Coenzyme $Q_{10}$ Production by Sphingomonas sp. ZUTE03 with Novel Precursors Isolated from Tobacco Waste in a Two-Phase Conversion System

  • Coenzyme $Q_{10}$ ($CoQ_{10}$) is a widely used supplement in heart diseases treatment or antioxidative dietary. The microbial production of $CoQ_{10}$ was enhanced by addition of solanesol and novel precursors recovered from waste tobacco. The novel precursors were separated by silica gel and identified as ${\alpha}$-linolenic acid (LNA) and butylated hydroxytoluene (BHT) based on the effect on $CoQ_{10}$ production and GC-MS. The effects of novel precursors on $CoQ_{10}$ production by Sphingomonas sp. ZUTE03 were further evaluated in a two-phase conversion system. The precursor's combination of solanesol (70 mg/l) with BHT (30 mg/l) showed the best effect on the improvement of $CoQ_{10}$ yield. A maximal $CoQ_{10}$ productivity (9.5 mg $l^{-1}$ $h^{-1}$) was achieved after 8 h conversion, with a molar conversion rate of 92.6% and 92.4% on BHT and solanesol, respectively. The novel precursors, BHT and LNA in crude extracts from waste tobacco leaves, might become potential candidates for application in the industrial production of $CoQ_{10}$ by microbes.

Effect of Incorporation of Pomegranate Peel and Bagasse Powder and Their Extracts on Quality Characteristics of Chicken Meat Patties

  • This study was conducted to develop chicken meat patties by incorporating pomegranate peel and bagasse powders and their extracts. Patties were developed by incorporating pomegranate peel powder (PPP, 2 g), pomegranate aril bagasse powder (PABP, 4 g), pomegranate peel powder aqueous extract (PPAE, 6 g) and pomegranate aril bagasse powder aqueous extract (PABAE, 9 g) individually per 100 g of minced meat. Both types of powders and extracts treated patties had significantly higher total phenolic content than control and butylated hydroxytoluene (BHT) treated patties. Both types of powder (PPP and PABP) treated patties had significantly higher water holding capacity, ash, crude fibre content, and hardness values, and significantly lower moisture content and lightness values in comparison to control patties. Emulsion stability and cooking yield of PABP treated patties were significantly higher than control. Addition of extracts and BHT did not influence the physico-chemical properties and proximate composition of chicken patties. Both types of powders and extracts provided better protection to chicken meat patties against oxidative rancidity and microbial proliferation in comparison to control and BHT treated patties during refrigerated storage. It is concluded that pomegranate fruit byproducts in the form of peel powder, aril bagasse powder and their extracts can be successfully utilised in development of healthier chicken meat patties and these byproducts can also be effectively used as a replacement of synthetic antioxidants such as BHT.

Efficiency of Antioxidants on Types of Substrate Systems (기질 system의 유형에 따른 항산화제의 효과에 관한 연구)

  • The purpose of this study was to investigate any differences in the efficiency of various antioxidants for the three types of substrates such as corn oil in water (O/W) emulsion, water in com oil (W/O) emulsion, and bulky corn oil. ${\alpha}$-Tocopherol (${\alpha}$-Toc) at 0.01 or 0.02%, ascorbic acid (AsA), ascorbyl palmitate (AP), and BHT at 0.02% were added separately to the prepared O/W emulsion, W/O emulsion, and bulk oil, and their antioxidative effects were compared. The mixture of ${\alpha}$-Toc ind AsA or AP at the level of 0.02% also was tested to observe any synergistic effect. Oxidation was made by storing at 42${\pm}$1$^{\circ}C$ for 25 days and the oxidative stability was determined by peroxide value and conjugated dienoic acid with time fluctuation of storage. The results were as follows: 1. In case of O/W emulsion, the order of antioxidative effect was AP> ${\alpha}$-Toc+AP>${\alpha}$-Toc+AsA>AsA>BHT. 2. In case of W/O emulsion, the order of antioxidative effect was AsA>AP>${\alpha}$-Toc+AsA>BHT. ${\alpha}$-Toc+AP mixture showed the prooxidant effect rather than synergistic effect. 3. In case of bulk oil, the order of antioxidative effect was AsA>AP>${\alpha}$-Toc+AsA>${\alpha}$-Toc+AP\ulcornerBHT. Therefore, AsA, a hydrophilic antioxidant, was more effective in W/O emulsion system than in O/W emulsion system, while the opposite trend was found in AP, a lipophilic antioxidant. AsA, a hydrophilic antioxidant, was more efficient in bulk oil of anhydrous substrate. ${\alpha}$-Toc showed prooxidant effects in all substrates.

Effects of Antioxidants and Packaging Methods on the repression of Lipid Oxidation in Plain Dried Large Anchovy (소건 대멸치 지질의 산화억제에 미치는 항산화제와 포장의 병행효과)

  • The antioxidants and packaging methods affecting on lipid oxidation of the plain dried large anchovy were investigated. Antioxidants resulted in decrease of lipid oxidation of dried large anchovy. The addition of BHT remarkably repressed lipid oxidation during storage of the dried large anchovy at $37^{\circ}C$, followed by ${\alpha}-tocopherol$, rosemary extract and sage extract. The addition of rosemary extract mixed with 0.1{\%} {\alpha}-tocopherol$ showed similar effect to BHT during storaBe at $25^{\circ}C\;in\;0.1{\%}$ rosemary extract mixed with $0.1{\%}\;{\alpha}-tocopherol$. When the dried anchovy were packed in polyethylene film after treatment with antioxidants, the vacuum package increased the antioxidative effect in repression of lipid oxidation during storage of dried large anchovy.

  • Antioxidative activities of ethanol extracts of fruits from three cultivars of Prunus mune with different harvest times were determined. Extracts of P. mune cutivar Okyoung showed the strongest activity at 97.5%(compared to BHT) using DPPH or thiocyanate method, although no statistically significant differences were observed among the cultivars. Antioxidative activities of the extracts measured with glutathione-S-transferase method was over 70% of that of BHT. Survival rates of cells in mouse spinal cord culture challenged by superoxide anion were 89, 79, and 62% at 40 mg/ml of Okyoung, Namgo, and Kochunme extracts, respectively, whereas 90% at 6mM glutathion.

  • Recently, there is a growing interest about unsaturated lecithin having excellent characteristics of skin affinity and absorbency. Accordingly, this study intended to develop a natural sulfuration material in order to enhance the stability of oxidation of unsaturated lecithin and substitute existing sulfuration materials which indicate variability and toxicity. As sulfuration components, plenol acid family, 3,5-dihydroxybenzoic acid, and flavanone were analyzed. Total polyphenol content was higher in the root extracts (133.85 mg/g) than in the fruit extracts (53.5 mg/g). Above 100 ppm polyphenol content, the free radical removal efficiency and lipid oxidation prevention of the root extracts were 20.1 and 19.2% superior compared with BHT respectively. Also, the extracts indicated high survival rate of more than 95% below 1250 ppm, showing the stability. For the stability of liposome made from an unsaturated lecithin, the root extracts were superior to the fruit extracts. Especially, 15.1 and 13.9% of sulfuration effect and zeta potential were improved with 9.3% reduced particle size compared with BHT as the control group, respectively.

Studies on the Antioxidant Effect of Korean Propolis (국산 프로폴리스의 합산화 효과에 관한 연구)

  • The studies was conducted to investigate the antioxidation effect of Korean propolis. Oxidation period of soy oil was 17.5 hr, soy oil with propolis(1,000 ppm) was extended the length of oxidation period. The length of oxidation periods of soy oil with Yecheon, Youngwol, Brazilian, Chinese, and Australian propolis were 33.5, 35.4, 32.0, 33.1, and 27.1hours respectively. The length of oxidation period of lard with propolis 1,000ppm of Yecheon, Youngwol, Brazilian, Chinese and Australian were 191, 167, 296, 230, and 207hours, respectively compared to control(22.5hours). When compared to tocopherol as natural antioxidant and BHT as synthetic antioxidant were revealed 16.7 and 20.5hours, respectively. The length of oxidation period was not different when compared to tocopherol and Youngwol propolis with soy oil. Ascorbic acid and citric acid had synergistic effect to propolis with soy oil when 200ppm of propolis was added.

Antimicrobial and Antioxidative Activities of Opuntia ficus-indica var. Saboten Extracts (백년초 추출물의 항균 및 항산화성)

  • The antimicrobial and antioxidative effects of Opuntia ficus-indica vur. saboten (OP) extract with different solvents and extraction methods were investigated. In general, antimicrobial activity of OP was higher in extracts by reflux than in extracts by shake. However, no significant difference in antimicrobial activity was observed between methanol, ethanol and water extract. The antimicrobial activity was significant against all microbes tested, especially against Pseudomonas aeruginosa by shake extract and against Escherichia coli O-157 by reflux extract. A severe morphological change with destruction of outer layer was observed in Salmonella enteritidis treated with water extract. Though OP extract in hydrogen activity was lower than 0.1% BHT, the activity of OP extract showed very high effectivenss than the activity of control. OP extract showed a similar extent of antioxidative activity with BHT.

  • A procedure based on liquid liquid extraction(LLE)-GC/MS has been developed for the simultaneous analysis of antioxidants and insecticides known as endocrine disrupters. The endocrine disrupters investigated in this study are butylated hydroxy anisole(BHA) and butylated hydroxy toluene(BHT), and the insecticides are allethrin and fenvalerate. The limit of detection(LOD) was 0.071~0.159 ng/ml with RSD of 1.41~5.34% for the standard sample. From water reservior of Han river, Geum river, Nakdong river and Sumjin river, these compounds were not detected. For the synthesized sample, the LOD is 0.051~0.132 ng/ml with RSD of 6.39~13.4%.

Antioxidative Properties of Phenolic Compounds Extracted from Black Rice (흑미 색소물질에 함유된 페놀화합물의 항산화 특성)

  • The composition and antioxidative effects of phenolic compounds in black rice were studied. The contents of free and bound phenolic compounds extracted from black rice were 845.4 and 401.6 mg respectively per 100g sample weight. Free phenolic compounds had higher antioxidation ability than those of bound phenolic compounds. Solvent fractionation of free phenolic compounds revealed that butanol fraction had the highest phenolic compounds contents and antioxidative activity among other solvent fractions. Although butanol fraction showed lower lipid peroxidation inhibition (LPI) ability than that of $\alpha$-tocopherol and BHT, free radical scavenging ability was much higher than that of $\alpha$-tocopherol and BHT, as evidenced by electron donating ability (EDA) and benzoic acid hydroxylation inhibition (BAHI) assays.
