나프탈렌 분해균주인 Pseudomonas aeruginosa CZ6을 오염된 토양에서 분리하였으며 분리된 균주는 결정상태의 나프탈렌에 부착하고 그 주변에 extracellular polymeric substance를 분비하는 특성을 가졌다. LB, YM과 MSM 배지를 사용하여 배지의 종류에 따른 biofilm 생성량을 측정한 결과, LB 배지에서 biofilm이 가장 많이 생성되는 것으로 나타났다. 나프탈렌을 기질로 한 배양조건에서 균주는 기질의 농도 영향을 크게 받지 않고 0.10% 기질, 150 rpm 조건에서 최적 배양조건은 $30^{\circ}C$, pH 7로 나타났다. 두 가지 토양에서 배지의 종류에 따른 나프탈렌의 분해특성을 관찰한 결과 초기에는 MSM 배지에서 나프탈렌이 가장 많이 분해가 되었다. 그러나 생물이용성이 제한을 받는 조건에서 LB 배지의 나프탈렌이 가장 빨리 제거가 되었다. 이런 결과는 biofilm의 형성과 extracellular polymeric substance 생성이 토양에 흡착된 잔류 나프탈렌의 생물학적 이용성을 향상시키기 때문인 것으로 사료된다.
Biofilm 내부의 항생제 내성 박테리아의 성장형태는 만성감염과 질병을 발생한다. 인위적 형성한 biofilm의 체외실험 모델 시스템 통한 항생제 침투 실험을 수행하였다. 항생제 내성 균주 (E. coli, S. aureus)는 항생제 내성균주 은행으로부터 획득하였다 Ca-alginate bead를 인위적 biofilm으로 사용하였고, 세포 생존률을 향상시키기 위해 공기 압축을 이용한 세포 포획 실험도 측정되었다. biofilm의 항생제 감수성은 항생제의 농도 따라 최저 저해 농도 (MIC)를 이용하여 측정되었다. bead생성에 따른 안정성은 bead를 형성하지 않은 세포와 비교하여 감소하였다. 항생제에 민감한 E. coli의 경우 시간이 지남에 따라 균체수도 감소하였으나, 항생제 내성 E. coli는 일정한 균체수를 유지하였다. bead형성 후 항생제 투여 효과는 항생제 내성이 있고, 낮은 농도의 항생제를 처리할수록 더 높은 생존률을 보였다.
본 연구는 Planktonic growth inhibition test와 CDC Biofilm Reactor를 사용한 Biofilm assay를 통해 S-PRG filler를 함유하는 치면열구전색제의 S. mutans에 대한 항미생물 효과를 기존의 치면열구전색제와 비교하고자 하였다. S-PRG 필러를 함유하는 치면열구전색제인 BeautiSealant, 불소를 방출하는 치면열구전색제인 Clinpro$^{TM}$ sealant, 불소 미방출 치면열구전색제인 Concise$^{TM}$ sealant를 실험군으로 선정하였다. 성장억제평가를 위해 치면열구전색제를 사용하지 않은 군을 음성 대조군으로 설정하였으며, 3개의 실험군 모두 대조군보다 유의할 정도로 낮은 집락 형성 단위를 보였고, Clinpro$^{TM}$ sealant가 BeautiSealant와 Concise$^{TM}$ sealant보다 유의할 정도로 낮은 집락 형성 단위를 보였다. BeautiSealant와 Concise$^{TM}$ sealant 군간에는 유의한 차이가 관찰되지 않았다. 바이오 필름 평가에서도 Clinpro$^{TM}$ sealant군이 BeautiSealant와 Concise$^{TM}$ sealant군들에 비해 유의할 정도로 낮은 집락 형성을 보였으며, BeautiSealant와 Concise$^{TM}$ sealant 군간에 유의한 차이는 관찰되지 않았다. 본 연구 결과 S-PRG filler를 포함하는 치면열구전색제인 BeautiSealant는 기존의 불소방출 치면열구전색제에 비하여 낮은 항미생물 효과와 높은 바이오 필름 형성능을 보였다.
Biofilms are aggregates of microorganisms present in a self-produced matrix of extracellular polymeric substance (EPS) adhered to a surface. Formation of a biofilm in the environment on farms and in dairy plants comprises several stages: attachment, growth (development), and detachment. Generally, biofilms are harmful to humans and need to be controlled. Stainless steel (SS) surfaces that are untreated or are scratched comprise substrata that are especially vulnerable to biofilm formation; therefore, SS surfaces should be polished and sanitized. Various approaches are available for the destruction of biofilms; cleaning-in-place (CIP) is the method mainly used in dairy plants. Further study on optimum detergents, cleaning conditions, and methods for this purpose is needed.
수서 생태계에서 생물막 성장을 고찰하기 위해 2차원 셀룰라오토마톤 (CA) 모델을 구현하였다. 영양물 농도와 연계하여 CA의 단순한 지역 규칙을 세균 개체군의 성장을 나타내는데 적용하였다. 초기 세균 분포는 낮은 영양물 농도에서 개체군 크기와 생물막 형태를 결정하는데 중요한 역할을 하였다. 집중분포에서는 균일분포나 마구잡이분포에 비해 개체군 크기가 느리게 증가하였다. 반면 공극률은 다른 초기 분포에 비해 균일분포에서 높게 나타나는 경향을 보였다.
Most oral microorganisms exist as biofilms which initiate formation via the attachment of an early colonizer to host proteins on the tooth surface. Fusobacterium nucleatum act as a bridge between early and late colonizers. Dental biofilms eventually comprise dental pathogens such as Porphyromonas gingivalis, Treponema denticola and Tannerella forsythia. To evaluate the effects of mutual interactions between oral bacteria on the growth of biofilms, periodontopathogens were co-cultured with a $0.4\;{\mu}m$ barrier. Streptococcus gordonii inhibited the growth of F. nucleatum and periodontopathogens. However, F. nucleatum, P. gingivalis and T. denticola activated the growth of other bacteria. A co-culture system of early and late colonizers could be a useful tool to further understand bacterial interactions during the development of dental biofilm.
Lactic acid bacteria (LAB) growth in processed meat products produces slime. In this study, 10 different biofilm-forming LAB, including Leuconostoc mesenteroides, Lacticaseibacillus paracasei, Levilactobacillus brevis, Lactiplantibacillus plantarum, Leuconostoc citreum, Weissella viridescens, and Latilactobacillus sakei, were isolated from various meat products and identified based on 16S rRNA gene analysis. To inhibit biofilm formation by LABs, Eisenia bicycles methanolic extract (EB) and ethyl acetate soluble fraction (EA) were used as antibacterial and antibiofilm agents, respectively. Furthermore, EA and EB were employed to synthesize gold nanoparticles (AuNPs) such as EB-AuNPs and EA-AuNPs, which could serve as antibiofilm agents against the isolated LAB. These findings demonstrate that EA, EB-AuNPs, and EA-AuNPs exhibit significant antibacterial activity against the isolated LAB. Furthermore, EB-AuNPs reduced L. citreum biofilm production, whereas EA-AuNPs inhibited L. mesenteroides and L. brevis biofilm formation. The current results suggest that EB-AuNPs and EA-AuNPs can be used as nanomaterials to inhibit LAB that form biofilms on meat products.
본 연구에서는 홍삼의 항충치 효과를 조사하기 위하여 홍삼박 n-hexane 추출물의 충치 원인균 Streptococcus mutans에 대한 항균 효과, 균체 부착과 생물막 생성 저해 효과를 측정하였다. 홍삼박 n-hexane 추출물의 농도에 비례하여 S. mutans의 생육이 저해되었으며, 125 ㎍/mL 이상의 농도에서는 거의 관찰되지 않았다(MIC=125 ㎍/mL). 홍삼박 n-hexane 추출물이 세포막에 작용하여 세포의 핵산 성분이 유출시키는 것으로 나타났다. 그리고, 홍삼박 n-hexane 추출물은 125 ㎍/mL 이상의 농도에서 S. mutans의 부착과 생물막 형성을 90% 이상 저해하였다. GTase 활성은 50 ㎍/mL 농도에서 완전히 저해되었다. 결론적으로 홍삼박 n-hexane 추출물은 S. mutans의 생육과 생물막 형성을 공통적으로 저해하는 것으로 나타났다.
Candidiasis involving the biofilms of Candida albicans is a threat to immunocompromised patients. Candida biofilms are intrinsically resistant to the antifungal drugs and hence novel treatment strategies are desired. The study intended to evaluate the anti-Candida activity of allyl isothiocyanate (AITC) alone and with fluconazole (FLC), particularly against the biofilms. Results revealed the concentration-dependent activity of AITC against the planktonic growth and virulence factors of C. albicans. Significant (p <0.05) inhibition of the biofilms was evident at ${\leq}1mg/ml$ concentrations of AITC. Notably, a combination of 0.004 mg/ml of FLC and 0.125 mg/ml of AITC prevented the biofilm formation. Similarly, the preformed biofilms were significantly (p <0.05) inhibited by the AITC-FLC combination. The fractional inhibitory concentration indices ranging from 0.132 to 0.312 indicated the synergistic activity of AITC and FLC against the biofilm formation and the preformed biofilms. No hemolytic activity at the biofilm inhibitory concentrations of AITC and the AITC-FLC combination suggested the absence of cytotoxic effects. The recognizable synergy between AITC and FLC offers a potential therapeutic strategy against biofilm-associated Candida infections.
This study investigated possibility of the nitrification and denitrification in one counter-current column with the growth of biofilm attached to its media. This experiment was performed through use of the lab scale reactor composed of the column and settler. The column used was packed with the small size of plastic rings called PALL($1.5{\times}1.5{\;}cm$) with a cylindrical shape. Synthetic wastewater was used in the experiment. The loading rates of carbon (C) and total nitrogen (TN) furnished to the reactor were 0.23 to 1.0 kg COD/m3.d and 0.023 to 1.0 kg N/m3.d, respectively. Major factors controlling the removal efficiencies of COD and TN were the different air flux and volumetric loading rates of COD and TN. The experimental results obtained from this study demonstrated that the removal efficiencies of COD ranged from 90 to 95% and those of TN were from 80 to 83% under the N loading rate of 0.035 and $0.058{\;}kg{\;}N/m^3{\cdot}d$, respectively. The patterns of TN removed were distinctively different on the limit of 50cm of column in depth. This indicated that the nitrification and denitrification occurred near the surface zone of and inside the biofilm respectively, upto the 50cm of the column in depth.
