Electrogenerated Chemiluminescence Sensor Based on Tris(2,2'-bipyridyl) ruthenium(II) Immobilized in the Composite Film of Multi-walled Carbon Nanotube/Sol-gel Zinc oxide/Nafion

  • A composite film of multi-walled carbon nanotube (MWCNT)/sol-gel-derived zinc oxide(ZnO)/Nafion has been utilized as an efficient immobilization matrix for the construction of a highly sensitive and stable tris(2,2'-bipyridyl) ruthenium(II) (Ru(${bpy)_3}^{2+})$ electrogenerated chemiluminescence (ECL) sensor. The electrochemical and ECL behaviors of Ru(${bpy)_3}^{2+})$ ion-exchanged into the composite film were strongly dependent upon the sol-gel preparation condition, the amount of MWCNT incorporated into the ZnO/Nafion composite film, and the buffer solution pH. The synergistic effect of MWCNTs and ZnO in the composite films increased not only the sensitivity but also the long-term stability of the ECL sensor. The present ECL sensor based on the MWCNT/ZnO/Nafion gave a linear response ($R^2$ = 0.999) for tripropylamine concentration from 500 nM to 1.0 mM with a remarkable detection limit (S/N = 3) of 15 nM. The present ECL sensor showed outstanding long-term stability (94% initial signal retained for 5 weeks). Since the present ECL sensor exhibits large response towards NADH, it could be applied as a transduction platform for the ECL biosensor in which the NADH is produced from the dehydrogenase-based enzymatic reaction in the presence of NA$D^+$ cofactor.

Electrochemical Behaviors of Hydroquinone on a Carbon Paste Electrode with Ionic Liquid as Binder

  • In this paper the electrochemical behaviors of hydroquinone ($H_2Q$) were investigated on a carbon paste electrode using room temperature ionic liquid N-butylpyridinium hexafluorophosphate ($BPPF_6$) as binder (ILCPE) and further applied to $H_2Q$ determination. In pH 2.5 phosphate buffer solution (PBS), the electrochemical response of H2Q was greatly improved on the IL-CPE with a pair of well-defined quasi-reversible redox peaks appeared, which was attributed to the electrocatalytic activity of IL-CPE to the $H_2Q$. The redox peak potentials were located at 0.340 V (Epa) and 0.240 V (Epc) (vs. the saturated calomel electrode, SCE), respectively. The formal potential ($E^0$') was calculated as 0.290 V and the peak-to-peak separation (${\Delta}E_p$) was 0.100 V. The electrochemical parameters of $H_2Q$ on the IL-CPE were further calculated by cyclic voltammetry. Under the selected conditions the anodic peak current was linear with $H_2Q$ concentration over the range from $5.0\;{{\times}}\;10^{-6}$ to $5.0\;{\times}\;10^{-3}\;mol\;L^{-1}$ with the detection limit as $2.5\;{\times}\;10^{-6}\;mol\;L^{-1}$ (3$\sigma$ ) by cyclic voltammetry. The proposed method was successful applied to determination of $H_2Q$ content in a synthetic wastewater sample without the interferences of commonly coexisting substances.

Species characterization of animal by DNA hybridization (DNA hybridization을 이용한 축종특이성 구명)

  • DNA hybridization assay using probes prepared from liver was carried out to identify species characterization of the domestic animals. Gel electrophoresis showed that the target DNA extracted from raw muscle were 1kb and uniform pattern while fragments size of heated muscle were irregular. Hybridization was performed by adding 200ng/ml probe in hybridization solution and incubating for 12 hours at $68^{\circ}C$. To obtain good discrimination, applied washing buffer and washing step differently depending on the species. The probes of pig, horse and dog formed the specific hybrids with each target DNA respectively. Although cross reaction was detected in cattle, goat and sheep but signal intensity among these species made the discrimination possible each other. Such pattern was the same in the cases of chicken, turkey and duck. The hybridization pattern of heated muscle was similar to that of raw muscle in general, but the signal intensity was inferior to that of raw muscle. Species identification between closely related animal species, hybridized using the target DNA of such closely related animal species as a blocking agent, remarkable increase of discrimination from the evident decrease of non specific reaction compared with the control group. In addition, in the admixture where certain meat was included in the beef, pork, chicken meat, we could find whether any unjust meat was admixed or not. In this case, detection limit of certain meat in admixture was 1%.

  • Historically, IP-based internets have been able to provide a simple best-effort delivery service to all applications they carry. Best effort treats all packets equally, with no service level, packet loss, and delay. But the needs of users have changed. The want to use the new real-time, multimedia, and multicasting applications. Thus, there is a strong need to be able to support a variety of traffic with a variety of quality-of-service requirements. The DiffServ architecture, proposed by the Internet Engineering Task Force(IETF), has become the most viable solution for provising QoS over IP networks. The DiffServ architecture does not specify any handling method between AF out-profile packets and BE packets. This paper propose a mechanism for supporting inter class fairness in the DiffServ architecture. Ⅰ proposed a modified Weighted Round Robin method to protect the BE traffic from AF out-profile packets in the core routers. The proposed technique is evaluated through simulation. Simulation results indicate that the proposed method provides better protection not only for BE packets from AF out-profile packets, but also for the AF in-profile packets in congested networks.

  • Membranes are used in most processes of biopharmaceutical production. It is used for pretreatment of other processes, separation of impurities in the process, virus removal, control of products concentration and buffer solution exchange. Virus filters play an important role in ensuring product efficacy and stability because viral contamination of biopharmaceuticals for humans is a sensitive issue that is directly related to serious clinical outcomes. Virus filters typically have complex multilayer structures made of various polymers such as surface-modified PVDF, PES, CRC. Depending on the manufacturer, filters have different pore structures and shapes, such as symmetric or asymmetric, and is used in the form of pleated membrane, flat sheets or hollow fibers. Virus filters are exclusively supplied by few foreign companies such as Asahi Kasei, Millipore, Pall and Sartorius. Replacing virus filters can be time consuming and expensive, including approval from regulatory agencies through validation. As localization has become important due to Japan's recent export regulations, it is necessary to increase the degree of technical independence.

Development of Polymer Coating Method for Stable Stent Coating Using Chemical Bond Between Metal Surface and Polymer (안정된 스텐트 코팅막을 형성하기 위해 금속표면과 고분자 사이의 화학적 결합을 이용한 고분자 코팅법 개발)

  • To produce stable polymer coating layer using the interaction between metal stent and polymer layer, Ahx-HSAB was synthesized by coupling 6-aminoheanoic acid (Ahx) with N-Hydroxy succinimidyl 4-azidobenzonate (HSAB) containing photo reactive group. Then, Ahx-HSAB was applied to self·assembled monolayer (SAM) on $TiO_2$-coated surface, since one end of Ahx-HSAB was carboxyl acid which was known to be able to interact with $TiO_2$ surface. That SAM layer was incubated in 1% polycaprolacton (PCL) solution and photoreacted by ultraviolet light (254 nm) to produce the chemical bond between SAM and polymer layer, followed by PCL polymer coating ({\sim}5\;{\mu}m$) by the method of spray coating. The surface change was investigated by measuring of contact angle of the surface. The contact angle values of stainless steel (SS) surface, $TiO_2$-coated surface, SAM layer by Ahx-HSAB, photoreacted surface with PCL and PCL layer by spray coating were 70.48${\pm}$1.89, 38.57${\pm}$3.31, 60.14${\pm}$2.21, 54.91${\pm}$2.70 and 56.47${\pm}$2.12, respectively. The stability of polymer layers was tested by incubation of PCL-coated plates in 0.1M PBS buffer (pH 7.4, 0.05%, Tween 80) with vigorous shaking (200 rpm). While the poiymer layer prepared by these processes showed the intact surface morphology over 3 days, the polymer layers prepared by spray coating of PCL onto SS plate (control 1) and $TiO_2$-coated SS plate (control 2) were Peeled off in 3 days. Thus, the polymer coating method using SAM and photoreaction seems to be a effective method to obtain the stable polymer layer onto SS surface.

Effects of Sodium Concentration and Osmolality on Renin Release of the Renal Cortical Slice ($Na^+$농도 및 삼투압의 변화가 신피질 절편에서의 Renin분비에 미치는 영향)

  • Most investigators have come to stress two different concepts of mechanism controlling renin release; intrarenal baroreceptor theory and the macula densa theory(Vander 1967, Thurau and Masson 1974). In the macula densa theory, the specific macula densa parameter, most commonly suggested as a possible signal, is either the osmolality or the concentration of sodium in the tubular fluid (Thurau 1964, Vander and Miller 1964, Reeves and Sommers 1965). It has been shown that sodium plays an important role in the release of renin either in vivo (Thurau 1964, Vander and Miller 1964, Thurau et al 1972) or in vitro experiments(Oelkers et al 1970, Hammerson et al 1971, Michelakis 1971). On the other hand the osmolality appears to have no effect on the release of renin in vivo (Vander 1967, Thurau and Masson 1974). However, there has been little attempt to study the effect of osmolality on in vitro renin release. We therefore undertook the present investigation to elucidate the effect of osmolality on renin release and to further test the sodium influence upon the release of renin from isolated kidney slice preparations. Isolated renal cortical slices were washed with normal Krebs-Hensenleit bicarbonate buffer solution and incubated for 30 minutes in a medium containing an appropriate concentration of sodium and osmolality. The renin released into the medium was measured by the method of radioimmunoassay(Haber et al 1969). The results obtained are as follows; 1. The release of renin from renal cortical slices was progressively inhibited as the sodium concentration in the medium increased. 2. No significant alteration in renin release was observed when osmolality of the medium was changed. These results suggest that the release of renin from the renal cortical slices is directly affected by the changes in sodium concentration in the medium, but is not influenced by the alterations in osmolality.

Water Soluble Cyclosporine Monomethoxy Poly(ethyleneglycol) Conjugates as Potential Prod rugs

  • The highly water-soluble monomethoxypoly(ethyleneglycol) (mPEG) prod rugs of cyciosporin A(CsA) were synthesized. These prod rugs were prepared by initially preparing intermediate in the form of carbonate at the 3'-positions of CsA with chloromethyl chloroformate, in the pres-ence of a base to provide a 3'-carbonated CsA intermediate. Reaction of the CsA intermediate with mPEG derivative in the presence of a base provides the desired water-soluble prod rugs. As a model, we chose molecular weight 5 kDa mPEG in the reaction with CsA to give water soluble prodrugs. To prove that the prod rug is decomposed in the body to produce CsA, the enzymatic hydrolysis test was conducted using human liver homogenate at $37^{\circ}C$. The prodrug was decomposed in human liver homogenate to produce the active material, CsA, and the hydrolysis half-life ($t_{1/2}$) of the prodrug, KI-306 was 2.2 minutes at $37^{\circ}C$. However, a demon-stration of non-enzymatic conversion in pH 7.4 phosphate buffer was provided by the fact that the half-life ($t_{1/2}$) is 21 hours at 37$^{\circ}C$. The hydrolysis test in rat whole blood was also conducted. The hydrolysis was seen with half-life ($t_{1/2}$) of about 9.9, 65.0, 14.2, 3.4, 2.1 9.5, and 1.6 minutes for KI-306, 309, 312, 313, 315, 316, and 317, respectively. This is the ideal for CsA prodrug. The pharmacokinetic study of the prodrug, KI-306, in comparison to the commer-cial product (Sandimmune Neoral Solution) was also carried out after single oral dose. Each rat received 7 mg/kg of CsA equivalent dose. Especially, the prodrug KI-306 exhibits higher AUC and $C_{max}$ than the conventional Neoral. The AUC and $C_{max}$ were increased nearly 1.5 fold. The kinetic value was also seen with $T_{max}$ of about 1.43 and 2.44 hours for KI-306 and Neoral, respectively.

Synthesis, Characterization and in Vitro Identification of $N^7-Guanine$ Adduct of 2-Bromopropane

  • Recently, we have reported that 2-bromopropane might have an immunotoxic potential in rats when exposed for 28 days. In the present studies, the possibility of 2i-deoxyguanosine abduct formation by 2- bromopropane was investigated in vitro to elucidate molecular mechanism of 2-bromopropane-induced immunosuppression. $N^7-Guanine adduct$ of 2'-bromopropane (i.e., $N^7-isopropyl$ guanine) was chemically synthesized and structurally characterized by analysis of UV,$^1H-NMR,{\;}^{13}C-NMR$, COSY and fast atom bombardment mass spectrometry to use as a reference material. Incubation of 2'-deoxyguanosine with an excess amount of 2-bromopropane in PBS buffer solution, pH 7.4, at $37^{\circ}C$ for 16 h, followed by a thermal hydrolysis, produced a detectable amount of $N^7-isopropyl$ guanine by an HPLC and UV analysis. The present results suggest that 2-bromopropane might form a DNA adduct in $N^7-position$ of 2'-deoxyguanosine at 3 Physiological condition.

Inhibitory Effects of Constituents of Gastrodia elata Bl. on Glutamate-Induced Apoptosis in MIR-32 Human Neuroblastoma Cells

  • The inhibitory effects of the constituents of Gastrodia elata Bl. (GE) on glutamate-induced apoptosis in human neuronal cells were investigated using IMR32 human neuroblastoma cells. Glutamate (GLU) induced DNA fragmentation, a hallmark of apoptosis, in a dose-dependent manner. GLU also induced a slow and sustained increase in intracellular $Ca^{2+}$ concentration. Treatment with EGTA, an extracellular $Ca^{2+}$ chelator, in a nominal $Ca^{2+}$ -free buffer solution abolished the GLU-induced intracellular $Ca^{2+}$ increase, indicating that GLU stimulated Ca2+ influx pathway in the IMR32 cells. BAPTA, an intracellualr $Ca^{2+}$ chelator, significantly inhibited the GLU-induced apoptosis assessed by the flow cytometry measuring hypodiploid DNA content indicative of apoptosis, implying that intracellular $Ca^{2+}$ rise may mediate the apoptotic action of GLU. Vanillin (VAN) and p-hydroxybenzaldehyde(p-HB), known constituents of GE, significantly inhibited both intracellular $Ca^{2+}$ rise and apoptosis induced by GLU. These results suggest that the apoptosis-inhibitory actions of the constituents of GE may account, at least in part, for the basis of their antiepileptic activities. These results further suggest that intracelluarl $Ca^{2+}$ signaling pathway may be a molecular target of the constituents of GE.
