 10 human bone remains from Joseon Dynasty. The result of crystallinity index (CI) is $4.25{\pm}0.78$, carbonate to carbonate ratio (C/C) is $0.91{\pm}0.04$ and cabonate content (C/P) is $0.19{\pm}0.06$. The higher histological index (HI) confirmed CI and C/P value was increased and C/C value was reduced. While C/C or C/P values analysis is possible. While DNA analysis can be extracted from the bone, C/C values are lower or C/P values are higher was found to the analysis is possible. Chemical assessment of FTIR and histological index consequence is expected to be applicable as a basis for comprehensive understanding of the conservation status excavated bones.

  • X-linked adrenoleukodystrophy (X-ALD) is a rare peroxisomal disorder, that is rapidly progressive, neurodegenerative, and recessive, and characteristically primary affects the central nervous system white matter and the adrenal cortex. X-ALD is diagnosed basaed on clinical, radiological, and serological parameters, including elevated plasma levels of very long chain fatty acids (VLCFA), such as C24:0 and C26:0, and high C24:0/C22:0 and C26:0/C22:0 ratios. These tests are complemented with genetic analyses. A 7.5-year-old boy was admitted to Department of Pediatrics, Chungnam National University Hospital with progressive weakness of the bilateral lower extremities. Brain magnetic resonance imaging confirmed clinically suspected ALD. A low dose adrenocorticotropic hormone stimulation test revealed parital adrenal insufficiency. His fasting plasma levels of VLCFA showed that his C24:0/C22:0 and C26:0/C22:0 ratios were significantly elevated to 1.609 (normal, 0-1.390) and 0.075 (normal, 0-0.023), respectively. Genomic DNA was extracted from peripheral whole blood samples collected from the patient and his family. All exons of ABCD1 gene were amplified by polymerase chain reaction (PCR) using specific primers. Amplified PCR products were sequenced using the same primer pairs according to the manufacturer's instructions. We identified a missense mutation (p.Arg163Leu) in the ABCD1 gene of the proband caused by the nucleotide change 488G>T in exon 1. His asymptomatic mother carried the same mutation. We have reported an unpublished mutation in the ABCD1 gene in a patient with X-ALD, who showed increased ratio of C24:0/C22:0 and C26:0/C22:0, despite a normal VLCFA concentrations.

Discrimination of the geographical origin of commercial sesame oils using fatty acids composition combined with linear discriminant analysis (지방산 조성과 선형판별분석을 활용한 유통판매 참기름의 원산지 판별)

  • In this study, the fatty acid (FA) composition of commercial sesame oils (n = 62) was investigated using gas chromatography with flame ionization detector (GC-FID). Multivariate statistical techniques, including principal component analysis (PCA) and linear discriminant analysis (LDA), were applied to the chromatographic data of the FAs to discriminate the geographical origin of sesame oils. A statistically significant difference was observed in the content of C16:0, C18:0, C18:1, and C18:2 between domestic and imported sesame oils. A satisfactory recovery rate of 82.8-100.2 % was achieved for C16:0, C18:0, C18:1, C18:2, and C18:3. The correlation of C16:0, C18:1, and C18:2 in domestic sesame oils showed opposite trends compared to imported oils. The PCA plot demonstrated that sesame oils were clustered in distinct groups according to their origin. LDA was used to predict sesame oil samples in one of the two groups. C16:0 (Wilks λ = 0.361) and C18:1 (Wilks λ = 0.637) demonstrated the highest discriminant power for classifying the origin of the samples. The correct prediction rates were 88.9 % and 100 % for the domestic and imported samples, respectively. Further, 60 of the 62 sesame oil samples (96.8 %) were correctly classified, indicating that this approach can be used as a valuable tool to predict and classify the geographical origin of sesame oils.

  • We studied to find out the effect of water temperature on the egg development of pearl oyster, Pincata fucata martensii and Pacific oyster, Crassostrea gigas. The optimum water temperatures for egg development were $20{\~}25^{\circ}C$ in P. fucata martensii and $15{\~}25^{\circ}C$ in C. gigas. The required time from fertilization to D-shaped lana was $41.7\;hours\;at\;20^{\circ}C$ and 27.5 hours at $25^{\circ}C$ in P. fucata martensii, and 35.3 hours at $15^{\circ}C$, 26.3 hours at $20^{\circ}C$ and 17.6 hours at$ 25^{\circ}C$ in C. gigas, respectively.   • Effect of predation of starfish Asterias amurensis and Asterina pectinifera under different water temperature in the recirculating seawater vessel was conducted at the invertebrate culture laboratory of Yosu National University from February 1 to February 25, 2000. The highest activity of Asterias amurensis and Asterina pectinifera according to water temperature was obtained at 14$\^{C}$ and 22$\^{C}$. The average individual numbers of Anadara broughtonii predated by Asterias amurensis and Asterina pectinifera were 2.20 and 1.60 at 6$\^{C}$ , 3.60 and 2.00 at 10$\^{C}$, 7.20 and 2.50 at 14$\^{C}$, 3.40 and 6.80 at 22$\^{C}$, 0.80 and 3.90 at 26$\^{C}$, respectively.

  • The study was conducted to find out the gene effects on body weights at weaning and at the age of 60 days in mice, with 343 progenies from full-dialell crosses of four lines of BALB/c, CBA, C3H and C57BL. The progenies were reared at the Experimental Animal Farm, College of Agriculture, Kyungpook National University from November, 1984 to February, 1985, and data collected from the progenies were analyzed into general combining ability, maternal effects, specific combining ability and reciprocal effects with Harvey's model. General combining ability effects estimated in line-crosses were -0.6033~0.5298 for weaning weights and -0.5086~1.0012 for body weights at the age of 60 days. General combining ability for BALB/c and C57BL were significantly better than general combining ability for CBA and C3H for both traits (P<0.05). Maternal effects for C3H were significantly larger than the maternal effects of BALB/c for both traits (P<0.05). The estimates of maternal effects were -0.9678~0.4609 for weaning weights and -1.1886~0.0729 for body weights at the age of 60 days. Specific combining ability effects were estimated to be significant (P<0.05), and the estimates were -0.1999~0.3380 for weaning weights and -0.4056~0.3317 for body weights at the age of 60 days. Reciprocal effects were found to be largest in BALB/c${\times}$C57BL and BALB/c${\times}$C3H. The estimates were -0.5049 from BALB/c${\times}$C57BL and 0.4972 from BALB/c${\times}$C3H form weaning weights, and -1.0336 from BALB/c${\times}$C57BL and 1.2793 from BALB/c${\times}$C3H for body weights at the age of 60 days.

Effects of Full-Fat Soybeans and Linseed as Dietary Fat Sources on In Vitro Ruminal Disappearances of Dry Matter and C18-Unsaturated Fatty Acids and Fatty Acids Profile (지방원으로 전지대두와 아마종실의 첨가가 반추위내 건물과 C18계-불포화지방산의 조성과 소실율에 미치는 영향)

  • This study was conducted to investigate the effects of dietary full-fat soybeans and linseed as fat sources on in vitro ruminal disappearances of dry matter and unsaturated fatty acids and fatty acids profile. The full-fat soybeans and linseed were high in linoleic acid (C18:2n-6) and $\alpha$-linolenic acid (C18:3n-3), respectively. The incubation times were 0, 3, 6, 12, 24, 48 and 72 h. After each time of incubation, medium digesta was lyophilized for analyzing its DM and fatty acids contents. DM disappearance was significantly higher in linseed treatment compared to full-fat soybeans treatment on 6 h (p<0.01), 12 h (p<0.05) and 24 h (p<0.01), but cumulative gas production was not significantly different between both treatments. Stearic acid (C18:0) content in medium digesta was increased in both soybeans and linseed as a result of complete biohydrogenation with increased incubation time and C18:0 and C18:1 contents of full-fat soybeans were significantly higher than those of linseed (p<0.05). The content of C18:2 and C18:3 in digesta of each treatment were decreased by biohydrogenation as incubation time was increased. The content of C18:2 in full-fat soybeans was significantly higher than that of linseed (p<0.05) while the content of C18:3 in linseed was significantly higher than that of full-fat soybeans (p<0.001). Net C18:0 production was significantly higher in full-fat soybeans (332.24%) than linseed (133.16%) on 72 h. Disappearance of C18:1 was significantly lower in full-fat soybeans than linseed (p<0.05), especially full-fat soybeans showed negative (-) values on 3, 6, 12 and 24 h. The disappearance of C18:3 was significantly higher in linseed than full-fat soybeans (p<0.05). The disappearance of C18-unsaturated fatty acid was significantly higher in linseed than full-fat soybeans. In conclusion, polyunsaturated fatty acid (PUFA) in both full-fat soybeans and linseed were extensively biohydrogenated. In addition, biohydrogenation of PUFA was more completed to C18:0 in full-fat soybeans than linseed, reflecting dietary PUFA composition.

  • Stability of the anthocyanin pigment isolated from 'Comet' radish was decreased with increase of pH from 1.0 to 8.0. The destruction rate of anthocyanin at early stage was faster at pH 1.0 than pH 2.0 and 3.0, increased with increase of storage temperature from $0^{\circ}C\;to\;40^{\circ}C$. Thus the pigment preservability at $0^{\circ}C\;and\;40^{\circ}C$ after 10 days was 92.5% and 48.1%, and the half life at $0^{\circ}C\;and\;40^{\circ}C$ was 94.9 and 12.1 days, respectively Pigment was decreased with increase of ascorbic acid content from 0.015g to 0.05g.

Effects of Green Tea Catechin on Serum Lipid Composition and Angiotensin- Converting Enzyme Activity in Rats Exposed to Microwave

  • This study aims to investigate the changes in the lipid composition and angiotensin-converting enzyme activity in serum of rats exposed to microwave radiation, and to examine improving effects of green tea catechin to the lipid composition. The microwave-exposed rats received the normal and one of 3 diets: catechin free (MW-0C), 0.25% catechin (MW-0.25C) or 0.5% catechin (MW-0.5C). Rats were sacrificed 6th day after microwave radiation (2.45 ㎓, 15 min). The concentration of serum triglyceridein MW-0C group was increased by 85%, compared with the normal group, but that of MW-0.25C group with 0.25% catechin supplementation was to 17% lower, compared with the MW-0C group. There was no significant difference between normal group and MW-0.5C group. Total-cholesterol and LDL-cholesterol concentrations were increased by 21.4% and 38.6%, respectively, by microwave irradiation. The concentration of HDL-cholesterol in MW-0C group was lower to 29.3%, but it was maintained at the normal level by catechin supplementation. There was no significant difference among four groups in HLD-cholesterol/LDL-cholesterol. Atherogenic index in MW-0C group was increased by 56%, compared with the normal group. Angiotensin-converting enzyme (ACE) activity in lung tissue of MW-0C group was increased by 59 %, while that of MW -0.25C and MW -0.5C group were maintained at the normal level. ACE activity of MW-0C group in serum was increased by 122.8%, compared with the normal group. Catechin supplementation group was significantly reduced, compared with the MW-0C group ACE activity. In conclusion, microwave irradiation increased the serum triglyceride and cholesterol concentrations and ACE activity which are considered as the blood pressure increasing agents. However, catechin supplementation decreased the level of triglyceride, total cholesterol, LDL cholesterol, and ACE activity, which maybe consider catechin as being agent of lowering effect for blood lipid profile for athero-genesis.
