Amniotic Fluid Bisphenol A Level and Its Relationship with Pregnancy Outcomes (임신 중 내분비계 장애물질 Bisphenol A의 양수 내 농도와 임신결과와의 상관관계)

  • Bisphenol A (BPA), a chemical with weak estrogenic activity is reported to affect preimplantation embryos, fetuses and alter their postnatal development. This study amied to determine the relation between the levels of BPA in the amniotic fluid and pregnancy outcomes. ELISA was used to measure amniotic fluid BPA in 120 pregnant women who underwent genetic amniocentesis at 15~20 weeks gestation. The most common indication for amniocentesis was advanced maternal age (35 yrs or older). BPA was detected in all amniotic fluid. The range of amniotic fluid BPA concentrations was from 0.89 ng/mL to 37.13 ng/mL with a mean level of 7.24 ng/mL. We compared the means of amniotic fluid BPA concentrations according to maternal age (${\geq}35$ vs. <35 yrs), fetal sex (male vs. female), gestational age at birth (${\geq}37$ vs. <37 weeks), and infant birth weight (${\geq}2.5$ vs. <2.5 kg). No significant differences were found in these outcomes. This is the first report of amniotic fluid BPA levels in Korean pregnant women. Our findings suggest that BPA may not affect the pregnancy outcomes such as fetal sex, preterm delivery and low birth weight. Whether prenatal exposure to BPA can have teratogenic effect on developing embryo needs to be studied.

Electrochemical Determination of Bisphenol A at Carbon Nanotube-Doped Titania-Nafion Composite Modified Electrode

  • A highly sensitive electrochemical detection method for bisphenol A (BPA) has been developed by using multi-walled carbon nanotube (CNT)-doped titania-Nafion composite modified glassy carbon (GC) electrode. The CNT-titania-Nafion/GC electrode exhibited excellent electrocatalytic activity towards BPA. Therefore, the CNT-titania-Nafion/GC electrode showed improved voltammetric responses for BPA compared to that obtained with bare GC electrode. In addition, cetyltrimethylammonium bromide (CTAB), a cationic surfactant, was added into the BPA sample solution in order to accumulate BPA through hydrophobic interaction between CTAB and BPA. The CNT-titania-Nafion/GC electrode gave a linear response ($r^2$ = 0.999) for BPA from $1.0{\times}10^{-8}$ M to $5.0{\times}10^{-6}$ M with a detection limit of $9.0{\times}10^{-10}$ M (S/N = 3). The modified electrode showed good selectivity against interfering species and also exhibited good reproducibility. The present electrochemical sensor based on the CNT-titania-Nafion/GC electrode was applied to the determination of BPA in food package samples.

Adsorption Behavior of Environmental Hormone Bisphenol A onto Mesoporous Silicon Dioxide

  • Mesoporous silicon dioxide (meso-$SiO_2$) was prepared using cetyltrimethylammonium bromide as the structure-directing reagent and tetraethyl orthosicate as the silicon source. The influence of pH value on the adsorption behavior of bisphenol A (BPA) was investigated. The adsorption capacity of BPA onto meso-$SiO_2$ increases slightly with pH value from 2 to 6, and then gradually decreases as further improving pH value. The effect of temperature was also studied, and the adsorption capacity of BPA gradually declines with increasing temperature. The adsorption kinetics and thermodynamics of BPA were examined. It is found that the adsorption of BPA onto meso-$SiO_2$ is in good agreement with Langmuir adsorption model. The rate constant of adsorption is $5.17{\times}10^{-3}g\;mg^{-1}\;min^{-1}$, and the maximum adsorption capacity is as high as 353.4 $mg\;g^{-1}$ at 20 $^{\circ}C$.

Effects of Bisphenol A on Vitellogenin Synthesis in the Hepatocyte Primary Culture in the Olive Flounder, Paralichthys olivaceus (넙치 Paralichthys olivaceus 초대 배양 간세포의 Vitellogenin합성에 미치는 Bisphenol A의 영향)

  • The effects of bisphenol-A(BPA), a monomer of plastics used in many consumer products, on vitellogenin(VTG) synthesis were examined in primary hepatocyte culture of olive flounder, Paralichthys olivaceus. The hepatocytes were precultured for 2 days, and then estradiol-$17\beta(10^{-6}M)$ and BPA were simultaneously added to the incubation medium. The hepatocytes were cultured for 6 more days and then spent medium was analyzed by SDS-PAGE. BPA increased the rate of VTG to total protein concentrations in a concentration-dependent way, and a significant difference was obtained at concentrations of $10^{-6}M$ and $10^{-5}M$ (P<0.05). In particular, the rate of VTG to total protein concentrations was 26.36% at $10^{-5}M$ of BPA, and its level did not differ from the control level with $E_2$ alone. $E_2$ and/or BPA-primed VTG synthesis was markedly inhibited to about 80% of the control(with $E_2$) by the addition of tamoxifen($10^{-6}M$) to the incubation medium. Furthermore, In vivo $E_2$-primed VTG synthesis was significantly inhibited by in vitro $E_2$-free incubation of hepatocyte to about 22% of the control (with $E_2$) on Day 6. The effect of reducing was delayed in a BPA concentration-dependent way. These results suggest that BPA induce VTG synthesis by estrogenic activity through estrogen receptor mediated response and prolong VTG synthesis on vitellogenesis in olive flounder.

Set, a Putative Oncogene, As a Biomarker for Prenatal Exposure to Bisphenol A

  • Background: Bisphenol A (BPA), an endocrine disrupting chemical, has been suspected to pose carcinogenic risks. However, likely mechanisms are obscure and there are difficulties to estimating its real significance for cancer development. Methods: We therefore studied BPA-induced proteomic alterations in immune organs of ICR mice offspring that were prenatally exposed to BPA (15 and 300 mg/L of drinking water). We performed 2D-gel analyses of samples, considering differences in spleen, exposure levels, sex, and ages. Results: From proteomic analyses, we found various proteins were up- or down-regulated by BPA. Among them, SET, a putative oncogene and inhibitor of phosphatase 2A, was significantly down-regulated in a BPA dose-dependent manner. We also confirmed down-regulation of SET in western blot and real time PCR analyses. From gene network analysis, SET is predicted to communicate with other genes including CYP17, which is involved in biosynthesis and metabolism of sex-hormones. Conclusions: This study provided evidence that SET can be applied as a new biomarker for prenatal BPA exposure and suggests a potential new mechanism of action in that BPA may disrupt CYP17 via SET.

Spectrophotometric Determination of Bisphenol A by Complexation with Ferricyanide and Ferric chloride solution (Ferricyanide와 ferric chloride 혼합액을 사용한 Bisphenol A의 비색 정량법 개발)

  • Bisphenol A (BPA) has been widely used as a monomer for production of epoxy resins and polycarbonate plastics. The annual production of BPA exceeds 640,000 metric tons in worldwide. BPA, a suspected phenolic endocrine disrupter, is moderately soluble and frequently detected in industrial wastewater. To date, HPLC and GC has been used for BPA analysis. However, HPLC and GC-analysis need high operation lost, experts, and an elaborate pre-treatment of samples, and is difficult to apply on-time and mass analysis. Therefore, simple, mass and rapid detection of BPA in environments is necessary. In the present study, spectrophotometric method of BPA quantification was developed. Based on blue-color product formation with BPA and ferric chloride/ferricyanide under the optimized conditions, the standard curve was acquired $({\lambda}_{750}=0.061\;BPA\;[{\mu}M]+0.07155,\;R^2=0.992)$. Using an established method, the BPA contents in the soil extract, and different water samples and living products, including disposable syringe, cup and plastic tube, were analyzed. The results suggested that the method is useful for BPA determination from different massive samples. Since the BPA metabolites, nontoxic 4-hydroxyacetophenone or 4-hydroxybenzaldehyde, did not form blue-color product, this method is also useful to screen a microorganism for BPA bioremediation.

Alternation in the Cell-mediated Immune Reaction by Bisphenol A Exposure in vitro (In vitro에서 Bisphenol A 노출에 의한 세포성 면역반응의 변화)

  • In order to investigate the effects of bisphenol A (BPA) on cell mediated immune reaction in vitro we examined the allogenic mixed lymphocyte reaction (MLR), splenocytes proliferation (SP) to T cell mitogens and IFN-${\gamma}\;production$. Splenocytes of Balb/c mice ($1.5{\times}10^5$ cells/well) were co-cultured with different numbers of mitomycin C-treated mature dentritic cells (DCs) in presence of BPA (25, 50, 100 ${\mu}M$) and $[^{3}H]$thymidine incorporation (cpm) was measured by scintilation counting. Splenocytes ($2{\times}10^6$ cells/well) were cultured with mitogens, Con A ($2\;{\mu}g/ml$), PHA ($5\;{\mu}g/ml$) and IL-2 ($0.1\;{\mu}g/ml$), or PMA ($5\;{\mu}g/ml$) and INO ($1\;{\mu}g/ml$) in presence of BPA (1, 10, 25, 50, 100 ${\mu}M$) and SP was assessed by MTT assay. $IFN-{\gamma}$ levels in culture supernant were determined by ELISA. At low concentration, BPA slightly increased MLR, SP and $IFN-{\gamma}$ levels, but at higher concentration it showed significant inhibitory effects on these immunological parameters. These results indicate that BPA is able to alternate cell-mediated immune reaction.

Effects of Bisphenol A and Nonylphenol on In Vitro Steroid Production in Matured Oocyte of Greenlings, Hexagrammos agrammus (Bisphenol A와 Nonylphenol이 노래미, Hexagrammos agrammus 성숙기 난모세포의 스테로이드 생성과정에 미치는 영향)

  • Endocrine disrupting chemiclas (EDCs) such as bisphenol A (BPA) and nonylphenol (NP) have estrogenic activity and can alter reproduction in fish. In the present study, the effects of BPA and NP on in vitro steroid production from oocytes of maturation stage (oocyte diameter$\fallingdotseq$1.88 mm) from the greenling (Hexagrammos agrammus) were evaluated. Oocytes were incubated with different concentrations of BPA and NP (0.1, 1, 10, 100 and 1,000 ng/$m{\ell}$) in the presence or absence of 50 IU human chorionic gonadotropin (HCG) for 48 hours. After incubation, levels of $17{\alpha},\;20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}OHP$), estradiol-$17{\beta}(E_2)$ and testosterone (T) from incubated media were quantified by radioimmunoassay (RIA). In BPA treatment, 100 ng/$m{\ell}$ of BPA stimulated $E_2$ production regardless HCG supplement. Every concentration of BPA inhibited T production without HCG although 0.1 ng/$m{\ell}$ of BPA stimulated T production with HCG. In NP treatment, 10 ng/$m{\ell}$ of NP stimulated $17{\alpha}20{\beta}OHP$ and T production without HCG. 1 ng/$m{\ell}$ of NP inhibited $E_2$ production. Taken toghther, these results suggest that BPA might have weak estrogen-agonistic effect and NP has estrogenantagonistic effect at final oocyte maturation stage of H. agrammus.

Molecular Modeling of Bisphenol-A Polycarbonate and Tetramethyl Bisphenol-A Polycarbonate

  • To efficiently demonstrate the molecular motion, physical properties, and mechanical properties of polycarbonates, we studied the differences between bisphenol-A polycarbonate(BPA-PC) and tetramethyl bisphenol-A-polycarbonate(TMBPA-PC) using molecular modeling techniques. To investigate the conformations of BPA-PC and TMBPA-PC and the effect of the conformation on mechanical properties, we performed conformational energy calculation, molecular dynamics calculation, and stress-strain curves based on molecular mechanics method. From the result obtained from conformational energy calculations of each segment, the molecular motions of the carbonate and the phenylene group in BPA-PC were seen to be more vigorous and have lower restriction to mobility than those in TMBPA-PC, respectively. In addition, from the results of radial distribution function, velocity autocorrelation function, and power spectrum, BPA-PC appeared to have higher diffusion constant than TMBPA-PC and is easier to have various conformations because of the less severe restrictions in molecular motion. The result of stress-strain calculation for TMBPA-PC seemed to be in accordance with the experimental value of strain-to-failure ∼4%. From these results of conformational energy calculations of segments, molecular dynamics, and mechanical properties, it can be concluded that TMBPA-PC has higher modulus and brittleness than BPA-PC because the former has no efficient relaxation mode against the external deformations.

Bisphenol-A Concentrations from Leiomyoma Patients by LC/MS

  • The aim of this study is to investigate how many leiomyoma patients are exposed to bisphenol-A (BPA), an endocrine disruptor, and whether the serum concentration of BPA is related to leiomyoma growth. Initially, 128 patients were divided into one control and three leiomyoma groups (mild, moderate and severe) according to the size of the leiomyomas. Serum BPA concentrations were measured by liquid chromatography and mass spectrometry (LC/MS). Nearly two-thirds of leiomyoma patients were exposed to BPA and the range of BPA was from non-detection to 2.603 ng/ml. The mean BPA concentrations in the groups were $1.015{\pm}0.775\;ng/ml$ (control), $0.774{\pm}0.834\;ng/ml$ (mild), $1.261{\pm}0.797\;ng/ml$ (moderate) and $1.244{\pm}0.860\;ng/ml$ (severe) (p = 0.158). After recombination into two group, Group 1 (control plus mild) vs. Group 2 (moderate plus severe), higher level was found in Group 2 even with no statistical significance (p = 0.06). In conclusion, about two-thirds of leiomyoma patients were exposed to BPA, but it may not have growth promoting effect on leiomyoma.
