Determination of Boron by Ion Pair Liquid Chromatography with Chromotropic Acid (Chromotropic Acid를 착화제로 이용한 이온쌍 액체 크로마토그래피에 의한 붕소의 분리와 정량)

  • The separation and determination of boron with chromotropic acid (1,8-Dihydroxynaphthalene-3,6-disulfonic acid) as a complex agent has been studied using ion pair liquid chromatography. The use of tetrabutylammonium bromide added as an ion pair reagent to mobile phase (MeOH 61%, phosphate buffer 39% pH=8.5) allowed good separation of boron-chromotrophic acid complex anion and chromotrophic acid on poly(styrene-divinylbenzene) based reversed phase column (PRP-1, 15 $cm{\times}4.6$ mm i.d.). The complex formation between boric acid and chromotrophic acid was enhanced in the presence of 0.1 M tetrabutylammonium bromide, resulting in high sensitivity. The linear calibration was achieved over the boron concentration range of 0.5∼1000 ${\mu}g/L.$ The detection limit was 0.5 ${\mu}g/L$ (S/N=2). The proposed method was applied to the determination of boron in commercially available chemicals, $Na_2SO_4$, NaOH, KCl.

The Stability Appraisement on Cultural Property Material with the Replacing Fumigation Gas of Methyl Bromide (Methyl Bromide를 대체하는 훈증 가스의 문화재 재질 안정성 평가)

  • Methyl Bromide that was used as fumigation gas turned out to be the substance of destroying the ozone layer. For that reason, at the Montreal Protocol in 1987 the use of methyl bromide was forbidden starting 2005 in the advanced country. Also according to the 2007 Bali Protocolly methyl bromide is expected to be forbidden and therefore the purpose of this study is to find out the effects of substitution fumigation gas (Ethylene Oxide+HFC 134a, Methyl Iodide, Cyanogen and Argon) on the metal(silver, copper and iron), wood(oregon pine), pigment(yellow, red, blue, white and black), textile(hemp, ramie, jute, silk1 and silk2 / indigo, safflower and cork) and paper. After the fumigation test, ethylene oxide+HFC 134a did not have changes in the weight and color of the material itself before and after the experiment. On exterior alteration, color change occurred partly on paper and metal. Also, in most materials color change extent was 0.5 to 1.5 on the average and showed scanty difference. The materials after the fumigation test with methyl iodide did not show weight changes after the test. However, color changes more than 1.0 was shown in most of the materials especially in dyed textile material. In blue pigment, the discoloration on the surface could be seen by naked eyes. Fumigation test with cyanogen gas did not show weight changes and discoloration is more than 1.5 before and after the test. The weight changes of test materials with the argon gas was decreased about 3 to 6%. It can be observed that discoloration on paper was generated. Color changes can be seen on jute dyed with safflower and cork for two weeks with argon gas and the extent was 6.3 and 6.0.

Rapid Isolation of Mitochondrial DNA-Depleted Mammalian Cells by Ethidium Bromide and Dideoxycytidine Treatments

  • Mitochondrial DNA (mtDNA)-depleted (${\rho}^0$) cells are often used as mtDNA recipients to study the interaction between the nucleus and mitochondria in mammalian cells. Therefore, it is crucial to obtain mtDNA-depleted cells with many different nuclear backgrounds for the study. Here, we demonstrate a rapid and reliable method to isolate mammalian mtDNA-depleted cells involving treatment with the antimitochondrial agents ethidium bromide (EtBr) and 2',3'-dideoxycytidine (ddC). After a short exposure to EtBr or ddC, followed by rapid clonal isolation, we were able to generate viable mtDNA-depleted cells from mouse and human cells and were able to successfully repopulate them with exogenous mitochondria from platelets isolated from mouse and human blood samples. These mtDNA-depleted cells can be used to characterize the nuclear mitochondrial interactions and to study mtDNA-associated defects in mammalian cells. Our method of isolating mtDNA-depleted cells is practical and applicable to a variety of cell types.

Lead bromide crystal growth from the melt and characterization: the effects of nonlinear thermal boundary conditions on convection during physical vapor crystal growth of mercurous bromide

  • We investigate the effects of solutal convection on the crystal growth rate in a horizontal configuration for diffusive-convection conditions and purely diffusion conditions achievable in a low gravity environment for a nonlinear thermal gradient. It is concluded that the solutally-driven convection due to the disparity in the molecular weights of the component A $(Hg_2Br_2)$ and B (CO) is stronger than thermally-driven convection for both the nonlinear and the linear thermal profiles, corresponding to $Gr_t= 8.5{\times}10^3,\; Gr_s = 1.05{\times}10^5$. For both solutal and thermal convection processes, the growth rates for the linear thermal profile (conducting walls) are greater than for the nonlinear case. With the temperature humps, there are found to be observed in undersaturation for diffusive-convection processes ranging from $D_{AB}$ = 0.087 to 0.87. For the vertical configurations, the diffusion mode is so much dominated that the growth rate and interfacial distribution is nearly regardless of the gravitational accelerations. Also, the diffusion mode is predominant over the convection for the gravity levels less than 0.1 $g_0$ for the horizontally oriented configuration.

  • Mercurous bromide ($Hg_{2}0Br_{2}$) crystals hold promise for many acousto-optic and opto-electronic applications. This material is prepared in closed ampoules by the physical vapor transport (PVT) growth method. Due to the temperature gradient between the source and the growing crystal region, the buoyancy-driven convection may occur. The effects of thermal convection on the crystal growth rate was investigated in this study in a horizontal configuration for conditions ranging from typical laboratory conditions to conditions achievable only in a low gravity environment. The results showed that the growth rate increases linearly with Grashof number, and for 0.2 $\leq$ Ar (transport length-to-height, L/H)$\leq$1.0 sharply for Ar=5 and $\Delta$T=30 K. We have also shown that the magnitude of convection decreases with the Ar. For gravity levels of less than $10^{-2}$g the non-uniformity of interfacial distribution is negligible.

  • This study was carried out to find out the relationship between qualities and contraction phenomenon of silk fibers by treatment of concentrated neutral salts. The contraction effects of silk fibers showed the critical point on the treatment conditions of concentration, temperature and time, among three kinds of neutral salts such as calcium nitrate, calcium chloride and lithium bromide. But, The silk fibers, pretreated with bromide and/or formaldehyde, did not show the contraction upon treating with calcium nitrate. This indicates that tyrosine and serine can be correlated with the contraction reaction because of coupling these amino acids with bromide and formaldehyde. In conclusion, a mechanism for the contraction of silk fiber with highly concentrated calcium nitrate solution is supposed as follows. At the initial stage of ration, the water was penetrated into the amorphous regions and fibers swollen, therefore, the contraction took place mainly in amorphous regions, which have plenty of functional groups with hydroxyl residues. Then, as the calcium nitrate is penetrated into the microfibril, the gydrogen bonds of tyrosine and serine residues and broken and crystalline regions are more and more influenced by increasing concentration of calcium nitrate solution. Microgibrils of crystalline regions become entangled, contracted to linear direction and rearranged to form new stable hydrogen bonds.

  • Ipratropium bromide(IP) is a new anticholinergic bronchodilator To evaluate its effect on bronchial asthma which is still unkown in Korea, a double blind and randomized study was done on all patients of bronchial asthma who visit out-patients clinic of our department from June to September 1987 and showed 75 to 100% of FEV1 / FVC ratio (On pre bronchodilator spirometry(pre BD). The selected patients were given 2 puffs of Fenoterol(FE) or Ipratropium inhalator blindly and Spirometry The repeated results are: 1. In both FE and IP groups, there was a significant bronchodilatory effect on 5 and 60 minutes after administration. 2. On 5 minutes, effect of FE was significantly greater than IP.(FVC p<0.05, FEV1 p<0.01) 3. On 60 minutes, effect of IP was slightly less than FE but statistically non-significant. On the basis of above results, we concluded that onset of effect of IP is slower than FE, but its effect is significant and nearly comparable to FE.

Conductivity and Surface Tension Behavior for Binary Mixtures over a Various Concentration of TTAB

  • The conductances of trimethyltetradecylammonium bromide (TTAB) plus triphenyltetradecyl phosphonium bromide (TTPB) and TTAB plus trimethylhexadecylammonium bromide (HTAB) over the entire mole fraction range of TTAB were measured in water and in cyclodextrin plus water mixtures at fixed 4 and 8 mM of cyclodextrin at $30^{\circ}C$. The conductivity plot for both binary mixtures shows a single break from which the mixed critical micelle concentration and degree of micelle ionization were computed. From the slope of the conductivity curve, the equivalent ionic conductivities of the monomeric, associated, and the micelle states were calculated and discussed with respect to the surfactant-cyclodextrin complexation in the whole mole fraction range of both surfactant binary mixtures. The association constant K between the respective monomeric surfactant and cyclodextrin cavity of fixed 4 mM cyclodextrin was computed by considering 1:1 association from the surface tension measurement. A comparison among the K values for HTAB-cyclodetrin, TTAB-cyclodextrin, and TTPB-cyclodextrin shows that the former complexation is significantly stronger in comparison to the other ones due to the longer hydrophobic tail.

Protective Effect of Acetylbenzoylaconine Against Gamma-radiation

  • The protective effects of acetylbenzoylaconine, 2-aminoethylisothiouronium bromide hydrobromide, $\beta$-mercaptoethylamine HCI, and L-thiazolidine-4-carboxylic acid were studied on the white male mice, aged 5-6 weeks. The toxicity test of acetylbenzoylaconine revealed that the LD$_{50}$ was 2.5 mg/kg of body weight. After the administration of test substances, mice were irradiated with whole body dose of 800 rad by the Co-60 source. Observing the number of surviving mice for 30 days, the survival coefficients for the test groups were calculated and with these the protective coefficients against radiation injury, PCR, were also calculated. The PCR values are 2.24, 2.95, 2.78, and 1.23 for acetylbenzoylaconine, 2-aminoethylisothiouronium bromide hydrobromiide, $\beta$-mercaptoethylamine HCI, and L-thiazolidine-4-carboxylic acid respectively. These values reveal that the acetylbenzoylaconine has protective potency against radiation injury on white male mice.

The Effect of Physical Chemistry Factors on Formation of Disinfection by-products (소독부산물 생성에 미치는 물리화학적인 인자 영향)

  • This research studied the effect of factors that are able to form disinfection by-products (DBPs) of chlorination, including natural organic matter (NOM) with sewage, bromide ions, pH and contact time. Trihalomethane (THMs) yield of $0.95{\mu}mol/mg$ was higher than other DBPs yield for the chlorinated humic acid samples. THMs yield of sewage sample was $0.14{\mu}mol/mg$ and haloacetonitriles (HANs) yield in the sewage samples were $0.13{\mu}mol/mg$ but only $0.02{\mu}mol/mg$ for the humic acid samples. As the concentration of bromide ions increased, brominated DBPs increased while chlorinated DBPs decreased, because bromide ions produce brominated DBPs. THMs were highest $(55.55{\mu}g/L)$ at a pH of 7.9 and haloacetic acids (HAAs) were highest $(34.98{\mu}g/L)$ at a pH of 5. Also THMs increased with increasing pH while HAAs decreased with increasing pH. After chlorination, the rate of THMs and HAA formation are faster at initial contact time and then reaches a nearly constant value after 24 hours. This study considers ways to reduce DBP formation by chlorination.
