Comparison of Gene Coding Clones Content in In vivo and In vitro Methyl-Filtration Libraries of Maize(Zea may)

  • It has been hypothesized that efficient exclusion of methylated retrotransposons and repeated DNA region is one of the rapid and cost-effective approaches for comprehensive gene discovery in large genome size of maize. Three kinds of methylation-sensitive restriction enzymes, HapII, MspI and McrBC, were used to identify the restriction frequency of cytosine methylation sites in maize genome. Roughly 60% of total maize genomic DNA was restricted less than 500bp by McrBC, and the most of restricted small size fraction was composed retrotransposon. In order to validate the efficient construction of gene-rich shotgun library, we compare two gene-rich methyl-filtration shotgun libraries using in vivo and in vitro methyl-filtration system. The size selected DNA fraction by Sau3A-McrBC enzyme treated was very stable and has not appeared modification in E. coli, but most insert DNA size of partially digested with Sau3A were decrease less than 500bp by bacterial methylation-modification system. In compare of retroelements portion, A 44.6% of the sequences were retroelement in unmethyl-filtered library, and the most of them was Copia type, such as Prem, Opie and Ji. The portion of retroelement was drastically decreased to 25% and 20% by in vivo and in vitro filtration system, respectively.

Anti-Helicobacter pylori Effect of Costunolide Isolated from the Stem Bark of Mgnolia Sieboldii

  • Helicobacter pylori (H. pylorl) infection is now established as the major pathogenic factor in chronic gastritis and peptic ulcer disease. in addition, there is accumulating evidence that H. pylori plays an important role in the process of gastric carcinogenesis. On the other hand, oriental traditional medicines have been used for stomach disease for thousands of years. In the present study, methanol extract from the stem bark of Magnolia sieboldii (M. sieboldii) and its components were investigated on their inhibitory effects against urease activity and growth of H. pylori in vitro. The methanol extract of M. sieboldii significantly inhibited the growth of H. pylori ATCC 43504 at 5 mg/ml. From the further fractionation, the chloroform fraction inhibited the bacterial growth dose-dependently. Among four fractions separated from the chloroform fraction by silica gel column chromatography, MS-C-2 was the most potent. Costunolide was isolated from the MS-C-2 subtraction by preparative TLC and recrystallization using n-hexane. Anti-H. pylori effect of costunolide was investigated using one commercial strain (H. pylori ATCC 43504) and three clinical strains (H. pylon 4, 43, 82548). Costunolide exhibited potent anti-H. pylori activity, and the MIC was around $100-200{\mu}g/ml$. However, costunolide had no inhibitory effect of H. pylori urease activity at the concentration used for the growth inhibition assay. From these results, we conclude that costunolide inhibits the, growth of H. pylori by the independent manner of H. pylori urease inhibition.

효모에서 포자형성 특이 글루코아밀라제의 분비서열에 의한 세균 endo-1,4-β-D-glucanase의 분비 (The Signal Sequence of Sporulation-Specific Glucoamylase Directs the Secretion of Bacterial Endo-1,4-β-D-Glucanase in Yeast)

  • 효모 Saccharomyces diastaticus가 포자형성기에 세포질에서 생산된다고 알려진 포자형성 특이 glucoamylase (SGA)가 세포 외로 분비되는 단백질임을 증명하고자 S. dastaticus의 SGA promoter와 예상되는 분비신호서열 다음에 reporter gene으로 사용한 고초균의 CMCase 구조유전자를 융합한 재조합 플라스미드 pYSC25를 제작하고 수주세포인 S. diastaticus YIY345에 형질전환 하였다. 형질전환체를 1% CMC를 포함하는 최소한천배지에서 배양한 후 Congo red 염료로 염색하여 생성된 투명환으로부터 SGA의 분비서열에 의해 세균의 CMCase가 효모세포외로 분비되는 것을 확인하였다. 효모세포부위 별 CMCase의 활성분포를 측정하여 SGA 분비서열의 분비효율을 추정하기 위해 효모세포 배양액을 배양상등액, periplasmic 및 세포질 분획으로 나눈 다음 효소활성을 측정한 결과 CMCase 활성의 76%가 배양상등액과 periplasmic 부위에 존재하였으며 N-연결형 당쇄가 일어났으므로 SGA 분비서열은 효과적으로 작용함을 알 수 있었다. 대조균인 고초균에서 생산된 CMCase에서는 당쇄가 일어나지 않은 것을 확인하였다. 이상의 결과로부터 SGA는 아미노 말단에 존재하는, 24개의 아미노산으로 구성된 분비서열을 보유한 분비성 단백질임을 확인하였다.

Long-Term Starvation Induces the Viable-but-Nonculturable Condition in Lactobacillus crispatus KLB46

  • In a previous study, we have isolated a number of lactobacilli from Korean women, and one of them (KLB46) was identified as Lactobacillus crispatus by 16S rRNA gene sequencing. For the ecological treatment of bacterial vaginosis (BV) cell suspension of L. crispatus KLB46 was instillated into BV patients. L. crispatus KLB46 was found to persist for several days in cell suspension with no nutrients. In this study, in order to assess the influence of starvation on physiological activity, we compared the viability and culturability of KLB46 following suspension in various buffer solutions. A pair of in situ fluorescent dye was used to assess viability (i.e. membrane integrity) and the culturability was examined by plate count assay. A rapid epifluorescence staining method using the LIVE/DEAD Bacterial Viability Kit $(BacLight^{TM})$ was applied to estimate both viable and total counts of bacteria in cell suspension. $BacLight^{TM}$ is composed of two nucleic acid-binding stains ($SYTO\;9^{TM}$ and propidium iodide). $SYTO\;9^{TM}$ penetrates all bacterial membranes and stains the cells green while propidium iodide only penetrates cells with damaged membranes, therefore the combination of the two stains produces red fluorescing cells. Optimal staining conditions for $BacLight^{TM}$ were found to be with 0.0835M $SYTO\;9^{TM}$ and 0.05M propidium iodide for 15 min incubation at room temperature in dark. When cells were microscopically examined during 140 hours of starvation, the culturability decreased markedly while the viability remained relatively constant, which suggests that large fraction of KLB46 cells became viable but non-culturable (VBNC) upon starvation.

천연소독제 Clean Natural의 Salmonella typhimurium에 대한 복귀돌연변이시험 (Bacterial Reverse Mutation Test of Clean Natural using Salmonella typhimurium)

  • Clean Watural은 주성분이 참나무로부터 추출정제한 목초액과 프로폴리스로 구성된 새로운 천연 살균소독제이다. Salmonella typhimurium TA98, TA100, TA102, TA1535 및 TA1537 등의 5종의 균주를 이용한 복귀돌연변이성 시험을 수행한 결과, 모든 균주에서 대조군과 비교하여 시험용량군 사이에 돌연변이 유발성이 관찰되지 않았다. 그러나, 앙성대조물질에서는 대조군과 비교하여 통계학적으로 유의한 균주의 증가가 관찰되었다. 따라서, 시험물질인 천연 살균소독제, Clean Natulat은 돌연변이 유발 가능성이 없는 것으로 평가되었다.

산삼배양추출물의 세균을 이용한 복귀돌연변이시험 (Bacterial Reverse Mutation Test of Wild Ginseng Culture Extract)

  • 산삼배양추출물의 세균에서의 돌연변이 유발성 검색을 위하여 Salmonella typhimurium의 히스티딘 요구성 균주 LA100, TA1535, TA98 및 TA1537의 4개의 균주와 대장균 Escherichia coli의 트립토판 요구성 균주인 WP2 uvrA를 이용해 복귀돌연변이 시험을 실시하였다 시험물질은 멸균생리식염수에 용해하여 처리하였다. 대사활성계 적용 및 미적용시 모든 균주에 대해 0, 62, 185, 556, 1,667 및 5,000{\mu}g/plate의 범위를 설정하고 각각 음성 및 양성대 조군으로 시험군을 구성해 본 시험을 실시하였다. 시험 결과 모든 균주에서 최고농도에 이르기까지 집락수의 일관성 있는 증가는 나타나지 않았다. 이상의 결과를 종합할 때, 시험물질 산삼배양추출물은 본 시험조건 하에 사용한 시험균주들의 복귀돌연변이를 유발하지 않는 것으로 사료된다.

Screening and isolation of antibacterial proteinaceous compounds from flower tissues: Alternatives for treatment of healthcare-associated infections

  • Healthcare-associated infection represents a frequent cause of mortality that increases hospital costs. Due to increasing microbial resistance to antibiotics, it is necessary to search for alternative therapies. Consequently, novel alternatives for the control of resistant microorganisms have been studied. Among them, plant antimicrobial protein presents enormous potential, with flowers being a new source of antimicrobial molecules. In this work, the antimicrobial activity of protein-rich fractions from flower tissues from 18 different species was evaluated against several human pathogenic bacteria. The results showed that protein-rich fractions of 12 species were able to control bacterial development. Due its broad inhibition spectrum and high antibacterial activity, the protein-rich fraction of Hibiscus rosa-sinensis was subjected to DEAE-Sepharose chromatography, yielding a retained fraction and a non-retained fraction. The retained fraction inhibits 29.5% of Klebsiella pneumoniae growth, and the non-retained fraction showed 31.5% of growth inhibition against the same bacteria. The protein profile of the chromatography fractions was analyzed by using SDS-PAGE, revealing the presence of two major protein bands in the retained fraction, of 20 and 15 kDa. The results indicate that medicinal plants have the biotechnological potential to increase knowledge about antimicrobial protein structure and action mechanisms, assisting in the rational design of antimicrobial compounds for the development of new antibiotic drugs.

산사(Crataegifructus) 메탄올 추출물로부터 용매분획된 분획물의 생리활성 (Biological Activities of Solvent Fractions from Methanolic Extract of Crataegi fructus)

  • 산사추출물의 DPPH free radical 소거법에 의한 항산화활성은 ethyl acetate 분획($RC_{50}=4.39{\mu}g/m{\ell}$), butanol 분획($RC_{50}=6.25{\mu}g/m{\ell}$)에서 강한 항산화 활성을 보였으며, ${\alpha}$-tocopherol이나 BHA보다 유사하거나 강한 항산화 활성을 나타냈다. Linoleic acid에 대한 항지질과산화 활성 실험은 15일 째에 물층을 제외한 추출물, 분획물에서 높은 활성을 나타내었다. 박테리아에 대한 항균실험은 Staphylococcus aureus에서만 활성을 보이지 않았을 뿐 다른 피검균에서는 높은 활성을 보였다. 다만 fungi strain인 Candida albicands에 대해 각각 250 및 $500{\mu}g/m{\ell}$의 생육 억제 농도를 나타내었다. 항보체 활성화능을 측정한 결과, 물 층 18.4%를 제외한 다른 분획물은 10% 이하의낮은 억제효과를 보이거나 활성이 나타나지 않았다. 따라서 산사 추출물은 항산화 활성, 항지질과산화 활성, 항미생물 활성이 우수하였다. 이러한 결과를 바탕으로 산사 추출물을 이용하여 식품 및 음료 등과 같은 기능성 식품으로서의 다양한 산업적 응용분야에 널리 응용될 수 있을 것으로 생각된다.

괴화((槐花), Flos Sophora japonica Linne) 추출물의 생리활성 (Biological Activities in the Extract of Flos Sophora japonica L.)

Antibacterial Activity of Essential Oil from Abies holophylla against Respiratory Tract Bacteria

  • We extracted essential oils from four species (Pinus densiflora, Larix kaempferi, Pinus koraiensis, and Abies holophylla) in the family Pinaceae to investigate their antibacterial activities against respiratory tract bacteria (Klebsiella pneumoniae, Haemophillus influenzae, Streptococcus pyogenes, Streptococcus pneumonia, and Neisseria meningitides). Among the tested oils, that from A. holophylla was showed strong activity based on disc diffusion and broth medium dilution (minimum inhibitory concentration, MIC) assays. Qualitative analysis of A. holophylla oil was carried out by GC-MS; ${\alpha}$-pinene, camphene, ${\beta}$-pinene, 3-carene, limonene, bornyl acetate, borneol, ${\beta}$-caryophyllene, ${\alpha}$-caryophyllene, caryophyllene oxide, and ${\alpha}$-bisabolol were identified as its major constituents. Fractionation by silica gel chromatography was performed to analyze the active constituents of the crude oil. In particular, one fraction containing caryophyllene oxide as the major constituent showed stronger antibacterial activity than the crude oil of A. holophylla. Growth rates of bacterial strains exposed to fraction D were explored by optical density (OD600) measurements while morphology was examined by optical microscopy observations ( ${\times}1000$). OD600 of K. pneumoniae decreased from 0.2582 to 0.005 in response to treatment with fraction D at a MIC value of $0.31{\mu}{\ell}/m{\ell}$.
