The Effects of HyungBangSaBaek-San(JingFangXieBaiSan) on the Alzheimer's Disease Model Induced by βA (βA로 유도된 Alzheimer's Disease 동물모델에 대한 형방사백산(荊防瀉白散)의 효과)

  • Objectives : This research investigates the effect of the HBSBS on Alzheimer's disease. Specifically, the effects of the HBSBS extract on (1) the behavior (2) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's disease mice induced with $\beta$A were investigated. Methods : The effects of the HBSBS extract suppressed the expression of IL-1$\beta$, IL-6, TNF-$\alpha$ and NOS-II mRNA in BV2 microglial cell line treated with LPS plus $\beta$A were investigated. The effects of the HBSBS extract on the behavior of the memory deficit mice induced by scopolamine were investigated. Results : 1. The HBSBS extract suppressed the expression of IL-1$\beta$, IL-6, TNF-$\alpha$ and NOS-II mRNA in BV2 microglial cell line treated with LPS plus $\beta$A. 2. The HBSBS extract suppressed the expression of $\beta$A protein production in BV2 microglial cell line treated with LPS plus $\beta$A. 3. The HBSBS extract showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. 4. The HBSBS group suppressed the over-expression of IL-1$\beta$ protein, TNF-$\alpha$ protein significantly in the mice with Alzheimer's disease induced by $\beta$A. 5. The HBSBS group reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by $\beta$A. 6. The HBSBS group reduced tau protein, and GFAP in the brain tissue of the mice with AD induced by $\beta$A. Conclusions : These results suggest that the HBSBS group may be effective for the treatment of AD. Thus, HBSBS could be considered among the future therapeutic drugs indicated for the treatment of AD.

Anti-neuroinflammatory effects of ethanolic extract of black chokeberry (Aronia melanocapa L.) in lipopolysaccharide-stimulated BV2 cells and ICR mice

  • BACKGROUND/OBJECTIVES: One of the mechanisms considered to be prevalent in the development of Alzheimer's disease (AD) is hyper-stimulation of microglia. Black chokeberry (Aronia melanocapa L.) is widely used to treat diabetes and atherosclerosis, and is known to exert anti-oxidant and anti-inflammatory effects; however, its neuroprotective effects have not been elucidated thus far. MATERIALS/METHODS: We undertook to assess the anti-inflammatory effect of the ethanolic extract of black chokeberry friut (BCE) in BV2 cells, and evaluate its neuroprotective effect in the lipopolysaccharide (LPS)-induced mouse model of AD. RESULTS: Following stimulation of BV2 cells by LPS, exposure to BCE significantly reduced the generation of nitric oxide as well as mRNA levels of numerous inflammatory factors such as inducible nitric oxide synthase (iNOS), cyclooxygenase 2 (COX-2), interleukin 1 beta ($IL-1{\beta}$), and tumor necrosis factor alpha ($TNF-{\alpha}$). In addition, AD was induced in a mouse model by intraperitoneal injection of LPS ($250{\mu}g/kg$), subsequent to which we investigated the neuroprotective effects of BCE (50 mg/kg) on brain damage. We observed that BCE significantly reduced tissue damage in the hippocampus by downregulating iNOS, COX-2, and $TNF-{\alpha}$ levels. We further identified the quinic acids in BCE using liquid chromatography-mass spectrometry (LCMS). Furthermore, we confirmed the neuroprotective effect of BCE and quinic acid on amyloid beta-induced cell death in rat hippocampal primary neurons. CONCLUSIONS: Our findings suggest that black chokeberry has protective effects against the development of AD.

Antioxidant effect and inhibitory activities of ethyl acetate fraction from Gardenia jasminoides extract on nitric oxide production and pancreatic cancer cell proliferation (치자 에틸아세테이트 분획의 산화방지, 산화질소 제거 및 암세포증식 억제 활성)

  • To evaluate the radical scavenging activity of phenolic-rich fractions of Gardenia jasminoides, we first measured the levels of total polyphenols in hexane, ethyl acetate, and butanol fractions from the extract of G. jasminoides. The ethyl acetate fraction of G. jasminoides extract (GJ-EA) showed high level of phenolics, potent reducing power, and 2,2-diphenyl-1-picrylhydrazyl/2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid radical scavenging effect. In addition, GJ-EA inhibited the overproduction of nitric oxide in lipopolysaccharide-activated BV-2 microglia. Furthermore, we found that GJ-EA suppressed $H_2O_2$-induced PANC-1 pancreatic cancer cell proliferation in a concentration-dependent manner and also reduced their migratory ability. These results suggest that GJ-EA may be a good source for functional foods with antioxidant and chemo-preventive activities.

Effects of Houttuynia cordata Extracts of Different Aerial Parts on Antioxidants and Anti-inflammatory

  • The current study was carried out to determine the effects of the leaf and root of Houttuynia cordata Thunb on antioxidant and anti-inflammatory. Total polyphenol contents of leaf and root ethanol extracts were found to be 59.32 and 12.07 mg/g, respectively. Also, total flavonoid contents of leaf and root ethanol extracts were found to be 10.85 and 8.55 mg/g, respectively. The $RC_{50}$ values of DPPH radical scavenging of leaf and root ethanol extracts were 23.51 and $154.72{\mu}g/mL$, respectively. The $RC_{50}$ values of ABTS radical scavenging of leaf and root ethanol extracts were 35.42 and $233.89{\mu}g/mL$, respectively. The antioxidant activities in leaf ethanol extracts were higher in root. Also, to confirm anti-inflammatory activity of ethanol extract, we treat leaf and root of Houttuynia cordata Thunb extract on BV-2 cell with LPS. The NO inhibition effects in of Houttuynia cordata Thunb leaf ethanol extracts showed higher values compared with the root ethanol extracts. These results indicate that Houttuynia cordata Thunb ethanol extracts may play a positive role in antioxidant and anti-inflammatory.

Improvement of Menopausal Signs by Isoflavones Derived from Sophorae fructus in Ovariectomized Female Rats and the Antioxidant Potentials in BV2 Cells

  • The aim of this study was to determine if the isoflavones from Sophorae fructus (SISO) have potential clinical benefit in hormone replacement therapy (HRT) for the treat ment of menopausal signs, such as the levels of total cholesterol (TC), high density lipoprotein (HDL), low density lipoprotein (LDL) and follicle stimulating hormone (FSH). An additional aim was to present the potential antioxidant effect of SISO in a microglial cell line. For the animal model, the ovaries were removed from adult rats and the indicators of menopause were measured at the pre- and post-administration time points. Although no statistically significant correlation was found, SISO tended to decrease the TC level (p=0.15) and the FSH level (p=0.36), but to increase the HDL level (p=0.303).SISO (< 5${\mu}g$/mL) also exerted antioxidant activity on BV-2 microglial cells by inhibiting lipopolysaccharide-induced nitric oxide. This cytoprotective effect was confirmed by trypan blue staining, which was used to test for cellular damage from H$_2O_2$. In conclusion, this study highlights the anti-menopausal and antioxidant effect of SISO in an ovariectomized rat model, as well as in microglial cells, and provides new clinical targets for the screening of phytoestrogens as potential candidates for HRT in menopausal women.

Comparative Study of Bang-poong (root of Saposhnikovia divaricata Schischkin) and Related Species on Neuroprotective and Acetylcholinesterase Inhibitory Effects (방풍류(防風類) 약재(藥材)의 신경세포보호효과 및 아세틸콜린에스터라제 저해 효과 비교)

  • Objectives : Bang-poong (Saposhnikovia divaricata; SD) was traditionally used to treat inflammatory disorders. In this study, we aimed to investigate whether Bang-poong and related species including SD, Glehnia littoralis (GL), and Peucedanum japonicum (PJ) possess neuroprotective effects and acetylcholinesterase (AChE) inhibitory activities. Methods : Roots of SD, GL and PJ were extracted with distilled water (DW) or 70% ethanol (EtOH). We assessed 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activities of the extracts. To examine neuroprotective effects, we measured cell viability in PC12 or HT22 cells after treatment of the extracts with $H_2O_2$ or amyloid-beta ($A{\beta}$). To assess anti-neuroinflammatory effects, we measured the nitric oxide (NO) levels after treatment with the extracts and lipopolysaccharide (LPS) in BV2 microglial cells. In addition, we performed AChE inhibition assay to explore effects of the extracts on the cholinergic system. Results : DW and EtOH extracts of SD, GL and PJ showed mild DPPH free radical scavenging activities. Also, DW extracts of GL and PJ showed protective effects against $H_2O_2$-induced toxicity in PC12 cells. In LPS-activated BV2 cells, EtOH extracts of SD, GL and PJ exerted inhibitory effects on NO production. Meanwhile, DW extracts of SD, GL and PJ inhibited the $A{\beta}$-induced cell death in HT22 cells. In addition, DW and EtOH extracts of GL exhibited remarkable inhibitory activities on AChE. Conclusions : We demonstrated that SD, GL and PJ exert anti-oxidative, anti-neuroinflammatory and AChE inhibitory activities. These results indicate that SD, GL and PJ could be potential candidates for neurological disorders.

The effects of the CMT and MCMT hot water extract on expression of IL-l${\beta}$, IL-6, TNF-${\alpha}$, NOS-II, COX-2, IL-10, TGF-${\beta}$1 mRNA and production of IL-lP, IL-6, TNF-a, NO, ROS in BV2 microglial cell line treated by lipopolysacchaide(LPS) ; serum glucose, uric acid, AChE activity of the memory deficit mice induced by scopolamine , behavior of the memory deficit mice induced by scopolamine and were investigated, respectively. The MCMT hot water extract & ultra-fine powder increased glucose, decreased uric acid and AChE significantly in the serum of the memory deficit mice induced by scopolamine. The CMT and MCMT hotwater extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. According to the above result, it is suggested that the CMT and MCMT hot water extract & ultra-fine powder might be usefully applied for prevention and treatment of dementia.

Anti-neuroinflammatory Effects of a Locusta migratoria Ethanol Extract in LPS-stimulated BV-2 Microglia (LPS로 자극된 미세아교세포에서 풀무치 에탄올 추출물의 신경염증 억제 효능)

  • Activated microglia, induced by various pathogens, protect neurons and maintain homeostasis of the central nervous system (CNS). However, severe activation causes neurodegenerative disorders such as Alzheimer's disease and Parkinson's disease because of the secretion of various neurotoxic molecules, such as nitric oxide (NO), prostaglandin (PG), and pro-inflammatory cytokines. Because chronic microglial activation endangers neuronal survival, negative regulators of microglial activation have been identified as potential therapeutic candidates for treatment of many neurological diseases. One potential source of these regulators is Locusta migratoria, a grasshopper of the Acrididae, usually 4-6 cm in size, belonging to the family of large insects in Acrididae. This grasshopper is an edible insect resource that can be consumed by humans as protein source or used for animal feed. The aim of the present study was to examine the inhibitory effects of a L. migratoria ethanol extract (LME) on the production of inflammatory mediators in LPS-stimulated BV-2 microglia cells. The extract significantly inhibited the NO, iNOS, COX-2, and pro-inflammatory cytokine ($TNF-{\alpha}$, IL-6 and $IL-1{\beta}$) levels in BV-2 microglia cell. Because the inhibition of microglial activation may be an effective solution for treating brain disorders like Alzheimer's and Parkinson's diseases, these results suggest that LME may be a potential therapeutic agent for the treatment of brain disorders induced by neuroinflammation.

The Effects of OJaJiHwangEumJa(OJJHEJ) Hot water extract & Ultra-fine Powder on Proinflammatory Cytokine of Microglia and Memory Deficit Model (오자지황음자(五子地黃飮子) 열수추출물과 초미세분말이 싸이토카인과 건망증 생쥐모델 기억력감퇴에 미치는 영향)

  • Background: Microglia produces a barrage of factors (IL-l, TNF-$\alpha$, NO, superoxide) that are toxic to neurons and playa major role in the cellular immune response associated with the pathology of Alzheimer's disease(AD). OJaJiHwangEumJa(OJJHEJ) has been usually used for the treatment of senile disorders. For enhancing efficacy and convenience, the change of the drug delivery device of oriental herbal medicine is required. Objective: This experiment was designed to investigate the effect of the OJJHEJ hot water extract & ultra-fine powder on proinflammatory cytokine of microglia and memory deficit model. Method: The effects of the OJJHEJ hot water extract on production of IL-1$\beta$, IL-6, TNF-$\alpha$, in BV2 microglial cell line treated by lipopolysacchaide(LPS) were investigated. The effects of the OJJHEJ hot water extract & ultra-fine powder on the behavior of the memory deficit mice induced by scopolamine and AChE in serum of the memory deficit mice induced by scopolamine were investigated. Results: 1. The OJJHEJ hot water extract suppressed the production of IL-1$\beta$, IL-6, TNF-$\alpha$ in BV2 microglial cell line and the production of IL-6 was suppressed significantly. 2. The OJJHEJ hot water extract & ultra-fine powder decreased AChE significantly in the serum of the memory deficit mice induced by scopolamine. 3. The OJJHEJ hot water extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine-induced impairment of memory in the experiment of Morris water maze. Conclusions: This experiment shows that the OJJHEJ hot water extract & ultra-fine powder might be effective for the prevention and treatment of memory impairment diseases. Investigation into the clinical use of the OJJHEJ hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.
