A New White Waxy Corn 'Daedukchal 1' Hybrid with High Yield and High Table Quality

  • A new waxy corn hybrid 'Daedukchal 1' was developed from single cross between CNU 504 and CNU 2396 at Chungnam National University in 2007. Inbred CNU 504 used as a seed parent and inbred CNU 2396 as a pollen parent were collected throughout domestic regions. Tasseling date of this hybrid was seven days later than that of check hybrid, 'Chalok 1'. 'Daedukchal 1' was 19.7 cm in ear length and 4.5 cm in ear diameter. On yield trial, ear length of this hybrid was increased 29.7% compared with a check hybrid, 'Chalok 1'. 'Daedukchal 1' had white kernels and good eating quality due to high sugar content and thinner pericarp. The ratio of kernel set length/ear length was similar to 'Chalok 1'. It is moderately resistant to southern leaf blight and northern leaf blight but susceptible to corn borer. The yields of 'Daedukchal 1' in fresh ear weight and in number of fresh ear were 16% and 8%, respectively, higher than those of a check hybrid in regional yield trials for two years. Seed production for this hybrid was better because of the consistency of tasseling dates.

Development of "L2L Teaching Model" Integrating a Hospital to School Laboratory to Strengthen Practical Education for Preliminary Clinical Laboratory Technologist (예비 임상병리사의 실무중심 교육의 강화를 위한 병원-학교 검사실 융합형(L2L) 교수학습 모형의 개발)

  • The development of teaching models to strengthen practical education and actively participate in the classes of students is now required in recent education situations. This study examined the participation and satisfaction level of students' classes after applying a teaching model-integrated hospital and school laboratory (named L2L). A total of 33 students who took the subject of a clinical microbiology experiments were involved in this study. Each group representative participating in a pre-class field exercise in the hospital microbiology laboratory was asked to conduct the experiment. After applying the L2L teaching models, the academic self-efficacy ($2.87{\pm}0.58{\rightarrow}3.38{\pm}0.55$), class participation ($2.60{\pm}0.92{\rightarrow}3.62{\pm}0.78$), and class satisfaction ($2.48{\pm}1.01{\rightarrow}3.85{\pm}0.87$) increased significantly (P<0.05). This means that pre-class field exercises created interest in the student's class and boosted self-confidence, leading to increased participation and satisfaction for the class. In conclusion, the L2L teaching model is an effective teaching method to enhance the practical training for preliminary clinical laboratory technologists.

Alum as a Chemical Amendment for Reducing Ammonia Emission and Stabilizing Pig Manure Phosphorus during Composting (돈분뇨 퇴비화 과정에서 Alum 처리가 암모니아 휘산과 인산 안정화에 미치는 영향)

  • Composting is a good strategy for management of livestock manure. However, it leads to large ammonia emissions and has a potential phosphorus runoff due to high content of soluble phosphorus. The objective of this study was to evaluate the efficiency of alum on reducing ammonia emissions and stabilizing phosphorus during composting of pig manure. For this study, alum was applied at rates of 0 (No-Alum), 1.0 (Alum-L), and 3.0 (Alum-H) g Al $kg^{-1}$ pig manure and sawdust mixture (fresh matter basis). The thermophilic stage was quickly achieved in Alum-L and No-alum treatment, but it was delayed to 5 days in Alum-H treatment. The thermophilic stage was maintained for 2 weeks in all treatment. The pH of compost treated with alum remained below 8.0 for the 35 d but it was above 8.0 in No-Alum treatment. For the first 15 days of composting process, 93, 87, and 58% of total ammonia emissions were occurred in No-Alum-L and Alum-H, respectively. The Alum-H and Alum-L treatments reduced $NH_3$ volatilization by 31 and 78% compared with No-Alum treatment. Alum treatments shifted manure P form $H_2O$ and $NaHCO_3$ extractable P into NaOH extractable P which is very stable under acid and alkaline condition. Therefore, alum is a good chemical amendment for reducing ammonia emission during composting and potential losses of P following compost applications.

The Semantic System in Late Korean-English Bilinguals (후기 한국어-영어 이중언어자의 의미체계)

  • The present study was aimed to compare the semantic systems represented by the lexicon between L1 and L2 in late Korean-English bilinguals. The participants performed the word-picture matching task. the task was to decide whether the pictures represent the previously presented words' meaning. The words were the basic level categories. The stimuli were consisted of common object belonged to two different semantic categories (natural and artificial). To control the translation strategies, the SOA were manipulated as 650ms(Exp. 1) and 250ms(Exp. 2). No translation effort was found in the comparison of the two experiments. In both experiment, the RTs were faster in L1 rendition, and it took longer to decide the stimuli in natural categories than with artificial ones in L1. However, this category effect was not observed in L2. The results showed the differences in the organization of semantic representations in the brain through the bilinguals' two languages. While L1 semantic knowledge might be more systematically organized, that of L2 seems to be less well organized, at least by late bilinguals who participated in the present study.

$10^{-10}$ Pa 영역에서의 스퍼터 이온펌프와 Non-Evaporable Getter (NEG) 펌프조합의 배기 특성

  • 스퍼터 이온펌프(Sputter Ion Pump)는 주로 화학흡착으로 동작하며 기계적 진동이 없고, 기름 등의 오염 물질을 배출하지 않으며, 수명이 길어 초고청정 진공이 요구되는 표면실험장치, 표면분석계, 입자가속기 등에서 널리 사용 되고 있다. 일정한 지름을 갖는 다수의 원통 양극과 그 양단에 두개의 음극판을 배치시킨 후, 양극과 음극 사이에 수 kV의 전압을 걸고 원통의 축방향으로 자장을 인가하면 페닝 방전이 발생한다. 냉음극에서 방출된 전자는 양극으로 비행하면서 가스를 이온화한다. 이온분자는 가스흡수성 게터재료로 된 음극에 충돌하여 스퍼터링을 일으키며 게터막를 주변에 증착시킨다. 이온 및 중성 가스는 게터 고체막 속에 주입 포획되는 형태로 배기된다. 스퍼터 이온펌프는 $10^{-5}$ Pa 부근에서 최대 배기속도를 가지며, 압력이 낮아질 수록, 특히 $10^{-10}$ Pa영역 이하에서는 그 배기속도가 급격히 저하되며, $10^{-10}$ Pa영역에서는 배기능력을 거의 상실한다. 따라서 스퍼터 이온펌프 단독으로 진공시스템을 배기할 때 도달압력은 $10^{-9}$ Pa 영역에 머무르게 되며, $10^{-10}$ Pa 이하의 극고진공을 얻기 위해서는, $10^{-8}$ Pa 이하의 압력에서 배기 속도가 압력과 무관한 흡착펌프(getter pump)와 이온펌프를 조합하여 사용한다. 본 실험에서는 $600^{\circ}C$ 이상의 온도로 진공로에서 탈개스시킨 진공용기를 배기속도 450, 60, 30, 20, 5, 3 l/s의 6종류의 이온펌프와 배기속도 400 l/s, 100 l/s의 non-evaporable getter (NEG) 펌프를 조합시켜 배기하여 그 배기 특성을 비교하였다. 도달 압력은 이온펌프의 배기속도가 클수록 낮아지는 경향을 보여주었다. 450 l/s 이온펌프와 400 l/s NEG를 조합하여 배기시킬 때 도달 압력은 ~$2{\times}10^{-10}$ Pa을 기록하여 가장 낮았으며, 3 l/s 이온펌프와 400 l/s NEG를 조합하였을 때는 $ 2{\sim}3{\times}10^{-8}$ Pa을 기록하였다. 450 l/s 이온펌프와 400 l/s NEG를 조합한 경우 잔류가스의 대부분이 수소였으나, 3 l/s 이온펌프와 400 l/s NEG의 조합한 경우에는 메탄의 잔류량이 수소 보다 많았다. 이 결과는 메탄을 배기하지 못하는 NEG의 배기 특성을 보완하기 위해서는 일정 배기속도 이상의 이온 펌프가 필요함을 보여준다.

MDP-Lys (L18), a Synthetic Muramyl Dipeptide Derivative, Enhances Antitumor Activity of an Inactivated Tumor Vaccine

  • The adjuvant effect of a muramyl dipeptide (MDP) derivative, MDP-Lys(L18), on enhancing of antitumor immunity induced by X-irradiated tumor cells against highly metastatic B16-BL6 melanoma cells was examined in mice. Mice immunized intradermally (i.d.) with a mixture of X-irradiated B16-BL6 cells and MDP-Lys (L18) [Vac+MDP-Lys (L18)] followed by an intravenous (i.v.)inoculation of $10^4$ viable tumor cells 7 days after immunization, showed a significant inhibition of experimental lung metastasis of B16-BL6 melanoma cells. The most effective immunization for the prophylactic inhibition of tumor metastasis was obtained from the mixture of $100{\;}\mu\textrm{g}$ of MDP-Lys (L18) and $10^4$ X-irradiatied tumor vaccine. Furthermore, immunization of mice with Vac+MDP-Lys(L18), 3 days after tumor challenge, resulted in a significant inhibition of lung metastasis of B16-BL6 melanoma cells in an experimental lung metastasis model. Similarly, the administration of Vac+MDP-Lys(L18), 1 or 7 days after tumor removal, markedly inhibited tumor metastasis of B16-BL6 in a spontaneous lung metastasis model. When Vac+MDP-Lys (L18) was i.d. administered 3 days after subcutaneous (s.c.) inoculation of tumor cells ($5{\times}10^5/site$) on the back, mice treated with Vac+MDP-Lys(L18) showed inhibition of significantly tumor growth on day 20. These results suggest that MDP-Lys (L18) is able to enhance antitumor activity induced by X-irradiated tumor vaccine to reduce lung metastasis of tumor cells, and is a potent immunomodulating agent which may be applied prophylactically as well as therapeutically to treatment of cancer metastasis.

  • Aseptic meristem of Fragaria ananassa 'Hokowase' were inoculated on Murashige and Skoog medium containing various levels of Benzylamiro purine(BA), IAA, and 2.4-D. Formations of shoots plantlets, roots and callus depend on hormone levels used. On medium containing high level of BA 1.0mg/l, multiple plantlets were formed, however, elongation of shoots was inhibited than on BA 0.5 mg/l. 1.0mg/l IAA induced root formation and 1.0mg/l+1.0mg/l BA inhibited root formation. Callus formation was occurred on the medium added 2.4-D. When plantlets were subcultured, formation of callus or shoot depend on BA/2.4-D ratio. 2.0mg/l 2.4-D+0.2mg/l BA and 0.5 mg/l 2.4-D+0.2mg/l BA induced callus formation and 2.0mg/l BA induced plantlet and shoot vigorously.

Microbial β-Galactosidase of Pediococcus pentosaceus ID-7: Isolation, Cloning, and Molecular Characterization

  • Pediococcus pentosaceus ID-7 was isolated from kimchi, a Korean fermented food, and it showed high activity for lactose hydrolysis. The ${\beta}$-galactosidase of P. pentosaceus ID-7 belongs to the GH2 group, which is composed of two distinct proteins. The heterodimeric LacLM type of ${\beta}$-galactosidase found in P. pentosaceus ID-7 consists of two genes partially overlapped, lacL and lacM encoding LacL (72.2 kDa) and LacM (35.4 kDa). In this study, Escherichia coli MM294 was used for the production of LacL, LacM, and LacLM. These three types of recombinant proteins were expressed, purified, and characterized. The specific activities of LacLM and LacL were 339 and 31 U/mg, respectively. However, activity was not detected with LacM alone. The optimal pH of LacLM and LacL was pH 7.5 and pH 7.0, and the optimal temperature of LacLM and LacL was $40^{\circ}C$ and $50^{\circ}C$, respectively. The optimal temperature changes indicate that LacLM is able to achieve higher activity at a relatively lower temperature. LacLM was strongly activated by $Mg^{2+}$, $Mn^{2+}$, and $Zn^{2+}$, which was not true for LacL. Consistent with this, EDTA strongly inactivated LacLM and LacL, but the presence of reducing agents did not dramatically alter the activity. Taken together, multiple alignment of amino acid sequences and phylogenetic analysis results of LacL and LacM of P. pentosaceus ID-7 suggest the evolution of LacL into LacLM and that the use of divalent metal ions results in higher activity.

In vivo evaluation of preventive effect of Lactobacillus reuteri on porcine epidemic diarrhea in suckling piglets

  • Lactic acid bacteria have been reported their beneficial roles on host including reduction of infectious diarrhea problems. In this study, preventive effect of Lactobacillus (L.) reuteri HY25101 and L. johnsonii HY25103 on porcine epidemic diarrhea virus (PEDV) was investigated in suckling piglets. Two groups of one day old PEDV naïve piglets were orally administered L. reuteri HY25101 and L. johnsonii HY25103 for three days respectively before challenge with lethal dose of PEDV. In second experiment, passive immunized one day old piglets using colostrums containing PEDV specific IgA were used. The survival rates of the L. reuteri HY25101 administered group were significantly higher than that of L. johnsonii HY25103 administered group and viral shedding was rapidly diminished in L. reuteri HY25101 administered group. Interestingly piglets born from the sow immunized with attenuated PEDV vaccine were not completely protected from PEDV challenge, however coadministeration of L. reuteri HY25101 and colostrums containing PEDV specific IgA were more effectively prevent PEDV infection. These results suggested that dietary treatment using L. reuteri HY25101 could reduce diarrheal problem and mortality rate caused by PEDV in suckling pigs. In addition, L. reuteri HY25101 could be used as one of effective compensation treatment with attenuated live vaccine for PED.

Controlled Fed-Batch Cultivation of Escherichia coli Mutant for L-Tryptophan Production (대장균 변이주의 조절식 유가배양법에 의한 L-트립토판 생산)

  • For optimal production of L-tryptophan using a regulatory mutant of Escherichia coli the relationship between product formation and acid production was investigated. Experimental results showed that the production level of L-tryptophan was lowered as the specific acid production rate increased. In order to reduce the amount of acid produced during the fermentation, a controlled fed-batch fermentation was employed. In this fed-batch process, the feed rate of the nutrient feed medium was controlled in relation to the oxygen level in the culture and thus the growth of the cells was regulated in such n way that the oxygen demand of the culture could not exceed the oxygen sup-ply. When E. coli cells were cultivated in a controlled fed-batch mode of tormentor operation, the specific acid production rate was significantly reduced and L-tryptophan production was increased as much as five times that obtained in a conventional fed-batch fermentation.
