Although the majority of surviving pigs cloned by somatic cell nuclear transfer (SCNT) appear to be physiologically normal, there is a general lack of detailed hemato-physiologic studies for the period of early adulthood to substantiate this claim. In the present study, we investigated variation in blood chemistry and endocrinological parameters between mesenchymal stem cells (MSCs) derived from cloned and normal age-matched female and male miniature pigs. Cloned females and males showed normal ranges for complete blood count assessments. Biochemical assessments showed that ${\gamma}$-GGT, ALT and cholesterol levels of male and female clones were significantly (P<0.05 or P<0.01, respectively) higher than that of age-matched control miniature pigs. Variations in insulin and IGF-1 were higher in female clones than in male clones and controls. Thus, although female and male cloned miniature pigs may be physiologically similar to normal animals, or at least within normal ranges, a greater degree of physiological and endocrinological variation was found in cloned pigs. The above variation must be taken into account before considering cloned female or male miniature pigs for various biomedical applications.
The liver extract of chicks, Gallus domesticus, suffering from hydropericardium syndrome (HPS) was inoculated intraperitoneally into healthy chicks to produce HPS. After inoculation the blood serum and the pericardial fluid of the newly infected chicks were analyzed, at regular intervals for a total period of 72 hours for concentration of proteins, $K^+$ ions, $Na^+$ ions and the LDH activity. The protein content was significantly decreased both in the blood serum (30%) and the pericardial fluid (39%) within 24 hours of inoculation, which was then maintained during the subsequent period. The $K^+$ ions and the LDH activity, on the other hand, were significantly increased in the blood serum (26% and 169%, respectively) as well as the pericardial fluid (131% and 217%, respectively) within 24 hours of inoculation. After 72 hours this increase was, respectively, 43% and 191% in blood serum, and 153% and 200% in the pericardial fluid. Accumulation of $K^+$ ions, and decrease of protein and $Na^+$ ions in the pericardial fluid indicate homoestatic imbalance, which may prove fatal. The increased LDH activity is indicative of heptocytic damage.
To investigate dietary habits and health and nutritional status of the elderly with cerebrovascular disease(CVD), 31 hospitalized patients and 28 old people who had experienced stroke and were living at home were interviewed for the dietary habits and drinking and smoking habits. Their blood pressure and triceps skinfold thickness were measured and biochemical assessment of blood samples were conducted. The results were compared with those from 30 healthy elderly. Ca Index reflecting ca intake and intake of dietary fiber were significantly lower in the patient elderly groups than in the healthy elderly. Cholesterol intakes were not significantly different among the groups. Although salt intakes were almost similar among the groups, patient elderly tended to like salty taste as compared to the healthy elderly. Systolic and diastolic blood pressures of the patient elderly were significantly higher than those of the healthy elderly. Triceps skinfold thickness and serum total protein and albumin were significantly lower in patient groups than the healthy elderly. Serum cholesterol and hemoglobin concentrations were not different among groups. Among the above variables only Ca index was inversely correlated with blood pressure significantly.
The purpose of the present study was to establish the mechanism of the generation of atherosclerosis by analyzing the hemodynamic variables in the coronary artery where atherosclerosis occurs frequently. From the previous results, the stenosis phenomena due to atherosclerosis were related to not only biochemical reaction between blood and blood vessel but also the hemodynamic factors like flow separation and oscillatory wall shear stress. The present study aimed to investigate the causes of the generation and progression of atherosclerosis in the coronary artery. This study also aimed to develop the softwares which generate automatically three dimensional vascular models obtained by the angiogram images and the computer vision techniques. In the present study, the flow patterns for full three-dimensional hemodynamic characteristics were analyzed. To understand the three-dimensional hemodynamic characteristics, the wall shear stress distributions and secondary flows were investigated quantitatively.
It has been known that the pronounced hypotension resulting from hemorrhage gives rise to compensatory stimulation of the adrenosympathetic system, which leads to an increase of liberation of catecholamines from sympathetic nervous system and adrenal medulla. It is obvious, therefore, that numerous physiological and biochemical changes during the hemorrhagic hypotention might be mediated through the increased liberation of catecholamines. Although an extensive studies have been reported on changes of protein and carbohydrate metabolism in hemorrhagic shock a few studies on the changes of lipid metabolism have been reported. Levenson(1961) observed a marked increase of serum lipids content during hemorrhagic shock and also noticed a marked elevation of serum free fatty acids. He suggested that these effects were due to mobilization and accelerated metabolic breakdown of lipids which might be resulted by sympathetic stimulation as a cause. To elucidate the mechanism of this, author studied the change of serum free fatty acids and blood sugar with relation to catecholamines during experimentally induced hemorrhagic shock in dog. Healthy male mongrel dogs weighing approximately 15kg were used. Under the general anesthesia with pentobarbital, rapid hemorrhage was produced from the femoral artery maintaining blood pressure level of 40 mmHg measured by the manometer connected with the opposite femoral artery throughout the experiment. Serum free fatty acids(FFA) and blood sugar were measured by the methods of Dole(1956) and Folin-wu,(1920) respectively. Tissue catecholamine was measured by Shore and Olin method(1958) using Aminco-Bowman spectrophotofluorometer.
This study was conducted to examine the hypoglycemic effects of complexcity extract(DB 55) from five oriental medicinal plants on the intake of food, body weight, blood glucose level, insulin content, serum GOP, GPT, BUN and hematocrit value in streptozotocin(STZ)-induced diabetic rats. General nutritional composition was also assessed. Thirty- five male Sprague-Dawley rats were assigned to five groups : normal control group(NC), STZ-diabetic control group (STZ-control), STZ-diabetic DB-1 group(DB 55-1), STZ-diabetic DB-2 group(DB 55-2) and STZ-diabetic DB-3 group(DB 55-3). The animals were fed an experimental diet for 12 weeks. Body weight gain of the STZ-diabetic group was significantly lower but diet intake was significantly higher than the NC group. Blood glucose levels of the DB-1, DB-2 and DB-3 rats were significantly lower than the STZ-control animals. Insulin level of STZ-control rats was lower than the NC serum GOT, GPT and BUN levels were significantly higher in the NC group, but hematocrit value was not significant as compare to the STZ-diabetic group. Blood glucose level was lower observed for the DB 55-1, DB 55-2 and DB 55-3 group. The results will be useful in oriental diet therapy and in the developing functional food resources.
This study was performed to investigate the urinary methyl hippuric acid (UMHA) excretion among aircraft maintenance workers exposed to xylene by examination of blood test, urine test and biochemical tests. Study subjects (301) were divided into 2 groups; non-exposed group (n=123, who were not exposed to xylene), and exposed group (n=178, who were exposed to xylene). The average of blood level and MHA level was normal. The ${\gamma}$-GTP level was 35.5 IU/L (45.7%) in the exposed group and 27.9 IU/L (44.7%) in the non-exposed (control) group. The proportion of total cholesterol (TG) level higher than 150 mg/dl were 34.3% in the exposed group and 21.1% in the non-exposed group. These results were showed statistically significant difference (${\chi}^2$ = 6.10, P<0.05). Other items showed no statistically significant differences. The results of urine tests were no statistically significant differences in protein, occult blood and glucose level.
This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on blood hemoglobin, $HbA_{1c}$, levels and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM) and Araliaceae water extracts supplemented groups: Aralia elata (AE), Acanthopanacis cortex (AC) and Ulmus davidiana (UD). The extracts were supplemented in diet base on 11.42 g of raw Araliaceae/kg diet for 7 weeks. The diabetes was induced by injecting 572 (55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. Relative weights of liver were significantly lowered in the DM group compared to the normal group, whereas those of kidney and heart were significantly increased in the DM group. Supplementation of the Araliaceae water extracts improved reduced liver weights in STZ-induced diabetic rats. Blood glucose level was significantly higher in the DM group than in the normal group, whereas insulin contents were significantly lowered in the DM groups. However, these parameters were normalized in the An, AC and UD supplemented groups, respectively. Blood hemoglobin and $HbA_{1c}$ levels were significantly higher in the DM group than in the normal group. When all of Araliaceae water extracts were supplemented to the diabetic rats lowered hemoglobin and $HbA_{1c}$ levels. Red blood cell, white blood cell and Lymphocyte were significantly higher in the DM group than in the normal group. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. MCV, MCH contents were declined in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts elevated of these contents in STZ-induced diabetic rats. Accordingly, these results indicate that Aralia elata, Aeanthopanacis corex and Ulmus davidiana water extracts would seem to improve the blood biomarkers in STZ-induced diabetic rats.
Poly-ADP-ribosylation (PAR) is a post-translational modification of mainly chromosomal proteins. It is known to be strongly involved in several molecular events, including nucleosome-remodelling and carcinogenesis. In this investigation, it was attempted to evaluate PAR level as a reliable biomarker for early detection of cancer in blood lymphocyte histones. PAR of isolated histone proteins was monitored in normal and dimethylnitrosamine (DMN)-exposed mice tissues using a novel ELISA-based immuno-probe assay developed in our laboratory. An inverse relationship was found between the level of PAR and period of DMN exposure in various histone proteins of blood lymphocytes and spleen cells. With the increase in the DMN exposure period, there was reduction in the PAR level of individual histones in both cases. It was also observed that the decrease in the level of PAR of histones resulted in progressive relaxation of genomic DNA, perhaps triggering activation of genes that are involved in initiation of transformation. The observed effect of carcinogen on the PAR of blood lymphocyte histones provided us with a handy tool for monitoring biochemical or physiological status of individuals exposed to carcinogens without obtaining biopsies of cancerous tissues, which involves several medical and ethical issues. Obtaining blood from any patient and separating blood lymphocytes are routine medical practices involving virtually no medical intervention, post-procedure medical care or trauma to a patient. Moreover, the immuno-probe assay is very simple, sensitive, reliable and cost-effective. Therefore, combined with the ease of preparation of blood lymphocytes and the simplicity of the technique, immuno-probe assay of PAR has the potential to be applied for mass screening of cancer. It appears to be a promising step in the ultimate goal of making cancer detection simple, sensitive and reliable in the near future.
Dehydration, electrolyte disturbance, and acid-base imbalance are the most significant consequences of diarrhea in calves. We aimed to determine blood gas, hematological, electrolyte, and biochemical values and investigate the relationship between the physical status and blood parameters in Korean native calves (KNCs) with diarrhea. One hundred eighty KNCs with diarrhea (age < 75 days) were investigated. Blood samples were collected from the external jugular vein and analyzed using a portable clinical blood gas analyzer. The measured parameters were statistically compared according to the status of physical activity, dehydration, or prognosis. The mean values of parameters in the Calves with diarrhea showed metabolic acidosis, hyponatremia, and azotemia. The mean values of potassium, chloride, hematocrit, and hemoglobin were in the upper limit of their reference ranges. More than 75% of the calves had metabolic acidosis caused by bicarbonate loss, and 63.6% had high blood urea nitrogen (BUN) values. Moreover, BUN showed the highest correlation with the physical activity status and dehydration. pH, base excess of the extracellular fluid (BE), anion gap, potassium, hematocrit, bicarbonate, and hemoglobin were closely correlated with physical deterioration and dehydration (p < 0.001). BUN, pH, BE, and anion gap were closely correlated with physical deterioration and dehydration. These correlations between clinical symptoms and blood gas parameters can be clinically relevant in predicting the status of parameters according to clinical symptoms.
