A Study on Measurements of Optical Absorption Coefficients of Biological tissue (생체조직의 광학적 흡수계수 측정에 관한 연구)

  • In this study, we measured the absorption coefficient of the tissues of mouse (brain, heart, liver, muscle and tumor) and human brain (normal and tumor) in the wavelength between 500nm~900nm. The optical coefficient is a representative of the characteristics of the materials. So, we can characterize the biological tissue with the optical coefficients. Using the spectrograph monometer and PDA(Photo Diode Array), we experimented with quick-frozen sectioned specimens. Because the optical coefficient is concerned with the conformation and biochemical component of the biological tissue, we experimented as the wavelength between 500nm~900nm on the normal and tumor samples of the animal and human. For the mouse, there are distinctive differences of the absorption coefficients between normal tissues and tumor. The absorption coefficient of the normal tissue varies 0.1~0.2cm$^{-1}$ with wavelength. But, the absorption coefficient of the brain tumor is changed round about 0.4~0.5cm$^{-1}$ as the wavelength. The absorption coefficients we measured can be a useful implement to detect diseases.

  • A non-invasive technique to measure absorption and scattering coefficients was investigated The reflected backscattered light from the surface of phantom and biological tissue was obtained by using a time-correlated single photon counting system in pico-second time domain. The absorption and scattering coefficients were acquired by the time of peak and asymptotic behavior of the time-resolved reflectance curve and agreed well the ones that is obtained with deconvolution method It was found that the approximation method was good for biological medium to calculate optical properties due to its convenience and accuracy.

  • To predict light propagation in biological tissues irradiated by laser, the optical properties such as absorption and scattering coefficients are required. There have been various techniques for measuring these coefficients. One method requires tissue samples, often a slab of thin tissue, is invasive. On the other hand, non-invasive method usually measures back-scattered light from a subject with no physical intervent ions. Advantages and disvantages of using different methods are investigated. A careful attention should be made in order to select the best method for a given experimental condition since, even either for invasive or non-invasive method, accuracy is subject to governing models and sample preparations.

  • Diffuse optical tomography (DOT) is a relatively new medical imaging modality which uses near infrared light to image large-sized tissues noninvasively. We constructed a frequency-domain DOT system to measure the optical properties of optical phantoms and human tissues. The FD-DOT uses the intensity-modulated infrared light source that illuminates the biological tissues. The phase shift and modulation changes at each detector site are separately processed to measure the optical properties. The absorption and scattering coefficients are separately estimated using inverse algorithms.

Moisture Absorption Properties of Liquid Type Epoxy Encapsulant with Nano-size Silica for Semiconductor Packaging Materials (나노크기 실리카를 사용한 반도체용 액상 에폭시 수지 성형재료의 흡습성질)

  • The moisture absorption properties such as diffusion coefficient and moisture content ratio of liquid type epoxy resin systems with the filler were investigated. Bisphenol A type and Bisphenol F type epoxy resin, Kayahard MCD as hardener and 2-methylimidazole as catalyst were used in these epoxy resin systems. The nano-sized spherical type fused silica as filler were used in order to study the moisture absorption properties of these liquid type epoxy encapsulant according to the change of filler size. The temperature of glass transition (Tg) of these epoxy resin systems was measured using Dynamic Scanning Calorimeter (DSC), and the moisture absorption properties of these epoxy resin systems according to the change of time were observed at $85^{\circ}C$ and 85% relative humidity condition using a thermo-hygrostat. The diffusion coefficients in these systems were calculated in terms of modified Crank equation based on Ficks' law. An increase of Tg and diffusion coefficient with filler size in these systems can be observed, which are attributed to the increase of free volume with Tg. The change of maximum moisture absorption ratio according to the filler size and filler content cannot be observed; however, the diffusion coefficients of these systems decreased with filler content. The diffusion via free volume is dominant in the epoxy resin systems with low nano-sized filler content; however, the diffusion with the interaction of absorption according the increase of the filler surface area is dominant in the liquid type epoxy encapsulant with high nano-sized filler content.

Biopharmaceutical studies on copper(II) chelates of sulfanilamide derivatives (Sulfanilamide유도체의 동(II)착화합물에 대한 생물약제학적 연구)

  • Cu(II) chelates of several sulfanilamide derivatives (Sulfa-Cu) were prepared and their effects on solubility, absorptivity in intestinal lumen, biding tendency with serum protein and erythrocytes, concentration in rabbit blood, and acetylation rate were studied in comparison with their free ligand forms. For solubility concerned, the partition coefficients of Sulfa-Cu are decreased as following order: Sulfadimethoxine Copper chelate (SDM-Cu), Sulfamethoxypyridazine Copper chelate (SD-Cu), Sulfamerazine Copper chelate (SM-Cu), Sulfaisoxazole Copper chelate (SIX-Cu). The partition coefficients of SDM-Cu and ST-Cu were much greater than those of ligands. this phenomenone acounts for the rapid absorption of SDM-Cu and ST-Cu in the rat small intestine (in situ). The Sulfa-Cu were absorbed at the intestinal lumen of a rat in the rate of first order and there was no difference between long acting sulfa drugs and their Cu0chelates in biological half lives. In binding experiments, sulfa-Cu binded with serum protein in lower ratio than their ligands except SIX-Cu. On other hand, acetylation rates of sulfa-Cu were higher than those of free sulfa drugs and the acetylation rate were higher than those of free sulfa drugs and powder. In a experiment on Sulfa-Cu concentration in rabbit blood, the half lives of SD-Cu, SIX-cu, ST-Cu, and SM-Cu were longer than those of their ligands. Above all, the half life of SD-Cu appeared to be approximately 3.5 times logner than that of corresponding ligand, SD. When absorption of sulfa drugs or sulfa-Cu at the small intestinal lumen of a rat and the concentration in rabbit blood after absorption were compared, it was found that there was not always conrrelated.

Measurement of optical coefficients of multiple scattering media by using frequency domain spectroscopy (주파수 영역 분광법을 이용한 다중산란 매질의 광학계수 측정)

  • A frequency domain spectroscopic system was constructed to investigate the optical properties of multiple scattering media. The alternating current (AC) and phase lag components of backscattered light were measured by using the heterodyne detection method. Absorption and transport scattering coefficients were computed from the values based on diffusion theory. Predictions showed excellent matches in comparison with actual values of absorption and scattering. Predictable ranges of the optical coefficients were analyzed in terms of the distance between light source and detector, and modulation frequencies. A proposed compact experimental set-up using laser diodes can be utilized to estimate non-invasively the optical properties of multiple scattering media such as biological tissues.

  • The study of $CO_2$ absorption in a packed column by 1,1,3,3-tetramethylguanidine (TMG) dissolved in ethylene glycol is presented. Absorption column of inner diameter 1 in and 0.6 m length was filled with Protruded-packing $0.16in{\times}0.16in$. We investigated the effect of operating conditions on overall mass transfer coefficients as well as on $CO_2$ removal efficiency. The loading values reached at about $1.0mol_{CO2}/mol_{TMG}$. In case of absorbent with lean $CO_2$ loading, the overall mass transfer coefficient was proportional to the concentration of TMG. However, in the range of more than ${\alpha}=0.5molCO_2/molTMG$, the overall mass transfer coefficients decreased with the concentration of TMG. It is due to the increasing of mass transfer resistance in liquid phase as increasing of viscosity at higher loading values.

Synthesis of a Novel Near-Infrared Fluorescent Dye: Applications for Fluorescence Imaging in Living Cells and Animals

  • Fluorescence imaging is considered as one of the most powerful techniques for monitoring biomolecule activities in living systems. Near-infrared (NIR) light is advantageous for minimum photodamage, deep tissue penetration, and minimum background autofluorescence interference. Herein, we have developed a new NIR fluorescent dye, namely, RB-1, based on the Rhodamine B scaffold. RB-1 exhibits excellent photophysical properties including large absorption extinction coefficients, high fluorescence quantum yields, and high photostability. In particular, RB-1 displays both absorption and emission in the NIR region of the "biological window" (650-900 nm) for imaging in biological samples. RB-1 shows absorption maximum at 614 nm (500-725 nm) and emission maximum at 712 nm (650-825 nm) in ethanol, which is superior to those of traditional rhodamine B in the selected spectral region. Furthermore, applications of RB-1 for fluorescence imaging in living cells and small animals were investigated using confocal fluorescence microscopy and in vivo imaging system with a high signal-to-noise ratio (SNR = 10.1).

  • A method for measuring optical properties of biological medium was investigated by using the frequency domain spectroscopy. When amplitude-modulated light with the frequency of several tens up to several hundred MHz propagates through a scattering medium, the phase lag and the amplitude reduction occur. The phase lag depends on the average of optical path lengths. The amplitude reduction with respect to the radial distance is influenced by the penetration depth. The mean of optical path length and penetration depth are related to optical coefficients. The phase lag and the amplitude reduction were measured based on the heterodyne detection method. The experimental data were fitted with the theoretical curves derived from diffusion theory and the absorption and scattering coefficients were calculated.
