An experimental study has been conducted to quantitatively characterize the motion of neutrally buoyant particles in 2-dimensional Poiseuille flow through the micron-sized circular capillaries in the range of Re (Reynolds number) $\approx0.1\sim100$. $A{\mu}-PTV$ (Particle Tracking Velocimetry) system is adopted, which consists of a double-headed Nd:YAG laser, an epi-fluorescence microscope and a cooled CCD camera. Since high shear rate can be induced due to the scale effect even at low Re, it is shown that in micro scale neutrally buoyant particles in Poiseuille flow drift away from the wall and away from the center of the capillary. Consequently, particles accumulate at the equilibrium position of $0.52\sim0.64R$ with R being the radius of the capillary, which is analogous to that of tube flow in macro scale. There is a plateau in equilibrium position at small Re, while equilibrium position starts increasing at $Re\approx30$. The outermost edge of particle cluster is closer to the center of the capillary than that in previous studies due to low Re effect. The present study quantitatively presents characteristics of particle motion in circular capillaries. Furthermore, it is expected to give optimum factors for designing microfluidic systems that are to be used fur plasma separation from the blood.
당뇨병 환자의 혈당검사시 통증을 최소화하는 새로운 진공 자동 채혈기법을 개발하였다. 특별히 고안, 제작된 채혈기구를 사용하여 전완부위에서 채혈침으로 피부를 관통한 직후 자동적으로 -100mmHg의 진공압력이 가해짐으로써 모세혈액이 흡입되는 자동화된 진공 채혈기법을 구현하였다. 진공 자동 채혈기법의 유용성을 검증하기 위하여 전완부위에서의 채혈용적을 측정하였다. 58명의 정상인을 대상으로 464회의 채혈을 시행한 결과 평균 채혈용적은 약 2.6$\muL$이었고 0.5$\muL$이상의 채혈빈도가 약 86%이었다. 최근 개발되고 있는 혈당계들은 0.3~0.5$\muL$의 모세혈액으로 혈당검사가 가능하므로 검사성공률 역시 약 86%로 추정되었다. 채혈통증 감소효과를 평가하기 위하여 손가락 및 전완부위 채혈 후 시각상사 통증척도를 사용하여 통증지수를 측정하였다. 전완부위 채혈시 통증은 손가락 채혈의 약 23%에 불과하였으며 거의 무통증 채혈이 가능하였다. 따라서 본 연구의 전완부위 진공 자동채혈기법은 전통적인 손가락 채혈에 비해 채혈통증이 25% 이하로 현저하게 감소하지만 검사실패율이 14%로써 경우에 따라 재채혈이 필요할 수 있다. 그러나 매일 수 차례 채혈해야 하는 만성 당뇨환자의 경우 검사기피를 방지할 수 있는 유용한 방법으로 생각된다.
To determine Pb level in blood, we usually .used to pull out about 5ml blood from venous vessel and this sample was digested with acids to decompose organic matter and then determined the Pb contents by Atomic Absorption Spectrophotometer with flame. But recent trend in quan titating Pb in small amount of sample is very much recommended in clinical chemistry specially pediatrics, and industrial hygiene and occupational health area. Authors tried to determine Pb contents in small amount blood of $25{\mu}l$ by using capillary tube method and got the possibility of determination of ng amount of Pb in $25{\mu}l$ whole blood sample without any pretreatment of sample.
The purpose of this study is to develop a new method which enables the goat ESR to be used as an effective clinical test. Blood samples were taken from 61 Korean native goats aged above one year old and the effect of tube inclinations, tube bores, tube lengths, environmental temperatures during tests and packed erythrocyte volumes (PCV) on the ESR were observed. The results were summarized as follows. 1. The ESR/hr using capillary hematocrit tube (Micro-Ht-tube) was gradually increased as the tube angle inclined from 90 (vertical) to 15 degrees and the best angle in view of both security and fast sedimentation rate was found to be an angle of 45 degrees. 2. The 45-degree angled ESR ($45^{\circ}$-ESR) increased as the diameter of tube bore decreased. 3. The tube length did not affect the $45^{\circ}$-ESR in percent. 4. The $45^{\circ}$-ESR increased with the increased environmental temperature during the ESR test. 5. The heparinized Micro-Ht tubes did not affect the $45^{\circ}$-ESR of EDTA-blood in healthy group but in anemic group. In the anemic group, the ESRs by the heparinized Micro-Ht-tubes were slightly higher than those by non-heparinized Micro-Ht-tubes. 6. By using the autologous plasma, PCV of the blood was adjusted to be 10, 20, 30, 40 and 50ml/100ml and $45^{\circ}$-ESRs were determined in the Micro-Ht-tubes. The $45^{\circ}$-ESRs increased as the values of PCV decreased. The regression of the $45^{\circ}ESR$ to PCV was curvilinear with the second degree polynomial, $Y=42.1838-1.7355X+0.0180X^2$(r=0.9997). 7. The $45^{\circ}$-ESR/hr, using non-heparinized Micro-Ht-tubes at $20^{\circ}C$, was determined in 35 healthy Korean native goats. The average PCV was $30.6{\pm}1.4ml/100ml$. The observed ESR values averaged $6.8{\pm}1.7%$ and the corrected ESR values to the standard PCV of 31ml/100ml averaged $6.5{\pm}1.2%$. From these results, the angled capillary tube method was found to be desirable ESR test of goat blood in which EDTA-blood is filled in nonheparinized Micro-Ht-tubes held at an angle of 45 degrees for an hour at $20^{\circ}C$.
Multiplex PCR-based short tandem repeat (STR) analysis is considered as a good tool for monitoring bone marrow engraftment after sex-mismatched allogeneic transplantation and provides a sensitive and accurate assessment of the contribution of both donor and/or recipient cells in post-transplantation specimens. Forensic STR analysis and quantitative real time PCR are used to determine the proportion of donor versus recipient each contained within the total DNA. The STR markers were co-amplified in a single reaction by using commercial $PowerPlex^{(R)}$ 16 system and $AmpFISTR^{(R)}$$Identifiler^{(R)}$ / $Yfiler^{(R)}$ PCR amplification kits. Separation of the PCR products and fluorescence detection were performed by ABI $PRIS^{(R)}$ 3100 Genetic Analyzer with capillary electrophoresis. The $GeneMapper^{TM}$ ID software were used for size calling and analysis of STR profiles. Extracted DNA was quantified by the $Quantifiler^{TM}$ Human DNA / Y Human Male DNA Quantification Kit The intent of this study was to analyze the ratio of donor versus recipient cells in the post-transplant peripheral blood, spleen, lung and kidney specimens. Specimens were taken from the traffic accident male victim who had been engrafted from bone marrow female donor. Blood and spleen specimens displayed female donor DNA profile. Kidney specimen showed male recipient DNA profile. Interestingly, lung tissue showed mixed profiles. The findings of this study indicate that the forensic STR analysis using fluorescence labeling PCR combined with capillary electrophoresis is quick and reliable enough to assess the ratio of donor versus recipient cells and to monitor the mixed chimeric patterns.
The blood-brain barrier (BBB) transport of acetylcholinesterase (AChE) inhibitors, donepezil and tacrine suggested to be mediated by choline transport system in our previous study. Therefore, in the present study, we investigated the interaction of other AChE inhibitors, rivastigmine and galantamine with choline transporter at the BBB. The effects of rivastigmine and galantamine on the transport of choline by conditionally immortalized rat brain capillary endothelial cell lines (TR-BBB cells) were characterized by cellular uptake study using radiolabeled choline. The uptake of [$^3H$]choline was inhibited by rivastigmine and galantamine, with $IC_{50}$ values (i.e. concentration necessary for 50% inhibition) for 1.13 and 1.15 mM, respectively. Rivastigmine inhibited the uptake of [$^3H$]choline competitively with $K_i$ of 1.01 mM, but galantamine inhibited noncompetitively. In addition, the efflux of [$^3H$]choline was significantly inhibited by rivastigmine and galantamine. Our results indicated that the BBB choline transporter may be involved in a part of the influx and efflux transport of rivastigmine across the BBB. These findings should be therapeutically relevant to the treatment of Alzheimer's disease (AD) with AChE inhibitors, and, more generally, to the BBB transport of CNS-acting cationic drugs via choline transporter.
It was confirmed that we got somewhat different results even though we performed same items with same methods from capillaries and venous blood of healthy 72 cases. Items which capillaries blood has higher value than venous blood are the numbers of erythrocyte, lymphocyte, basophil, hematocrit, MCHC and RDW. Total numbers of RBC (p<0.035), lymphocyte and basophil are shown statistically significance. Items which venous blood has higher value than capillary blood are the numbers of platelet (p<0.00) and neutrophil (p<0.01). Fallible items in clinics can be shown the numbers of RBC, platelet, lymphocyte and neutrophil because we got somewhat different results even though we performed same items with same methods from capillaries and venous blood, respectively. It is necessary to choose the clear criteria and normal value depends on clinical specimen as the number of platelets are measured with an abnormal value shown over 40% down. As normal difference according to each item and method is currently acceptable and applicable in clinics, it is considered that a new normal value depends on clinical specimen should be established and is to be useful positively in clinics.

So this system have developed for the improvement of flows in the capillary blood vessel of head and limbs. Also This system is able to measure brain blood pressure(BBP) which need diagnosis circulation state of brain blood. So, we extracted correlativity of blood pressure(BP) and BBP through clinical experiment. We confirmed that compare factor of BP with BBP is more useful factors for diagnosis state of brain hemokinesis.
Structure of the skin in amphibious fish, Periophthalmus modestus, was described in relation to cutaneous respiration. The epidermis has no gland cell. The epidermis consists of three regions: outermost layer of one to five layers of flattened epithelial cells, middle layer of swollen epithelial cells instead of glandular cells and stratum germinativum of cuboidal cells. There are numerous blood capillaries in the outermost layer of the epidermis and diffusion distance between the blood of capillaries and the epidermis is about 1.4 Um. The middle layer of the epidermis appears to be a web-like structure due to the swollen epithelial cells. The stratum germinativum has a well-developed lymphatic space containing lymphocytes. There are numerous blood capillaries and elliptical area with acid mucopolysaccharides in stratum laxum of the dermis. The skin of Periophthalmus modestus may be an accessory respiratory organ for oxygen uptake during terrestrial or aquatic life.
