Repeated Batch Production of Epothilone B by Immobilized Sorangium cellulosum

  • Production of extracellular epothilone B, one of the potent anticancer agents, by free and immobilized Sorangium cellulosum was studied using the repeated batch culture process. The concentration of alginate used in immobilization was directly related to the mass transfer rate of nutrients, mechanical stability, and the epothilone B production yield. With the optimized 3% (w/v) calcium alginate carrier, a prolonged repeated batch culture was investigated for the 5 repeated batches for 24 days. The maximum productivity of epothilone B obtained from the alginate-immobilized cells was 5.03 mg/l/day, which is 3 times higher than that of free cells (1.68 mg/l/day).

Production of Lactococcal Bacteriocin using Repeated-Batch and Continuous Cultures

  • Repeated-batch and continuous cultures of Lactococcus sp. 1112-1 were carried out for bacteriocin production using a glucose-casein medium. Repeated-batch culture did not efficiently enhanced the bacteriocin production. Continuous production was possible at the dilution rate of 0.4 $h^{-1}$. Maximum specific production rate ($Q^p$), bacteriocin production and biomass at the dilution rate were 347, 136 IU/g/h, 2, 121 IU/ml and 2.45 g/L, respectively.

한천분해효소를 생산하는 Agarivorans sp. JA-1의 배양조건 및 Fed-batch 배양 (Production of ${\beta}$-agarase in Batch and Fed-batch Culture by Agarivorans sp. JA-1)

  • 제주도 연안에서 분리 동정된 한천분해효소를 생산하는 Agarivorans sp. JA-1의 배양특성을 연구하였다. 균주가 기질로 이용하는 한천은 배지 내의 한천의 농도가 0.2%일 때 가장 효율적인 성장을 하였으며 4종의 탄소원에 대해서는 성장에 영향을 미치지 않는 것으로 나타났다. 배양시간에 따른 한천농도는 15시간 이후 모두 분해되었으며 점도의 변화도 이와 유사한 것으로 나타났다. 또한 fed-batch 배양에서 0.8 g 한천을 2회 첨가하였으며 한천을 첨가한 후 agarase생산이 증가되는 것으로 나타났다. 한천올리고당의 성분분석을 한 결과 중합도가 6인 당이 확인되었다.

  • 산업적 R3HB의 생산을 위한 재조합 대장균의 pilot규모에서의 유가식 배양과 연속식 배양을 연구하였다. Pilot 규모에서의 R3HB생산을 위하여 안전한 two plasmid system pBRRed와 pMCS 105를 제작하였으며, 제작된 plasmids을 이용하여 여러 다른 대장균을 형질 전환하였다. 얻어진 재조합 대장균들을 30 l의 발효기에서 회분식 배양한 결과 대장균 XL-10 Gold(pBRRed, pMCS105)가 가장 높은 R3HB 농도를 보였다 30 1 발효기에서 대장균 XL-10 Gold (pBRRed, PMCS105)을 유가식 배양한 결과 22.4 g/1의 R3HB가 얻어졌으며, 생산성은 0.97 g/1-h를 보였다. 고농도의 R3HB를 고생산성으로 얻기 위하여 유가식 배양으로 높은 균체 농도를 얻은 후 연속 배양으로 R3HB를 생산하는 전략을 개발하였다. 그 결과 0.2 $h^{-1}$ 의 dilution rate에서 R3HB 생산성은 5.06 g/1-h를 보였다. 이러한 결과는 산업적 규모에서 재조합 대장균을 이용하여 R3HB를 고농도, 고생산성으로 얻을 수 있다는 것을 보여준다.

에탄올 발효방법에 따른 음식물류 폐기물의 바이오에탄올 생산성 비교 (Comparison of Bio-ethanol Productivity Using Food Wastes by Various Culture Modes)

  • In order to improve bio-ethanol productivity by various cultivation methods in this paper, the culture modes using food wastes, such as batch culture, high-cell-density fermentation, SSF (simultaneous saccharification and fermentation) by fill & draw, continuous culture by fill & draw were performed and their productivities were compared. SSFs by fill & draw were performed by continuous decompression using 1 L evaporator system, and by 10 L bioreactor without decompression. In addition, the continuous cultures by fill & draw mode using SFW (saccharafied food wastes) medium were performed by changes of 40% culture broth with intervals of 12 h (0.03 $h^{-1}$), 6 h (0.07 $h^{-1}$), 3 h (0.13 $h^{-1}$). Consequently, productivities of bio-ethanol were 2.52 g/L-h and 1.30 g/L-h in batch culture and high- cell-density fermentation, respectively. The productivities of SSF by fill & draw showed 2.24 g/L-h and 2.03 g/L-h in continuous decompression with 1 L evaporator and 10 L bioreactor without decompression, respectively. Also, the productivities in continuous culture by fill & draw modes showed 2.02 g/L-h, 4.07 g/L-h and 6.25 g/L-h by medium change with intervals of 12 h, 6 h, and 3 h, respectively. In conclusion, the highest ethanol productivity was obtained in the continuous culture mode by fill & draw with dilution rate of 0.13 $h^{-1}$.

  • 유가식배양을 통한 고농도배양에서 계면활성제와 산소전달물질의 첨가는 세포의 고밀도와 배지의 고점도로 인해 발생하는 nutrient의 물질전달제한과 산소부족현상을 효과적으로 해결하였다. Pluronic 10R-5는 Pluronic F-68과 같은 triblock copolymer 구조이며 세포생장의 저해를 나타내지 않았다. 그러나 diblock copolymer구조이며 소수성 부분이 hydrocarbon block인 Plurafac A-38은 세포의 생장을 저해함을 관찰할 수 있었다. 따라서 장기간의 유가식배양이나 perfusion 배양을 수행할 경우 세포의 생존도를 증대시켜 유용물질의 생산성을 높일 수 있으며 여러 형태의 bioreactor에서 고농도 배양을 가능케 하여 유용물질 생산성을 높일수 있을 것이라고 기대된다.

Optimum Conditions for the Biological Production of Lactic Acid by a Newly Isolated Lactic Acid Bacterium, Lactobacillus sp. RKY2

  • Lactic acid is a green chemical that can be used as a raw material for biodegradable polymer. To produce lactic acid through microbial fermentation, we previously screened a novel lactic acid bacterium. In this work, we optimized lactic acid fermentation using a newly isolated and homofermentative lactic acid bacterium. The optimum medium components were found to be glucose, yeast extract, $(NH_4)_{2}HPO_4,\;and\;MnSO_4$. The optimum pH and temperature for a batch culture of Lactobacillus sp. RKY2 was found to be 6.0 and $36^{\circ}C$, respectively. Under the optimized culture conditions, the maximum lactic acid concentration (153.9 g/L) was obtained from 200 g/L of glucose and 15 g/L of yeast extract, and maximum lactic acid productivity ($6.21\;gL^{-1}h^{-1}$) was obtained from 100 g/L of glucose and 20 g/L of yeast extract. In all cases, the lactic acid yields were found to be above 0.91 g/g. This article provides the optimized conditions for a batch culture of Lactobacillus sp. RKY2, which resulted in highest productivity of lactic acid.

Optimization of cell growth and TAPS production by Pichia ciferrii mutant in batch culture

  • Batch culture of mutant derived from Pichia ciferrii ATCC 14091 was investigated for producing the intracellular tetraacetylphytosphingosine (TAPS). Composite experimental design was used to optimize the composition of the culture medium for maximizing the productivity of TAPS. In this experiment, various culture parameters were investigated that were the effects of temperature, the initial culture pH, the carbon-to-nitrogen ratio, the concentration of trace elements, and the concentration of cofactors. The optimal temperature for cell growth and TAPS synthesis appeared to be %25^{\circ}C$. An initial pH value of 7.5 gave the best results. Under the best condition, the maximum TAPS concentration indicated 7.2 g/L and its productivity was 0.06 g/L-hr in a 2.5 L jar

  • A process of organically bound germanium preparation was developed for healthy food using inorganic germanium adapted Saccharomyces cerevisiae. Adaptations of Saccharomyces cerevisiae against inorganic germanium were successively carried out through stepwise increase of GeO$_{2}$ concentration in order to produce high quantities of germanium bound yeast. Productivity of yeast and quantities of germanium in yeast were obtained 70.2 g/l and 9780 ppm, repectively, when adapted yeast and fed batch culture were used. Germanium taken-up yeast is to be organically bound germanium by evidence of no difference of germanium content after dialysis.
