The diseases caused by Xylella fastidiosa are associated with aggregation of the bacteria m xylem vessels, formation of a gummy matrix and subsequent blockage of water uptake. In the closely related pathogen, Xanthomonas campestris, xanthan gum is known to be an important virulence factor, probably contributing to bacterial adhesion, aggregation and plugging of xylem. Xanthan gum, produced by X. campestris, is an extra-cellular polysaccharide consisting of a cellulose backbone ($\bate$-1,4-linked D-glucose) with trisaccharide side chains composed of mannose, glucuronic acid and mannose attached to alternate glucose residues in the backbone. We had constructed a mutant of X. campestris lacking gumI gene that is responsible for adding the terminal mannose for producing modified xanthan gum which is similar to xanthan gum fromX. fastidiosa. The modified xanthan gum degrading endgphytic bacterium Acineto-bacter johnsonii GX123 isolated from the oleander infected with leaf scorch disease.

This work reports a facile and effective antibacterial coating for oxide substrates. As a coating material, a random copolymer, abbreviated as poly(TMSMA-r-PEGMA), was synthesized by radical polymerization of 3-(trimethoxysilyl)propyl methacrylate (TMSMA) and poly(ethylene glycol) methyl ether methacrylate (PEGMA). Polymeric self-assembled monolayers of poly(TMSMA-r-PEGMA) were formed on various inorganic oxide substrates, including silicon oxide, titanium dioxide, aluminum oxide, and glass, via the simple dip-coating process. The polymer-coated substrates were characterized by ellipsometry, contact angle measurements, and X-ray photoelectron spectroscopy. The bacterial adhesion on the polymer-coated substrates was completely suppressed compared to that on the uncoated substrates.

The aim of this work is to study the effect of aggregate type on the physico-mechanical properties and microstructure of bio-mortar (BM). Three different aggregates such as sand, dolomite and basalt were used. BM was prepared by mixing aggregates with bacterial cells (Sporosarcina Pasteurii) and one equimolar (1 M) of $urea/CaCl_2.2H_2O$. The results proved that the chemical composition and physical properties of aggregates play an important role in the microbial precipitation rate as well as size, morphology and crystallinity of the precipitated calcite, which strongly reflects on the properties of the prepared BM. The BM containing dolomite gave the highest compressive strength and lowest water absorption.
The human microbiome, which consists of a collective cluster of commensal, symbiotic, and pathogenic microorganisms living in the human body, plays a key role in host health and immunity. The human nasal cavity harbors commensal bacteria that suppress the colonization of opportunistic pathogens. However, dysbiosis of the nasal microbial community is associated with many diseases, such as acute respiratory infections including otitis media, sinusitis and bronchitis and allergic respiratory diseases including asthma. The nasopharyngeal acquisition of pneumococcus, which exists as a pathobiont in the nasal cavity, is the initial step in virtually all pneumococcal diseases. Although the factors influencing nasal colonization and elimination are not fully understood, the adhesion of opportunistic pathogens to nasopharyngeal mucosa receptors and the eliciting of immune responses in the host are implicated in addition to bacterial microbiota properties and colonization resistance dynamics. Probiotics or synbiotic interventions may show promising and effective roles in the adjunctive treatment of dysbiosis; however, more studies are needed to characterize how these interventions can be applied in clinical practice in the future.
복합레진 표면에 대한 APF gel 도포는 표면을 거칠게 하고 세균 부착을 증가시킨다. 본 연구는 Filtek Z250(FZ), Filtek Supreme Universal(FS)과, 실험적으로 나노충전재를 각각 0%, 3%, 6% 포함시켜 만든 복합레진(E0, E3, E6)으로 제작한 레진 시편을, APF gel을 적용한 군과 적용하지 않은 군으로 나누어 Streptococcus mutans의 부착량과 표면 조도를 측정하고 비교, 평가하여 다음과 같은 결과를 얻었다. 1. APF gel을 적용하지 않은 레진 시편에 대한 S. mutans 부착량은 FS에서 가장 적었고, FZ, E3, E6에 대한 부착량보다는 유의하게 낮았다(p<0.05). 2. 모든 레진군에서 APF gel을 적용한 레진 시편에 대한 S. mutans 부착량은 APF gel을 적용하지 않은 시편에 대한 부착량보다 유의하게 높았다(p<0.05). 3. APF gel을 적용한 레진 시편에 대한 S. mutans 부착량은 레진군간 유의한 차이를 보였으며(p<0.05), FS, FZ, E0, E3, E6의 순서로 높은 값을 보였다. 4. APF gel 적용 전이나 후에, 레진군간 표면조도의 차이는 유의하지 않았다(p>0.05).
치과용 재료의 표면특성은 세균 부착에 중요한 역할을 한다. 본 연구의 목적은 다섯 종류의 광중합 교정용 접착제(불소를 방출하지 않는 세 종류의 콤포지트 레진, 불소를 방출하는 콤포지트 레진 한 종류, 레진 변형 글래스아이오노머 시멘트 한 종류)의 표면특성을 평가하는 것이다. 표면거칠기는 공초점 레이저주사전자현미경을 이용하여 측정하였고, 접촉각과 표면에너지 요소는 sessile drop method를 이용하여 분석하였다. 본 연구의 결과 교정용 접착제간 표면거칠기는 각 재료간 표면거칠기 차이가 $0.05\;{\mu}m$ 이하로 상대적으로 적지만 각 재료 사이에 유의성 있는 차이를 보였다. Transbond XT와 Enlight는 Monolok2와 Lightbond 보다 유의하게 덜 거칠었다. 접촉각과 표면에너지 구성성분은 접착제 사이에 유의성 있는 큰 차이를 보였는데 특히 레진 변형 글래스아이오노머와 콤포지트 레진 접착제간에 접촉각과 표면에너지에서 커다란 차이를 보였다. 레진 변형 글래스아이오노머의 경우 콤포지트 레진 접착제에 비해 유의하게 작은 접촉각과 높은 표면에너지를 보였으며 콤포지트 레진 접착제보다 강한 극성, 특히 강한 염기성 경향을 보였다 본 연구는 레진 변형 글래스아이오노머가 콤포지트 레진 접착제에 비해 세균 부착에 유리한 환경을 제공한다는 것을 보여주었다.
천연물 황금으로부터 여러 유기용매 추출물을 얻어 치아우식증 원인균인 S. mutans 에 대한 항균활성, 배지 내 환경변화, 그리고 부착억제에 대해 알아보았다. 1. 구강 질병인 치아우식증의 대표적 원인균 S. mutans에 대한 황금 ethyl acetate 추출물 (IPK-3)의 최소억제농도는 125 mg/ml 이었다. 2. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때 균의 생장 량과 최대 생장시간은 대조군보다 지연되었다. 3. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때, pH 변화는 대조군은 18시간에서 pH 5.63으로 급격한 변화를 보였으며 황금 IPK-3 추출물이 50 mg/ml의 배지에서는 6.50이상을 유지하여 pH가 변화가 없었다. 4. 황금 IPK-3 추출물을 첨가한 배지 내 탄수화물, 단백질 및 균체 외 다당류의 변화는 대조군에 비해 매우 낮게 생산되었다. 5. 균체를 형광염료(DAPI)로 염색하여 S-HA 부착억제을 확인한 결과, 대조군은 시간이 경과함에 따라 S.mutans의 hydroxyapatite(HA)에 부착 정도가 증가되었으나, 황금 IPK-3 추출물이 첨가된 시험군에서는 S-HA에 균의 부착이 매우 적었다. 이같이 황금의 IPK-3 (ethyl acetate 추출물)이 치아우식증의 원인균인 S. mutans 에 대한 항균활성효과 뿐만아니라 hydroxyapatite에 부착 억제 효과도 있었다.
목적: 이번 연구는 다양한 pH의 불소 제재들이 티타늄 표면 거칠기에 미치는 영향을 평가하는 것이었다. 연구 재료 및 방법: 기계절삭형 티타늄 디스크를 시중에서 유통되는 세 가지 불소겔로 처리하였다. 불소겔의 종류와 처리 시간에 따라, 대조군, 1군(pH 3.5의 APF로 1분간 처리), 2군(pH 3.5의 APF로 30분간 처리), 3군(pH 4.0의 APF로 1분간 처리), 4군(pH 4.0의 APF로 30분간 처리), 5군(pH 7.0의 NaF로 1분간 처리), 6군(pH 7.0의 NaF로 30분간 처리)의 7군으로 분류하였다. 디스크의 표면 거칠기를 측정한 후, Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum을 배양하여 디스크에 부착하는 세균의 양을 측정하였다. 결과: 표면 거칠기는 그룹2에서만 유의하게 증가하였다(P < 0.0001). 세균의 부착량은 실험군과 대조군 사이에 유의한 차이를 보이지 않았다. 결론: pH 3.5의 APF를 30분간 처리한 그룹에서 표면 거칠기가 유의하게 증가하였지만, 세균의 부착에 대해서는 유의한 차이를 보이지 않았다.
병원성 대장균 O157:H7은 식중독 사고를 일으키는 주된 원인균으로서 항생제 내성 문제를 극복하기 위해서는 병원성 대장균 O157:H7을 제어하기 위한 새로운 방법이 개발되어야 한다. 따라서 본 연구에서는 식물화학물질을 스크리닝하여 병원성 대장균 O157:H7의 부착에 주된 역할을 하는 LEE 오페론의 발현을 감소시키는 물질을 찾고자 하였다. 스크리닝을 통해 선발된 식물화학물질 중 다이오스민은 사람 결장 상피세포 부착능을 3.62배 감소시킴으로써(p<0.01) 양성대조군으로 사용된 미리세틴과 유사한 정도의 효과를 나타냈다. 생물막 형성에 있어서는 다이오스민 처리 시 표현형이 25.6% 감소하여 유의적 차이가 확인되었고(p<0.05), curli 유전자의 발현 역시 미리세틴 처리 때보다 1.57-2.60배 더 유의미하게 감소하는 것으로 나타나 양성대조군보다 더 좋은 효과를 보였다. 한편 다이오스민은 병원성 대장균 O157:H7의 생장에는 아무런 영향을 미치지 않는 것으로 나타나 내성 발생률이 저감화된 새로운 항미생물제재로서 사용 가능하리라 판단된다.
Probiotics are microorganisms that have beneficial effects on the health of the host. The health promoting effect by probiotics influences suppressing harmful bacteria, prevention of constipation, blood cholesterol reduction and regulation of blood pressure. Prebiotics are used to promote the growth or activity of microorganisms. Synbiotics, which are a mixture of probiotics and prebiotics, synergize in the intestines by complementing each other. Synbiotics not only improves the viability of the probiotics while passing through the gastrointestinal tract, maintain intestinal homeostasis, but also regulate balance of harmful and useful bacterial growth. Glycine max Merr (GMM) has been widely used in Asian countries to treat cancer, obesity, oxidative stress and imbalanced immune diseases. In addition, it has been reported that dietary fiber-rich grains promote bowel movements and prevent constipation. In this study, we investigated the viability of LactobacillIus buchneri (L.buchneri) strains, known as lactic acid bacteria under conditions of gastric fluid and intestinal fluid to determine the suitability of L.buchneri as probiotics. The adhesion ability of L.buchneri to caco-2 cells was also confirmed. The present studies showed that GMM extract promoted the growth and activity of L.buchneri strains as prebiotics. Also, this results suggested that the mixture of L.buchneri and GMM extract can helps maintain intestinal health and healthy body as synbiotics and health functional food material.
