DNA Methylation Change of Oct-4 Gene Promoter Region during Bovine Preimplantation Early Embryos (소 착상 전 초기수정란에서 Oct-4 유전자 Promoter 영역의 DNA 메틸화 변화)

  • DNA methylation is involved in tissue-specific gene control and essential for normal embryo development Octamer-binding transcription factor 4 (Oct-4) is one of the most important transcription factors for early differentiation. This study was performed whether the bovine Oct-4 is tissue specific or developmental dependent epigenetic mark, we investigated transcripts and the methylation status of CpGs of 5'-promoter region of Oct-4 in bovine preimplantation embryos. Oct-4 transcripts were highly detected in morula and blastocyst, while they were present low levels in sperm and 2- to 8-cell stage embryos. These results suggest that de novo expression of Oct-4 initiates at morula stage of embryogenesis. Here we determined that there is a tissue-dependent differentially methylated region (T-DMR) in the 5'-promoter region of Oct-4. The methylation status of the Oct-4 T-DMR was distinctively different in the oocyte from that in the sperm and adult somatic tissues and changed from zygote to blastocyst stage, suggesting that active methylation and demethylation occur during preimplantation development. Based on these results, the 5'-promoter region of Oct-4 gene is target for DNA methylation and the methylation status changes variously during embryonic development in bovine.

Distribution of T and B lymphocytes in peripheral blood and lymphoid tissues of bovine (소의 순환혈액 및 림프조직내 T 및 B 림프구 분포)

  • This study was undertaken to identify the distribution of T and B lymphocytes in bovine peripheral blood and various lymphoid tissues by the method of ABPC using RABTS, $BLT_1$ and $_6E_{12}$ as primary antibodies. RABTS, $BLT_1$ and $_6E_{12}$ positive cells in PB-MNCs were 70.9${\pm}5.5%$, $59.0{\pm}8.7%$ and $23.0{\pm}8.7%$, respectively. $BLT_1$ and $_6E_{12}$ positive cells in nylon wool nonadherent cells of PB-MNCs were $91.6{\pm}1.0%$ and $9.6{\pm}0.8%$, respectively. In the lymphoid tissues such as inguinal lymph node, mesenteric lymph node, spleen and thymus, positive cells of RABTS were $76.3{\pm}3.4%$, $74.2{\pm}8.2%$, $73.6{\pm}5.5%$ and $95.6{\pm}2.8%$, those of $BLT_1$ were $56.4{\pm}6.2%$, $55.6{\pm}7.7%$, $48.6{\pm}5.1%$ and $23.0{\pm}4.8%$ and those of $_6E_{12}$ were $45.3{\pm}7.4%$, $42.3{\pm}5.8%$, $48.5{\pm}6.2%$ and $5.6{\pm}2.1%$, in order. These results are indicating that nylon wool column method is effective for separation of bovine ocytes.

Effects of Growth Hormone Gene Polymorphism on Lipogenic Gene Expression Levels in Diaphragm Tissues of Japanese Black Heifers

  • Two SNPs, i.e. L127V and T172M, of bovine growth hormone (GH) causing the presence of GH gene haplotypes A, B, and C was previously shown to alter intramuscular fatty acid (FA) composition in Japanese Black (JB) heifers. To determine the SNP effect on somatotropic hormone concentration and lipogenesis, we measured plasma GH, insulin, and insulin-like growth factor-1 (IGF-1) concentrations. We also measured mRNA levels of fatty acid synthase (FASN), stearoyl-coA desaturase (SCD), and sterol regulatory element binding proteins-1 (SREBP-1) and FA composition in diaphragm tissues. Heifers with genotype CC had the lowest plasma insulin concentration and FASN and SCD mRNA levels among genotypes. FASN mRNA levels in haplotype A tended to positively correlate with saturated FA (SFA) content and negatively correlated with C18:2 and unsaturated FA (USFA) contents. SCD mRNA levels in haplotype A positively correlated with monounsaturated FA (MUFA) contents and negatively correlated with C18:0 content. They also tended to positively correlate with C16:1, C18:1, and USFA contents and USFA/SFA ratio and negatively correlate with SFA content. Taken together, GH gene polymorphism affects the lipogenic genes expression levels and their relationships with fatty acid compositions in diaphragm tissues of JB heifers at 31 months of age.

TIME-DEPENDENT FRACTURE OF ARTICULAR CARTILAGE: PART 1 - THEORY & VALIDATION

  • A time-dependent large deformation fracture theory is developed for application to soft biological tissues. The theory uses the quasilinear viscoelastic theory of Fung, and particularizes it to constitutive assumptions on polyvinyl-chloride (PVC) (Part I) and cartilage (Part II). This constitutive theory is used in a general viscoelastic theory by Christensen and Naghdi and an energy balance to develop an expression for the fracture toughness of the materials. Experimental methods are developed for measuring the required constitutive parameters and fracture data for the materials. Elastic stress and reduced relaxation functions were determined using tensile and shear tests at high loading rates with rise times of 25-30 msec, and test times of 150 sec. The developed method was validated, using an engineering material, PVC to separate the error in the testing method from the inherent variation of the biological tissues. It was found that the the proposed constitutive modeling can predict the nonlinear stress-strain and the time-dependent behavior of the material. As an approximation method, a pseudo-elastic theory using the J-integral concept, assuming that the material is a time-independent large deformation elastic material, was also developed and compared with the time-dependent fracture theory. For PVC. the predicted fracture toughness is $1.2{\pm}0.41$ and $1.5{\pm}0.23\;kN/m$ for the time-dependent theory and the pseudo-elastic theory, respectively. The methods should be of value in quantifying fracture properties of soft biological tissues. In Part II, an application of the developed method to a biological soft tissue was made by using bovine humeral articular cartilage.

It is more effective and causes less side reactions than traditional ferrous sulfate, which has been used primarily to treat iron deficiency in pregnancy anemia. Currently, the ferritin extracted from bovine and equine spleens are sold as a commercial product. Its markets are several hundreds of million US dollars. However, because of recent warnings against the viral diseases of animal origins such as mad cow disease, a safer ferritin is sought after. In this study, a production process for human ferritin was successfully developed. The amount of its produced in yeast is high enough to be economically viable.

  • We have studied Meckel's diverticulum(MD) of the duck(Anas platyrhynchos platyrhyncos, Linne) by histological and immunohistochemical methods. Because MD were first observed in 2 weeks after hatching, tissues were taken from 2 weeks to 32 weeks after hatching groups. MD were observed as like any parts of intestinal tract that composed with simple columnar epithelium and numerous mucosal gland especially, cecum except that many lymphocytes were exist in this study. Also a number of bovine chromogranin(BCG)-, serotonin, and somatostatin(SOM)-immunoreactive cells were observed in epithelium and submucosal gland in this study, so it could be suggest that the MD of the duck serve as some digestive and lymphatic functions.

Presence of an Albumin-Like Protein on the Plasma Membrane of Adipocytes Isolated from Korean Cow

  • The main function of adipocytes in various species is to store nutrient energy in the form of triglycerides, and this function may he closely related with hormonal signaling through the plasma membrane of adipocytes. Using SDS-PAGE, two-dimensional gel electrophoresis, and a membrane biotinylation technique, we have identified a 55KDa protein (55K protein) from the plasma membrane fraction of adipocytes, with an isoelectric point (pI) of 8.1-8.3. However, this 55K protein was not observed with a two-dimensional gel electrophoresis carried out on plasma membrane fractions prepared from the liver, heart, and kidney tissues. An analysis of the 12 amino acids sequence at the N-terminal of the 55K protein showed that it has a similar sequence to that of bovine serum albumin.

  • Collagen is the most common structural protein in the connective tissues of vertebrates, playing a crucial role in maintaining the tissue structure and wound healing. Atelocollagen retains the original properties of collagen but does not induce inflammatory cytokines. Atelocollagen is extracted from various animals and humans, with bovine-derived collagen being the most commonly used. The safety and efficacy of atelocollagen have been demonstrated through multiple studies, but further research will be needed in the field of orthopedics regarding its range of use and effects. Foot and ankle diseases have attracted little research attention, highlighting the need for further studies.

  • This study was conducted to examine the utilization of immunohistochemistry using the bovine anti-brucella immunoglobulin G (IgG) antibody in the diagnosis of brucellosis and to develop a functional biomarker relation for the progress of the disease. Anti-brucella IgG antibody was purified from the affected bovine serum using an affinity chromatography. We performed our investigation on 17 cases of brucellosis and 19 control cases with negative Rose-Bengal test results. Our purified anti-brucella IgG antibody showed a positive immunoreactivity in cytoplasmic hepatocytes of the centrilobular region, and glomeruli and tubular epithelium of the kidney. The protein pattern of the affected liver versus control was analyzed by two-dimensional electrophoresis, showing a different expression pattern of proteins between the two. Five protein spots were up-regulated and another were five down-regulated in the brucellosis liver. Significant upregulaton of catalase and 3-hydroxyacyl-CoA dehydrogenase might be due to a compensatory reaction in response to the endotoxic shock of brucella. In conclusion, the anti-brucella IgG antibody may be a good tool for discriminative diagnosis of the affected tissues and proteomics data suggest new target proteins underlying a possible pathogenic mechanism of brucellosis.
