Effect of the supplement of the earthworm cast, earthworm and B. bassiana on the improvement of both the productivity of hens and nutritional constituents of eggs (지렁이, 분변토 및 백강균 첨가에 의한 산란계의 생산성 및 계란의 영양적 조성 향상)

  • Effects of the supplement of earthworm cast produced from the feeding of organic wastes and earthworm on the productivity and nutritional constituents of functional eggs were investigated. Compared with control experiments, the case supplemented with earthworm cast showed high ratios in egg production, selection and the reserved feed. According to the experiment with earthworm, both the number of jumbo eggs and the quantity of reserved feed were increased. Therefore, the nutritional effect of earthworm in the feed was positive. The optimum percentage of earthworm cast in the feed was 10%: the average laying increased to 96.8, which was a 5% increase; the ratio of the large eggs increased by 5% although the ratio of jumbo eggs and of extra large eggs decreased by 5% and 1.1%, respectively; the average reserved feed was 662.5g. Also, Beauveria bassiana was inoculated into the feed as valuable microorganisms to prevent the growth of pathogen and to obtain essential amino acid. With the inoculatio of B. bassiana KACC 40039, the average laying was 0.82/hen and with B. bassiana HYB, it was 0.77/hen. Those numbers were three to eight percentage over the control. As for the effect of inoculation of B. bassiana in the feed on the production of broken eggs, B. bassiana KACC 40039 produced no broken eggs. Analysis of nutritional contents of eggs showed the increase in protein content and decrease in lipid content when compared with the control. According to these results, increase in the income of farmers can be expected.

Field evaluation of conidia of Beauveria bassiana (Balsamo) Vuillemin strain CS-1 against diamondback moth larvae (포장에서 배추좀나방에 대한 백강균 CS-1의 방제효과)

  • The efficacy of conidia of Beauveria bassiana (Balsamo) Vuillemin strain CS-1 that showed high mortality in laboratory and net house was examined against diamondback moth larvae in the field. Conidia ($4{\times}10^{7}$ conidia/ml) were applied to larvae on chinese cabbage in a 1.5% emulsifiable oil-emulsion amended with 4% clay at a volume of 0.55 L/3.3 $m^{2}$. There were no significant differences among plants and replicates in the deposition of spray droplets on water-sensitive papers or of conidia on leaves and larvae. Weather conditions were rainy and cool during first few days, and then hot and dry. Persistence of conidia was equally short on both leaves and larvae. Nevertheless, treatment of B. bassiana potentially reduced larval populations. But the field efficacy was lower than the laboratory efficacy. This reduced efficacy was ascribed to unfavorable environmental conditions.

Selection of Optimal Culture Medium for Four Entomopathogenic Fungal Isolates with Dual Activity and Evaluation of Their Antimicrobial Activity against Several Phytopathogens (이중 활성 곤충병원성 곰팡이 4균주에 대한 최적 배양 배지 선발 및 다양한 항균활성 평가)

  • Selection of the optimal culture medium and evaluation of the antimicrobial activity against various phytopathogens were performed for four entomopathogenic fungal isolates with excellent insecticidal and antimicrobial activity against the two-spotted spider mite (Tetranychus urticae), green peach aphid (Myzus persicae), and gray mold (Botrytis cinerea). The optimal medium was selected by measuring the amount of blastospore production and the antifungal activity of the culture medium. On the basis of these experiments, GY medium was selected for Beauveria bassiana 2R-3-3-1 and Metarhizium anisopliae 4-2, SD3, and PDB medium for B. bassiana SD15. The antimicrobial activity test against other phytopathogens indicated that all four isolates showed high antifungal activities against Colletotrichum acutatum and Sclerotinia sclerotiorum. However, for Phytophthora capsici and C. fructicola, only M. anisopliae SD3 showed a high antifungal activity against P. capsici, and the other three isolates had little activity. Antibacterial activity against Clavibacter michiganensis subsp. michiganensis was high in two isolates of M. anisopliae but not in two isolates of B. bassiana. Thus, it was confirmed that entomopathogenic fungi effective for pest control could be effectively used as a control agent for various plant diseases.

  • In this study, nucleotide sequence structures of intergenic transcribed spacer (ITS) 1, complete 5.8S ribosomal RNA gene and ITS 2 region were analyzed to identify three Peruvian entomopathogenic fungal isolates. The isolates had highly conserved sequence region in 5.8S rRNA gene and unique sequences in ITS 1 and 2 region among them. 5.8S rRNA gene regions were highly conserved and showed high homoloies among tested isolates. In contrast, ITS region showed species-specific sequence region, resulting in inter-genus differencies. Scanning electron microscopic images of these isolates supported the result of ITS-based identification. From these result, Peruvian entomopathogenic fungal isolate J270, J278, were identified as Beauveria bassiana and J271 was identified as Lecanicillium attenuatum.

Microbial Control of Fly Maggots with Entomopathogenic Nematodes and Fungus in Outhouses of Farmhouses (곤충병원선충과 곰팡이를 이용한 농가화장실 파리의 미생물적 방제)

  • Infectivity of entomopathogenic nematodes and fungus was evaluated against fly larvae in the laboratory and outhouses. Mortalities of Muscina stabulans larvae were 96.7f 2.8% in Steinernema glaseri Dongrae strain, 90.0+0.0% in S. carpocapsae All strain, 86.7f 2.7% in Heterorhabditis bacteriophora Hamyang strain, and 70.0+9.4% in S. carpocapsae Pocheon strain on the filter paper. When 260, 000 nem\ulcornertodes were sprayed into the outhouses, H. bacterwphora Hamyang strain killed 100%, S. glaseri Dongrae strain killed 76.9+3.9%, and S. carpocapsae Pocheon strain killed 58.5+6.1% of maggots. When 130, 000 nematodes and 7.0X lo9 cfu of entomopathogenic fungus, Beauveria brongniartii were sprayed alone or combined into outhouses, mortalities of maggots were 73.6+0.1% in B. brongniartii alone, 77.8+3.9% in S. carpocapsae Pocheon strain plus B. brongniartii, and 77.7f 5.1% in H. bacteriophora Hamyang strain plus B. brongniartii. Entomopathogenic nematodes and fungus were potential biological control agents in this study.

Effect of Synnemata of Beauveria bassiana on the Properties of Noodle (백강균 자실체의 첨가가 제면적성에 미치는 영향)

  • This study was conducted to investigate the properties of noodle which was made of composite flour blended with the powder of synnemata of Beauveria bassiana. The characteristics of cooked-noodle including color, cooking properties, mechanical texture properties were measured, and sensory evaluation was performed. The L-value of dry and wet-noodle which made of composite flour was similar to that of 100% wheat flour, but a and b-value were higher than those of 100% wheat flour. There were no differences in the weight, volume and water absorption of the cooked-noodle made of composite flour and that of 100% wheat flour, but the turbidity of the cooked-noodle made of composite flour was higher than that of 100% wheat flour. The breaking force of dry-noodle which was made of composite flour was same as that of 100% wheat flour. Hardness and Gumminess of the cooked-noodle which was made of composite flour blended with synnemata powder at level of 1∼5% were same as those of 100% wheat flour. Springiness, chewiness, cohesiveness and adhesiveness of the cooked-noodle which was made of composite flour blended with synnemata powder at level of 1% were similar to those 100% wheat flour. The results of sensory evaluation showed that the cooked-noodles containing 1% and 3% synnemata powder were acceptable as much as those of 100% wheat flour in terms of color, taste, flavor, texture and overall acceptance.

Characteristics of an Entomopathogenic Fungus Infecting Corythucha ciliata (Hemiptera: Tingidae) (버즘나무방패벌레 기생성 곰팡이의 특성 구명)

  • Overwintering adults of sycamore lace bug (Corythucha ciliata) infected by an unidentified pathogenic fungus were found on the stems of street trees of sycamore in Cheongju city. The objective of this study was to describe this entomopathogenic fungus infecting overwintering sycamore lace bug adults. This unidentified fungus colonized the insect adult body and formed white colony with subglobose clusters of conidiocarps. The size of conidiocarps was 300 to $400{\mu}m$ and each conidium was 15 to $20{\mu}m$. The conidiospore was globus and 2.5 to $3.0{\mu}m$ in diameter, and the hyphae were 1 to $5{\mu}m$ thick. This fungus was successfully isolated and cultivated on potato dextrose agar medium (PDA). The fungal colony was white and then became light yellow. When conidia from this pure culture were inoculated into the overwintering adults, the fungus formed conidiocarps with the same morphology on the insect body and the lethal rate by the fungus was $88{\pm}16%$. This fungus has over 99% homology with Cordyceps bassiana (imperfect fungal name is Beauveria bassiana) in ITS-5.8s rDNA base sequence. The fungal ecology and the infection process of the fungus into its host need to be clarified before using this fungus as a biological control agent.

Effect of Synnemata of Beauveria bassiana on the Baking Qualities of Bread (백강균 자실체의 첨가가 제빵적성에 미치는 영향)

  • Addition of synnemata of Beauveria bassiana on the baking qualities of bread was studied. There were no differences in the loaf volume of bread made of composite flour blended with the synnemata powder at level of 1%, 3% from that of control, but that of 5% was lower than the control. The addition of synnemata powder caused a decrease in the lightness of bread crumb and an increase in the redness and yellowness. The hardness of bread made of composite flour blended with synnemata powder at level of 1% and 3% was similar to that of control, while 5% addition was higher than the control. There were no differences in the springiness, gumminess and chewiness of bread made with synnemata powder at level of 1%, 3% from those of control. In sensory evaluation, the addition of the synnemata powder at level of 1% and 3% did not affect appearance, flavor, taste, texture and overall acceptability, while 5% addition significantly decreased those values.

Turfgrass Insect Pests and Natural Enemies in Golf Courses (골프장 잔디 해충과 천적의 종류)

  • - Turfgrass insect pests and natura.l enemies for biological control were investigated to develop pest management effectively in golf courses at several golf clubs. Twenty eight insect pest species of 10 families in 6 orders were collected from golf courses. The zoysiagrass mite, Eriophyes zoysiae and root-knot nematode, Meloidogyne incognita were also collected from zoysiagrass. White grubs of several scarab beetles and cutworms (Agrotis spp.) damaged seriously at most surveyed golf clubs. In addition, bluegrass webworm (Crambus sp.), Japanese lawngrass cutworm (Spodoptera depravata), scale insects, Tipula sp., and ants (Camponitus japonicus, Formica japonica, and Lasins japonicus) damaged turfgrasses directly or indirectly in golf courses. The entomopathogenic nematodes, Heterorhabditis spp., Steinernema glaseri, and S. longicaudum, entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae, and milky disease, Paenibacil/us popil/iae were isolated from white grubs or turfgrass soil as microbial control agents. Besides, dipteran predators, Cophinopoda chinensis, Philonicus albiceps, and Promachus yesonicus and hymenopteran parasitoid, Tiphia sp. were also collected. The P. yesonicus was the most active in golf courses. The root-knot nematode, M. incognita was found from Zoysia japonica, Z. matrella. and Cynodon dactylon.

Effects of an Antimicrobial Substance from Bombycis corpus on Antibiotic Resistant Microbes (백강잠으로부터 분리한 항균물질의 항생제 내성균에 대한 효과)

  • Bombycis corpus, a batryticated silkworm and white-stiff silkworm, is an oriental drug consisting of the dried larva of silkworm, dead and stiffened due to the infection of Beauveria. An peptidyl antimicrobial molecule was purified from B. corpus by reverse phase-column chromatography and HPLC. Its molecular weight was determined to be 2295.45 by using matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometry. Its antimicrobial activity was diminished by trypsin digestion. It exhibited a broad antimicrobial spectrum against not only Gram-negative, but also Gram- positive bacteria. Furthermore, it was found to have an antimicrobial activity against vancomycin-resistant enterococci (VRE), methicillin-resistant S. arureus (MRSA), and vancomycin-intermediate resistant S. arureus (VISA). It may be a useful molecule for a new antibiotic development, especially against antibiotic resistant microbe. This substance may play a role in the defense system of this animal against Beauveria bassiana. This is the first report of a peptidyl antimicrobial substance from B. corpus.
