버섯병원균에 대한 길항세균 Bacillus sp. SD-10이 생산하는 항균물질의 분리 (Isolation of Antimicrobial Substance by Produced Bacillus sp. SD-10 with Antagonistic Activity Towards Mushroom Pathogens)

  • Bacillus sp. SD-10이 생산하는 항균물질을 이용하여 버섯의 갈색무늬병균 및 푸른곰팡이병균에 대한 천연물 버섯병해 방제약재 개발을 위한 기초적 연구를 행하였다. Bacillus sp. SD-10은 35∼4$0^{\circ}C$의 배양온도에서 30∼40 hrs 배양하였을 때 가장 높은 항균활성을 나타내었다. 이 배양액을 이용하여 T. virens의 균사 생육저해 및 포자발아 억제 농도를 검토한 결과, 균사생육은 30 ul 이상 처리 시 뚜렷하게 저해되었으며 포자발아는 평판배지에 5%(v/v) 이상 첨가 시 완전하게 억제되었다. Bacillus sp. SD-10의 배양액으로부터 항균물질을 순수분리하여 UPX-1으로 명명하고, UPX-1을 $^1H-$NMR과 $^{13}C$-NMR 스펙트럼으로 구조를 동정한 결과 탄소 6개로 구성된 당 2개가 2당류 구조로 결합되어 있는 화합물임이 밝혀졌다. UPX-1은 T. virens 뿐 아니라 Pse. tolaassi에도 아주 강한 항균효과를 나타내었다.

  • In this study, we have studied the fabrication and the performance evaluation of digital radiation detector of the based on selenium (a-Se) prototype which is widely researched about recently. The detector was fabricated using amorphous selenium in the specification of active area size $7{\times}8.5"$, pixel pitch $139{\mu}m$, and 12 bit ADC. In order for the performance evaluation of the fabricated detector, we used radiation quality RQA 5 that is suggested by the International Electrotechnical Commission (IEC), and evaluated modulation transfer function (MTF), noise power spectrum (NPS), and detective quantum efficiency (DQE). Concerning MTF measurement, we used slit camera (Nuclear Associates, Model : 07-624-2222), and evaluated in the slit method. Also so as to compare the performance evaluation on the detector fabricated in this study, we used Hologic Direct-Ray (DR-1000) and GE Revolution XQ/I system, and evaluated and compared in the same method MTF, NPS, and DQE which are image quality factors. And as a result, the MTF of each detector In Nyquist frequency were evaluated to be 58% (at 3.5 lp/mm) in the case of DR-1000 and 65% (at 2.5 lp/mm) in the case of XQ/I, and that for the detector fabricated in this study was evaluated to be 36% (at 3.51 lp/mm). Also in the case of DQE(0), the detector fabricated in this study, DR-1000 of Hologic company, and XQ/I system of GE company respectively were evaluated as 36%, 32%, and 50%.

Regulation of Nicotinic Acetylcholine Receptor by Tyrosine Kinase in Autonomic Major Pelvic Ganglion Neurons

  • It is widely known that protein tyrosine kinases (PTKs) are involved in controlling many biological processes such as cell growth, differentiation, proliferation, survival and apoptosis. An $\alpha3\beta4$ subunit combination acts as a major functional acetylcholine receptor (nAChRs) in male rat major pelvic ganglion (MPG) neurons, and their activation induces fast inward currents and intracellular calcium increases. Recently it has been reported that the activity of acetylcholine receptors (AChRs) in some neurons can be negatively regulated by PTKs. However, the exact mechanism of regulation of nAChRs by PTKs is poorly understood. Therefore, we examined the potential role particular in nAChR by PTK using electrophysiology and calcium imaging in male rat MPG neurons. ACh induced inward currents and $(Ca^{2+})_i$ increases in MPG neurons, concomitantly. These responses were inhibited by more than 90% in $Na^+$- or $Ca^{2+}$- free solution. $\alpha$-conotoxin AuIB, a selective $\alpha3\beta4$ nAChR blocket, inhibited ACh-induced inward currents. Genistein (10 $\mu$M), a broad-spectrum tyrosine kinase inhibitor, markedly decreased ACh-induced currents and $Ca^{2+}$ transients, whereas 10 $\mu$M genistin, an inactive analogue, had little effect. Overall these data suggest that the activities of $\alpha3\beta4$ AChRs in MPG neurons are positively regulated by PTK. In conclusion, trosine kinase may be one of the key factors in the regulation of $\alpha3\beta4$ nAChRs in rat MPG neurons, which may play an important roles in the autonomic neuronal function such as synaptic transmission, autonomic reflex, and neuronal plasticity.

Production of Surfactin and Iturin by Bacillus licheniformis N1 Responsible for Plant Disease Control Activity

  • Bacillus licheniformis N1, previously developed as a biofungicide formulation N1E to control gray mold disease of plants, was investigated to study the bacterial traits that may be involved in its biological control activity. Two N1E based formulations, bacterial cell based formulation PN1E and culture supernatant based formulation SN1E, were evaluated for disease control activity against gray mold disease of tomato and strawberry plants. Neither PN1E nor SN1E was as effective as the original formulation N1E. Fractionation of antifungal compounds from the bacterial culture supernatant of B. licheniformis N1 indicated that two different cyclic lipopeptides were responsible for the antimicrobial activity of the N1 strain. These two purified compounds were identified as iturin A and surfactin by HPLC and LCMS. The purified lipopeptides were evaluated for plant disease control activity against seven plant diseases. Crude extracts and purified compounds applied at 500 ${\mu}g/ml$ concentration controlled tomato gray mold, tomato late blight and pepper anthracnose effectively with over 70% disease control value. While iturin showed broad spectrum activity against all tested plant diseases, the control activity by surfactin was limited to tomato gray mold, tomato late blight, and pepper anthracnose. Although antifungal compounds from B. licheniformis N1 exhibited disease control activity, our results suggested that bacterial cells present in the N1E formulation also contribute to the disease control activity together with the antifungal compounds.

양식산 굴로부터 쓴맛 성분의 분리 및 성질 (Isolation and Some Properties of Bitter Taste Compounds from Cultured Oyster, Crassostrea gigas)

  • 1989년 11월부터 1990년 1월 사이에 전남 가막만에서 채취한 양식 굴 및 이를 원료로 제조된 훈제 기름담금 통조림, 보일드 통조림에서 강한 쓴맛이 나타나 품질상 문제가 발생하였는바, 본 연구에서는 이러한 쓴맛의 원인 물질을 규명하고저 시도하였다. 이들 시료의 아세톤 추출액을 액-액 분배, 알루미나, 규산 및 ODS 등의 각종 칼럼을 이용하여 정제한 결과, 5개의 쓴맛성분을 분리하였으며, 이들은 질량분석(FAB-MS)으로 부터 분자량이 각각, OY-22 : 837, OY-23 : 851, OY-24 : 821, OY-25, OY-26 : 835 이었으며, proton NMR spectrum 및 각종 기기분석 결과, 6-7개의 아미노산으로 이루어진 환상 peptide로 추정되었다. 또한, 아미노산 분석으로부터 OY-24에서는 Val, Leu 이, OY-25에서는 Leu, Ile이, OY-26에서는 Leu, Leu이 각각 확인되었으며, 나머지는 이상 아미노산으로 생각되었다. 이들 성분들은 마우스에 대하여 급성독성은 없었으며 $(100{\mu}g/20g\;mice,\;I.p.)$, Asp. niger E. coli, Bac. subtilis 에 대하여는 항균성도 없었다$(10{\mu}g/disk).$ 현재, 각종 생리활성 및 정확한 구조를 검토 중에 있다.

막걸리 유래 미생물의 활용을 위한 연구 동향 (Application of Functional Microbial Strains Isolated from Traditional Rice Wine in Korea)

  • 우리나라는 예전부터 전통적인 방법으로 누룩을 제조하고, 이를 발효제로 막걸리를 만들어 왔다. 막걸리는 여과 또는 살균 과정없이 다양한 미생물을 살아 있는 상태로 섭취하기 때문에 영양학적으로나 기능적인 측면에서 가치가 높다. 최근 많은 연구에서 막걸리로부터 미생물을 분리동정하고 다양한 기능성에 대해 스크리닝한 결과, 높은 프로바이오틱스 활성과 다양한 스펙트럼의 항균활성을 가진 균주들이 선별되었다. 특히 일부 유산균들은 GABA와 EPS 등의 기능성 물질을 생성하기도 했다. 또한, 일부 유산균과 효모의 경우 각각 bacteriocin 및 killer toxin을 통해 항균활성을 나타내는 것으로 여겨진다. 이러한 막걸리 유래 기능성 미생물과 그 대사산물은 기능성 식품 소재나 안전한 식품첨가물 및 다양한 산업분야에서 활용될 수 있을 것이다.

안정상태 시각유발전위 기반의 기능적 전기자극 재활훈련 시스템 (Steady-State Visual Evoked Potential (SSVEP)-based Rehabilitation Training System with Functional Electrical Stimulation)

  • The purpose of the brain-computer (machine) interface (BCI or BMI) is to provide a method for people with damaged sensory and motor functions to use their brain to control artificial devices and restore lost ability via the devices. Functional electrical stimulation (FES) is a method of applying low level electrical currents to the body to restore or to improve motor function. The purpose of this study was to develop a SSVEP-based BCI rehabilitation training system with FES for spinal cord injured individuals. Six electrodes were attached on the subjects' scalp ($PO_Z$, $PO_3$, $PO_4$, $O_z$, $O_1$ and $O_2$) according to the extended international 10-20 system, and reference electrodes placed at A1 and A2. EEG signals were recorded at the sampling rate of 256Hz with 10-bit resolution using a BIOPAC system. Fast Fourier transform(FFT) based spectrum estimation method was applied to control the rehabilitation system. FES control signals were digitized and transferred from PC to the microcontroller using Bluetooth communication. This study showed that a rehabilitation training system based on BCI technique could make successfully muscle movements, inducing electrical stimulation of forearm muscles in healthy volunteers.

신약전달기술체계인 유전자 치료의 현재까지의 개발동향 (Current Status of Gene Therapy as a New Drug Delivery System)

  • Gene therapy is fundamentally a sophisticated drug delivery technology to cure a disease by the transfer of genetic material to modify living cells. In other words, the gene is used as a therapeutic drug much like a chemical compound is employed in chemotherapy. Currently almost 600 clinical trials are underway worldwide since the first clinical trials carried out in 1990 to treat adenosine deaminase deficiency using retroviral vectors. Despite the great progress still is there no gene therapy product being approved as a new drug. This is partly due to a lack of an ideal gene delivery system that is safe and can provide stable, optimal level production of the therapeutic proteins in the cell. This review covers the current status of several different biological and physico-chemical agents that are being developed as gene delivery vehicles. Although gene therapy promises great hopes toward the cure of a broad spectrum of genetic and acquired diseases, the success of gene therapy heavily asks for the development of vector systems for safe and efficient application in humans.

서포트 벡터 머신을 이용한 심폐소생술 변이의 변화에 따른 제세동 성공률 분석 (Analysis of the Likelihood of Successful Defibrillation as a Change of Cardiopulmonary Resuscitation Transition using Support Vector Machine)

  • Unsatisfied results of return of spontaneous circulation (ROSC) estimates were caused by the fact that the predictability of the predictors was insufficient. This unmet estimate of the predictors may be affected by transitional events due to behaviors which occur during cardiopulmonary resuscitation (CPR). We thus hypothesized that the discrepancy of ROSC estimates found in statistical characteristics due to transitional CPR events, may affect the performance of the predictors, and that the performance of the classifier dichotomizing between ROSC and No-ROSC might be different during CPR. In a canine model (n=18) of prolonged ventricular fibrillation (VF), standard CPR was provided with administration of two doses of epinephrine 0 min or 3 min later of the onset of CPR. For the analysis of the likelihood of a successful defibrillation during CPR, Support Vector Classification was adopted to evaluate statistical peculiarity combining time and frequency based predictors: median frequency, frequency band-limited power spectrum, mean segment amplitude, and zero crossing rates. The worst predictable period showed below about 1 min after the onset of CPR, and the best predictable period could be observed from about 1.5 min later of the administering epinephrine through 2.0-2.2 min. As hypothesized, the discrepancy of statistical characteristics of the predictors was reflected in the differences of the classification performance during CPR. These results represent a major improvement in defibrillation prediction can be achieved by a specific timing of the analysis, as a change in CPR transition.

Synthesis of Biosurfactant-Based Silver Nanoparticles with Purified Rhamnolipids Isolated from Pseudomonas aeruginosa BS-161R

  • The biological synthesis of nanoparticles has gained considerable attention in view of their excellent biocompatibility and low toxicity. We isolated and purified rhamnolipids from Pseudomonas aeruginosa strain BS-161R, and these purified rhamnolipids were used to synthesize silver nanoparticles. The purified rhamnolipids were further characterized and the structure was elucidated based on one- and two-dimensional $^1H$ and $^{13}C$ NMR, FT-IR, and HR-MS spectral data. Purified rhamnolipids in a pseudoternary system of n-heptane and water system along with n-butanol as a cosurfactant were added to the aqueous solutions of silver nitrate and sodium borohydride to form reverse micelles. When these micelles were mixed, they resulted in the rapid formation of silver nanoparticles. The synthesized nanoparticles were characterized by UV-Visible spectroscopy, transmission electron microscopy, and energy dispersive X-ray spectroscopy (EDS). The nanoparticles formed had a sharp adsorption peak at 410 nm, which is characteristic of surface plasmon resonance of the silver nanoparticles. The nanoparticles were monodispersed, with an average particle size of 15.1 nm (${\sigma}={\pm}5.82$ nm), and spherical in shape. The EDS analysis revealed the presence of elemental silver signal in the synthesized nanoparticles. The formed silver nanoparticles exhibited good antibiotic activity against both Grampositive and Gram-negative pathogens and Candida albicans, suggesting their broad-spectrum antimicrobial activity.
