Study on the Estimation of Genetic Parameters and Selection Efficiency on Early Part-record for Improving Egg Production in Layer (산난계의 유전적 모수 추정과 조기선발효율에 관한 연구)

  • Data on egg number from time of first lay to 71 week of age were obtained from samples of single comb white leghorn breeder and brown layer breeder populations which were raised at manny breeding Co. These data were used to estimate the heritabilities of age at sexual maturity, early egg number, residual egg number and annual egg number. Also, the genetic correlation coefficients between these traits were estimated and selection efficiencies of each segment in early part record were estimated by use of the heritabilities and generic correlations. The estimated heritabilities and standard errors in two lines(W and B) were $0.30\pm0.07$ and $0.33\pm0.08$ at early records, $0.19\pm0.06$ and $0.18\pm$0.05 at residual records and $0.37\pm0.09$ and $0.49\pm0.10$ at time of first lay. Those at annual records were $0.24\pm0.06$ equally. The estimated correlation coefficients and standard errors between early record and annual record were $0.76\pm0.08$ and $0.77\pm0.07$. Those between early record and residual record were $0.46\pm0.15$ and $0.39\pm0.16$ respectively. At improving annual egg production per year, selection based on early part record(SM to 40 week of age) would be 69% more efficient than on annual record in line W and 80% more efficient than on annual record in line B. But, if it were considered that egg weight decreased due to sexual maturity and cost for data collection, use of selected segment from 35~40week of age in line W and from 31~40 week of age in line B would be desired in spite of a. small loss in relative selection efficiency. The generation interval would not be shortened.

  • 'Moulinrouge' was selected as the best regenerating cultivar among 18 different spray-type chrysanthemum cultivars bred in the Gyeongnam Flowers Breeding Research Institute. When the leaf explants from standard- and spray-type chrysanthemum 'Jinba' and 'Moulinrouge' were incubated on MS basal medium supplemented with $0.5mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA, both 'Jinba' and 'Moulinrouge' induced adventitious shoots that can be regenerated into plantlets. Based on these regeneration conditions, we developed an efficient $Agrobacterium$-mediated chrysanthemum 'Moulinrouge' transformation method by using sequential selection of shoots from low ($10mg{\cdot}L^{-1}$) to high ($30mg{\cdot}L^{-1}$) concentrations of kanamycin after co-cultivation of leaf explants with $Agrobacterium$ for 10 days and induction of shoots. All kanamycin resistant plants investigated with genomic PCR analysis carried the report gene, $AtSICKLE$, in their genome. Although expression levels of the report gene in the transgenic plants investigated with RT-PCR were relatively low because of inefficiency of CaMV 35S promoter in chrysanthemum, transgenic lines expressing $AtSICKLE$ efficiently showed leaf epinasty phenotype. We expect that our results will provide a useful method that can perform a high-throughput investigation of genes isolated and studied well in model plants for molecular breeding of chrysanthemum.

A New Naked Oat Cultivar for Human Food, "Daeyang" with High-Yielding and Good-Quality (조숙 대립 양질 다수성 식용 쌀귀리 "대양")

  • A new naked oat cultivar Daeyang (Avena sativa L.) was developed by Department of Rice and Winter Cereal Crop, NICS, RDA in 2007. It was derived from a cross between 'FLX446-1-84-Q1'. and 'SO92004-B-3-3-5-7'. The FLX446-1-84-Q1, a naked oat cultivar from USA, is early heading and has good seed quality, while the SO92004-B-3-3-5-7, a covered oat breeding line, has a high yield with large grain. Subsequent generations were handled in a bulk method and pedigree selection program, and the SO97013-B-16-4 was selected based on agronomic performance in 2001. The line showed both high yield and good husking rate of seed in the yield trial tested at Suwon from 2002 to 2003, being designated as Gwiri51. The Gwiri51 was subsequently evaluated for winter hardiness, earliness, and yield in four locations, Gimje, Iksan, Jeongeup, and Jinju, from 2004 to 2007 and was designated as "Daeyang" and released. Its heading date was May 8 and maturing time was June 14 in a paddy field condition. The new cultivar Daeyang had 97 cm of culm length and 25.2 cm of spike length, 644 spikes per $m^2$, 65 grains per spike, 30.3 g of 1,000-grain weight, and 635 g of test weight. Daeyang showed better winter hardiness than that of the check cultivar 'Sunyang', and similar seed quality to the check cultivar in respect to percent content crude protein and $\beta$-glucan. However, it showed higher husking rate than the check cultivar. Grain yield of Daeyang in the regional yield trial for 4 years were averaged 4.18 MT $ha^{-1}$, which was 20% higher than that of the check cultivar Sunyang. Fall sowing cropping is recommended only in a south area where daily minimum mean temperatures are averaged higher than $-4^{\circ}C$ in January, and should be excluded in mountain area where frost damage is presumable.

  • Twenty soybean cultivars developed recently were assessed using 27 insertion and deletion (InDel) markers derived from dense variation blocks (dVBs) of soybean genome. The objective of this study is to identify the distinctness and genetic relationships among a total of 169 soybean accessions including new cultivars. The genetic homology between 149 accessions in the soybean barcode system and 20 new cultivars was 61.3% on average with the range from 25.9% to 96.3%, demonstrating the versatile application of these markers for cultivars identification. The phylogenic analysis revealed four subgroups related to their usage. The 80% of cultivars for vegetable and early maturity and the 65.9% of cultivars for bean sprouts were clustered in subgroup I-2 and II-2, respectively, indicating of the limited gene pools of their crossing parents in breeding. On the other hands, the cultivars for soy sauce and tofu with considerable gene flow by genome reshuffling were distributed evenly to several subgroups, I-1 (44.4%), I-2 (26.4%) and II-2 (23.6%). We believe that the 27 InDel markers specific to dVBs can be used not only for cultivar identification and genetic diversity, but also in breeding purposes such as introduction of genetic resources and selection of breeding lines with target traits.

Studies on the Selection for Grain Yield and Components of Yield in Wheat Breeding (소맥육종에 있어서 수량 및 수량구성형질의 선발을 위한 기초적 연구)

  • A series of experiments were conducted at Crop Experiment Station from 1968 to 1971 to obtain basic information on effects of selection on yield of wheat varieties. Heritability estimates, correlation and path coefficients computed for yield and yield related characters from fixed variety groups-Korean, U.S., and Japan; early and late-$F_2$ and $F_3$ generations were studied. The same estimates for fixed variety groups grown under different fertilizer levels and years were also studied. The results were summarized as follow: 1. Three variety groups classified by their origins as Korea, United States and Japan showed high heritability estimates for heading date, plant height and spike length. The heritability estimates for grain number per spike and 1, 000 grain weight were moderate and those for the number of spikes per plant and grain yield were low. Very low estimates of heritability were obtained for grain number per spike and yield of variety group from the Unites States. 2. High genotypic correlation coefficients between 1, 000 grain weight and yield were obtained for all variety groups originated from Korea, United States and Japan and early variety group, except late variety group. The genotypic correlation coefficients between grain number per spike and yield were also high for all variety groups except variety group originated from the United States. 3. The direct effects of 1, 000 grain weight in terms of path-coefficients to yield were high for all variety groups except late variety group. 4. High genotypic correlation coefficients were obtained between 1, 000 grain weight and yield in $F_2$ from. two crosses. The same degree of genotypic correlation coefficients were obtained between grain number per spike and yield, although slight differences in its magnitude were found depending upon the cross combination. 5. The analysis of path-coefficients in $F_2$ shows that the direct effects of yield components to yield were negligible. 6. The characters that showed high genotypic correlation coefficients with yield in $F_3$ were 1, 000 gnain weight and grain number per spike. These characters showed also high direct effects to yield. 7. No great variations of heritability estimates for heading date, plant height and spike length were obtained for either fertilizer responsive or non responsive variety group due to fertilizer levels applied. 8. Heritability estimates of 1, 000 grain weight in fertilizer responsive group and yield in fertilizer nonresponsive group were high as level of fertilizer increased. 9. Heritability estimates for grain number per spike and 1, 000 grain weight of fertilizer non-responsive-group were higher than those of fertilizer. responsive group. 10. Genotypic corretation coefficients between yield and 1, 000 grain weight in fertilizer responsive group were getting lower as the level of fertilizer increased and those in fertilizer non-responsive group were vice versa. 11. Genotypic correlation coefficient between yield and spike number per plant in fertilizer responsive group was high. However, the genotypic correlation coefficient between yield and spike number per plant in fertilizer non-responsive group was low. 12. The direct effects of 1, 000 grain weight to yield were higher than other yield components either in fertilizer responsive or non-responsive group regardless of levels of fertilizer applied. The spike number per plant, however, was high only when high level of fertilizers were applied to fitilizer responsive group. 13. Slight variations of heritability estimates for heading dates, plant height, spike length, grain number per spike and 1, 000 grain weight were obtained between years. However, the spike number per plant with low heritabilility showed great variation between years. 14. The character that showed high genotypic correlation coefficients with yield in two years was 1, 000 grain weight, and this character was also high in direct effect to yield in terms of path-coefficients. 15. From the above experimental results, it might be concluded that 1, 000 grain weight would be one of the most important characters to increase the effects of selection for yield in wheat breeding in Korea.

  • A 6${\times}$4 top-cross set was made from the ten silkworm parental lines selected by the first principal component scores. They were also analysed for the relationship between the combining ability and the first principal component score. The highest general combining ability effects were detected in the parental lines of Japanese, N39 and chinese, C46, for the most quantitative characters in the study. The first principal component score of factors related to silk productivity in the parents was significantly and positively correlated to the general combining ability of the twelve characters such as cocoon yield, cocoon weight, cocoon shell weight, cocoon shell percentage, duration of the 5th instar larvae, total larval period, length of a bave, weight of a have, non-breaking length of a bave, non-breaking weight of a have, raw silk percentage, and neatness. Similarity distance (D$^2$) was related to the specific combining ability of the characters such as cocoon yield, non-breaking length of a bave, non-breaking weight of a have, non-breaking ratio of a bave, raw silk percentage, neatness. From the results, it is possible to predict the general combining ability effects by the principal component scores for the 12 characters of the parents related to silk productivity.

Development of molecular marker to select resistant lines and to differentiate the races related to powdery mildew in melon (Cucumis melo L.) (멜론 흰가루병의 race 분화 및 저항성 계통 선발을 위한 분자마커 개발)

  • Powdery mildew (Podosphaera xanthii) commonly occurs in cultivated fields of melon (Cucumis melo L.). It inflicts a lot of damages. Therefore, breeding resistant lines is essential. Development of a resistant line by integrating resistance gene takes a long time. In addition, break down of developed resistance by generating new virulent fungus strains increases disease susceptibility. This phenomenon was related to races of powdery mildew. Therefore, it is important to develop a DNA marker to genetically analyze race-specific resistance genes of melon powdery mildew to breed resistant lines. To date, a total of 28 races of Podosphaera xanthii have been reported in the literature. In Japan, 10 races have been reported in the Ibaraki region. We developed a system to characterize the races of Podosphaera xanthii and confirmed eight out of those 10 races in the Ibaraki region. In Korea, only one race has been characterized to date. However, some different races were detected. Through genetic analysis of resistant lines and susceptible lines of powdery mildew, resistance genes of race1 (Pm-X, PXB, and Pm-R 1), race N1 (PXA), race 2 (Pm-w and Pm-R 2), race 3 (Pm-X3), and race 5 (Pm-X5 and Pm-R5) were identified in melon. These related genes of race 1, 3, N1, 5, and race 1, 2, 5 were located at linkage group II and V, respectively. In race 1, resistance gene was located in the linkage group XII. In addition, each race-specific marker related to specific resistance gene was developed. Using race information and race selection system obtained in this study, resistant line can be bred to develop resistant cultivar for several areas. Furthermore, this will make it more easily and economically to breed resistant lines by using selected markers.

Evaluation of resistance to Pierce's disease among grapevine cultivars by using the culture filtrates produced from Xylella fastidiosa (Xylella fastidiosa의 배양여액을 이용한 포도나무 피어스병 품종 저항성 검정)

  • This study investigated whether culture filtrates produced by Xylella fastidiosa can be used to determine the varietal susceptibility to Pierce's disease in grapevines (Vitis spp.) as a substitute for pathogen inoculation or field screening. A bioassay of grape leaves with culture filtrates from the pathogen showed that their phytotoxicities were active and host-selective. Ethyl acetate extracts from them also showed toxicities and host selectivity in both bunches of grapes and muscadine grapes. The sensitive range of plants to the culture filtrates and their ethyl acetate extracts was consistent with the host range of the Pierce's disease pathogen. Susceptible cultivars are sensitive to even highly diluted culture filtrates, while resistant cultivars were not affected even at their original culture filtrates. Susceptible cultivars were more sensitive to the undiluted culture filtrate than were highly diluted culture filtrates, and the younger leaves were the most sensitive to the culture filtrates in grapes. Although some European grape cultivars showed moderately susceptibility in this study, the determination of varietal resistance to Pierce's disease by the treatment of culture filtrates of pathogens could provide valuable information for the preliminary selection of genetic resources and seedlings from hybridization in a disease resistant grape breeding program.

  • Principal component analysis and cluster analysis were performed on the nine quantitative characters of the one hundred and forty eight silkworm genetic stocks. The six major quantitative characters such as cocoon yield, cocoon weight, cocoon shell weight, cocoon shell percentage, larval period of the 5th instar silkworm, and total larval period showed significantly positive correlation between them. The first three principal components extracted form the initial nine variables by principal component analysis accounted for about eighty percent of original information. The first and second principal components were characterized as factors related to silk productivity, and cocoon productivity, respectively. On the basis of multivariate analysis using city block distance determined from the first three principal components to measure the phenotypic diversity, the one hundred and forty eight silkworm genetic stocks could be clustered into seven varietal groups, and the phenotypic diversity between the varietal groups was partly related to their geographical origins. Among 7 varietal group, group II and IV revealed higher silk and cocoon productivity.

Selection of a Triploid Poplar by Flow Cytometric Analysis and Growth Characteristics of its in vitro Grown Plants (유세포 분석을 통한 현사시나무 3배체 선발 및 계통별 기내생장 특성)

  • Triploids are a useful tool for biomass production and molecular breeding of trees with a long life span. Triploids of the poplar 'Hyunsasi' (Populus alba ${\times}$ P. glandulosa) have been developed by crossing between female diploids and a male tetraploid. The tetraploid was developed around the 1970s at Korea Forest Research Institute by colchicine-induced chromosome doubling. Seedlings of the $F_1$ generation were analyzed using flow cytometry to verify their ploidy status. The mean relative fluorescence index of 3 F1 poplars, labeled as Line- 1, Line-17, Line-18, were approximately 1.5 times higher than those of diploid poplars, and the results clearly indicated that they were triploids. The phenotype of the F1 poplars included larger leaves and thicker stem than diploids, and abnormal leaf morphology, especially in the triploid 'Line-18'. Three triploid lines developed roots more slowly and had less roots than diploid. However, 3 poplar cytotypes (2x, Line-1, Line-17) rooted within 10 days on MS medium. In contrast, compared with the 3 cytotypes, the Line-18 showed about 80% and 70% in the rooting rate and the number of roots. The triploid poplars could be directly utilized for biomass production and with their sterility, they could serve as basic material for genetic transformation. In addition, flow cytometric analysis proved to be an effective and reliable method for screening forest trees for their ploidy level.
