Molecular Evidence for the Presence of CYP2E1 Retropseudogene in Human Genome (사람의 게놈에 존재하는 Cytochrome P450 2E1의 Retropseudogene에 대한 분자유전학적 증거)

  • We have carried out polymerase chain reaction (PCR) to investigate if retropseudogene for CYP2El is present in human genome. PCR primers were designed based on the structure of functional CYP2El gene and used to amplify both functional gene and retropseudogene in one reaction. From the repeated experiments we were able to amplify a previously unidentified CYP2El retropseudogene that was present in human genome. Its detailed structure was confirmed by Southern blotting and DNA sequencing. Nucleotide sequence of this retropseudogene was completely matched up to human liver CYP2El mRNA suggesting that the development of this retropseudogene might be a relatively recent event.

  • Vascular endothelial cell injury or dysfunction has been implicated in the onset and' progression of cardiovascular diseases including atherosclerosis. A number of previous studies have demonstrated that the pro-oxidative and pro-inflammatory pathways within vascular endothelium play an important role in the initiation and progression of atherosclerosis, Recent evidence has provided compelling evidence to indicate that interleukin-4 (IL-4) can induce proc inflammatory environment via oxidative stress-mediated up-regulation of inflammatory mediators such as cytokine, chemokine, and adhesion molecules in vascular endothelial cells. In addition, apoptotic cell death within vascular endothelium has been hypothesized to be involved in the development of atherosclerosis. Emerging evidence has demonstrated that IL-4 can induce apoptosis of human vascular endothelial cells through the caspase-3-dependent pathway, suggesting that IL-4 can increase endothelial cell turnover by accelerated apoptosis, the event which may cause the dysfunction of the vascular endothelium. These studies will have a high probability of revealing new directions that lead to the development of clinical strategies toward the prevention and/or treatment for individuals with inflammatory vascular diseases including atherosclerosis.

A Study on Real-Time Sports Activity Classification & Monitoring Using a Tri-axial Accelerometer (가속도 센서를 이용한 실시간 스포츠 동작 분류.모니터링에 관한 연구)

This study was conducted to study the real-time sports activity classification and monitoring using single waist mounted tri-axial accelerometer. This monitoring system detects events of sports activities such as walking, running, cycling, transitions between movements, resting and emergency event of falls. Accelerometer module was developed small and easily attachable on waist using wireless communication system which does not constrain sports activities. The sensor signal was transferred to PC and each movement pattern was classified using the developed algorithm in real-time environment. To evaluate proposed algorithm, experiment was performed with several sports activities such as walking, running, cycling movement for 100sec each and falls, transition movements(sit to stand, lie to stand, stand to sit, lie to sit, stand to lie and sit to lie) for 20 times each with 5 healthy subjects. The results showed that successful detection rate of the system for all activities was 95.4%. In this study, through sports activity monitoring. it was possible to classify accurate sports activities and to notify emergency event such as falls. For further study, the accurate energy consumption algorithm for each sports activity is under development.

Expression of BMP6 is Associated with its Methylation Status in Colorectal Cancer Tissue but Lacks Prognostic Significance

  • Background: The study aimed to evaluate the incidence of CpG island promoter methylation of BMP6, a member of the transforming growth factor beta family, in tissue samples from colorectal cancers (CRC) and look for its association with BMP6 expression and clinicopathological correlation. Materials and Methods: Methylation specific PCR for the BMP6 promoter region was performed with 85 frozen tissue samples of CRC and 45 of normal colon. Methylation status of MLH1 was also determined by the same method. Expression of BMP6 was evaluated by immunohistochemistry (IHC), using Allred's scoring system. The methylation status was analyzed against clinical and pathological parameters in CRC. Results: The study revealed BMP6 hypermethylation in 34 of 85 tumor specimens (40%), and 15 out of 45 normal tissue samples from CRC (33%). The incidence of hypermethylation was inversely correlated with IHC score. Allred's scores of 7 or more were correlated with lower frequency of BMP6 hypermethylation (29% compared to 50% in the remaining, p-value 0.049). However, there was no association between hypermethylation status and any clinicopathological parameters. The methylation status of BMP6 was not correlated with that of MLH1, a key methylation determinant in CRC. On survival analysis, there was no significant difference in progress-free survival (PFS) between the cases with and without hypermethylation (2-year PFS 74% and 76%, respectively). Conclusions: CpG island methylation of BMP6 is found in high frequency in CRC and this epigenetic event is associated with suppressed protein expression in the tumor tissue. However, the marker is not associated with tumor progression of the disease.

NEUROD1 Intrinsically Initiates Differentiation of Induced Pluripotent Stem Cells into Neural Progenitor Cells

  • Cell type specification is a delicate biological event in which every step is under tight regulation. From a molecular point of view, cell fate commitment begins with chromatin alteration, which kickstarts lineage-determining factors to initiate a series of genes required for cell specification. Several important neuronal differentiation factors have been identified from ectopic over-expression studies. However, there is scarce information on which DNA regions are modified during induced pluripotent stem cell (iPSC) to neuronal progenitor cell (NPC) differentiation, the cis regulatory factors that attach to these accessible regions, or the genes that are initially expressed. In this study, we identified the DNA accessible regions of iPSCs and NPCs via the Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq). We identified which chromatin regions were modified after neuronal differentiation and found that the enhancer regions had more active histone modification changes than the promoters. Through motif enrichment analysis, we found that NEUROD1 controls iPSC differentiation to NPC by binding to the accessible regions of enhancers in cooperation with other factors such as the Hox proteins. Finally, by using Hi-C data, we categorized the genes that directly interacted with the enhancers under the control of NEUROD1 during iPSC to NPC differentiation.

A COMPARATIVE STUDY UPON EVENT-RELATED POTENTIALS OF THE PATIENTS WITH ADHD AND NORMAL CHILDREN USING FOURIER TRANSFORMATION AND WAVELET ANALYSIS (푸리에 변환과 웨이브렛 분석을 통한 주의력결핍 ${\cdot}$ 과잉운동장애 아동과 정상 아동의 사건관련전위 비교 연구)

  • Using Fourier transformation and wavelet analysis, we compared the auditory event-related potentials of the patients with attention deficit-hyperactivity disorders(abbr. ADHD, 13 boys) and normal control children(8 boys). Amplitudes of the event-related potentials which were calculated via Fourier transformation were compared between the groups and between conditions(non-target versus target) in each group. To the non-target stimuli, the patients with ADHD showed significantly greater amplitudes across almost all of the electrode sites and frequencies. To the target stimuli, the incidents which ADHD patients showed much higher amplitudes than normal controls significantly decreased, while those of the reverse results increased significantly. These results were consistent with the comparison results about negative difference wave(abbr. Nd wave) using Fourier transformation. In summary, it was proved that non-target stimulus which should be ignored elicited more robust electrical response from the patients with ADHD than normal children, but the target stimulus which reguired active processing did much less electrical activity in the patients. For the patients, they showed much inhibited electrical response to the target stimuli in some electrodes and frequency ranges. Normal children were more strongly stimulated by the target stimuli in almost all electrodes and frequency ranges than the patients, but less in prefrontal leads and frontal leads. Wavelet analysis results proved that early responses(0-300msec) to the nontarget stimuli of the patients were significantly greater than the normal controls in prefrontal, anterior frontal, some parts of temporal, and occipital lobes and that late response(300-370msec) were significantly lesser than normal children in parietal and central electrodes. Target stimuli elicited significantly higher electrical activity in both group than non-target stimuli did. Prefrontal and frontal lobes showed stronger responses in the patients than normal children irrespective of stimulus condition, but parietal and temporal lobes did higher activities in normal children than the patients only to the target stimuli. In conclusion, the patients with ADHD showed much greater responses to the stimuli which should be ignored, but failed to activated the necessary processes to the target stimuli. Also, we found that the frequency-dimension analysis and wavelet analysis were useful for the signal processing such as event related potentials.

  • 생명과학 관련 문서에서의 이벤트 추출은 관련 연구자들의 연구에 많은 도움을 줄 수 있다. 기존의 연구에서는 주로 이벤트 동사에 대해 패턴을 정의한 후에 정의된 패턴에 의해서만 이벤트를 추출하고자하였다. 그러나 모든 패턴을 수동으로 정의하는 것은 너무 많은 비용이 들기 때문에 패턴을 자동 추출 또는 확장하는 방법이 필요하다. 또한 학습을 하기 위해서는 상당수의 학습 말뭉치가 있어야 하는데 그것 또한 충분하지 않은 실정이다. 본 논문에서는 초기 패턴에 의해 생성된 소량의 정답 이벤트로부터 학습한 후 공기정보와 패턴정보를 이용한 Co-training방법으로 패턴 확장 및 이벤트 추출을 시도하였다. 실험 결과, 이벤트 동사의 패턴 정보가 유용한 정보라는 것을 확인할 수 있었고, 후보 이벤트 내의 개체간 공기정보와 문법관계정보 또한 매우 중요한 정보라는 것을 새롭게 보일 수 있었다. GENIA 말뭉치에서 162개의 이벤트 동사에 대해 실험한 결과, 88.02%의 정확률, 79.25%의 재현율을 얻었다.

Management of the First-time Traumatic Anterior Shoulder Dislocation

  • Traumatic anterior dislocation of the shoulder is one of the most common directions of instability following a traumatic event. Although the incidence of shoulder dislocation is similar between young and elderly patients, most studies have traditionally focused on young patients due to relatively high rates of recurrent dislocations in this population. However, shoulder dislocations in older patients also require careful evaluation and treatment selection because they can lead to persistent pain and disability due to rotator cuff tears and nerve injuries. This article provides an overview of the nature and pathology of acute primary anterior shoulder dislocation, widely accepted management modalities, and differences in treatment for young and elderly patients.

  • In this paper, we present the Computerized Polysomnographic System, which we developed for aiding manual polysomnographic diagnonis. This system is composed of four sub parts: patient information management tool, polysomno-graphic signal acquisition tool, signal analysis tool, and result-reporting tool. We calculated the parameters of sleep diagnosis, both on the sleep stage hypnogram and event lists. We developed this system on the Windows NT 4.0 and Pentium Pro 200Mhz. We are testing this system on the clinical site.

A Study on the Developement of Digital Polysomnograph System (디지탈 수면다원검사 시스템 개발)

  • We have developed the digital polysomnograph system to enhance the manual sleep study. This system is composed of host PC and target board. The host computer is IBM-PC Pentium 133 MHz and the target board is PC31 (processor type: TMS320C31). These two Processors communicate with each other by dualport RAM. Target board acquire 16 channel sleep signals from the Polysomnographic system (GRASS model 78, USA) and digitize them. We used high resolution monitor$(1600{\times}1280)$ to simulate the paper-recording quality of polysomnographic signal. We also implemented the GUI based polysomnographic staging program on the windows environment. Clinicians can score the sleep stage, and edit and mark the event efficiently. Finally it can support making patient database.
