Comparison of fatty acid composition of Staphylococcus sp isolated from bovine mastitis milk (유방염 감염 우유에서 분리된 Staphylococcus sp의 지방산 조성 비교)

  • The result of API staph-ident system was compared with cellular fatty acid composition for the identification of Staphylococcus species isolated from cattle. Isolated strains from cattle were correctly identified to S aureus, S intermedius, S hyicus, S simulans, S saprophyticus, S epidemis, S sciuri and S xylosus by API staph-ident system. The correlation between bacterial cellular fatty acid profile and Staphylococcus species isolated to API STAPH-IDENT system were. In conclusion, the result presented indicate that Staphylococci can be indentified to the species level by the cellular fatty acid profiles. Moreover, computerized fatty acid profile correlative anaylsis can be applied for determining identify of Staphylococcus species.

  • 젖소 농가에서 생산성을 높이려면 젖소의 번식성능을 최상으로 유지해야 한다. 성공적인 번식관리는 효율적이고 정확한 발정 감지 기술에 크게 의존한다. 발정감지에 실패하게 되면 공태기간이 길어져 유량은 물론 송아지 생산이 감소된다. 발정감지 실패로 인한 공태기간이 한 주기일 경우, 국내 연간 손실이 85억원(2400원/두/일 $\times$ 562,000 두 $\times$ 30% $\times$ 21일)에 이를 것으로 추정된다(Senger, 1994). 공태기간이 지나치게 길어진 저 능력소는 대개의 경우 도태시켜야 하므로 이에 따른 추가손실이 발생되며, 인공수정 횟수 증가에 따른 손실과 수의사 비용 등의 추가 비용은 전체 손실의 10%에 해당된다 (Ruiz et al., 1992, Britt, 1985).   • A simplified model for the colloidal calcium phosphate (CCP) nanocluster was developed from an active role of phosphate in the precipitation of casein (CN)/Ca mixtures and the composition of casein micelles (CM). The possible shape of the CCP nanocluster was selected as a tetrahedron, and we estimated that 4 CN molecules were involved in crosslinking a single CCP nanocluster. Similar values were obtained for the number of CN molecules involved in stabilizing the nanocluster when the number of CNs attached onto each nanocluster surface was deduced from the composition of CM. If one phosphoserine cluster consisted of 3 phosphoserine residues, the theoretical molecular weight and volume for the nanocluster were estimated to be 4,898 g/mol and 2.88 $nm^3$, respectively. It was also shown that the position of Ca present in our model were reasonably located to accommodate the serine phosphate in CN molecule.

  • A total of 593 mastitic pathogens were isolated from the clinical mastitic milk of dairy cattle in Gyeonggi area from March 1984 to February 1988. The mastitic pathogens were further studied bacteriologically and examined for susceptibility patterns to 10 antibiotics. The results obtained were summerized as follows: 1. One hundred and seventy three pathogens were isolated in the first year of studies, 205 pathogens in the second year, 122 pathogens in the third year and 93 pathogens in the last year. 2. The pathogens isolated from clinical mastitis were more in summer than other seasons. 3. Staphylococcus spp (50%) and Streptococcus spp (18%) were the main pathogens in the first year of studies but coliforms (15%) and other bacteria (40%) were the main pathogens in the last year of studies. 4. Coliform mastitis was occurred only in summer and autumn. 5. Antibiotic susceptibility patterns of the same genuses from clinical mastitis infections in different herds and in different years varied greatly. Therefore, treatment should be selected on the basis of susceptibility test results.

  • Bovine angiogenin (bAng) is a potent blood vessel inducing protein purified from cow milk. Fluorescence spectroscopy has been used to study the interaction of bAng with actin in 50 mM Tris-HCl, pH 7.5, and 1 mM CaCl$_2$ at 25$^{\circ}C$. Actin contains four tryptophans but bAng contains no tryptophans. A 50% decrease in intrinsic fluorescence accompanied formation of the bAng/actin complex. (omitted)

  • To compare the survival rate of periodontal ligament cells preserved in storage media with good availability at the time of an avulsion injury, periodontal ligament cells were incubated in ${\alpha}-MEM$ culture medium containing 10% FBS in condition of $37^{\circ}C$, 5% $CO_2$. These cells were then cultured in HBSS, ${\alpha}-MEM$, milk(S co., P. co.) and tap water at the temperature of 4, 25, $37^{\circ}C$ each in 60 min. The groups were measured by MTT assay. The results were as follows : 1. Among the storage media at $4^{\circ}C$, ${\alpha}-MEM$ and P-milk had the highest preserving ability of periodontal ligament cells, while that of HBSS S-milk and tap was low in order. 2. Among the storage media at $25^{\circ}C$, ${\alpha}-MEM$ had the highest preserving ability of periodontal ligament cells, while that of P-milk, HBSS, S-milk, tap water was low in order. 3. Among the storage media at $37^{\circ}C$, the preserving ability of periodontal ligament cells was very high in ${\alpha}-MEM$, P-milk, HBSS and S-milk, it's lowest in tap water. 4. The preserving ability of periodontal ligament cells in ${\alpha}-MEM$ was high at $4^{\circ}C$ and it's low in order of $25^{\circ}C$, $37^{\circ}C$, but in HBSS was high at $4^{\circ}C$ and it's low at $25^{\circ}C$, $37^{\circ}C$ 5. The preserving ability of periodontal ligament cells in S-milk and P-milk was high at $4^{\circ}C$, $25^{\circ}C$ and it s low at $37^{\circ}C$. In conclusion, HBSS is the storage medium of choice in an avulsion, but in this study it is preferable to choose milk at $4^{\circ}C$ for tooth since it is easy to get and affect cell viability.

  • Forty- nine quarters from 24 lactating cows with chronic mastitis were selected. The cows were raised on dairy farms in Gongju, Jochiwon and Yeongi in Chungnam province, and Iksan in Jeonbuk province, Korea. The 49 quarters with bovine mastitis were divided into control (7 quarters) and experimental (42 quarters) groups. The experimental quarters were assigned to experimental group A (10 quarters, somatic cell count: $50-100\times10^4/ml)$, experimental group B (14 quarters, somatic cells count: $100-300{\times}10^4/ml)$, and experimental group C (18 quarters, somatic cells count: $>300\times10^4/ml$), according to the number of the somatic cells in their milk. The quarters of control group were treated with norfloxacin ointment (10 g/tube) based on the result of sensitivity, twice a day for 3 days. The quarters or experimental groups were infused 10ml or ozonated oils twice a day for 3 days. After treatment, the milk of the control group contained non-significantly lower numbers of somatic cells and bacteria on day 7, compared with pretreatment levels. Experimental groups A, B and C had lower somatic and bacterial cells in their milk on day 7, compared with pretreatment levels. Experimental group B and C had significantly lower numbers of somatic cells in their milk ell day 7 than before treatment (p<0.01). However, no significant difference in somatic cell numbers was detected between the control alld experimental groups. It was concluded that ozone therapy with ozonated oil applied on bovine mastitis might be effective.

  • The potential therapeutic effect of ozone gas on bovine mastitis was investigated. Eighteen quarters from 18 lactating cows with chronic mastitis were included. The 18 quarters were assigned to the control group (treatment with antibiotics for 3 days), experimental group I (0.1 ppm ozone treatment, for 7 days) and experimental group II (1ppm ozone treatment, for 3days). In experimental group I, milk somatic cell counts were lower on day 7 after ozone treatment, compared to the pretreated counts, but were higher than the control counts. In experimental group II, somatic cell counts were significantly decreased (p<0.05) on day 7 compared to the pretreatment counts, and they were lower than the control counts. There were no changes in leukocyte, neutrophil, or lymphocyte numbers, N/L ratios, N/L ratios, or serum total protein in the control and experimental groups. We concluded that ozone gas treatment(1ppm for 3days) might be effective for treatment of bovine mastitis.

Antibacterial effect of bee venom against Gram-positive and negative bacteria isolated from mastitis in dairy cattle (봉독의 젖소 유방염 유래 그람 양성 및 음성 세균별 항균효과 분석)

  • Mastitis is an inflammatory condition of the mammary gland, most often caused by bacterial infections, resulting in significant economic losses to the dairy industry. Antimicrobial resistance has been of great concern because of the extensive clinical use of antibiotics. For this reason, the development of new compounds as an alternative treatment to bovine mastitis is needed. Bee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The aim of the present study was to evaluate the antimicrobial activity of bee venom on bacteria isolated from bovine mastitis. A total of 107 isolates from bovine mastitic milk samples collected in 2019 and 2020 in Jeonbuk province. All bacterial isolates were tested for susceptibility to bee venom of the honey bee (Apis mellifera). In order to obtain comprehensive antibacterial activities of the bee venom, we measured the minimal inhibitory concentration (MIC) of the bee venom against bacterial strains. Bee venom showed significant inhibition of bacterial growth of Gram-negative bacteria Citrobacter spp., Escherchia coli, Klebsiella spp., Pseudomonas spp., Serratia spp. and Raoultella with MIC values of 96, 81, 72, 230, and 85 ㎍/mL, respectively, and Gram-positive bacterial Enterococcus spp., Staphylococcus spp. and Streptococcus spp. with MIC values of 29, 21 and 16 ㎍/mL, respectively. The results indicated that the MIC values were different depending on the bacterial strains, and those of Gram-positive bacteria were lower than those of Gram-negative bacteria for bee venom. These findings suggested that bee venom could be an effective antimicrobial treatment for bovine mastitis; however, further research is necessary to evaluate the mechanism underlying the antimicrobial action, its effectiveness/safety in vivo and effective application for therapeutic use.
