Aluminum (Al) stress in acidic pH is known to decrease the growth and productivity of alfalfa. However, not much is known about how the application of silicon (Si) affects the Al stress response in alfalfa. This study was conducted to evaluate the effect of exogenous application of Si on the growth of alfalfa seedlings exposed to Al stress in pots. Alfalfa seedlings grown in pots for 2 weeks were treated either Al stress (pH 4.0, 0.2 mM Al) or Al stress + Si (1 mM) for 5 days, lengths and biomass of shoot and root, and chlorophyll and carotenoid contents in leaf tissues were analyzed respectively. Al stress treatment inhibited shoot and root growth, and decreased fresh and dry weights, and chlorophyll content in leaves, but increased carotenoid content. In contrast, when alfalfa seedlings treated with Al stress combined with Si, delayed growth caused by Al stress of shoot and root of alfalfa seedlings was restored, dry weight was increased and chlorophyll content of leaf tissue was increased, but carotenoid content was decreased. These results suggest that Si has a function of alleviating Al toxicity in alfalfa, of which it exhibits a mitigating effect by a function that overlaps with some of the intracellular functions of carotenoids.
L-Malic acid (L-MA) is widely used in food and non-food products. However, few microorganisms have been able to efficiently produce L-MA from xylose derived from lignocellulosic biomass (LB). The objective of this work is to convert LB into L-MA with the concept of a bioeconomy and environmentally friendly process. The unique trifunctional xylanolytic enzyme, PcAxy43A from Paenibacillus curdlanolyticus B-6, effectively hydrolyzed xylan in untreated LB, especially corn hull to xylose, in one step. Furthermore, the newly isolated, Acetobacter tropicalis strain H1 was able to convert high concentrations of xylose derived from corn hull into L-MA as the main product, which can be easily purified. The strain H1 successfully produced a high L-MA titer of 77.09 g/l, with a yield of 0.77 g/g and a productivity of 0.64 g/l/h from the xylose derived from corn hull. The process presented in this research is an efficient, low-cost and environmentally friendly biological process for the green production of L-MA from LB.
Although engineered Saccharomyces cerevisiae fermenting cellobiose is useful for the production of biofuels from cellulosic biomass, cellodextrin accumulation is one of the main problems reducing ethanol yield and productivity in cellobiose fermentation with S. cerevisiae expressing cellodextrin transporter (CDT) and intracellular β-glucosidase (GH1-1). In this study, we investigated the reason for the cellodextrin accumulation and how to alleviate its formation during cellobiose fermentation using engineered S. cerevisiae fermenting cellobiose. From the series of cellobiose fermentation using S. cerevisiae expressing only GH1-1 under several culture conditions, it was discovered that small amounts of GH1-1 were secreted and cellodextrin was generated through trans-glycosylation activity of the secreted GH1-1. As GH1-1 does not have a secretion signal peptide, non-conventional protein secretion might facilitate the secretion of GH1-1. In cellobiose fermentations with S. cerevisiae expressing only GH1-1, knockout of TLG2 gene involved in non-conventional protein secretion pathway significantly delayed cellodextrin formation by reducing the secretion of GH1-1 by more than 50%. However, in cellobiose fermentations with S. cerevisiae expressing both GH1-1 and CDT-1, TLG2 knockout did not show a significant effect on cellodextrin formation, although secretion of GH1-1 was reduced by more than 40%. These results suggest that the development of new intracellular β-glucosidase, not influenced by non-conventional protein secretion, is required for better cellobiose fermentation performances of engineered S. cerevisiae fermenting cellobiose.

Process-based model (PBM), based on the interactions between endogenous physiological processes and many environmental factors, can be a powerful tool for estimating crop growth and productivity. Carbon acquisition and biomass accumulation are the main components in PBM, so it has become important to understand and integrate gas exchange process in crop model. This study aimed to assess the applicability of FvCB model (a leaf model of C3 photosynthesis proposed by Farquhar, von C aemmerer, and Berry (1980)) in onion (Allium cepa L.). For parameterization, two early-maturing onion cultivars, 'Singsingball' and 'Thunderball', grown in a temperature gradient plastic film house, were used in measuring leaf net CO2 assimilation rate (A), and then, parameter estimation was carried out for four parameters including Vcmax (maximum rate of carboxylation), Jmax (maximum rate of electron transport), TPU (rate of triose phosphate utilization), and Rd (Dark respiration rate). The gas-exchange model calibrated in this research is expected to be able to explain the photosynthetic responses of onion under various environmental conditions (R2=0.95***).
Background: Culturing wild ginseng adventitious root using plant factory technology provides genetic safety and high productivity. This production technology is drawing attention in the fields of functional raw materials and product development. The cultivation method using elicitors is key technology for controlling biomass and increasing secondary metabolites. Methods and Results: Elicitor treatments using methyl jasmonate, pyruvic acid, squalene, β-sistosterol were performed to amplify total ginsenosides (Rb1, Rc, Rb2, Rb3, and Rd) of cultured wild ginseng adventitious root. Thereafter, fermentation and steaming processes were performed to convert total ginsenosides into minor molecular ginsenosides (Rg3, Rk1, and Rg5). The result indicated that methyl jasmonate minimizes the reduction in fresh weight of cultured wild ginseng adventitious root and maximizes total ginsenosides (sum of Rb1, Rc, Rb2, Rb3, and Rd). Ginsenoside conversion results showed a maximum degree of conversion of 131 mg/g. Conclusions: In this study, we demonstrated that the optimal elicitor treatment method increased the content of total ginsenosides, while the steaming and fermentation processing method increased the content of minor ginsenosides.

Rye, whole-crop barley and Italian Ryegrass are major winter forage species in Korea, and yield monitoring of winter forage species is important to improve forage productivity by precision management of forage. Forage monitoring using Unmanned Aerial Vehicle (UAV) has offered cost effective and real-time applications for site-specific data collection. To monitor forage crop by multispectral camera with UAV, we tested four types of vegetation index (Normalized Difference Vegetation Index; NDVI, Green Normalized Difference Vegetation Index; GNDVI, Normalized Green Red Difference Index; NGRDI and Normalized Difference Red Edge Index; NDREI). Field measurements were conducted on paddy field at Naju City, Jeollanam-do, Korea between February to April 2019. Aerial photos were obtained by an UAV system and NDVI, GNDVI, NGRDI and NDREI were calculated from aerial photos. About rye, whole-crop barley and Italian Ryegrass, regression analysis showed that the correlation coefficients between dry matter and NDVI were 0.91~0.92, GNDVI were 0.92~0.94, NGRDI were 0.71~0.85 and NDREI were 0.84~0.91. Therefore, GNDVI were the best effective vegetation index to predict dry matter of rye, wholecrop barley and Italian Ryegrass by UAV system.

Bio-based alternative leathers may be produced from biomass fiber, protein polymers, bacterial cellulose, and mushroom mycelia. Of these components, mushroom mycelia are of greatest interest. In this study, the potential of Fomes fomentariusas a mushroom mycelial mat was confirmed, and the optimal strain for the development of the mycelial mat was determined. Moreover, the quality of the mycelial mat was improved by identifying an efficient culture method to increase productivity. Mutant strains whose independence was verified were obtained by treatment with gamma irradiation under various conditions. Biofilm formation by the resulting strains was examined in sawdust and liquid media and the characteristics of the biofilms were analyzed. The biofilm of the mutant strains showed results that were similar to or better than the biofilms of longevity and cypress mushrooms. These findings are expected to be utilized in future research aimed at discovering new biomaterials using mushroom mycelia.
Crop productivity can be obstructed by various biotic and abiotic stresses and thus these stresses are a threat to universal food security. The information on the use of viruses providing efficacy to plants facing growth challenges owing to stress is lacking. The role of induction of pathogen-related genes by microbes is also colossal in drought-endurance acquisition. Studies put forward the importance of viruses as sustainable means for defending plants against dual stress. A fundamental part of research focuses on a positive interplay between viruses and plants. Notably, the tomato yellow leaf curl virus (TYLCV) and tomato chlorosis virus (ToCV) possess the capacity to safeguard tomato host plants against severe drought conditions. This study aims to explore the combined effects of TYLCV, ToCV, and drought stress on two tomato cultivars, Money Maker (MK, UK) and Shalala (SH, Azerbaijan). The expression of pathogen-related four cellulose synthase gene families (CesA/Csl) which have been implicated in drought and virus resistance based on gene expression analysis, was assessed using the quantitative real-time polymerase chain reaction method. The molecular tests revealed significant upregulation of Ces-A2, Csl-D3,2, and Csl-D3,1 genes in TYLCV and ToCV-infected tomato plants. CesA/Csl genes, responsible for biosynthesis within the MK and SH tomato cultivars, play a role in defending against TYLCV and ToCV. Additionally, physiological parameters such as "relative water content," "specific leaf weight," "leaf area," and "dry biomass" were measured in dual-stressed tomatoes. Using these features, it might be possible to cultivate TYLCV-resistant plants during seasons characterized by water scarcity.

Faced with aging populations, declining resources, and limited agricultural productivity, rural areas in South Korea require innovative solutions. This study investigated the potential of drone-based vegetation indices (VIs) to analyze soybean growth patterns in open-field smart agriculture in Goesan-gun, Chungbuk Province, South Korea. We monitored multi-seasonal normalized difference vegetation index (NDVI) and the normalized difference red edge (NDRE) data for three soybean lots with different irrigation methods (subsurface drainage, conventional, subsurface drip irrigation) using drone remote sensing. Combining NDVI (photosynthetically active biomass, PAB) and NDRE (chlorophyll) offered a comprehensive analysis of soybean growth, capturing both overall health and stress responses. Our analysis revealed distinct growth patterns for each lot. LotA(subsurface drainage) displayed early vigor and efficient resource utilization (peaking at NDVI 0.971 and NDRE 0.686), likely due to the drainage system. Lot B (conventional cultivation) showed slower growth and potential limitations (peaking at NDVI 0.963 and NDRE 0.681), suggesting resource constraints or stress. Lot C (subsurface drip irrigation) exhibited rapid initial growth but faced later resource limitations(peaking at NDVI 0.970 and NDRE 0.695). By monitoring NDVI and NDRE variations, farmers can gain valuable insights to optimize resource allocation (reducing costs and environmental impact), improve crop yield and quality (maximizing yield potential), and address rural challenges in South Korea. This study demonstrates the promise of drone-based VIs for revitalizing open-field agriculture, boosting farm income, and attracting young talent, ultimately contributing to a more sustainable and prosperous future for rural communities. Further research integrating additional data and investigating physiological mechanisms can lead to even more effective management strategies and a deeper understanding of VI variations for optimized crop performance.
The potential polyhydroxyalkanoates (PHA)-producing bacteria, Bacillus megaterium PP-10, was successfully isolated and studied its feasibility for utilization of pineapple peel waste (PPW) as a cheap carbon substrate. The PPW was pretreated with 1% (v/v) H2SO4 under steam sterilization and about 26.4 g/l of total reducing sugar (TRS) in pineapple peel hydrolysate (PPH) was generated and main fermentable sugars were glucose and fructose. A maximum cell growth and PHA concentration of 3.63 ± 0.07 g/l and 1.98 ± 0.09 g/l (about 54.58 ± 2.39%DCW) were received in only 12 h when grown in PPH. Interestingly, PHA productivity and biomass yield (Yx/s) in PPH was about 4 times and 1.5 times higher than in glucose. To achieve the highest DCW and PHA production, the optimal culture conditions e.g. carbon to nitrogen ratios of 40 mole/mole, incubation temperature at 35℃ and shaking speed of 200 rpm were performed and a maximum DCW up to 4.24 ± 0.04 g/l and PHA concentration of 2.68 ± 0.02 g/l (61% DCW) were obtained. The produced PHA was further examined its monomer composition and found to contain only 3-hydroxybutyrate (3HB). This finding corresponded with the presence of class IV PHA synthase gene. Finally, certain thermal properties of the produced PHA i.e. the melting temperature (Tm) and the glass transition temperature (Tg) were about 176℃ and -4℃, respectively whereas the Mw was about 1.07 KDa ; therefore, the newly isolated B. megaterium PP-10 is a promising bacterial candidate for the efficient conversion of low-cost PPH to PHA.
