Antitumor Activity of Fermented Antler on Sarcoma 180 in Mice (복수암 생쥐에 대한 발효녹용의 항암작용)

  • Some components and antitumor effect of antler and fermented antler with Bacillus P-92 were determined. By fermenting the antler with Bacillus P-92, the amount of free amino acids, polypeptides and other compounds were increased. The free amino acid of the fermented antler was 6-fold higher than those of the untreated antler. Sarcoma 180 cells were intraperitoneally implanted into ICR mice to determine the effect of fermented antler and untreated antler on life span prolongation. The life span of ICR mouse treated with antler and fermented antler were prolonged to 24.4% and 39.2%(p<0.05), and survival rates were 22% and 33%, respectively. In addition, the induction of neutrophil was 2.3-fold and 3.4-fold, respectively.

  • With the population growth and industrialization, the characteristics of discharged waste water and sewage have become more diverse. The removal of phosphorus (P) in the wastewater is essential for the prevention of eutrophication in the river and stream. This study was performed in order to estimate the field application of the Bacillus sp. 3434 BRRJ. Bacillus sp. 3434 BRRJ was cultured in the raw wastewater and synthetic medium at the 5 L reactor. The best optimum conditions for P removal by Bacillus sp. 3434BRRJ in the synthetic medium at the 5 L reactor were as follows: temperature, $30^{\circ}C$; P concentration, 20 mg/L; carbon sources, glucose + acetate (1:1); oxygen concentration, alternatively anaerobic and aerobic conditions. P removal efficiency under the optimum condition was 89.4%. In case of wastewater, P removal efficiency was 95.5% under controlled at $30^{\circ}C$. Through this study we confirmed that P removal by Bacillus sp. 3434BRRJ in case of wastewater was as effective as the synthetic medium. It is considered that Bacillus sp. 3434 BRRJ can be applied to the treatment of wastewater in order to biologically remove P from the wastewater on a large scale.

  • A Baczllus circdans KCTC3004 $\alpha$-amylase gene contained in a recombinant plasmid pAL850 was transferred into a new shuttle vector plasmid pALSIlI by ligating linearlzed DNAs of pUC19 and pUB110. B. subtilis RM125 and B. megatenurn ATCC14945 transfonned with pALS111 produced the $\alpha$-amylase substantially Most of the enzyme was produced during the exponential growth period. The maxiinurn activities of the $\alpha$-amylase produced by the Bucillus transformants were compared with that of the B. circulans gene donor strain. The B. subtilis RM125(pALS111) enzyme showed the actlvicy 95 times higher than that of the gene donor cells, followed by the B, nzegaterium ATCC14945(pALSlll) enzyme with activity 34 limes higher than that of the gene donor cells. While E coli secreted about 10% of the produced enzyme, B. subtilis excreted the enzyme inlo the medium wholly and B. megaterirun about 98% ofthe total product. The plasmid pALSI11 was quite stable inB. nzegaterium (92%), inoderately stable in B. subtilis (76%), but was unstable in E. coli (38%). The SDS-PAGE and zymogram of this enzyme produced in E. coli(pALS111), B. subtilis( pALS111) or B. megateril~m (pALS111) indicated a molecular weight of 55,000. The enzymes overproduced in three different host cells hydrolyzed starch to produce mainly maltoaiose and mallooligosaccharides.

Probiotic Potential of Indigenous Bacillus sp. BCNU 9028 Isolated from Meju (메주로부터 분리한 토착 Bacillus sp. BCNU 9028의 프로바이오틱스로서 이용 가능성)

  • Spore-forming bacteria are being used as probiotic supplements for human and animal use, due to their low pH stability and ability to survive the gastric barrier. In this study, the BCNU 9028 strain was screened from meju, a Korean fermented soybean food starter. Biochemical and physiological characteristics, as well as 16S rDNA sequence analyses, indicate that this strain belongs to the genus $Bacillus$. $Bacillus$ sp. BCNU 9028 showed a 92% survivability at pH 2.5 and could also withstand 0.3% ox bile. Furthermore, it was postulated that $Bacillus$ sp. BCNU 9028 could prevent biofilm formation and adherence of food-borne pathogens such as $Listeria$ $monocytogenes$, $S.$ $aureus$ and $E.$ $coli$ on the basis of its autoaggregation and coaggregation capacity with food-borne pathogens. It was shown that BCNU 9028 has good abilities to adhere to the intestinal tract from its hydrophobic character (63.3%). The $Bacillus$ sp. BCNU 9028 strain especially elicited antibacterial activity against both Gram-positive and -negative pathogens. These findings suggested that the $Bacillus$ sp. BCNU 9028 strain could be used as a potential probiotic.

Distribution of Beneficial Bacteria in the Intestines after Enzamin Ingestion of Bacillus subtilis AK Strain Fermentation (Bacillus subtilis AK균 발효액(Enzamin)의 섭취 후 장내 유익세균의 분포조사)

  • Purpose: The purpose of this study was to investigate whether intestinal proliferation is promoted in beneficial intestinal bacteria or decreased in harmful bacteria before and after ingesting Bacillus fermentation broth (ENM) for 8 weeks in the 16 subjects. Method: Intestinal bacteria were identified by PCR amplification using specific 16S rRNA primers. Results: The Bifidobacterium gene index(%)(gi%) increased to 58.92% in the control group and 69.53% in the test group after the ingestion of ENM, but there was no significant difference. Lactobacillus gi% increased significantly (49.37% in the control and 66.43% in the test) (p<.029). Clostridium gi% was significantly decreased after treatment (83.16% in the control and 67.76% in the test) (p<.077). Bacteroides gi% increased significantly (12.58% in the control and 20.87% in the test) after ingesting (p<.095). Prevotella gi% increased significantly (7.55% in the control and 17.28% in the test) after ingesting (p<.005). After ingesting, the median bacteria increased significantly in the control (20.06%) and the test (35.88%) (p<.001). Conclusions: After ingestion of the ENM, the number of beneficial bacteria increased and the number of harmful bacteria Clostridium tended to decrease. This suggests that ingestion of the Bacillus fermented beverage ENM has an effect on the proliferation of intestinal bacteria.

  • Bacillus P-92 which fermented antler was isolated from intestinal bacteria. The biological activites, carbon clearance and growth activity of lactic acid bacteria, of fermented antler was better than those of untreated antler. The enyzmes activities, ${\beta}-glucosidase,\;{\beta}-glucuronidase$ and tryptophanase, of intestinal bacteria of mice treated with fermented antler were lower than those of mice treated with untreated antler, although those of mice treated with fermented antler or untreated antler were higher than those of control. Biological activity of the antler seems to be increased by fermentation.

Characteriaation of BmaI methylase from bacillus macerans (Bacillus macerans의 BmaI methylase의 특성)

  • The isolation and characterization of a new type II methylase, BmaI methylase, from Bacillus macerans ATCC 8244 were described. BmaI methylase was isolated by procedures of ammonium sulfate fractionation, DEAE-cellulose chromatography and phosphocellulose chromatography. Two types of methylases were present in this strain and only one of the two was a site specific BmaI methylase. The pBR322 DNA methylated by BmaI methylase was not cleaved by BmaI endonuclease, and pBR322 DNA cleaved by BmaI endonuclease was not methylated by BmaI methylase. The optimal pH for the BmaI methylase activity was 7.5, and optimal NaCl concentration was about 50 mM. BmaI methylase could methylate single-stranded M13mp18 DNA.

Isolation and Biochemical Characterization of Bacillus pumilus Lipases from the Antarctic

  • Lipase-producing bacterial strains were isolated from Antarctic soil samples using the tricaprylin agar plate method. Seven strains with relatively strong lipase activities were selected. All of them turned out to be Bacillus pumilus strains by the 16S rRNA gene sequence analysis. Their corresponding lipase genes were cloned, sequenced, and compared. Finally, three different Bacillus pumilus lipases (BPL1, BPL2, and BPL3) were chosen. Their amino acid sequence identities were in the range of 92-98% with the previous Bacillus pumilus lipases. Their optimum temperatures and pHs were measured to be $40^{\circ}C$ and pH 9. Lipase BPL1 and lipase BPL2 were stable up to $30^{\circ}C$, whereas lipase BPL3 was stable up to $20^{\circ}C$. Lipase BPL2 was stable within a pH range of 6-10, whereas lipase BPL1 and lipase BPL3 were stable within a pH range of 5-11, showing strong alkaline tolerance. All these lipases exhibited high hydrolytic activity toward p-nitrophenyl caprylate ($C_8$). In addition, lipase BPL1 showed high hydrolytic activity toward tributyrin, whereas lipase BPL2 and lipase BPL3 hydrolyzed tricaprylin and castor oil preferentially. These results demonstrated that the three Antarctic Bacillus lipases were alkaliphilic and had a substrate preference toward short- and medium-chain triglycerides. These Antarctic Bacillus lipases might be used in detergent and food industries.

Overexpression and characterization of thermostable chitinase from Bacillus atrophaeus SC081 in Escherichia coli

  • The chitinase-producing strain SC081 was isolated from Korean traditional soy sauce and identified as Bacillus atrophaeus based on a phylogenetic analysis of the 16S rDNA sequence and a phenotypic analysis. A gene encoding chitinase from B. atrophaeus SC081 was cloned in Escherichia coli and was named SCChi-1 (GQ360078). The SCChi-1 nucleotide sequences were composed of 1788 base pairs and 596 amino acids, which were 92.6, 89.6, 89.3, and 78.9% identical to those of Bacillus subtilis (ABG57262), Bacillus pumilus (ABI15082), Bacillus amyloliquefaciens (ABO15008), and Bacillus licheniformis (ACF40833), respectively. A recombinant SCChi-1 containing a hexahistidine tag at the amino-terminus was constructed, overexpressed, and purified in E. coli to characterize SCChi-1. $H_6SCChi$-1 revealed a hydrolytic band on zymograms containing 0.1% glycol chitin and showed the highest lytic activity on colloidal chitin and acidic chitosan. The optimal temperature and pH for chitinolytic activity were $50^{\circ}C$ and pH 8.0, respectively.

Advanced Wastewater Treatment using Sequencing Batch Reactor on ship's sewage (연속 회분식 공정(SBR)을 이용한 선박 오·폐수의 고도처리)

  • Lab scale experimental study was carried out for SBR process, to investigate the effects of influent ship sewage organic compound removal and Bacillus sp. state on design parameters. This process was able to remove nitrogen and phosphorus as well as organic matter efficiently. More than $92.0\%$ of chemical oxygen demand(COD) were removed. In addition, about $84.0\%$ of total nitrogen (T-N) was reduced. The total phosphorus(T-P) reduction averaged $93\%$. The performance load of SBR process was shown to be $0.095kg{\cdot}TOC/m3{\cdot}day$. The pH was decreased from 8.1 to 7.0 within 30 min and increased to 7.3 at the end of anoxic stage, and these phenomena were explained. The sludge produced in the SBR process is characterized by low generation rate (about $0.36kg{\cdot}MLSS/kg{\cdot}TOC$) and excellent settleability. The number of Bacillus sp. in the SBR was $24.2\%$, indicating that Bacillus sp. was a predominant species in the reactor.
