화학적 허혈에 의해 손상된 마우스 간세포에 대한 hydrogen sulfide의 간세포 보호 효과 (Protection of Primary Cultured Mouse Hepatocytes from Chemical Hypoxia-induced Injury by Hydrogen Sulfide)

  • 본 연구는 화학적 허혈에 의해 손상된 마우스 간세포에서 hydrogen sulfide ($H_2S$)의 효과를 규명하기 위해 수행되었다. 본 연구에서 허혈 모방 화합물로 알려져 있는 cobalt chloride ($CoCl_2$)는 간세포 손상을 시간 및 농도 의존적으로 유의성 있게 증가 시켰다. $CoCl_2$에 의한 간세포 손상은 Sodium sulfide (NaHS, $H_2S$ 공여제)의 전처리에 의해 유의적으로 감소 되었다. $CoCl_2$는 세포 내 활성산소(reactive oxygen species, ROS)의 농도를 증가시켰으며, 이는 NaHS 및 N-acetyl-cysteine (NAC, a ROS 제거제)에 의해 감소하였다. 또한, $CoCl_2$에 의해 증가된 p38 MAPK 인산화가 NaHS 및 NAC에 의해 억제되었다. $CoCl_2$에 의해 증가된 Bax/Bcl-2 비율은 NaHS, NAC 및 SB 203580 (p38 MAPK 저해제)에 의해 차단되었으며, $CoCl_2$에 의해 유발된 간세포의 손상 또한 NaHS, NAC 및 SB 203580의 전처리에 의해 억제되었다. NaHS는 $CoCl_2$에 의해 증가된 COX-2의 발현을 억제하였다. 또한, NaHS의 효과와 유사하게 $CoCl_2$에 의해 증가된 COX-2의 발현이 NAC에 의해 억제되었다. 더욱이, NS-398 (COX-2 선택적 억제제)는 $CoCl_2$에 의한 Bax/Bcl-2 비율의 증가를 억제하였을 뿐 아니라, 간세포의 세포 손상 또한 억제하였다. 결론적으로, $H_2S$는 초대배양 된 마우스 간세포에서 $CoCl_2$에 의해 유발된 간세포의 손상을 ROS에 의해 유발된 p38 MAPK 및 COX-2 경로의 활성화를 억제함으로써 세포보호효과를 수행하는 것을 알 수 있었다.

Apoptotic Effects of Cordycepin Through the Extrinsic Pathway and p38 MAPK Activation in Human Glioblastoma U87MG Cells

  • We first demonstrated that cordycepin inhibited cell growth and triggered apoptosis in U87MG cells with wild-type p53, but not in T98G cells with mutant-type p53. Western blot data revealed that the levels of procaspase-8, -3, and Bcl-2 were downregulated in cordycepin-treated U87MG cells, whereas the levels of Fas, FasL, Bak, cleaved caspase-3, -8, and cleaved PARP were upregulated, indicating that cordycepin induces apoptosis by activating the death receptor-mediated pathway in U87MG cells. Cordycepin-induced apoptosis could be suppressed by only SB203580, a p38 MAPK-specific inhibitor. These results suggest that cordycepin triggered apoptosis in U87MG cells through p38 MAPK activation and inhibition of the Akt survival pathway.

Cisplatin의 난소암 세포 증식 억제에 관한 신호 전달 기전 (Cisplatin Suppresses Proliferation of Ovarian Cancer Cells through Inhibition Akt and Modulation MAPK Pathways)

  • Cisplatin (CDDP)은 난소암 치료에 사용되는 화학 요법제로 암세포에 따라 그리고 치료 용량에 따라 다중 신호경로를 활성화하여 세포 반응을 다르게 일으킬 수 있다. Cisplatin이 세포에 작용하는 신호전달 기전은 분명하지 않아 더 많은 연구가 필요해 보인다. 이에 본 연구는 cisplatin을 난소암 세포(SKOV3)에 처리하여 세포사멸 유도 과정에서 나타나는 신호 단백질의 역할을 밝히고자 하였다. 결과는 cisplatin으로 처리한 난소암 세포에서 TUNEL assay와 유세포 분석을 통해 대조군과 비교하여 세포 사멸수가 증가하였다. 세포 사멸 단계에서 나타나는 PARP 및 caspase-3 활성도 증가하였다. 그러나 Bcl-2의 발현은 감소하였다. 신호 전달 경로에서 나타나는 단백질의 발현은 ERK1/2의 활성은 시간 의존적으로 감소하였으나 Akt 활성은 24시간에 감소하다 48시간에서의 활성은 일정하였다. p38과 p-JUN의 활성은 24시간에 증가하는 것으로 나타났으나 48시간에서 p38의 활성은 감소하였으며 p-JUN의 활성은 일정하였다. 이상의 결과들을 토대로 결론은 cisplatin이 SKOV3 세포에서 Akt 활성을 감소하여 세포 증식을 억제하고 MAPK의 p38 발현을 조절하여 세포사멸을 유도하는 것으로 판단된다. 향후, 암치료 전략에 도움이 되는 cisplatin을 포함한 백금기반 화학요법제의 신호전달 기전을 밝히기 위한 더 많은 연구가 필요할 것으로 생각된다. 본 실험을 통해 제시한 결과는 MAPK 신호 경로를 타겟으로 하는 암 치료 전략에 유용하게 사용 될 수 있기를 기대한다.

마치현(馬齒莧)이 자궁경부암세포(子宮頸部癌細胞)(HeLa Cell)에 미치는 영향(影響) (Herba Portulacae induced Apoptosis in Human CervicalCarcinoma HeLa Cells)

  • To address the ability of Herba Portulacae(HP) to induce cell death, we investigated the effect of HP on cell viability. Twenty-four hours later, loss of viability occurred following HP exposure in a dose-dependent manner. The treatment of HP, a commonly used herb formulation in Korea, Japan and China, caused a decrease in cell viability. HP also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that 2mg/ml HP induces mitochondria membrane potential collapse. Immunoblotting data also shows that the expression of Bcl-2, antiaoptotic protein, decrease by the addition of HP. This GFP-Bax overexpression system shows that an important pro-apoptotic Bcl-2-family protein, Bax is translocated to mitochondria by the addition of 2mg/ml HP. Inerestingly, MAPK inhibitor study shows that p38 MAPK inhibitor, SB203580 inhibits HP-induced cell death and caspase-3 activation in HP-treated HeLa cells. Furthermore, HP transiently but significantly induces p38 activation. But P38 MAPK inhibitor does not have any effect on the translocation of Bax. Considering these results, HP induces apoptosis via p38 MAPK activation. But the pathway does not involve the translocation of Bax.

Neuroprotective effects of urolithin A on H2O2-induced oxidative stress-mediated apoptosis in SK-N-MC cells

  • BACKGROUND/OBJECTIVES: Oxidative stress causes cell damage and death, which contribute to the pathogenesis of neurodegenerative diseases. Urolithin A (UA), a gut microbial-derived metabolite of ellagitannins and ellagic acid, has high bioavailability and various health benefits such as antioxidant and anti-inflammatory effects. However, it is unknown whether it has protective effects against oxidative stress-induced cell death. We investigated whether UA ameliorates H2O2-induced neuronal cell death. MATERIALS/METHODS: We induced oxidative damage with 300 μM H2O2 after UA pretreatment at concentrations of 1.25, 2.5, and 5 μM in SK-N-MC cells. Cytotoxicity and cell viability were determined using the CCK-8 assay. The formation of reactive oxygen species (ROS) was measured using a 2,7-dichlorofluorescein diacetate assay. Hoechst 33342 staining was used to characterize morphological changes in apoptotic cells. The expressions of apoptosis proteins were measured using Western blotting. RESULTS: UA significantly increased cell viability and decreased intracellular ROS production in a dose-dependent manner in SK-N-MC cells. It also decreased the Bax/Bcl-2 ratio and the expressions of cytochrome c, cleaved caspase-9, cleaved caspase-3, and cleaved PARP. In addition, it suppressed the phosphorylation of the p38 mitogen-activated protein kinase (MAPK) pathway. CONCLUSIONS: UA attenuates oxidative stress-induced apoptosis via inhibiting the mitochondrial-related apoptosis pathway and modulating the p38 MAPK pathway, suggesting that it may be an effective neuroprotective agent.

TIMP-1 in the regulation of ECM and apoptosis

  • The importance of apoptosis in normal development and pathogenesis has been well recognized, and explosive progress towards dissecting its commitment step has been made during the past decade. Mitochondria, Apaf-1, caspase, and bcl-2 family members play central roles in the commitment step. However, it is still unclear how upstream cell survival pathways regulate apoptosis. It is also unknown whether the bcl-2 family members have any effect on the upstream survival pathways. We have demonstrated that the anti-apoptotic gene product bcl-2 greatly induces expression of the tissue inhibitor of metalloproteinase-1 (TIMP-1) in human breast epithelial cells. Surprisingly, we found that TIMP-1, like bcl-2, is a potent inhibitor of apoptosis induced by a variety of stimuli. Functional studies indicate that TIMP-1 inhibits a classical apoptotic pathway mediated by caspases, and that focal adhesion kinase (FAK)/Pl 3-kinase and mitogen activated protein kinase (MAPK) are critical for TIMP- 1 -mediated cell survival. We also showed specific association of TIMP-1 with the cell surface. Consistently, a 150-H)a surface protein was identified in MCF10A cells that specifically binds TIMP-1. Taken together, we hypothesize that TIMP-I binding on the cell surface induces a cell survival pathway that regulates the common apoptosis commitment step. The results of these studies will address a new paradigm in the regulation of apoptosis by an extracellular molecule TIMP-1, and also greatly enhance our understanding of TIMP-1's pleiotropic activity in many physiological and pathological processes. This information may also be useful in designing more rational therapeutic interventions aimed at modulating the anti-apoptotic activity of TIMP-1 .

Angelica polymorpha Maxim Induces Apoptosis of Human SH-SY5Y Neuroblastoma Cells by Regulating an Intrinsic Caspase Pathway

  • Angelica polymorpha Maxim root extract (APRE) is a popular herbal medicine used for treating stomachache, abdominal pain, stomach ulcers, and rheumatism; however the effect of APRE on cancer cells has not yet been explored. Here, we examined APRE cytotoxicity seen on target neuroblastoma cells (NB) using cell viability assays, DAPI visualization of fragmented DNA, and Western blotting analysis of candidate signaling pathways involved in proliferation and apoptosis. We demonstrated that APRE reduced cell viability in NB to a greater extent than in fibroblast cells. In addition, we found that APRE could inhibit the three classes of MAPK proteins and could also down-regulate the PI3K/AKT/GSK-$3{\beta}$ activity all being relevant for proliferation and survival. APRE could also up-regulate Bax expression and down-regulate Bcl-2 and Mcl-1. With APRE treatment, depolarization of mitochondria membrane potential and activation of caspase-3 was demonstrated in the SH-SY5Y cells. We could not found increased activity of death receptor and caspase-8 as markers of the extrinsic apoptosis pathway for the APRE treated cells. In presence of a caspase-3 siRNA and a pan-caspase inhibitor, APRE could not reduce the viability of NB cells to a significant degree. So we predicted that with APRE, the intrinsic pathway was solely responsible for inducing apoptosis as we also showed that the non-caspase autophagy pathway or ER stress-ROS mediated pathways were not involved. These findings demonstrate that an intrinsic mitochondria-mediated apoptosis pathway mediates the apoptotic effects of APRE on SH-SY5Y cells, and that APRE shows promise as a novel agent for neuroblastoma therapy.

Berberine Inhibited Radioresistant Effects and Enhanced Anti-Tumor Effects in the Irradiated-Human Prostate Cancer Cells

  • The purpose of this study was to elucidate the mechanism underlying enhanced radiosensitivity to $^{60}Co\;{\gamma}$-irradiation in human prostate PC-3 cells pretreated with berberine. The cytotoxic effect of the combination of berberine and irradiation was superior to that of berberine or irradiation alone. Cell death and Apoptosis increased significantly with the combination of berberine and irradiation. Additionally, ROS generation was elevated by berberine with or without irradiation. The antioxidant NAC inhibited berberine and radiation-induced cell death. Bax, caspase-3, p53, p38, and JNK activation increased, but activation of Bcl-2, ERK, and HO-1 decreased with berberine treatment with or without irradiation. Berberine inhibited the anti-apoptotic signal pathway involving the activation of the HO-1/NF-${\kappa}B$-mediated survival pathway, which prevents radiation-induced cell death. Our data demonstrate that berberine inhibited the radioresistant effects and enhanced the radiosensitivity effects in human prostate cancer cells via the MAPK/caspase-3 and ROS pathways.

만성적인 에탄올 섭취로 인한 쥐의 위 조직 손상에서 방사무늬 김(Pyropia yezoensis)의 보호효과 (Protective Effects of Pyropia yezoensis Glycoprotein against Ethanol-induced Chronic Gastric Injury in the Rat)

  • We examined the protective effects of Pyropia yezoensis glycoprotein (PYGP) against ethanol-induced gastric damage. The experimental animals were divided into four groups. They were treated with distilled water (control), ethanol alone (EtOH), ethanol + PYGP 150 mg/kg BW (EtOH+150), or ethanol + PYGP 300 mg/kg BW (EtOH+300). The groups were treated for 4 weeks. We measured mitogen-activated protein kinase (MAPK), the apoptotic signaling pathway, and PARP activity in gastric tissues obtained from the rats. Ethanol consumption increased apoptotic signal activity and ERK, JNK, and p38 phosphorylation. PYGP reduced the apoptotic signaling pathway activity and ERK, JNK, and p38 phosphorylation. Furthermore, PYGP regulated Bcl-2 family expression. In light of these findings, PYGP appears to prevent ethanol-induced gastric injury and oxidative stress.
