Chemosensitization of Fusarium graminearum to Chemical Fungicides Using Cyclic Lipopeptides Produced by Bacillus amyloliquefaciens Strain JCK-12

  • Fusarium head blight (FHB) caused by infection with Fusarium graminearum leads to enormous losses to crop growers, and may contaminate grains with a number of Fusarium mycotoxins that pose serious risks to human and animal health. Antagonistic bacteria that are used to prevent FHB offer attractive alternatives or supplements to synthetic fungicides for controlling FHB without the negative effects of chemical management. Out of 500 bacterial strains isolated from soil, Bacillus amyloliquefaciens JCK-12 showed strong antifungal activity and was considered a potential source for control strategies to reduce FHB. B. amyloliquefaciens JCK-12 produces several cyclic lipopeptides (CLPs) including iturin A, fengycin, and surfactin. Iturin A inhibits spore germination of F. graminearum. Fengycin or surfactin alone did not display any inhibitory activity against spore germination at concentrations less than 30 ug/ml, but a mixture of iturin A, fengycin, and surfactin showed a remarkable synergistic inhibitory effect on F. graminearum spore germination. The fermentation broth and formulation of B. amyloliquefaciens JCK-12 strain reduced the disease incidence of FHB in wheat. Furthermore, co-application of B. amyloliquefaciens JCK-12 and chemical fungicides resulted in synergistic in vitro antifungal effects and significant disease control efficacy against FHB under greenhouse and field conditions, suggesting that B. amyloliquefaciens JCK-12 has a strong chemosensitizing effect. The synergistic antifungal effect of B. amyloliquefaciens JCK-12 and chemical fungicides in combination may result from the cell wall damage and altered cell membrane permeability in the phytopathogenic fungi caused by the CLP mixtures and subsequent increased sensitivity of F. graminearum to fungicides. In addition, B. amyloliquefaciens JCK-12 showed the potential to reduce trichothecenes mycotoxin production. The results of this study indicate that B. amyloliquefaciens JCK-12 could be used as an available biocontrol agent or as a chemosensitizer to chemical fungicides for controlling FHB disease and as a strategy for preventing the contamination of harvested crops with mycotoxins.

Characterization of the Unrecorded Wild Yeasts from the Water and Riverside Soils of Daejeoncheon and Gapcheon in Daejeon Metropolitan City, Korea (대전광역시 대전천과 갑천의 물과 주변 토양들로부터 분리한 국내 미기록 야생효모들의 특성)

  • We isolated 120 strains of wild yeasts from the water and riverside soils of Daejeoncheon and Gapcheon in Daejeon, Korea. We identified Debaryomyces udenii JSF601, Kazachstania telluris JSF602, Trichosporon faecale JSF614, Candida infanticola WJSL0039, Candida palmioleophila WJSL0048, Pichia spartinae WJSL0087, and Trichosporon coprophilum WJSL0093 from Daejeoncheon, and Leucosporidium golubevii WJSL0108 and Ustilentyloma graminis WJSL0118 from Gapcheon, as newly recorded yeast strains in Korea and investigated their microbiological characteristics. All of these previously unrecorded yeasts were oval- or ellipsoidal-shaped with ascospores, except C. infanticola WJSL0039, C. palmioleophila WJSL0048, and D. udenii JSF601. L. golubevii WJSL0108 and U. graminis WJSL0118 grew in vitamin-free medium, and C. infanticola WJSL0039 was halotolerant and grew in 10% NaCl-containing YPD broth. K. telluris JSF602 and P. spartinae WJSL0087 were thermophilic yeasts, which grew at $37^{\circ}C$.

The antibacterial effect of photo-catalytic titanium dioxide on canine skin (광 촉매 이산화 티타늄의 개 피부에 대한 항균효과)

  • Photo-catalytic products have been widely used at home and hospital to prevent bacteria, virus and fungus. Activities of anti-bacteria, anti-viruses and anti-fungi are based upon direct contact of crystals and particles of titanium dioxide with pathogens, into which titanium is catalyzed by photo. Those antimicrobial activities of the photo-catalytic titanium dioxide have been proved in vitro. However, in vivo tests of those activities have not been carried out on dog skin. Aim of this study was to evaluate the antimicrobial activities of the catalytic titanium dioxide in vivo. Ten beagle dogs were divided into two groups. One group was sprayed with 10ml of titanium dioxide(1 mg/ml) whereas the other was not. The treated dogs were exposed under the sunlight for 120 min. A set of three hairs was taken 15, 30, 60 and 120 min after the exposure and the bacteria contaminated in hairs were amplified in, Muller Hilton broth at $35^{\circ}C{\pm}1$ for 3 h. The supernatant of the bacterial culture was diluted 1 : 10 in phosphafe-buffered saline. One milliliter of the diluents was transferred into triphenyltetrazolium medium(TTC) and incubated at $35^{\circ}C{\pm}1$ for 2 days. The number of bacteria was counted. The number of bacteria colonies was decreased compared to control group. To further investigate the longevity effect of titanium dioxide, the dogs were kept in indoor without sun light for 6 and 12 h, 1, 2, 3, 7, 14 days after exposure of the chemical during each 15, 30, 60 min. The number of bacteria colony in 1ml was counted. The number of bacterial colonies was decreased. Treated group is exposured by sun light during 15 min, the longevity effect of titanium dioxide is continued by 1 week. Treated group is exposured by sun light during 30, 60 min, the longevity effect of titanium dioxide is continued over 2 weeks. These data indicated that the photo-catalytic titanium dioxide may be used for prevent bacteria on dog skin.

The Inhibitory Effects of Intestine-oriented Lactobacillus sp. KP-3 on the Accumulation of Heavy Metals in Sprague Dawley rats (Sprague Dawley 쥐에서 장내 유래 Lactobacillus sp. KP-3의 중금속 축적 저해 효과)

  • To investigate the effect of lactic acid bacteria on the heavy metal adsorption from internal organs and blood, lactic acid bacteria were isolated from human feces. Some strains resistant to heavy metals were selected by incubation in agar media containing each of chrome and cadmium salts. Among them, a strain named KP-3 was ultimately chosen due to its higher growth rate in selective broth medium containing the heavy metals at the concentration of 0.01%. The strain was identified as Lactobacillus sp. based on its morphological, cultural and physiological characteristics. For evaluating the ability to prevent accumulation of heavy metals by selected Lactobacillus sp. strain in vivo, Sprague Dawley rats were fed with heavy metal salts (cadmium, chrome and lead) with or without cultured whole cells for 7 days. The amounts of heavy metals accumulated in liver, kidney and blood were analyzed. As a result, chrome was accumulated to kidney mostly, and lead was frequently found in liver and kidney. Experimental group (rats fed with lactic acid bacteria) showed less accumulation of heavy metal than control group (rats fed with saline solution). The inhibition rates of heavy metal accumulation were calculated to 41.8% (Cd), 33.4% (Cr) and 44.2% (Pb). Especially, feeding lactic acid bacteria strongly reduced accumulation of cadmium in blood. The results showed that feeding Lactobacillus sp. KP-3 could prevent the bioaccumulation of heavy metals in the living body.

Isolation and Characterization of Lactobacillus brevis AML15 Producing γ-Aminobutyric acid ((γ-Aminobutyric acid를 생산하는 Lactobacillus brevis AML15의 분리 및 특성)

  • For the screening of ${\gamma}-aminobutyric$ acid (CABA)-producing bacteria, 86 bacterial strains which produce GABA were isolated from Kimchi and Salted fisk .Among these, three strains designated AML15, AML45-1, AML72 with relatively high GABA productivity were selecled by thin layer chromatography (TLC). To elucidate the relationship between isolated strains and the genus Lactobacillus, their 16S rDNA sequence were examined. The result of their DNA sequences showed 99% similarity with Lactobacillus brevis ATCC 367. On the basis of the these results, isolated strains were identified as Lactobacillus brevis and designated L. brevis AML15. In order to determine the optimum conditions for GABA production, the isolated strains were cultivated in pyridoxal phosphate (PLP) and monosodium glutami. acid (MSG). Results showed that L. brevis AML15 had the highest CABA productivity with 10,424 $nM/{\mu}l$ concentration in MRS broth containing 5% (w/v) MSG and 10 ${\mu}M$ PLP at pH 5.0. The results imply that L. brevis AML15 has the potential to be developed as a strain for GABA hyper-production.

Toxicity and Characteristics of Antifungal Substances Produced by Bacillus amyloliquefaciens IUB158-03 (Bacillus amyloliquefaciens IUB158-03이 생산하는 항진균물질의 생화학적 특성 및 독성)

  • The purified antifungal substances produced by Bacillus amyloliquefaciens IUB158-03 was positive to ninhydrin but negative to aniline, suggesting that the antifungal substance could be a peptide. FAB-MS, UV adsorption spectrum, and amino acid composition analysis revealed that the molecular weight of the antifungal substance was 1042 and that maximal adsorption was at 220 nm and 277 nm. The antifungal substance was composed of $Asn_3$, $Gln_2$, $Ser_1$, $Gly_1$, and $Tyr_1$. The composition and structural characteristics of antifungal substance were analysed by $^1H$-NMR spectrum, $^1H$-COSY, HMQC, which revealed that the compound belongs to the iturin A family. Temperature and pH had little effect on the stability of the antifungal substance in the ranges of $-70{\sim}121^{\circ}C$ and pH 6.0~10.0, respectively. It showed strong antibiotic activity against fungi. An in vitro cytotoxicity test using NIH3T3 cell showed that the antifungal substance does not have cytotoxicity. The number of circulating leukocytes and the hematobiological analysis of the mice administered with the antifungal substances was similar to those of the control group, indicating no cytotoxicity in vivo. Therefore, the antifungal substances extracted from culture broth of Bacillus amyloliquefaciens IUB158-03 have future potential as biocontrol agents against plant diseases caused by fungi.

Characterization of Bacillus licheniformis KJ-9 Isolated from Soil (토양으로부터 분리한 Bacillus licheniformis KJ 9의 특성)

  • In order to produce high-quality fermenting composts, a microorganism was isolated from the natural world. The bacterium has not only in high enzyme activities but also had good antimicrobial activities against phytopathogenic microorganisms. Its cultivating characteristics were then investigated. Bacterium KJ-9, which contains high CMCase, protease and chitinase activities and excellent antimicrobial activities against phytopathogenic microorganisms, was separated from leaf mold and identified as Bacillus licheniformis by two methods: Bergey's Manual of Systematic Bacteriology and API 50 CHL Carbohydrate Test Kit (Bio Merieux, France) using an ATB (Automated Identification) computer system (Bio Merieux, France). Optimal medium for cultivation of B. licheniformis was 2% soluble starch as a carbon source, 0.5% yeast extract as a nitrogen source and 0.05% $MgSO_4{\cdot}7H_2O$. Optimal growth conditions of pH, temperature and shake speed were pH 7.0, $50^{\circ}C$ and 180 rpm, respectively. Culture broth of B. licheniformis KJ-9 cultured for 36~60 hr was effective in fungicidal activities against plant pathogens including Botrytis cinerea, Corynespora cassicola, Fusarium oxysporum, and Rhizoctonia solani.

Production of Inhibitory Compounds against Helicobacter pylori by Culture Condition of Morus alba cv. Cheongmoknosang Callus (청목노상(Morus alba cv. Cheongmoknosang) callus의 배양조건에 따른 Helicobacter pylori 억제물질의 생산)

  • The optimal condition for Morus alba cv was an MS culture medium at $27^{\circ}C$ for 20 days. Cheongmoknosang callus showed inhibitory activity against Helicobacter pylori at 1.05 g of wet weight of the cultured callus. The callus formation of Morus alba cv. Cheongmoknosang was influenced by naphthalene acetic acid (NAA), 2,4-dichlorophenoxy acetic acid (2,4-D), 6-benzylaminopurine (BA) and kinetin at concentrations of 2 mg/l. The growth rate of callus was higher than it was when these hormones were mixed with a single hormone. Thus, the optimal condition for direct callogenesis was to incubate with mixture (2,4-D/NAA) of 2 mg/l concentration at $27^{\circ}C$ for 20 days. Moreover, the optimal culture condition of the biomass in the mass production of inhibitory compounds against Helicobacter pylori from Morus alba cv. Cheongmoknosang callus was to incubate in an MS broth (each concentration 1 mg/l of 2,4-D and BA). When Morus alba cv. Cheongmoknosang callus were incubated for 20 days in a bioreactor, Helicobacter pylori inhibition of callus extracts was the highest at a clear zone of 16 mm.

Plant Growth Promotion by Purple Nonsulfur Rhodopseudomonas faecalis Strains (자색비유황세균 Rhodopseudomonas faecalis의 식물생장촉진능)

  • Photosynthetic purple nonsulfur bacterial strains were isolated from the sediments collected from rice paddy fields and sludges of wastewater treatment plant, and their plant growth promoting capabilities were examined. Most well known phytohormones, auxin such as indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) and 5'-aminolevulinic acid (ALA) were detected by HPLC in the culture broth of these isolates. Among the isolated bacteria, Rhodopseudomonas faecalis D15 showed the highest production rate of 769.8 ${\mu}g$/mg protein of IAA, 1323 ${\mu}g$/mg protein of IBA and 7.4 mM/mg protein of ALA in the modified Biebl and Pfennig's medium. R. faecalis C9 showed the highest production rate of 20.82 ${\mu}g$/mg protein of gibberellin. In consequence, the root length and dry weight of the germinated tomato seedling treated with R. faecalis isolates were longer and heavier than those of uninoculated control after 15 days of incubation in the soil. Especially, the dry weight of germinated tomato seedling increased by 119.4% in C9-treated samples after 15 days. These purple nonsulfur bacteria may be utilized as environment-friendly biofertilizer in the agriculture.

Research Trend of Cronobacter Species Detection Methods: A Review (Cronobacter Species의 검출에 관한 연구동향: 총설)

  • Cronobacter species (Cronobacter spp.), previously known as Enterobacter sakazakii, are gram negative food borne pathogenic bacteria. They pose a very high risk of infection to neonates and immuno-compromised individuals and can affect the human central nervous system. Consequently, survivors often suffer from severe neurological impairment including hydrocephalus, quadriplegia, and developmental delays. Cronobacter spp. were not only isolated from plant food and products such as cereals, fruits, vegetables, legume products, herbs, and spices but also from animal source foods such as milk, meat, fish, and products made from these foods. Therefore, rapid detection of Cronobacter spp. is essential for food safety. Many detection methods have been developed since the Cronobacter spp. were first reported. However, the development of more rapid, sensitive, and easy-to-use detection methods for the Cronobacter spp. is required. In this review, our aim was to study and compare the available detection methods for Cronobacter spp., including culture-based, molecular biology-based, and immunology-based methods. This study will contribute to the development of new and rapid detection method for Cronobacter spp.
