An Effective Method of Diagnosis of Potato Leafroll Virus by RT-PCR (RT-PCR 방법을 이용한 효과적인 감자 잎말림 바이러스의 검정)

  • 감자 잎말림 바이러스를 검정하기 위하여 ELISA 및 전자현미경에 의해 바이러스 감염이 확인된 기내 배양중인 감장의 줄기로부터 RT-PCR 분석을 수행하였다. 분리된 총 RNA들로부터 바이러스 cDNA를 합성하고 감자 잎말림 바이러스 외피단백질의 일부인 465bp를 특이하게 증폭하도록 고안한 두 primer를 사용하여 PCR 반응을 하였다. 증폭된 465pb의 DNA 절편의 염기서열을 분석한 결과 역시 감자 잎말림 바이러스임을 확인하였다. 바이러스 검정에 있어서 EL-ISA 방법과 RT-PCR 방법간의 민감도를 조사한 결과 RT-PCR 방법간의 민감도를 조사한 결과 RT-PCR 방법이 ELISA 방법보다 감자 잎말림 바이러스검정에 있어서보다 정확한 방법인 것으로 사료된다.

Effects of Oral Rutaecarpine on the Pharmacokinetics of Intravenous Chlorzoxazone in Rats

  • It has been reported that hepatic microsomal cytochrome P450(CYP) 2E1 is responsible for the metabolism of chlorzoxazone(CZX) to 6-hydroxychlorzoxazone. The present study was undertaken to assess the possible interaction of rutaecarpine, an alkaloid originally isolated from the unripe fruit of Evodia rutaecarpa, with CZX. Male Spraque-Dawley rats were administered with 80 mg/kg/day of oral rutaecarpine for three consecutive days. Twenty four hr after the pre-treatment with rutaecarpine, the rats were treated with 20 mg/kg of intravenous CZX. Rat hepatic microsomes isolated from rutaecarpine-treated rats showed greater(50% increase) activity of p-nitrophenol hydroxylase(a marker of CYP2E1) when compared with the control rats. Compared with control rats, the AUC of CZX was significantly smaller(84% decrease) possibly due to significantly faster CL(646% increase) in rats pretreated with rutaecarpine. This could be, at least partially, due to induction of CYP2E1 by rutaecarpine.

  • Opuntia ficus-indica var. saboten (Cactaceae) is widely cultivated in Jeju Island (South Korea) for use in the manufacture of health foods such as tea, jam and juice. Its fruit and stem have been traditionally used as oriental folk medicine to treat diabetes, hypertension, asthma, burns, edema and indigestion. During the search for naturally occurring antioxidative neuroprotective agents, it was found that the ethyl acetate fraction of the stems and fruits of Opuntia ficus-indica var. saboten exhibited potent antioxidant effect. Using a chromatographic fractionation method, several constituents were isolated from this fraction. Among isolates, KYS50182 showed the most potent in vitro and in vivo neuroprotective activities, suggesting that the flavonoid can serve as a lead structure for the development of neuroprotective agents by providing neuroprotection against oxidative and focal ischemic neuronal injuries.

Ginsan Improved Th1 Immune Response Inhibited by Gamma Radiation

  • Gamma radiation causes suppression of the immune function, and immune properties are related to cytokine production. In the present study, the polysaccharide, Ginsan, purified from an ethanol-insoluble fraction of Ginseng (Panax ginseng C.A. Meyer, Araliaceae) water extract was studied to assess its effects on the immunosuppressive activities of gamma radiation. Gin­san was found to stimulate murine normal splenocytes by inducing the mRNA expressions of Th1 and Th2 type cytokines, and also restore the mRNA expression of IFN-$/gamma$, Th1 cytokine, after its inhibition by whole-body gamma irradiation. Therefore, Ginsan was found to restore the T lymphocytes function that had been suppressed by gamma irradiation in allogenic MLR (mixed lymphocyte reactions). However, Ginsan exhibited no excessive stimulatory effects on the control group. The above results indicated that Ginsan may constitute a new noble agent for the improvement of gamma radiation-induced immunosuppression.

Rapid Determination of Chlorostyrenes in Fish by Freezing-Lipid Filtration, Solid-Phase Extraction and Gas Chromatography-Mass Spectrometry

  • An analytical method has been developed for measuring chlorostyrenes in fish tissue sample. Extraction of chlorostyrenes from fish tissue was carried out by ultrasonication using acetone/n-hexane (5:2, v/v) mixture. Most of the lipids in the extract were eliminated by freezing-lipid filtration, prior to solid-phase extraction (SPE) cleanup. During freezing-lipid filtration, about 90% of the lipids extracted from the fish samples were easily removed without any significant losses of chlorostyrenes. For purification, SPE using Florisil was used for the rapid and effective cleanup. Quantification was performed using gas chromatography-mass spectrometry in the selected ion monitoring mode. Spiking experiments were carried out to determine the recovery, precision, and limits of detection (LODs) of the method. The overall recovery was above 80% in the spiked fish tissue sample at 10 and 100 ng/g levels, respectively. The detection limits for chlorostyrenes were ranged from 0.05 to 0.1 ng/g. This developed method is demonstrated to give efficient recoveries and LODs for detecting chlorostyrenes spiked into fish tissue with high lipid content.

  • Protein modifications of recombinant pharmaceuticals have been observed both in vitro and in vivo. These modifications may result in lower efficacy, as well as bioavailability changes and antigenicity among the protein pharmaceuticals. Therefore, the contents of modification should be monitored for the quality and efficacy of protein pharmaceuticals. The interface of EPO and its receptor was visualized, and potential amino acids interacting on the interface were also listed. Two different types of modifications on the interface were identified in the preparation of rHu-EPO BRP. A UPLC/Q-TOF MS method was used to evaluate the modification at those variants. The modification of the oxidized variant was localized on the Met54 and the deamidated variants were localized on the Asn47 and Asn147. The extent of oxidation at Met54 was 3.0% and those of deamidation at Asn47 and Asn147 were 2.9% and 4.8%, respectively.

  • The in vitro metabolism of a new herbicide pyribenzoxim, {benzophenone O-[2,6-bis[(4,6-dimethoxy-2-pyrimidinyl)oxy]benzoyl]oxime} was studied using rice, barnyardgrass and rat liver microsomes. No metabolism of pyribenzoxim was observed with rice and barnyardgrass microsomes though the cvtochrome P450 was active, which was evidenced by the metabolism of cinnamic acid. With rat liver microsomes, four metabolites (M1, M2, M3, and M4) were produced while parent compound decreased. M1 and M2 were from the hydrolysis reactions and NADPH-dependent metabolites were M3 and M4 (major metabolite) which were hydroxylated by cytochrome P450. They were identified as bispyribac-sodium (M1), benzophenone oxime (M2), {benzophenone O-[2,6-bis[(5-hydroxy-4,6-dimethoxy-2-pyrimidinyl)oxy]-benzoyl]oxime}(M3), and {benzophenone O-[2[(5-hydroxy-4,6-dimethoxy-2l-pyrimidinyl)6-(4,.6dimethoxy-2-pyrimidinyl)oxy]benzoyl]oxime} (M4) through LC/MS/MS analyses. Based on the results obtained metabolic map of pyribenzoxim is proposed.

Flavonol Glycosides with Antioxidant Activity from the Aerial Parts of Epimedium koreanum Nakai

  • The aerial parts of Epimedium koreanum Nakai have been used to stimulate hormone secretion in treating impotence. Ten flavonol glycosides, 3,4,5-trihydroxy-8-prenylflavone 7-O-[${\beta}$-D-glucopyranosyl($1{\rightarrow}2$)-${\beta}$-D-glucopyranoside] (1), hyperoside (2), icarisid II (3), 2"-O-Rhamnosylicarisid II (4), epimedin A (5), epimedin B (6), epimedin C (7), icariin (8), hexandraside E (9), and epimedoside A (10) were isolated from the an ethylacetate soluble extracts of the aerial parts of Epimedium koreanum Nakai through activity-monitord fractionation and isolation method. The structures of compounds 1 - 10 were elucidated by high resolution fast atom bombardment mass spectrometry and two dimentional nuclear magnetic resonance spectroscopy analysis. Compounds 1 and 4 showed potent antioxidant activity, with $IC_{50}$ values of 19.7 and 11.5 ${\mu}g$/mL and 88.2 and 90.5 ${\mu}M$, respectively.

A Sensitive and Rapid LC-MS/MS Method for Determination of Berberine in Human Plasma

  • Coptidis Rhizoma (CR) has been used widely in traditional medicine to treat common diseases. This study aimed to develop a high-sensitivity liquid chromatography-tandem mass (LC-MS) spectrometry method for the evaluation of the pharmacokinetics of a new natural product that contain CR extract with the main bioactive compound, berberine, at trace concentrations. Human plasma samples were pretreated with methanol by a protein precipitation method. Berberine was analyzed on a Kinetex C18 column ($2.1mm{\times}50mm$, $100{\AA}$, $1.7{\mu}m$) using a mobile phase of 10 mM ammonium formate/0.1% formic acid in water (A) and acetonitrile (B) (50:50, v/v) with a flow rate of 0.25 mL/min. The analyte was detected by using electrospray ionization in positive mode with multiple reaction monitoring (MRM). The method was sensitive, with a lower limit of quantification of 1 pg/mL, which has not been previously obtained. The method was validated (over the range of 1-50 pg/mL) and applied successfully for the pharmacokinetic study of human plasma samples.

Proposal for AI Video Interview Using Image Data Analysis

  • In this paper, the necessity of AI video interview arises when conducting an interview for acquisition of excellent talent in a non-face-to-face situation due to similar situations such as Covid-19. As a matter to be supplemented in general AI interviews, it is difficult to evaluate the reliability and qualitative factors. In addition, the AI interview is conducted not in a two-way Q&A, rather in a one-sided Q&A process. This paper intends to fuse the advantages of existing AI interviews and video interviews. When conducting an interview using AI image analysis technology, it supplements subjective information that evaluates interview management and provides quantitative analysis data and HR expert data. In this paper, image-based multi-modal AI image analysis technology, bioanalysis-based HR analysis technology, and web RTC-based P2P image communication technology are applied. The goal of applying this technology is to propose a method in which biological analysis results (gaze, posture, voice, gesture, landmark) and HR information (opinions or features based on user propensity) can be processed on a single screen to select the right person for the hire.
