In this study, Saccharomyces cerevisiae culture fluid (SCCF) has been added to a diet of lactating dairy cows to attempt to improve the ruminal fermentation and potentially increase the dry matter intake (DMI) and milk yield. This study was conducted to investigate the effects of SCCF on the milk yield and blood biochemistry in lactating cows during the summer. Twenty-four Holstein dairy cows were randomly assigned to one of four treatments: (1) total mixed ration (TMR-1) (Control); (2) TMR-1 supplemented with SCCF (T1); (3) TMR-2 (containing alfalfa hay) (T2); and (4) TMR-2 supplemented with SCCF (T3). SCCF (5 ml/head, 2.0×107 CFU/mL) was mixed with TMRs daily before feeding to dairy cows. The mean daily temperature-humidity index (THI) during this trial was 76.92 ± 0.51 on average and ranged from 73.04 to 81.19. For particle size distribution, TMR-2 had a lower >19 mm fraction and a higher 8-9 mm fraction than TMR-1 (p < 0.05). The type of TMR did not influence the DMI, body weight (BW), milk yield and composition, or blood metabolites. The milk yield and composition were not affected by the SCCF supplementation, but somatic cell counts were reduced by feeding SCCF (p < 0.05). Feeding SCCF significantly increased the DMI but did not affect the milk yield of dairy cows. The NEFA concentration was slightly decreased compared to that in the control and T2 groups without SCCF. Feeding a yeast culture of S. cerevisiae may improve the feed intake, milk quality and energy balance of dairy cows under heat stress.
The aim of this study was to evaluate the growth performance between two different crossbred strains of Korean native chickens denoted as 1A and 2A compared to white semi broilers and Ross (RR) from hatching to 40 days. In total, 576 one-day-old chicks were allocated in a completely randomized design to give 18 replicate cages each (eight chicks per cage). The two-phase feeding program consisted of the starter (day 1 - 21: Crude protein [CP] 20% and metabolizable energy [ME], 3,050 kcal·kg-1) and the grower (day 22 - 40: CP 18%, 3,100 kcal·kg-1). The results revealed that 1A and 2A had a lower (p < 0.05) body weight (BW) from day 1 to day 40 compared with the other groups. A lower (p < 0.05) average daily gain (ADG) was observed in 1A and 2A during the entire period when compared with the other groups. The 1A and 2A groups had the minimum (p < 0.05) average daily feed intake (ADFI) compared with the other groups for the whole period. The RR had an improved (p < 0.05) FCR compared with the other groups during the whole period. The 1A and 2A groups were observed to have the lowest (p < 0.05) shank length compared with the other groups during the entire period.
The objective of this study was to evaluate in vitro antimicrobial and anti-biofilm activity of sodium long chain polyphosphate (SLCPP) and effect of dietary supplementation of SLCPP on growth performance, organ characteristics, blood metabolites, and intestinal microflora of broilers. Antimicrobial activities of SLCPP were observed against Escherichia coli O157:H7, Listeria monocytogenes, Salmonella enterica ser. Pullorum, Shigella sonnei, Klebsiella pneumonia, Pseudomonas aeruginosa in agar well diffusion assay. In addition, SLCPP demonstrated good anti-biofilm activity against K. pneumonia and P. aeruginosa. Furthermore, to investigate the dietary effect of SLCPP, a total of 480 1-day-old male Ross 308 broiler chicks were randomly allotted to three dietary treatment groups (4 replicates per group, 40 birds in each replicate): an antibiotic-free corn-soybean meal basal diet (NC); basal diet + enramycin 0.01% (PC); and basal diet + 0.1% SLCPP (SPP). The experiment lasted for 35 days. Results showed that birds fed with SLCPP had higher body weight (BW) and average daily gain (ADG), and lower feed conversion ratio (FCR) during the grower phase (days 7 to 21) (p < 0.05). Except for blood urea nitrogen, all other blood biochemical parameters remained unaffected by the dietary supplementation of SLCPP. Compared to the control group, lengths of the duodenum and ileum in the SPP group were significantly shorter (p < 0.05). Moreover, counts of lactic acid bacteria (LAB), total aerobes, and Streptococcus spp. in jejunum as well as LAB in cecum were increased in the SPP group than in the PC group (p < 0.05). These results suggest that dietary supplementation of SLCPP might promote the growth of broilers in their early growth phase.
The purpose of this study is to examine whether spraying an anti-microbial agent into the slurry pit will reduce the noxious odor substances from piggery barns. For this, a total of 200 crossbred ([Landrace × Yorkshire] × Duroc) growing pigs with an initial average body weight (BW) of 23.58 ± 1.47 kg were selected and housed in two different rooms, i.e. control (CON) and treatment (TRT). Each room has 100 pigs (60 gilts and 40 borrows). For a period of 42 days, all pigs were fed with corn-soybean meal-based basal diet. Later the noxious odor substances were measured by the following methods. First, fecal samples were randomly collected and stored in sealed and unsealed containers, and sprayed with the non-anti-microbial agent (NAMA) (saline water) and multi-bacterial spraying (MBS) agent (200 :1, mixing ratio-fecal sample : probiotic), Second, the slurry pit of CON and TRT rooms were directly sprayed with NAMA and MBS, respectively. The fecal sample that was stored in sealed and un-sealed containers and sprayed with MBS significantly reduced NH3 and CO2 concentration at the end of day 7. However, at the end of day 42, the fecal sample showed a lower H2S, methyl mercaptans, acetic acid, and CO2 concentration compared to the unsealed container. Moreover, at the end of days 7, 14, 21, 28, 35, and 42 compared to the CON room and TRT room slurry pit emits lower concentrations of NH3, acetic acid, H2S, and methyl mercaptans, and CO2 into the atmosphere. Based on the current findings, we infer that spraying anti-microbial agents on pig dung would be one of the better approaches to suppress the odor emission from the barn in the future.
Objectives : This study was performed to investigate the effect of root extract of Pueraria thunbergiana Bentham (Puerariae Radix, PR) in diabetic mice as similar as emaciation-thirst disease in Oriental medicine. Methods : C57BL/6 mice were fed high fat (HF) and high sucrose (HS) for 8 weeks, and then administrated with 90 mg/kg body weight (bw) of streptozotocin (STZ) for induction of diabetes which is similar to the middle emaciation stage. After 5 days, blood glucose levels were measured, and selected the mice with ranges above $250mg/d{\ell}$. PR water extract was administrated orally once a day for 4 weeks with high fat and high sucrose. The levels of glucose, insulin, total cholesterol, triglyceride, ${\gamma}glutamyl$ transpeptidase (${\gamma}GTP$), glutamic oxaloacetic transaminase (GOT) and glutamate pyruvate transaminase (GPT) were analysed in the serum. Also, observed their histological changes by hematoxylin and eosin (H&E) of different organs, lung, heart, pancreas, stomach, liver, and kidney. Results : PR extract significantly decreased the levels of serum glucose and insulin in diabetic mice. PR extract significantly increased the levels of triglyceride, total cholesterol, GOT and GPT in diabetic mice. In H&E stain, PR extract inhibited the histopathological changes of lung (as a channel of the upper emaciation stage in the channel-tropism theory), pancreas (as a channel of the middle emaciation stage) and kidney (as a channel of the lower emaciation stage) in diabetic damage. Conclusions : PR extract has an anti-diabetic effect in HF/HS and low-dose STZ-induced diabetic mice. This result suggests that PR follows the channel-tropism theory in the emaciation-thirst disease through the protection of lung, pancreas and kidney.
Objectives : The root bark of Morus alba L. (Mori Cortex Radidus; MCR) has been traditionally used to reduce heat from the lungs, soothe asthma, and edema and to promote urination. In this study, we investigated the effect of MCR ethanol extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at day 0, 7 and 14 with 0.2% OVA and then airway challenged at day 21, 23, 25, 27 and 29 to induce allergic asthma. MCR extracts at doses of 50 and 100 mg/kg body weight (bw) were orally administered during OVA challenge once per a day. The levels of allergic mediators such as histamine, OVA-specific IgE, IFN-${\gamma}$ and IL-4 were measured in the sera of mice by ELISA. The histopathological change of lung tissues was observed with hematoxylin and eosin (H&E) staining. Results : MCR extract significantly decreased not only the serum levels of histamine, OVA-specific IgE, and IL-4 compared with those of OVA control group, but significantly increased the serum level of IFN-${\gamma}$. In H&E staining, MCR extract inhibited the infiltration of inflammatory cells and bronchiolar damage with epithelial thickening in lung tissues of OVA-induced asthma mice. Conclusions : These results indicate that MCR extract inhibits lung damage by asthma through regulating the allergic immune response, suggesting that MCR may be used as a useful agent for the treatment of allergic asthma.
Objectives : The root of Peucedanum japonicum Thunberg (Peucedani Japonici Radix; PJR) has been traditionally used as an herbal medicine for the treatment of anti-headache, anti-paralysis, anti-cancer, vascular protection, and blood pressure regulation. In this study, we investigated the anti-allergic effect of PJR water extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at days 1, 8 and 15 with OVA and airway challenged at days 22, 24, 26, 28, and 30 to induced allergic asthma. PJR-W extract at doses of 100 and 300 mg/kg/body weight (bw) was orally administered during OVA challenge once per a day. The levels of allergic mediators such as immunoglobulin (Ig) E, and Th1/Th2 cytokines (IFN-${\gamma}$ and IL-4) were measured in the sera of mice by ELISA. The histological change of lung tissue was observed with hematoxylin and eosin (H&E) staining. Results : The administration of PJR-W extract significantly decreased the serum levels of IgE, IL-4, and IFN-${\gamma}$ compared with those of OVA control group. In H&E staining, PJR-E extract inhibited OVA-induced airway inflammation and the inflammatory cells infiltration in the peribronchial regions of the lung. Conclusions : These results indicate that PJR-W extract has an anti-inflammatory and anti-allergic effect on allergic response through the down-regulation of allergic mediators, suggesting that this herb may be used as a useful source for the treatment of allergic inflammatory diseases such as asthma.
This study was conducted to evaluate the effects of mixed and separate gender feeding on the growth performance and carcass traits of Korean native chickens (KNC) compared to white semi-broilers (WSB) and a commercial broiler (CB) from hatching to 35 days. Here, 240 chicks were used with eight birds per cage in a randomized design with six replicates per breed. For the KNC lines, three groups of males (KNC-M), mixed-gender birds (KNC-FM) and females (KNC-F) were used. The two-phase feeding program used here consisted of a starter phase (days 1 - 21: crude protein [CP] 20% and metabolizable energy [ME], 3,050 kcal·kg-1) and a grower phase (days 22 - 35: CP 18%, 3,100 kcal·kg-1) for a commercial broiler. The WSB and CB were fed the only starter commercial diet. Fresh water and feed were provided ad-libitum. The results revealed that the KNC group had a lower (p < 0.05) body weight (BW) from day 7 and day 35 compared to that of the CB. Furthermore, the KNC group showed a lower (p > 0.05) average daily gain (ADG) relative to that of the CB during the entire experimental period. No difference (p > 0.05) was observed in the shank length among the treatments. There were also no significant differences in the growth performance or carcass traits between the KNC-M and WSB groups or in the genders of the KNC group.
Although Aster yomena has recently attracted attention for its potential anti-cancer, antimicrobial, and immune-boosting effects, there are concerns about residual pesticides because they are consumed in salad-like forms. This study investigated residual characteristics and human dietary risks of the imidazole fungicide prochloraz applied to the herbal medicine Aster yomena with different spray frequency and timing. Residual analysis of prochloraz and its three main metabolites (BTS44595, BTS44596, and 2,4,6-trichlorophenol) in Aster yomena samples was performed using the QuEChERS method and LC-MS/MS. Mean recovery rates of the fungicide and its metabolites were satisfactory in the range of 80.1 ± 1.2% to 108.2 ± 3.8%. The residual concentration of the fungicide calculated as the sum of prochloraz and its metabolites was the highest (4.14 mg·kg-1) in the Aster yomena sample applied three times at weekly intervals with the fungicide until immediately before harvest. The fungicide residue concentration in the Aster yomena was below the method-limit of quantification (MLOQ) when it was applied twice at the interval of 9 day until 21 days before harvest. The theoretical maximum daily intake of prochloraz, calculated based on the daily intake of Aster yomena, mean adult body weight, and the highest residue level analyzed in the this study, was safe at < 80% of the acceptable daily intake of the fungicide (0.01 mg-1·kg·bw-1·day). In conclusion, the triple application method with prochloraz at weekly intervals until the harvest day is recommended to produce safe Aster yomena from the fungicide residues and risks to humans.

This study was to investigate effects of different phytogenic feed additives (PFA) in grower finishing pigs with stressed by high stocking density. A total of 84 growing pigs ([Landrace × Yorkshire] × Duroc) with initial body weight (BW) of 28.23 ± 0.21 kg were used for 10 weeks (4 replicate pens with 3 pigs per pen). The dietary treatment consisted of basal diets in animal welfare density (positive control [PC]), basal diet in high stocking density (negative control [NC]), NC + 0.04% bitter citrus extract (PT1), NC + 0.01% microencapsulated blend of thymol & carvacrol (PT2), NC + 0.10% mixture of 40% bitter citrus extract and 10% microencapsulated blend of thymol and carvacrol (PT3), NC + 0.04% premixture of grape seed and grape marc extract, green tea and hops (PT4), and NC + 0.10% fenugreek seed powder (PT5). The reduction of space allowance significantly decreased (p < 0.05) growth performance (average daily gain, average daily feed intake, feed efficiency) and nutrient digestibility (dry matter, crude protein). Also, the fecal score of NC group increased (p < 0.05) compared with other groups. In blood profiles, lymphocyte decreased (p < 0.05), and neutrophil, cortisol, TNF- α increased (p < 0.05) when pigs were in high stocking density. Basic behaviors (feed intake, standing, lying) were inactive (p < 0.05) and singularity behavior (biting) were increased (p < 0.05) under high stocking density. However, PFA groups alleviated the negative effects such as reducing growth performance, nutrient digestibility, increasing stress indicators in blood and animal behavior. In conclusion, PFA groups improved the health of pigs with stressed by high stocking density and PT3 is the most effective.
