This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.
Activated carbons were used to examine their performance for the separation of undesirable colored materials from culture broth containing hyaluronic acid. Six local samples and a NORIT ROX 08 were tested, whereas the latter was mainly studied under batch and continuous modes. The optimal wavelength for the detection of colored materials was 330nm. The optimal choice of NORIT ROX 08 provided 30% colored residuals with 96% hyaluronic acid recovery of original broth in batch experiments. The nonlinear adsorption behavior of protein and colored materials with activated carbon (C) was correlated by a Langmuir equation to give 18C/24+C and 500C/892+C for protein and colored materials, respectively. It appears that colored materials were composed of 78% protein and 22% glucose residuals on the basis of clearance results. A microscopic study using a scanning electron microscope suggests that regeneration of used activated carbon with 0.1N NaOH and hot water was not satisfactory. The present study proposes that the continuous monitoring of colored materials during purification can be accomplished by Installation of a UV monitor commonly used for continuous detection of protein during the process, as resulted from the significant correlation of color (A330)=0.353protein(mg/ml)+0.1(R=99.7%).
The antioxidative ability on the basis of reduced glutathione sulphydryl level, the inhibition activities of linoleic acid peroxidation of cell free extract of Lactobacillus spp. and the effects of types of media and growth phase of the cells on the cellular GSH level have been determined. Correlation between reduced glutathione sulphydryl level and antioxidative ability of Lactobacillus spp. was analyzed: Lactobacillus casei HY 2782 contained 25.15 $\mu$mole/g of GSH, the cellular GSH level of L. casei HY 2782 reached maximum after 24 h of cultivation and tended to decrease on further cultivation up to 72 h. There was a significantly higher level of cellular GSH when grown in de Man Rogosa and Sharpe (MRS) broth than in tryptone phytone yeast extract (TPY) broth or bromcresol pruple dextrose (BCP) broth (p<0.05). The antioxidant activity of cell free extract of Lactobacillus spp. have been shown to be significantly different among strains in the inhibition of linoleic acid peroxidation by thiobarbituric acid (TBA) test (p<0.01). L. casei HY 2782 and L. acidophilus ATCC 4356 revealed a high degree of antioxidative effect in linoleic acid oxidation system. Spearmans' rank correlation coefficient between inhibitory activity on linoleic acid peroxidation and cellular GSH levels of Lactobacillus spp. was 0.65, which means a significant positive correlation.
During the cultivation of Penkillium uerrmulosum in the media containing cellobiose octaacetate (COA), avicel, or KC-flock as an inducer and as a sole carbon source for 21 days, cellulolytic activity and SDS-PAGE pattern of proteins in the culture broth were investigated. Protein concentration and cellulolytic activity were highest in the COA medium. As cultivation period was increased, protein content and avicel hydrolytic activity of culture broth were increased as similar extent but neither $\beta$-glucosidase nor CMC hydrolytic activity was correlated to protein content. When crude proteins from the culture broth were separated on DEAE column by HPLC, distribution of avicel-hydrolytic activities were well correlated with that of major proteins. From those results it was suggested that three major proteins having 60 K, 68 K, and 76 K of Mr. were avicel-hydrolytic enzymes.
The mycelia of Tricholoma matsutake DGUM 26001, 26101, 26210 and FRI 91024 were used to determine the extracellular enzyme activity in mycelia. When the filtrate of culture broth after 30-day cultivation at $24^{\circ}C$ was used as a crude solution of extracellular enzyme, the average specific activity of ${\alpha}-amylase$ was 6142.3 unit/mg protein. The specific activity of xylanase was comparatively high. However, little or no enzyme activities were found for ${\beta}-glucosidase$, ligninase, CMCase, chitinase, protease, and lipase.
Gluconacetobacter diazotrophicus strain PA15 exhibited a minimum inhibitory concentration value of 11 mM in an LGI medium amended with $ZnCl_2$. When an LGI medium was amended with Zn metal, solubilization halos were observed in a plate assay, and further solubilization was confirmed in a broth assay. The maximum solubilization was recorded after 120 h with a 0.1% Zn metal amendment. During solubilization, the culture growth and pH of the broth were indirectly correlated. Using a Fourier Transform Infrared Spectroscopy analysis, one of the agents solubilizing the Zn metal was identified as gluconic acid. When the Zn-amended broth was observed under a bright field microscope, long involution cells were observed, and further analysis with Atomic Force Microscopy revealed highly deformed, pleomorphic, aggregate-like cells.
A total of 60 growth curves were generated with combinations of temperature, pH, and potassium lactate (PL) (60% (v/v) commercial solution) to determine the lag time (LT) and specific growth rate (SGR) of L. monocytogenes in broth. LT and SGR were significantly (P<0.05) affected by temperature, pH, concentration of PL, or the combined interaction of these factors. LT was extended and SGR was reduced significantly (P<0.05) by increased concentration of PL at lower temperature and pH. Listericidal effect was observed in the broth containing 2, 3, and 4% PL at pH 5.0 and $4^{\circ}C$. The antimicrobial activity of PL against L. monocytogenes increased when the pH of the medium was decreased at all temperatures tested. The results suggest that PL has antimicrobial properties to suppress the growth of L. monocytogenes. Potassium lactate has many potential applications as an antimicrobial additive in variety of refrigerated ready-to-eat foods.
Synergistic effects of the falcarindiol mixed with monoglyceride in broth exhibited strong growth inhibition on all test strains. It was confirmed that the falcarindiol mixed with monoglyceride exhibited bactericidal effect in broth against L. monocytogenes ATCC 19111 and ATCC 19114 at the same level.
For a microbial control of a plant pathogenic Pseudomonas syringae, Bacillus sp. strain BT182-3 was isolated. The strain BT182-3 had a growth inhibition against P. syringae not only on agar plate but also on cultured broth. After heat treatment at $40^{\cird}C$ and $80^{\cird}C$ for 30min, the lytic substance from the strain BT182-3 had about 52% remaining activity and 17% remaining activity, respectively. The optimal pH and temperature of the lytic substance was 6.0 and $28^{\cird}C$, respectively. Germination ratio of healthy radish seeds was 87% at $25^{\cird}C$ for 5 days in 0.8% saline, and that of the radish seeds infected with P. syringae was 67%, while that of the radish seeds treated with cultured broth of the strain BT182-3 was 90%. The 5-days healthy radish seedlings were 3.90cm at high and the seedlings infected with P. syringae were 3.06cm at high, while the seedlings treated with cultured broth of the strain BT182-3 were 4.30cm at high. The growth of the radish seedlings infected with P. syringae was inhibited after cultivation for 40days on pots, while the growth of the infected radish seedlings with P. syringae was recovered at stem length, root length and total weight at the same as the healthy seedlings after treatment of a lytic substance from the strain BT182-3.

Low molecular weight RNA(LMW RNA : 5S rRNA and tRNAs, <150 nucleotides) profiles of several bacteriocin production lactic acid bacteria from pig meats and reference lactic acid bacteria were generated on 10% denaturing polyacrylamide gel electrophoresis. Data evaluation including three molecular weight markers enabled the calculation of relative nucleotide units(RNU) for every band. Gels profiles and RNU evaluations were effective for identification of lactic acid bacteria species. LMW RNA profiles of lactic acid bacteria showed no variation in dependence on APT(All Purpose Tryptone Broth), TSB(Tryptic Soy Broth), MRS(Lactobacilli MRS Broth) different cultural medium.
