Bioremediation of Diesel-Contaminated Soil by Bacterial Cells Transported by Electrokinetics

  • The electrokinetic technology was applied in bioremediation for the purpose of supplying a Pseudomonas strain capable of degrading diesel to contaminated soil bed, and their biodegradation of diesel was carried out after a desired cell distribution was obtained. Electrokinetic injection of the strain was made possible because the cells acted as negatively charged particles at neutral pH, and thus the cells were transported with a precise directionality through the soil mostly by the mechanism of electrophoresis and in part by electroosmosis. A severe pH change in the soil bed was formed due to the penetration of electrolysis products, which was harmful to the cell viability and cell transport. To achieve a desirable cell transport and distribution, the control of pH in soil bed by a recirculating buffer solution in electrode chambers was essential during the appliation of an electric field. The judicious selections of electrolyte concentration and conductivity were also important for achieving an efficient electrokinetic cell transport since a higher electrolyte concentration favored the maintenance of pH stability in soil bed, but lowered electrophoretic mobility on the other hand. With electrolyte solution of pH 7 phosphate buffer, a 0.05 M concentration showed a better cell transport buffer, a 0.05 M concentration showed a better cell transport than 0.02 M and 0.08 M. The cell under pH 8 were obtained, compared to the cells under pH 7 or pH 9 in a given time period Up to $60\%$ of diesel was degraded in 8 days by the Pseudomonas cell, which were distributed electrokinetically under the conditions of pH 8 ($1,800{\mu}S/cm$, a mixture of phosphate and ammonia buffers) and 40 mA in a soil bed of 15 cm length.

경안천과 팔당호에서 총세균수의 분포 및 동태 (Distribution and Dynamics of the Total Bacterial Number in the Kyongan Stream and Paltang Reservoir)

  • 경안천과 팔당호 주요 유입부에서 2000년 9월부터 2001년 2월까지 수중 총세균수의 분포 및 변동을 조사하였다. 경안천 본류에서 총세균수는 하수처리장 배출수가 유입될 때 뚜렷이 증가하였다. 경안천 본류에서 유하거리 (km)당 총세균의 소멸량은 0.13${\times}10^{6}$cells/ml로서 하류로 이송되면서 하상에 침강 소멸되는 양이 상당하였다. 총세균수의 변동은 9${\sim}$10월, 11월 및 12${\sim}$2월에 평균값이 각각 1.74${\sim}$3.10${\times}10^{6}$cells/ml, 1.86${\sim}$7.30${\times}10^{6}$cells/ml 및 4.56${\sim}$8.75${\times}10^{6}$cells/ml 범위로서 세균의 생물량은 고수온기에 적었고 저수온기에 오히려 증가하였다. 총세균수는 수온이 >$10^{\circ}C$인 시기(9${\sim}$10월)보다 <$10^{\circ}C$인 저수온기(12${\sim}$2월)에 2.1${\sim}$3.0배 풍부하였다. 총세균수로 평가하였을 때 수질은 부영양상태였고 하수처리장 배출수는 경안천과 팔당호의 미생물 오염에 대한 가장 큰 source로 평가되었다. 경안천 뿐만 아니라 팔당호의 상수원 수질을 보호하기 위해서는 하수처리장 배출수 관리에 대한 대책 마련이 매우 시급한 것으로 판단되었다.

Influence of Temperature on the Bacterial Community in Substrate and Extracellular Enzyme Activity of Auricularia cornea

  • Temperature is an important environmental factor that can greatly influence the cultivation of Auricularia cornea. In this study, lignin peroxidase, laccase, manganese peroxidase, and cellulose in A. cornea fruiting bodies were tested under five different temperatures ($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$, and $40^{\circ}C$) in three different culture periods (10 days, 20 days and 30 days). In addition, the V4 region of bacterial 16S rRNA genes in the substrate of A. cornea cultivated for 30 days at different temperatures were sequenced using next-generation sequencing technology to explore the structure and diversity of bacterial communities in the substrate. Temperature and culture days had a significant effect on the activities of the four enzymes, and changes in activity were not synchronized with changes in temperature and culture days. Overall, we obtained 487,694 sequences from 15 samples and assigned them to 16 bacterial phyla. Bacterial community composition and structure in the substrate changed when the temperature was above $35^{\circ}C$. The relative abundances of some bacteria were significantly affected by temperature. A total of 35 genera at five temperatures in the substrate were correlated, and 41 functional pathways were predicted in the study. Bacterial genes associated with the membrane transport pathway had the highest average abundance (16.16%), and this increased at $35^{\circ}C$ and $40^{\circ}C$. Generally, different temperatures had impacts on the physiological activity of A. cornea and the bacterial community in the substrate; therefore, the data presented herein should facilitate cultivation of A. cornea.

느린 전단흐름에서 편모운동에 의한 대장균의 거동 특성 (Swimming Motion of Flagellated Bacteria Under Low Shear Flow Conditions)

  • 본 연구의 목적은 낮은 전단흐름조건에서 편모 운동성이 박테리아의 거동 특성에 미치는 영향을 파악하는데 있다. 대다수의 미생물은 편모를 이용하여 수용액 내에서 운동할 수 있는 능력을 가지고 있으며, 이러한 운동성은 수계나 수처리 시스템에서 미생물의 거동에 있어서 중요한 역할을 한다. 그러나 현재까지 병원성 미생물의 이동 현상과 관련된 연구에서 편모에 의한 운동성은 거의 고려되지 않고 있는 실정이다. 본 연구에서는 미세유체장치를 이용하여 전단흐름이 낮은 조건에서 E. coli의 거동 특성을 파악하고자 하였다. 실험을 통하여 유속이 작은 경우에 E. coli는 포물선의 형태의 궤적들을 그리며 이동하는 것을 알 수 있었으며, 벽면 근처에서는 상류로 헤엄쳐 올라간다는 것을 파악하였다. 또한 유속과 종횡비(aspect ratio)에 따른 궤적의 변화를 분석하였는데, 유속이 작을수록 포물선 형태의 궤적을 그리게 되며, 길이가 짧을수록 보다 작은 회전 반경을 그리며 운동하는 것을 관찰할 수 있었다.

Isolation and ars Detoxification of Arsenite-Oxidizing Bacteria from Abandoned Arsenic-Contaminated Mines

  • The ecosystems of certain abandoned mines contain arsenic-resistant bacteria capable of performing detoxification when an ars gene is present in the bacterial genome. The ars gene has already been isolated from Pseudomonas putida and identified as a member of the membrane transport regulatory deoxyribonucleic acid family. The arsenite-oxidizing bacterial strains isolated in the present study were found to grow in the presence of 66.7 mM sodium arsenate($V;\;Na_2HAsO_4{\cdot}7H_2O$), yet experienced inhibited growth when the sodium arsenite($III;\;NaAsO_2$) concentration was higher than 26 mM. Batch experiment results showed that Pseudomonas putida strain OS-5 completely oxidized 1 mM of As(III) to As(V) within 35 h. An arsB gene encoding a membrane transport regulatory protein was observed in arsenite-oxidizing Pseudomonas putida strain OS-5, whereas arsB, arsH, and arrA were detected in strain OS-19, arsD and arsB were isolated from strain RW-18, and arsR, arsD, and arsB were found in E. coli strain OS-80. The leader gene of arsR, -arsD, was observed in a weak acid position. Thus, for bacteria exposed to weak acidity, the ars system may cause changes to the ecosystems of As-contaminated mines. Accordingly, the present results suggest that arsR, arsD, arsAB, arsA, arsB, arsC, arsH, arrA, arrB, aoxA, aoxB, aoxC, aoxD, aroA, and aroB may be useful for arsenite-oxidizing bacteria in abandoned arsenic-contaminated mines.

Gallium(III) Nitrate Inhibits Pathogenic Vibrio splendidus Vs by Interfering with the Iron Uptake Pathway

  • It is well known that iron is critical for bacterial growth and pathogenic virulence. Due to chemical similarity, $Ga^{3+}$ competes with $Fe^{3+}$ for binding to compounds that usually bind $Fe^{3+}$, thereby interfering with various essential biological reactions. In our present study, gallium(III) nitrate [$Ga(NO_3)_3$] could repress the growth of V. splendidus Vs without complete inhibition. In the presence of $Ga(NO_3)_3$, the secretion of homogentisic acid-melanin (HGA-melanin) in V. splendidus Vs cells could be increased by 4.8-fold, compared to that in the absence of $Ga(NO_3)_3$. HGA-melanin possessed the ability to reduce $Fe^{3+}$ to $Fe^{2+}$. In addition, HGA-melanin increased the mRNA levels of feoA and feoB, genes coding Fe2+ transport system proteins to 1.86- and 6.1-fold, respectively, and promoted bacterial growth to 139.2%. Similarly, the mRNA expression of feoA and feoB was upregulated 4.11-fold and 2.71-fold in the presence of $640{\mu}M$ $Ga(NO_3)_3$, respectively. In conclusion, our study suggested that although $Ga(NO_3)_3$ could interfere with the growth of V. splendidus Vs, it could also stimulate both the production of $Fe^{3+}$-reducing HGA-melanin and the expression of feoA and feoB, which facilitate $Fe^{2+}$ transport in V. splendidus Vs.

2중 구조의 PVA/alginate 겔 비드에서의 독립영양 단일공정 질소제거효율 시뮬레이션 (Simulated Nitrogen Removal for Double-Layered PVA/Alginate Structure for Autotrophic Single-Stage Nitrogen Removal)

  • Recently, an autotrophic single-stage nitrogen removal (ASSNR) process based on the anaerobic ammonium oxidation (ANAMMOX) reaction has been proven as an economical ammonia treatment. It is highly evident that double-layered gel beads are a promising alternative to the natural biofilm for ASSNR because of the high mechanical strength of poly(vinyl alcohol) (PVA)/alginate structure and efficient protection of ANAMMOX bacteria from dissolved oxygen (DO) due to the thick outer layer. However, the thick outer layer results in severe mass transport limitation and consequent lowered bacterial activity. Therefore, the effects of the thickness of the outer layer on the overall reaction rate were tested in the biofilm model using AQUASIM for ammonia-oxidizing bacteria (AOB), nitrite-oxidizing bacteria (NOB) and ANAMMOX bacteria. A thickness of 0.5~1.0 mm is preferred for the maximum total nitrogen (TN) removal. In addition, a DO of 0.5 mg/L resulted in the best total nitrogen removal. A higher DO induces NOB activity and consequent lower TN removal efficiency. The optimal density of AO B and NO B density was 1~10% for a 10% ANAMMOX bacterial in the double-layered PVA/alginate gel beads. The real effects of operating parameters of the thickness of the outer layer, DO and concentrations of biomass balance should be intensively investigated in the controlled experiments in batch and continuous modes.

L-arginine and N-carbamoylglutamic acid supplementation enhance young rabbit growth and immunity by regulating intestinal microbial community

  • Objective: An experiment was conducted to determine the effects of L-arginine (L-Arg) and N-carbamoylglutamic acid (NCG) on the growth, metabolism, immunity and community of cecal bacterial flora of weanling and young rabbits. Methods: Eighteen normal-grade male weanling Japanese White rabbits (JWR) were selected and randomly divided into 6 groups with or without L-Arg and NCG supplementation. The whole feeding process was divided into weanling stage (day 37 to 65) and young stage (day 66 to 85). The effects of L-Arg and NCG on the growth, metabolism, immunity and development of the ileum and jejunum were compared via nutrient metabolism experiments and histological assessment. The different communities of cecal bacterial flora affected by L-Arg and NCG were assessed using high-throughput sequencing technology and bioinformatics analysis. Results: The addition of L-Arg and NCG enhanced the growth of weanling and young rabbit by increasing the nitrogen metabolism, protein efficiency ratio, and biological value, as well as feed intake and daily weight gain. Both L-Arg and NCG increased the concentration of immunoglobulin A (IgA), IgM, and IgG. NCG was superior to L-Arg in promoting intestinal villus development by increasing villus height, villus height/crypt depth index, and reducing the crypt depth. The effects of L-Arg and NCG on the cecal bacterial flora were mainly concentrated in different genera, including Parabacteroides, Roseburia, dgA-11_gut_group, Alistipes, Bacteroides, and Ruminococcaceae_UCG-005. These bacteria function mainly in amino acid transport and metabolism, energy production and conversion, lipid transport and metabolism, recombination and repair, cell cycle control, cell division, and cell motility. Conclusion: L-Arg and NCG can promote the growth and immunity of weanling and young JWR, as well as effecting the jejunum and ileum villi. L-Arg and NCG have different effects in the promotion of nutrient utilization, relieving inflammation and enhancing adaptability through regulating microbial community.

Sandusky Bay 와 Lake Erie 의 상이한 동물 플랑크톤 군집의 구조에 대하여 (Contrasting Zooplankton Community Structure in Sandusky Bay and Lake Erie)

  • Zooplankton community structure and the factors correlated with community differences were examined in sandusky Bay (SB) and the open water of Lake Erie (LE, U.S.A.). SB zooplankton communities differed from those in LE by having a greater rotifer density and species richness. Keratella spp., Brachionus spp., and Pompholyx complanata dominated SB rotifers; Brachionus and Pompholyx were rarely seen in LE. Of 19 rotifer species observed, nine species were found only at SB sites. Ordination of zooplankton species abundance by detrended correspondence analysis (DCA) showed an overlap between SB and LE sites, but indicated a portion of the space that was occupied by only SB communities. The seasonal trajectories of zooplankton dynamics in the ordination space at SB sites differed from that of LE. The zooplankton most important in forcing site separation along a DCA Axis I at SB sites were Brachionus angularis, Pompholyx complanata, Keratella valga, Keratella quadrata, Filinia terminalis (rotifers), and Eubosmina coregoni and Daphnia (cladocerans). These species had axis scores which were significantly correlated (p<0.01) with bacterial density and bacterial phosphorus, total phosphorus, and algal density. Very high baterial density and very abundant bacterivorous rotifers in SB suggest that the transport of bacterial carbon through rotifers may be a relatively important link to higher trophic leaels. We believe that this "microbial carbon flow" from the base of the food web may be important in determining the suitability of SB as a spawning site and nursery for larval and juvenile fish.nile fish.

RNAseq-based Transcriptome Analysis of Burkholderia glumae Quorum Sensing

  • Burkholderia glumae causes rice grain rot and sheath rot by producing toxoflavin, the expression of which is regulated by quorum sensing (QS). The QS systems of B. glumae rely on N-octanoyl homoserine lactone, synthesized by TofI and its cognate receptor TofR, to activate the genes for toxoflavin biosynthesis and an IclR-type transcriptional regulator gene, qsmR. To understand genome-wide transcriptional profiling of QS signaling, we employed RNAseq of the wild-type B. glumae BGR1 with QS-defective mutant, BGS2 (BGR1 tofI::${\Omega}$) and QS-dependent transcriptional regulator mutant, BGS9 (BGR1 qsmR::${\Omega}$). A comparison of gene expression profiling among the wild-type BGR1 and the two mutants before and after QS onset as well as gene ontology (GO) enrichment analysis from differential expressed genes (DEGs) revealed that genes involved in motility were highly enriched in TofI-dependent DEGs, whereas genes for transport and DNA polymerase were highly enriched in QsmR-dependent DEGs. Further, a combination of pathways with these DEGs and phenotype analysis of mutants pointed to a couple of metabolic processes, which are dependent on QS in B. glumae, that were directly or indirectly related with bacterial motility. The consistency of observed bacterial phenotypes with GOs or metabolic pathways in QS-regulated genes implied that integration RNAseq with GO enrichment or pathways would be useful to study bacterial physiology and phenotypes.
