The radioprotective effects of radices herbs (대표적 근류 생약의 방사선 방호효과)

  • We performed this study to determine the effect of Jiegeng(Platycodon grandiflorum), Danggui(Angelica sinensis), Gancao(Glycyrrhiza glabla), Chaihu(Bupleurum falcatnosa), Shoudehuang(Rehmannia glutinosa), Huangqi(Satragalus membranaceus), Muxiang(Saussurea lappa), Yuanzhi(Polygala tenuifolia), Rensen(Panax ginseng) and Baishaoyao(Paeonia lactiflolia), as Oriental radices herbs, on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of ${\gamma}-radiation$. Jiegeng(p<0.005), Danggui(p<0.0005), Gancao(p<0.005), Chaihu(p<0.05), Muxiang(p<0.05), Rensen(p<0.005) and Baishaoyao(p<0.005) were effective in intestinal crypt survival. Danggui(p<0.05), Chaihu(p<0.05), Shoudehuang(p<0.05), Huangqi(p<0.05), Rensan(p<0.005) and Baishaoyao(p<0.05) increased the formation of endogenous spleen colony. The frequency of radiation induced apoptosis was also reduced by pretreatment with Chaihu(p<0.05), Muxiang(p<0.005), Yuanzhi(p<0.05), Rensan(p<0.05) and Baishaoyao(p<0.05). Although the mechanisms of this effect remain to be elucidated, these results indicated that Danggui, Chaihu, Muxiang, Rensan and Baishaiyao might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • To enhance functional properties of 4 different hoki frame protein hydrolysates (30K, 10K, 5K and 1K hydrolysate) fractionated through a series of 30, 10, 5 and 1 kDa molecular weight cut-off membranes in order to decrease pore size, all hydrolysates were phosphorylated with sodium trimetaphosphate and altered phosphorylated 30K, 10K, 5K and 1K (P-30K, P-10K, P-5K and P-1K), respectively. The covalent attachment of anionic phosphate groups to polypeptide chains improved the functional properties, such as solubility, emulsifying properties and foaming properties, of hoki frame protein hydrolysates. Especially, P-30K hydrolysate with the highest molecular weight fraction possessed the most excellent functional properties among 4 different phosphorylated hydrolysates.

Comparison of Gamma Irradiation and Sodium Hypochlorite Treatments to Inactivate Staphylococcus aureus and Pseudomonas aeruginosa Biofilms on Stainless Steel Surfaces

  • Biofilm formation on various surfaces is a well-known phenomenon and it has caused pollution problems, health and safety hazards, and substantial economic loss in many areas including the food industry. In the present study, Gamma irradiation at a dose of 2.0 kGy reduced the bacterial counts of Staphylococcus aureus and Pseudomonas aeruginosa suspensions by 6.7 and >6.5 log CFU/mL, respectively, and 30 ppm of sodium hypochlorite effectively reduced the counts of both bacterial suspensions to below the limit of detection ($<2\;log\;CFU/cm^2$). However, in bacterial biofilms attached to stainless steel, gamma irradiation at a dose of 10.0 kGy reduced the counts of S. aureus attached fur 1 hr and overnight by ${\geq}5.1\;and\;5.0\;log\;CFU/cm^2$, respectively. Gamma irradiation at a dose of 1.0 kGy reduced the counts of P. aeruginosa counts to below the limit of detection ($<2\;log\;CFU/cm^2$). On the contrary, S. aureus and P. aeruginosa cells attached to stainless steel chips were difficult to eliminate using sodium hypochlorite. Four hundred ppm of sodium hypochlorite reduced the counts of S. aureus and P. aeruginosa attached for 1 hr by 2.5 and $3.3\;log\;CFU/cm^2$, respectively.

Buckling analysis and optimal structural design of supercavitating vehicles using finite element technology

  • The supercavitating vehicle is an underwater vehicle that is surrounded almost completely by a supercavity to reduce hydrodynamic drag substantially. Since the cruise speed of the vehicle is much higher than that of conventional submarines, the drag force is huge and a buckling may occur. The buckling phenomenon is analyzed in this study through static and dynamic approaches. Critical buckling load and pressure as well as buckling mode shapes are calculated using static buckling analysis and a stability map is obtained from dynamic buckling analysis. When the finite element method (FEM) is used for the buckling analysis, the solver requires a linear static solver and an eigenvalue solver. In this study, these two solvers are integrated and a consolidated buckling analysis module is constructed. Furthermore, Particle Swarm Optimization (PSO) algorithm is combined in the buckling analysis module to perform a design optimization computation of a simplified supercavitating vehicle. The simplified configuration includes cylindrical shell structure with three stiffeners. The target for the design optimization process is to minimize total weight while maintaining the given structure buckling-free.

The radioprotective effects of Bu-Zhong-Yi-Qi-Tang and its major ingredients in irradiated mice (방사선 피폭 마우스에서 보중익기탕 및 구성단미의 효과)

  • We performed this study to determine the effect of Bu-Zhong-Yi-Qi-Tang, as a prescription of traditional Oriental medicine, and its major ingredients on jejunal crypt survival, endogenous spleen colony formation, apopotosis in jejunal crypt cells, lethality and hematological change of mice irradiated with high and low dose of Y-radiation. Bu-Zhong-Yi-Qi-Tang administration before irradiation protected the jejunal crypts (p<0.0001), increased the formation of endogenous spleen colony (p<0.05) and reduced the frequency of radiation-induced apoptosis (p<0.05). The survival rate and mean survival time of the groups treated with Bu-Zhong-Yi-Qi-Tang within 30 days after the treatment were far better than the irradiation control group. In the experiment on the effect of ingredients of Bu-Zhong-Yi-Qi-Tang, the result indicated that the extract of Rensan (Panax ginseng), Danggui (Angelica sinensis), Shengma (Cimicifuga heracleifolia) and Chaihu (Bupleurum falcatnosa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indicated that BU-Zhong-Yi-Qi-Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the protective nature of Bu-Zhong-Yi-Qi-Tang extract and its ingredients.

Effects of Ozone Treatment and Gamma Irradiation on the Quality Properties of Dried-Spirullina and Dried-Sea Tangle Powders (오존처리와 감마선 조사가 스피루리나와 다시마 분말의 품질특성에 미치는 영향)

  • For the purpose of improving hygienic quality of dried-spirullina and dried-sea tangle powders as raw materials of health food, the comparative effects of ozone treatment and gamma irradiation on the microbial and physicochemical properties were investigated. Gamma irradiation at 7.5 kGy resulted in sterilization of total aerobic bacteria, coliforms and molds below detective levels $(<10^{1}\;CFU/g\;sample)$, while ozone treatment for 8 hr up to 18 ppm could not sufficiently eliminate the microorganisms of the samples. Physicochemical properties including compositions of fatty acid and amino acid, minerals, pH and natural pigments were not changed by gamma irradaition up to 10 kGy, whereas, ozone treatment caused changes in pH, TBA value, natural pigments and fatty acid compositions. Especially, ozone treatment markedly decreased unsaturated fatty acid of the samples.

Protective Effects of a Herb, Menthae Herba, against Radiation-induced Oxidative DNA Damage

  • As utilization of radiation in medicine, industry and biochemical research increases, the protection against radiation damage has become an important issue. Natural products such as herbal medicines are beginning to receive attention as modifiers on the radiation response. In the present study, the protective effect of a herb, Menthae Herba, against radiation-induced DNA damage was evaluated using alkaline single-cell gel electrophoresis (SCGE; comet assay) in the mouse peripheral blood Iymphocytes and the micronucleus formation test in the Chinese hamster ovary (CHO) cells. The tail moment, which was a marker of DNA damage in the SCGE, and the frequency of micronuclei was decreased in groups treated with Mentae Herba extract before exposure to 200 cGy of gamma-ray. We also confirmed its activities to scavenge DPPH and hydroxyl radicals. These experiments demonstrated that Menthae Herba was effective at reducing the radiation-induced damage of DNA and scavenging free radicals. It is plausible that scavenging of free radicals by Menthae Herba may have played an important role in providing the protection against the radiation-induced damage to the DNA. These results indicated that Menthae Herba might be a useful radioprotector and that radical scavenging appears to be one of the mechanisms of radiation protection.

  • Cares should be taken when performing the P and S wave velocity loggings in engineering and environmental fields. Some of them are the effect of casing, which is installed to prevent the borehole collapsing when the drilling is done on the loose ground such as soil and/or soft rock, and the discrepancy of the velocities of the same media according to the difference of the source wave frequency spectrum. The elastic moduli obtained from the P and S wave velocity logging have the dynamic characteristics. To overcome these difficulties, the following suggestions are recommended; (1) develop and apply a careful drilling technique that can keep the borehole wall without a casing, and (2) apply the logging methods with the suitable frequency bandwidth for the object of the velocity logging. It is important to make the aseismological engineers understand the difference between the dynamic elastic moduli and the static ones obtained from mechanical test, and to advise them to use the information properly.

  • A new water-soluble and self-doped poly(styrenesulfonic acid-graft-aniline), PSSA-g-PANI, for dispersing carbon nanotubes (CNTs) in water was synthesized and its ability to stabilize aqueous CNT dispersions was examined. It was observed that the PANI in PSSA-g-PANI, which has benzoid and quinoid structure, was strongly adsorbed onto the nanotube surface via a strong ${\pi}-{\pi}$ interaction, and thus only gentle sonication causes exfoliation of CNT ropes to small bundles and the long-term stability of their resulting dispersions was much better than commercial surfactants. Furthermore, when thin films of PSSA-g-PANI/CNT are prepared from aqueous dispersion and their electrical conductivities are measured by the four probe technique, it is observed that their conductivities are in the range of 1.5-2.5 S/cm.

  • We performed this study to determine the radioprotective effect of Sam Ryung Baek Chul San (San Ling Bai Shu San), as a prescription of traditional Oriental medicine, and its major ingredients. Jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were investigated in irradiated mice with high and low dose of rays. Sam Ryung Baek Chul San administration before irradiation increased the formation of endogenous spleen colony(p<0.05) and reduced the frequency of radiation induced apoptosis(p<0.05). In the experiment on the effect of ingredients of Sam Ryung Baek Chul San, the result indicated that the extracts of Panax ginseng, Poria cocos and Coix lacryma jabi might have the major radioprotective effects. Although the mechanisms of these inhibitory effects remain to be elucidated, these results indicated that Sam Ryung Baek Chul San might be a useful radioprotector, especially since it is a relatively nontoxic natural product.
