  • We have expressed human erythropoietin (EPO) in transgenic mice using a recombinant EPO cDNA combined with a partial TPO construct. The gene was microinjected using standard techniques and five mice were detected as transgenic by PCR and further used as founders. The life span of the transgenic founders was much shorter than that of their normal littermates. Most of the tissues of the transgenic founders contained human EPO transcripts as judged by RT-PCR. Especially high expression levels were seen in the liver and lung. EPO protein levels in serum were examined by ELISA and ranged from 266-414 mIU/ml. The number of red blood cell, white blood cell and hemoglobin in the hEPO transgenic mice was higher than in normal mice. These results indicate that overexpression of hEPO is deleterious and can provoke lung failure and erythrocytosis.

  • Cancer is a major cause of death in dogs worldwide, and the incidence of cancer in dogs is increasing. The attenuated total reflection Fourier transform infrared spectroscopic (ATR-FTIR) technique is a powerful tool for the diagnosis of several diseases. This method enables samples to be examined directly without pre-preparation. In this study, we evaluated the diagnostic value of ATR-FTIR for the detection of cancer in dogs. Cancer-bearing dogs (n = 30) diagnosed by pathologists and clinically healthy dogs (n = 40) were enrolled in this study. Peripheral blood was collected for clinicopathological diagnosis. ATR-FTIR spectra were acquired, and principal component analysis was performed on the full wave number spectra (4,000-650 cm-1). The leave-one-out cross validation technique and partial least squares regression analysis were used to predict normal and cancer spectra. Red blood cell counts, hemoglobin levels and white blood cell counts were significantly lower in cancer-bearing dogs than in clinically healthy dogs (p < 0.01, p < 0.01 and p = 0.03, respectively). ATR-FTIR spectra showed significant differences between the clinically healthy and cancer-bearing groups. This finding demonstrates that ATR-FTIR can be applied as a screening technique to distinguish between cancer-bearing dogs and healthy dogs.

  • Multidrug resistance is a major obstacle in cancer chemotherapy. Cancer cells efflux chemotherapeutic drug out of cell by means of transporter and reduce the active concentration of it inside cell. Such transporters are member of the ATP binding cassettes (ABC) protein. It includes P-gp, multiple resistant protein (MRP), and breast cancer resistant protein (BCRP). These proteins are widely distributed in the human cells such as kidney, lung, endothelial cells of blood brain barrier etc. However, there are number of drugs developed for it, but most of them are getting transported by it. So, still there is necessity of a good modulator, which could effectively combat the transport of chemotherapeutic agents. Natural products origin modulators were found to be effective against transporter such as flavonoids, which belongs to third generation modulators. They have advantage over synthetic inhibitor in the sense that they have simple structure and abundant in nature. This review focuses on the P-gp structure its architecture, efflux mechanism, herbal inhibitors and their mechanism of action.

  • We treated KunYangDan (KYK) in SD rats in order toexamine the protective effect against cell damage induced by diethylhexyl phthalate (DEHP). KYD reduced DEHP toxicity by increasing the function of immune cell numbers weight increase of spleen.red blood cells,HB,and HCT content, sperm number and mobility, resulting in improving reproductive function by judging from the recovery of testosterone content. Interestingly the hormone change of testosterone by KYD significantly recovered the decreased its leve. Taken together, these results suggest that KYD specifically affect the reproductive function induced by DEHP,an endocrine disruptor.

  • Ligusticum chuanxiong (Umbelliferae) is a perennial herb that has been used for invigoration of blood in Korean traditional medicine. It is especially important in gynecological therapy of amenorrhea and dysmenorrhea. In this study, the essential oil of L. chuanxiong was obtained by steam distillation and its main components of L. chuanxiong, Z-ligustilide and butylidene phthalide, were isolated by silica gel column chromatography. We investigated the cytotoxic effects of the essential oil fraction of L. chuanxiong and its main components on MCF-7, HeLa and SK-Hep-1 cell lines by measuring the number of surviving cancer cells after treatment through direct cell counting and MTT analysis, and by examining the morphological changes under the microscope. The essential oil from the rhizomes of L. chuanxiong and its main components showed significant cytotoxic activities for all three tested cell lines. We also observed morphological changes of shrinking and blebbing in the membranes of the three cell lines, depending on the concentration of L. chaunxiong oil or its main components.

  • The purpose of this study was to compare the nutritional status and the immunocompetence of elderly women residing in urban and rural areas. Dietary food records and anthropometric measurements were used to evaluate the nutritional status of subjects. The immune function of subjects was assessed by total and differential white blood cell(WBC) counts. Total B and T Lymphocytes, and T cell subsets were quantified by flow-cytometer. Immunoglobulin G, A, and M concentrations were also measured as an index of humoral immunity. Elderly women in rural area showed a relatively lower dietary intake of total energy, protein, and iron than did urban elderly women. Total WBC, neutrophil counts, eosinophil counts, and the percentage of neutrophils among total leukocytes were significantly higher in urban elderly women than in rural women. Although the numbers of lymphocytes were not significantly different, the percentage of Lymphocytes among total leukocytes as greater in rural elderly women than in urban. Both groups did not show any significant differences in numbers of T cell subsets and NK cells. Immunoglobulin G, A, and M levels were not significantly different between the two groups, but the numbers of subjects placed under the deficient range of immunoglobulins were greater in rural than in urban elderly women. from the present study, it could be suggested that poor nutritional intake may selectively affect the number of immune cells, thereby influencing the immunocompetence of elderly women. • In various organisms, the Hippo signaling pathway has been identified as a master regulator of organ size determination and tissue homeostasis. The Hippo signaling coordinates embryonic development, tissue regeneration and differentiation, through regulating cell proliferation and survival. The YAP and TAZ (YAP/TAZ) act as core transducers of the Hippo pathway, and they are tightly and exquisitely regulated in response to various intrinsic and extrinsic stimuli. Abnormal regulation or genetic variation of the Hippo pathway causes a wide range of human diseases, including cancer. Recent studies have revealed that Hippo signaling plays a pivotal role in the immune system and cancer immunity. Due to pathophysiological importance, the emerging role of Hippo signaling in blood cell differentiation, known as hematopoiesis, is receiving much attention. A number of elegant studies using a genetically engineered mouse (GEM) model have shed light on the mechanistic and physiological insights into the Hippo pathway in the regulation of hematopoiesis. Here, we briefly review the function of Hippo signaling in the regulation of hematopoiesis and immune cell differentiation.
