Silk sericin, a natural protein from silkworm cocoons, is emerging as a multifunctional biomaterial in biomedicine, particularly in tissue engineering and wound healing. Recent studies have highlighted its biocompatibility, biodegradability, and potential for chemical modification, which allows it to be incorporated into various scaffold architectures. This review article synthesizes current research, including the development of sericin-based hydrogel scaffolds for tissue engineering and sericin's role in enhancing wound healing. Key findings demonstrate sericin's ability to refine scaffold porosity and mechanical strength, expedite tissue healing, and reduce bacterial load in wounds. The integration of sericin into novel bioactive dressings and its use in peripheral nerve injury repair are also discussed, showcasing its adaptability and efficacy. The convergence of these studies illustrates the broad applications of sericin, from scaffold design to clinical interventions, making it a promising material in regenerative medicine and tissue engineering, with the potential to improve patient outcomes significantly.

Exosomes are homogenous vesicles of 40-100 nm diameter produced endogenously. Exosomes are generated by inward budding into multi-vesicular bodies (MVB) and then released to extracellular space. Exosomes contain various nucleic acid and protein cargoes from their cells of origin and this endosomal cellular molecules are used for intracellular communication and for both promotion and suppression of immune responses. Recently, they are also considered as delivery vehicle for therapeutic proteins due to their characteristics of stability in body fluids and ability for target uptake. Also, they show less immune reactivity because the isolated exosome harboring therapeutic proteins can be from the same host. White-spotted flower chafer, Protaetia brevitarsis is one of the major insect commercially reared in Korea. There are bacterial and fungal pathogens causing diseases in the beetle, and these diseases incur economic loss to the larva-rearing farms. Due to their endosomal cargoes, exosomes are good candidates in use of disease diagnosis. In this study, we isolated insect exosome from the hemolymph of P. brevitarsis, and verified it by analysis of the exosome-specific surface proteins and RNA.
우리는 이전실험에서 배양세포를 사용하여 4종류의 천연물(Corydaline, (${\pm}$)-Car-3-ene-2,5-dione, Cinobufagin, Corilagin)이 ERAP1와 FOXO1 (DFA16) 유전자발현을 2배 이상 상승시키는 것을 증명하였다. 본 실험은 1% agar, 5% sucrose, natural compound $20{\mu}l$를 넣은 먹이를 만들어 4시간 starvation후에 4시간 동안 먹였다. Cinobufagin와 Corilagin를 먹이면 대조군에 비하여 6-8일 정도 더 생존하였다. RT-PCR 실험결과 ERAP1와 FOXO1 유전자 발현을 조절하는 것이 증명되었다. 산업곤충질병 진단과 치료에 사용될 것이며, 초파리를 대신한 생쥐에서도 동일한 결과를 실험하여 수명연장을 위한 신약으로 발전시킬 것이다.

This is the second catalog listing unrecorded invertebrates discovered during the research project 'Discovery of Korean Indigenous Species'. The data to compile the catalog were primarily gathered from the final reports of the project, between 2013 and 2021. We present 38 previously undocumented species, belonging to four phyla (Platyhelminthes, Annelida, Rotifera and Arthropoda). Samples were collected from intertidal coastal waters, soil, freshwater ponds, reservoirs and hosts in South Korea. In this study, we provide brief taxonomic information, including collection site (GPS), diagnosis, specimen vouchers, figures of representative individuals and the Korean name newly assigned, for each species. All data were reviewed and updated by experts working on the respective taxonomic group. The aim of the present study is to publish species that have been previously reported through the project. Upon publication, these species will be added to the 'National Species List of Korea', curated by the National Institute of Biological Resources(NIBR).

The Bombyx mori Microarray Database (BmMDB; http://silkworm.swu.edu.cn/microarray) provides information for tissue-specific gene expression by using the whole-genome oligonucleotide microarray in the silkworm. We analyzed the tissue-specific expression patterns in the silk gland, fat body, and midgut five days of fifth instar larvae during the development of B. mori. To verify the tissue-specific expression, analysis was conducted using quantitative Real-time RT-PCR and the highly expressed endogenous Actin RNA as an intrinsic reference. Finally, we confirmed five genes, (sw15872, sw00692, sw20990, sw05300,and sw2250), out of 18 candidates expressed in two different tissues, which was consistent with the data published by Dr. Xiang's group, thereby supporting the BmMDB. Further studies for promoter regions of candidate genes can be applied in creating transgenic silkworms as biomedical insects for use in producing biomaterials, and to serve as well-characterized models for understanding the mechanism for the genetic regulation of tissue-specific development.
Toxoplasma gondii infections occur throughout the world, and efforts are needed to develop various vaccine candidates expressing recombinant protein antigens. In this study, influenza matrix protein (M1) virus-like particles (VLPs) consisting of T. gondii rhoptry antigen 4 (ROP4 protein) were generated using baculovirus (rBV) expression system. Recombinant ROP4 protein with influenza M1 were cloned and expressed in rBV. SF9 insect cells were coinfected with recombinant rBVs expressing T. gondii ROP4 and influenza M1. As the results, influenza M1 VLPs showed spherical shapes, and T. gondii ROP4 protein exhibited as spikes on VLP surface under transmission electron microscopy (TEM). The M1 VLPs resemble virions in morphology and size. We found that M1 VLPs reacted with antibody from T. gondii-infected mice by western blot and ELISA. This study demonstrated that T. gondii ROP4 protein can be expressed on the surface of influenza M1 VLPs and the M1 VLPs containing T. gondii ROP4 reacted with T. gondii-infected sera, indicating the possibility that M1 VLPs could be used as a coating antigen for diagnostic and/or vaccine candidate against T. gondii infection.
Sf21 cells become popular as the host permissive cell line to support the baculovirus AcNPV replication and protein synthesis. The cells grow well on TC-100 medium that contains $0.1\%$ D-glucose as the major carbon source, strongly suggesting the presence of endogenous glucose transporters. However, unlike human glucose transporters, very little is known about the characteristics of the endogenoussugar transporter(s) in Sf21 cells. Thus, some kinetic properties of the sugar transport system were investigated, involving the uptake of 2-deoxy-D-glucose (2dG1c). In order to obtain a true measure of the initial rate of uptake, the uptake of $[^3H]2dGlc$ from both low $(100{\mu}M)$ and high (10 mM) extracellular concentrations was measured over periods ranging from 30 sec to30 min. The data obtained indicated that the uptake was linear for at least 2 min at both concentrations, suggesting that measurements made over a 1min time course would reflect initial rates of the jexpse uptake. To determine $K_m\;and\;V_{max}$ of the endogenous glucose transporter(s) in Sf21 cells, the uptake of 2dG1c was measured over a range of substrate concentrations $(50{\mu}M\~10mM)$ 2dG1c uptake by the Sf21 cells appeared to involve both saturable and non-saturable (or very low affinity) components. A saturable transport system for 2dG1c was relatively high, the $K_m$ value for uptake being < 0.45 mM. The $V_{max}$ value obtained for 2dG1c transport in the Sf21 cells was about 9.7-folds higher than that reported for Chinese hamster ovary cells, which contain a GLUT1 homologue. Thus, it appeared that the transport activity of the Sf21 cells was very high. 인류는 기후변화와 인구 구성 비율의 급격한 노령화 라는 두 가지 커다란 생존을 위협하는 문제점에 직면해 있다. 기후변화는 경제 발전과 운송 수단의 발달로 화석 연료 사용 증가에 따른 대기 중 온실가스 농도가 증가한 결과이고, 인구 구성 비율의 노령화는 선진국의 의 생명과학 기술 발전과 개인 위생의 증진으로 기대 수명이 증가한 결과이다. 돌이킬 수 없는 전 지구적인 기후변화를 피하기 위해서는 빠른 기간 내에 온실 가스의 배출이 없는 탄소 제로 경제로 전환을 해야 한다. 이 목표를 달성하기 위해서는 농업 중 온실가스의 발생이 가장 많은 낙농축산업을 저탄소 경영방식으로 전환하고 동시에 소비자들의 저탄소 식품들에 대한 인식의 변화가 필요하다. 현재 지구상 이용 가능 초지 중 77%가 가축용 사료 재배에 활용되지만, 인간이 섭취하는 전체 단백질의 37%와 총 열량의 18%만이 낙농축산업에서 얻어질 뿐이다. 그러므로, 가축보다 온실가스 배출량, 물의 사용량이 적고, 사육 공간이 작아도 되며 사료전환율이 높은 식용 곤충을 단백질원으로 활용해야 할 필요성이 있다. 이와 더불어 건강기능 증진 효과가 있다고 과학적으로 밝혀진 누에와 같은 곤충들의 기능성을 활용하여 현재 치료 방법이나 예방법이 확립되지 않은 퇴행성 질환들을 예방하고 치료를 촉진시킬 수 있는 기능성 식품 개발이 필요하다. 곤충은 동물 중 가장 오래 전에 지구상에 나타났고, 인간의 생존 유무와 상관없이 앞으로도 빠르게 진화를 하여 지구의 환경 변화에 적응하고 번성할 것이다. 그러므로, 다양한 식용 곤충과 누에를 포함한 약용 곤충을 이용한 산업은 현재 인류가 직면한 문제를 해결하여 미래에 인간이 지구에서 생존하고 번영할 수 있는 중요한 받침돌이 될 것이다.
Beta-amyloid peptide (A$\beta$) is a major component of senile plaques and its aggregation is considered to play a critical role in pathogenesis of Alzheimer's disease (AD). Aggregation of A$\beta$ could result from both increased synthesis and decreased degradation of A$\beta$. Our laboratory is interested in understanding the mechanism of A$\beta$ degradation in brain. Recently our laboratory identified a bacterial gene (SKAP) from Streptomyces sp KK565 whose protein product has an activity to cleave A$\beta$ and thus reduce the A$\beta$-induced neurotoxicity. The sequence analysis showed that this gene was closely related to aminopeptidase. Maldi-Tof analysis showed that the recombinant SKAP protein expressed in E. coli cleaves both A$\beta$ 40 and A$\beta$ 42 at the N-terminal of A$\beta$ while an aminopeptidase from Streptomyces griseus (SGAP) cleaves at the C-terminal. We also identified a mammalian homolog of SKAP and the recombinant mammalian protein expressed in Sf-9 insect cells showed a similar proteolytic activity to SGAP, cutting A$\beta$ at the C-terminus. I well discuss the detailed mechanism of the enzyme action and its functional implication in AD.
