An Anti-cancer experimental study using herbs (한약을 이용한 항암 실험 연구의 경향과 연구 방향)

  • After examining and analysing the medicinal herbs of fifty-three experimental papers, we studied their effects on immediate tumors in specific cancers. We did not study the influence on the life span of general cancerous cells. We looked to see if the combined usage of medicinal herbs and anticancer agents inhibited the tumor cell's growth. The serum test and blood cell count test showed if the medicinal herbs inhibited the side effects of the anticancer agent. The test showed that more than 80 percent of used medicinal herbs, brought anticancer activities. However, anticancer experimental studies using medicinal herbs have draw-backs. First, it is difficult to choose a prescription using the standards of Oriental Medicine because we are testing a mouse not a man. Second, because we only observed the indirect effect on the whole physiological regulation caused by the synergic effects of the complex prescription, we are not able to understand the detailed mechanism of the herbs. Therefore; if the anticancer effect of the herbs is proved by the experiment, we need to research the concrete medical action of medicinal herbs and the immunological analysis of herbal medicines on the body.

Immunomodulatory Effect of Methanol Extracts of Korean Oldenlandiae Herba and Radix (국내산 백화사설초 전초 및 뿌리 메타놀층의 면역조절 효과)

  • For the evaluation of immunomodulatory effect of Korean Oldenlandiae Herb (OH) and Radix (OR), our experiment was performed with methanol extracts of Korean Oldenlandiae Herba and Radix. After administration of methanol extracts of Korean OH and OR for 7 days, Balb/C mice were immunized with sheep red blood cells. Four days later, splenic leukocytes were isolated and immunological experiments were performed. Rosette forming cells and plaque forming cells were significantly increased in Korean OH and OR treated mice compared with PBS treated control. Korean OH and OR also enhanced T and B lymphocytes, macrophage and natural killer cells by flowcytometric analysis. LPS-induced TNF-α and IL-6 levels were increased by OH and OR compared with untreated control. These results suggest that Korean OH and OR have immunomodulatory activity through regulation of cell-mediated immune and humoral immune response.

Systemic Administration of the Potassium Channel Activator in the Polystyrene Latex Bead-Induced Cerebral Vasospasm (Polystyrene Latex Bead에 의한 뇌혈관연축 모델에서 K+ 통로활성제의 전신투여)

  • Objectives : It has been reported that the presence of a pharmacologically inactive foreign substance, polystyrene latex bead, in subarachnoid space activates a non-specific immunological response and elicits arterial narrowing. Recently the activation of potassium($K^+$) channels may be of benefit in relieving cerebral vasospasm. The present study examined the effects of systemic administration of a ATP-sensitive $K^+$ channel activator, cromakalim, on the polystyrene latex bead-induced cerebral vasospasm. Methods : The spasm models similar to that caused by subarachnoid blood injection were created by injection of bead into rabbit cisterna magna. Intravenous injections of cromakalim were administered twice daily(bid) 30 minutes after induction of vasospasm. Animals were killed by perfusion-fixation 2 days after vasospasm. Basilar arteries were removed and sectioned, and the luminal cross-sectional areas were measured. Results : Injection of bead elicited an arterial constriction, reducing arterial diameter to 33.3% of resting tone. Cromakalim inhibited bead-induced constriction at a dose of 0.3mg/kg(Mann-Whitney test, p<0.01). Conclusion : These results support the concept that the cellular events triggered by inactivation of ATP-sensitive $K^+$ channels are responsible for the pathogenesis of vasospasm. The findings also indicate that cromakalim represents a potential therapeutic agents for the treatment of cerebral vasospasm.

An update on necrotizing enterocolitis: pathogenesis and preventive strategies

  • Necrotizing enterocolitis (NEC) is one of the most critical morbidities in preterm infants. The incidence of NEC is 7% in very-low-birthweight infants, and its mortality is 15 to 30%. Infants who survive NEC have various complications, such as nosocomial infection, malnutrition, growth failure, bronchopulmonary dysplasia, retinopathy of prematurity, and neurodevelopmental delays. The most important etiology in the pathogenesis of NEC is structural and immunological intestinal immaturity. In preterm infants with immature gastrointestinal tracts, development of NEC may be associated with a variety of factors, such as colonization with pathogenic bacteria, secondary ischemia, genetic polymorphisms conferring NEC susceptibility, anemia with red blood cell transfusion, and sensitization to cow milk proteins. To date, a variety of preventive strategies has been accepted or attempted in clinical practice with regard to the pathogenesis of NEC. These strategies include the use of breast feeding, various feeding strategies, probiotics, prebiotics, glutamine and arginine, and lactoferrin. There is substantial evidence for the efficacy of breast feeding and the use of probiotics in infants with birth weights above 1,000 g, and these strategies are commonly used in clinical practice. Other preventive strategies, however, require further research to establish their effect on NEC.

  • It has long been known that Jahage(紫河車) extracts of Placenta hominis are effective for immunological and vascular diseases in human body and thus, was used a major constituent of traditional oriental medicines. From full-term human placenta, we have purified a new type anticoagulant protein, PP27, using different chromatographic techniques of a phenyl TSK gel 650M column, DEAE, HA and Mono-Q columns. PP27 showed single band on SDS-PAGE with a molecular mass (Mr) of 27 kDa under denaturing conditions and a calibrated Sepharose 4B column chromatography indicated a molecular mass of 23 kDa, indicating that the value is similar to those of other PP4 enzyme reported to date. Isoelectric point of PP27 was p15.2. The protein was found to inhibit the coagulation time in a concentration-dependent manner. PP27 was acted as a vascular anticoagulant of annexin type, inhibits the blood clotting process by binding of the essential lipids in a reaction which is dependent on $Ca2^+$ ions. In the presence of $Ca2^+$ ions, PP27 combines with platelet membranes neutralizing their procoagulant effect. Coagulation triggered by the addition of thromboplastin/ lipid- mixtures is extinguished by PP27.

Diagnostic Agents for Oral and Maxillofacial Diseases (구강 질환 진단용 제제)

  • The most important progress in diagnostic sciences is the increased sensitivity and specificity in diagnostic procedures due to the development of newer micromethodologies and increasing availability of immunological and molecular biological reagents. The outcome of researches in this field has already provided DNA probes and antibodies which can be used for diagnosing various kinds of diseases including inherited ones. This development can be also applied to diagnose diseases in oral and maxillofacial regions. Technological advances have yielded highly sensitive test methodologies so that low analyte concentration and small sample volume are no longer limiting factors. Therefore, saliva can be useful test fluid for an array of analytes. Salivary constituents of diagnostic significance include steroid hormones, antibodies, drugs, and tumor markers. Of the proteins present in saliva, viral-specific immunoglobulins are of the greatest diagnostic interest. The development of conjugates and antigens by recombinant DNA technique and peptide synthesis is necessary for clinical application. Several kits developed for the purpose of blood testing should be modified to permit their application to saliva. The final practical outcome of researches in diagnostic sciences will be various diagnostic agents which can be used for detection of bacteria and viruses, screening and diagnosis of diseases, genetic screening for forensic individual identification. For these purposes, collaboration researches and development between institutions and companies are essential.

Immunomodulatory Effect of Bio-Q, Bokiikhyul Prescription (보기익혈 복합방, 바이오 큐의 면역조절 효과)

  • Bio-Q is a modified prescription with the activities of supplementing Qi and blood in human body. In the present study, immunomodulatory effect of Bio-Q was examined. After oral administration of Bio-Q for 7 days to Balb/c mice, splenocytes were isolated and immunological experiments were performed. Bio-Q significantly increased the proliferation of splenocytes exposed to concanavalin A (Con A), while it did not in case of lipopolysaccharide (LPS) stimulation. Bio-Q also significantly increased CD3/CD19, CD4/CDB and NK cells by flow cytometric analysis. In addition, Bio-Q significantly enhanced the level of $INF-\gamma$ in splenocytes, but not $TNF-\alpha$ by ELISA. These results strongly suggest that Bio-Q has immunomodulatory activity through the regulation of T cell mediated immune pathway.

Effects of the Administration of Bezoar Bovis on Immune Responses of Mice (우황이 생쥐의 면역반응에 미치는 영향)

  • The effects of Bezoar Bovis on immune responses of ICR Mice were studied. In the present study, the Jerne hemolytic plaque assay (PFC) was used to evaluate the humoral immune response to sheep red blood cells, and macrophage regulatory function was measured by quantitating the production of molecules secreted by macrophages. Bezoar Bovis was given in single daily p.o doses for short term (1, 2 days) or long term (7, 14 days). PFC response for taken 3 days after short term exposure to Bezoar Bovis was increased. The production of nitirc oxide in macrophages was also stimulated. In contrast, long term exposure to Bezoar Bovis resulted in inhibitory effect of Bezoar Bovis on nitirc oxide, $TNF-{\alpha}$and IL-6 production in macrophages. These findings suggest that treatment with Bezoar Bovis may result in differential immunological effect depending on treatment schedule.

Identifying pathogenic variants related to systemic lupus erythematosus by integrating genomic databases and a bioinformatic approach

  • Systemic lupus erythematosus (SLE) is an inflammatory-autoimmune disease with a complex multi-organ pathogenesis, and it is known to be associated with significant morbidity and mortality. Various genetic, immunological, endocrine, and environmental factors contribute to SLE. Genomic variants have been identified as potential contributors to SLE susceptibility across multiple continents. However, the specific pathogenic variants that drive SLE remain largely undefined. In this study, we sought to identify these pathogenic variants across various continents using genomic and bioinformatic-based methodologies. We found that the variants rs35677470, rs34536443, rs17849502, and rs13306575 are likely damaging in SLE. Furthermore, these four variants appear to affect the gene expression of NCF2, TYK2, and DNASE1L3 in whole blood tissue. Our findings suggest that these genomic variants warrant further research for validation in functional studies and clinical trials involving SLE patients. We conclude that the integration of genomic and bioinformatic-based databases could enhance our understanding of disease susceptibility, including that of SLE.

Effect of Dietary Bacillus subtilis on Growth Performance, Immunological Cells Change, Fecal NH3-N Concentration and Carcass Meat Quality Characteristics in Finishing Pigs (비육돈 사료내 Bacillus subtilis의 첨가가 성장, 면역세포 변화, 분내 암모니아태질소 함량 및 도체 특성에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary Bacillus subtilis on growth performance, nutrient digestibility, immunological cells change, fecal noxious gas and carcass meat quality characteristics in finishing pigs. The dietary treatments were 1) CON(basal diet), 2) BS0.1(basal diet+ 0.1% Bacillus subtilis) and 3)BS0.2(basal diet+0.2% Bacillus subtilis). Sixty crossbred(Landrace$\time$ Yorkshire$\time$Duroc) pigs(89.5$\pm$0.11kg average initial body weight) were used in a 42 days growth trial. The pigs were assigned to the treatments according to body weight and each treatment had 5 replicates of 4 pigs per pen in a randomized complete block design. Through the entire experimental period, average daily gain(ADG) and average daily feed intake(ADFI) were not significantly different among the treatments. Pigs fed BS0.1 diet significantly increased their gain/feed compared to pigs fed CON and BS0.2 diets(P<0.05). Also, dry matter(DM) and nitrogen(N) digestibilities were greater in the pigs fed BS0.1 diet than those fed CON diet(P<0.05). There were no significant differences in fecal NH3-N concentration among the treatments. In blood assay for immunological cells change investigations, red blood cells(RBC) counts increased in the pigs fed BS0.2 diet compared to pigs fed CON and BS0.1 diets. There were no significant differences in carcass pH, drip loss, marbling and firmness. However, sensual color and a*(redness) value of meat in the pigs fed BS0.2 diet were higher than in pigs fed CON diet(P<0.05). Therefore, this experiment suggested that Bacillus subtilis supplementation could improve nutrient digestibility, RBC counts and carcass meat color of pigs.
