레이저 소결 적층 시스템과 실험 계획법을 이용한 3차원 바이오 세라믹 인공지지체의 제작 (Fabrication of 3D Bioceramic Scaffolds using Laser Sintering Deposition System and Design of Experiment)

  • In this study, we developed a novel laser sintering deposition system (LSDS) based on solid free-form fabrication (SFF) technology as it has the potential to fabricate complex geometries with controllable architecture for bone tissue engineering applications. The 3D biphasic calcium phosphate (BCP) scaffolds were fabricated with a pore size of 800㎛, a line width and height of 1000㎛, and an overall size of 8.2×8.2×8.0 mm3 according to the design of experiment (DOE) results. Additionally, an optimized manufacturing process using response surface analysis was established to fabricate 3D BCP scaffolds. The fabricated 3D BCP scaffolds were sintered at 950℃, 1050℃, 1150℃, and 1250℃ according to sintering processes with a furnace. As the sintering temperature increased, the porosity increased. Through the compressive strength test, the 3D BCP scaffolds sintered at 1050℃ presented good results of about 0.76 MPa. These results suggest that fabrication methods for 3D bioceramic scaffolds using LSDS may meet the basic requirements for bone tissue engineering.

Characterizations of nano-zinc doped hydroxyapatite to use as bone tissue engineering

  • Contamination by bacterial strands is a major problem after bone replacement surgeries, so there is a great need to develop low cost biocompatible antibacterial bioactive scaffolds to be used in bone tissue engineering. For this purpose, nano-zinc doped hydroxyapatite with different zinc-concentrations (5, 10 and 15 mol%) was successfully prepared by the wet chemical precipitation method. The prepared powders were used to form porous scaffolds containing biodegradable Ca-cross-linked alginate (5%) in order to enhance the properties of alginate scaffolds. The scaffolds were prepared using the freeze-gelation method. The prepared powders were tested by X-ray diffraction; transmission electron microscope and Fourier transform infrared analyses, while the prepared scaffolds were investigated by Fourier transform infrared analyses, thermogravimetric analyses and measurement of the antibacterial properties. Best results were obtained from scaffold containing 15% mol zinc-doped hydroxyapatite powders and 5% alginate concentration with ratio of 70:30.

  • Calcium-phosphate-based bioceramics are promising biomaterials for scaffolds because they can assist in bone regeneration. In this study, a laser sintering deposition system was developed, and 3D hydroxyapatite (HA) scaffolds were fabricated. The main process conditions of the HA scaffolds were laser power, table velocity, and laser focal distance. As the laser power increased, the line width, line height, and layer thickness also increased. Further, the line width, line height, and layer thickness decreased as the table velocity increased. As the laser focal distance increased, the line width increased, but the line height and layer thickness decreased. The fabricated green scaffolds were sintered at 1050 ℃ and 1150 ℃. The sintered scaffolds had a uniform and continuous interconnected shape, with pore sizes ranging from 850 to 950 ㎛ having 53% porosity. The compressive strength of the scaffolds decreased from 0.72 MPa (1050 ℃) to 0.53 MPa (1150 ℃). The biocompatibility of the scaffolds was investigated by analyzing the adhesion of osteoblast-like MG-63 cells cultured on the surfaces of the scaffolds. The results indicate that the scaffold sintered at 1050 ℃ had good mechanical and biological properties compared to that at 1150 ℃.

Growth and Osteoblastic Differentiation of Mesenchymal Stem Cells on Silk Scaffolds

  • In this study, we compared the efficiency of osteoblast differentiation media (ODM) containing three distinct reagent combinations in osteoblastic differentiation of human bone marrow-derived mesenchymal stem cells (hBMSCs) in monolayer culture. In addition, we analyzed growth and differentiation of hBMSCs on silk scaffolds and examined the bone-forming activity of a nanofibrous silk scaffold in a tibia diaphysis defect model of a rat hind limb with intramedullary nailing. Although all three ODM increased alkaline phosphatase activity to a comparable extent, the ODM containing bone morphogenetic protein-2 (BMP-2) was found to be significantly less effective in promoting mineral deposition than the others. Growth of hBMSCs on sponge-form silk scaffolds was faster than on nanofibrous ones, while osteoblastic differentiation was apparent in the cells grown on either type of scaffold. By contrast, bone formation was observed only at the edge of the nanofibrous scaffold implanted in the tibia diaphysis defect, suggesting that use of the silk scaffold alone is not sufficient for the reconstitution of the long bone defect. Since silk scaffolds can support cell growth and differentiation in vitro, loading MSCs on scaffolds might be necessary to improve the bone-forming activity of the scaffold in the long bone defect model.

  • Mesenchymal stem cells(MSCs) have been though to be multipotent cells that can replicate that have the potential to differentiate into lineages of mesenchymal tissue including the bone, cartilage, fat, tendon, muscle, and marrow stroma. Especially, scaffolds to support cell-based tissue engineering are critical determinants of clinical efforts to regenerate and repair the body. Selection of a matrix carrier imvolves consideration of the matrix's role as a scaffold for physical support and host tissue integration as well as its ability to support of synergize the osteoinductive program of the implanted mesenchymal stem cell. The aim of this study is to evaluate the effect of autobone and Bio-$Oss^{(R)}$ to adherent mesenchymal stem cells as scaffolds on sinus augmentation with fibrin glue mixture in a rabbit model. 16 New Zealand White rabbits were divided randomly into 4 groups based on their time of sacrifice(1, 2, 4 and 8 weeks). First, mesenchymal stem cells were isolated from iliac crest marrow of rabbits and expanded in vitro. Cell culture was performed in accordance with the technique described by Tsutsumi et al. In the present study, the animals were sacrificed at 1, 2, 4 and 8 weeks after transplantation, and the bone formation ability of each sides was evaluated clinically, radiologically, histologically and histomorphologically. According to the histological observations, autobone scaffolds group showed integrated graft bone with host bone from sinus wall. At 2 and 4 weeks, it showed active newly formed bone and neovascularization. At 8 weeks, lamellae bone was observed in sinus graft material area. Radiologically, autobone with stem cell showed more radiopaque than Bio-$Oss^{(R)}$ scaffolds group. there were significant differences in bone volume between 4 and 8 weeks(p<0.05).

  • Layer manufacturing technology has been recently spotlighted as a promising candidate to fabricate porous scaffolds for tissue engineering, because it can provide three dimensional interconnectivity and different pore structures and on-demand scaffold design. This study aims to fabricate HA/PCL composite scaffolds for bone tissue engineering by a layer manufacturing technology, paste extruding deposition, and to characterize in vitro and in vivo biocompatibilities of the scaffolds. This study discusses the mechnical properties, proliferation and differentiation of osteogenic cells, and tissue in-growth and bone regeneration behavior using animal models.

Preparation and Characterization of Demineralized Bone Particle Impregnated Poly(L-lactide) Scaffolds

  • In order to endow with new bioactive functionality from demineralized bone particle (DBP) as natural source to poly(L-lactide) (PLA) synthetic biodegradable polymer, porous DBP/PLA as natural/synthetic composite scaffolds were prepared and compared by means of the emulsion freeze drying and solvent casting/salt leaching methods for the possibility of the application of tissue engineered bone and cartilage. For the emulsion freeze drying method, it was observed that the pore size decreased in the order of 79$\mu\textrm{m}$ (PLA control) > 47$\mu\textrm{m}$ (20% of DBP) > 23 $\mu\textrm{m}$ (40% of DBP) > 15$\mu\textrm{m}$ (80% of DBP). Porosities as well as specific pore areas decreased with increasing the amount of DBR. It can be explained that DBP acts like emulsifier resulting in stabilizing water droplet in emulsion. For the solvent casting/salt leaching method, a uniform distribution of well interconnected pores from the surface to core region were observed the pore size of 80 ∼70 $\mu\textrm{m}$ independent with DBP amount. Porosities as well as specific pore areas also were almost same. For pore size distribution by the mercury intrusion porosimeter analysis between the two methods, the pore size distribution of the emulsion freeze drying method was broader than that of the solvent casting/salt leaching method due to the mechanism of emulsion formation. Scaffolds of PLA alone, DBP/PLA of 40 and 80%, and DBP powder were implanted on the back of athymic nude mouse to observe the effect of DBP on the induction of cells proliferation by hematoxylin and eosin staining for 8 weeks. It was observed that the effect of DBP/PLA scaffolds on bone induction are stronger than PLA scaffolds, even though the bone induction effect of DBP/PLA scaffold might be lowered than only DBP powder, that is to say, in the order of DBP only > DBP/PLA scaffolds of 40 and 80% DBP > PLA scaffolds only for osteoinduction activity. In conclusion, it seems that DBP plays an important role for bone induction in DBP/PLA scaffolds for the application of tissue engineering area.

조직공학적 골재생을 위한 탈미넬화된 골분을 함유한 다공성 지지체의 제조 및 그 특성 (Preparation and Characterization of Demineralized Bone Particle-loaded PLGA Scaffold for Tissue Engineered Bone)

  • 생체적합성 천연재료 중 하나인 탈미네랄화된 골분 (demineralized bone particle, DBP)은 골형성단백질 (BMP)을 함유하고 있어 골수간엽줄기세포 (BMSCs)의 분화를 유도한다. 본 연구에서는 DBP를 함유한 폴리 락타이드 (PLA)와 락타이드-글리콜라이드 공중합체 (PLGA) 다공성 지지체를 용매 캐스팅/염추출법으로 제조하였고, 수은다공측정계 및 전자주사현미경을 이용하여 특성결정 하였다. BMSCs는 골분화 배지를 이용하여 조골세포로 분화시켜 Wright-Giemsa, Alizarin red, von Kossa 및 ALP 염색으로 확인하였다. DBP가 함유된 지지체와 DBP가 함유되지 않은 지지체에 BMSCs를 파종한 후 면역결핍 누드마우스의 피하에 삽입하여 이들의 골형성 정도를 비교하여 보았다. 제조한 지지체의 다공도는 $90.2\%$ 이상이었고 평균 다공크기도 69.1$\mu$m 이상이었다. BMSCs는 Wright-Giemsa, Alizarin red, von Kossa 및 ALP 염색결과 조골세포로 분화가 가능했으며, 동물실험을 수행한 결과 DBP가 함유된 지지체에서 칼슘침착 영역을 확인할 수 있었지만 DBP가 함유되지 않은 지지체에서는 칼슘침착 영역을 확인하지 못하였다. 결론적으로 DBP를 함유한 지지체에서 DBP와 BMSCs가 골형성에 중요한 요인으로 작용한다고 사료된다.

Porous Hyaluronic Acid-Gelatin Loaded Sponge Biphasic Calcium Phosphate Scaffold for Bone Implant Application

  • In this study, hyaluronic acid (HyA) - Gelatin (Gel) hydrogels were prepared at ratio of 15:85 with the goal of obtaining a high uniform porosity and porous biocompatibility scaffold for bone tissue engineering applications. In order to develop a proper scaffold for bone implant application, a HyA-Gel hydrogel loaded in sponge Biphasic Calcium Phosphate (BCP) was prepared. To assay the cytocompatibility and cell behavior on the HyA-Gel hydrogel and HyA-Gel/BCP scaffold, cell attachment and spreading of MSCs seeded on the scaffolds were studied. An invivo study was performed for HyA-Gel/BCP scaffolds after 1 and 3 months implantation. Our results provide a novel and simple method to obtain an adequate scaffold for osteoblast cells and indicate that HyA-Gel hydrogel and HyA-Gel/BCP scaffold could be a good candidate for bone tissue engineering scaffolds.

해양무척추동물을 활용한 골 조직 재생용 바이오 메디컬 소재 (Biomedical Materials for Regenerating Bone Tissue Utilizing Marine Invertebrate)

  • Tissue engineering is an emerging, innovative technology to improve or replace the biological functions of damaged tissues and organs. Scaffolds are important materials for tissue engineering as they support cell attachment, migration, and differentiation. Marine sponges naturally contain scaffolds formed by extracellular matrix proteins (collagen and sponging) and strengthened by a siliceous or calcium carbonate skeleton. Coral skeletons are also derived naturally formed by essential calcium carbonate in the form of aragonite, and are similar to human bone. In addition, collagen extracted from jellyfish is a biosafe alternative to bovine and porcine collagen and gained attention as a potential source for tissue engineering. Moreover, cuttlefish bone is an excellent calcium source and can be used to generate bio-synthetic calcium phosphate. It has become a natural candidate for biomimetic scaffolds. This review describes the use of natural products derived from marine invertebrates for applications in bone tissue engineering based on studies from 2008 to 2014.
