Cryptotanshinone promotes brown fat activity by AMPK activation to inhibit obesity

  • BACKGROUND/OBJECTIVES: Activating brown adipose tissue (BAT) and browning of white adipose tissue (WAT) can protect against obesity and obesity-related metabolic conditions. Cryptotanshinone (CT) regulates lipid metabolism and significantly ameliorates insulin resistance. Adenosine-5'-monophosphate (AMP)-activated protein kinase (AMPK), a receptor for cellular energy metabolism, is believed to regulate brown fat activity in humans. MATERIALS/METHODS: The in vivo study included high-fat-fed obese mice administered orally 200/400 mg/kg/d CT. They were evaluated through weight measurement, the intraperitoneal glucose tolerance test (IPGTT), intraperitoneal insulin tolerance test (IPITT), cold stimulation test, serum lipid (total cholesterol, triglycerides, and low-density lipoprotein) measurement, hematoxylin and eosin staining, and immunohistochemistry. Furthermore, the in vitro study investigated primary adipose mesenchymal stem cells (MSCs) with incubation of CT and AMPK agonists (acadesine)/inhibitor (Compound C). Cells were evaluated using Oil Red O staining, Alizarin red staining, flow cytometry, and immunofluorescence staining to identify and observe the osteogenic versus adipogenic differentiation. Quantitative real-time polymerase chain reaction and the Western blot were used to observe related gene expression. RESULTS: In the diet-induced obesity mouse model mice CT suppressed body weight, food intake, glucose levels in the IPGTT and IPTT, serum lipids, the volume of adipose tissue, and increased thermogenesis, uncoupling protein 1, and the AMPK pathway expression. In the in vitro study, CT prevented the formation of lipid droplets from MSCs while activating brown genes and the AMPK pathway. AMPK activator enhanced CT's effects, while the AMPK inhibitor reversed the effects of CT. CONCLUSION: CT promotes adipose tissue browning to increase body thermogenesis and reduce obesity by activating the AMPK pathway. This study provides an experimental foundation for the use of CT in obesity treatment.

Taxonomical Classification of Bugog Series (부곡통의 분류)

  • This study was conducted to reclassify Bugog series based on the second edition of Soil Taxonomy : A Basic System of Soil Classification for Making and Interpreting Soil Surveys. Morphological properties of typifying pedon of Bugog series were investigated and physico-chemical properties were analyzed according to Soil Survey Laboratory Methods Manual. The typifying pedon of Bugog series has strong brown (7.5YR 4/6) loam Ap horizon (0~22 cm), brown (7.5YR 4/4) clay loam BAt horizon (22~41 cm), strong brown (7.5YR 4/6) silty clay loam Bt1 horizon (41~59 cm), strong brown (7.5YR 4/6) silty clay loam Bt2 horizon (59~78 cm), brown (7.5YR 4/4) silty clay loam Btx1 horizon(78~90 cm), and brown (7.5YR 4/4) Btx2 horizon(90~160 cm). That occurs on swale foot slope in area of mainly granite gneiss, granite, and schist rock materials. The typifying pedon has an argillic horizon from a depth of 22 to more than 160 cm and a base saturation (sum of cations) of less than 35% at 75 cm below the upper boundary of the fragipan. That can be classified as Ultisol, not as Alfisol. The pedon has udic soil moisture regime, and can be classified as Udult. That has a fragipan with an upper boundary within 100 cm of the mineral soil surface, and keys out as Fragiudult. Also that meets the requirements of Typic Fragiudult. That has 18% to 35% clay at the particle-size control section, and has mesic soil temperature regime. Bugog series can be classified as fine silty, mixed, mesic family of Typic Fragiudults, not as fine loamy, mixed, mesic family of Typic Fragiudalfs.

The Effects of Sex Education on the Primary Schooler's Knowledge and Attitude about Sex (성교육이 국민학생의 성지식과 성태도에 미치는 효과)

  • The confusion of the sense of value on sex is increasing because of the rapid change in social-cultural environment. Also due to a rise in the standard of living, the age of adolescence is getting lower, and so the second sexual marks appear to primary schoolers. At this time in the aspect of the education for the whole man, it's very important for primary schoolers to acquire right knowledge and desirable attitude on sex so that can overcome psychological instability caused by physical growth, be responsible for their behaviors and lead happy lives. This study was made to find out the effects of sex education for the primary schoolers' knowledge and attitude about sex. The study was designed as simulated control group pretest-posttest design, which only pretest was practiced to control group and after sex education only posttest was practiced to experimental group. The data was collected for 18 days from March 13, 1995 to March 31, 1995. The subjects of this study were 130 six-graders in a private primary school in seoul. The control group and experimental group were composed of 65 pupils, each. Sex education consisted of lecture and discussion and the materials were this researcher's own made, 'How do I grow? and related OHP film and video. This education was practiced 40 minutes at a time, at intervals of 2-4 days' six times during 3weeks. For sex knowledge tools, 20 item questionaries on the base of related reference books and contents of this study were used. For sex attitude tools, this researcher's own made 13 item questionaries were used which were revised and complemented and laying stress on the reference books. The data was analyzed through pc-SAS program. The homogeneity test in terms of the general characteristics of experimental and control group was analyzed through $x^2$-test and t-test. And the difference in the primary schoolers' score on knowledge and attitude about sex before and after the sex education was analyzed through t-test and ANCOVA. The results of this study are as follows : 1. The result of the homogeneity test in terms of the general characteristics of the experimental and control group showed that there were significant differences in economic standard($x^2$=2.92, P=0.052) and brotherly ties($x^2$=3.78, P=0.052). 2. Hypothesis 'After sex education, the score of primary schoolers' sex knowledge will be higher than before. 'showed statistically significant difference.(t=11.99, P=.0001) Sex education was practiced under control over subjective view of economic standard and brotherly ties which showed significant difference at the homogeneity test in terms of the general characteristics between the two groups. After this education, the results of comparing the score of primary schoolers' sex knowledge also showed significant difference between the two groups. So hypothesis I was supported since primary schoolers to have education showed noticeable results in the sex knowledge score. (F=16.52, P=.0001) 3. Hypothesis 'After sex education, primary schooler's sex attitude score will be higher than before' showed statistically significant(t=5.08, P=.0001) Sex education was practiced, too, under control over subjective view of economic standard and brotherly ties which showed significant difference at the homogeneity test in terms of the general characteristics between the two groups. Bat this time the results of comparing the primary schoolers' sex attitude showed no significant difference between the two groups. So hypothesis II was rejected since for primary schoolers to have sex education showed no noticeable results in sex attitude score. (F=3.52, P=.0628) These results of the study show that sex education gives affirmative change to the primary schooler's sex knowledge, bat short periods' sex education doesn't give any change to the sex attitude. For the establishment of the pupils' desirable sex attitude, systematic and concrete sex education fit for the pupils' developing stage should be practiced over a long period of time.

The Thermal Preference and the Selection of Hibernacula in Seven Cave-dwelling Bats (동굴성 박쥐 7종의 온도선호도와 동면처 선택)

  • Bats hibernating in caves and unused mines were surveyed during six hibernation periods (from December to February, 2005 and 2011) in Korea. We recorded 13,288 individuals of 11 species at 140 hibernacula (60 caves and 80 abandoned mines): Rhinolophus ferrumequinum (n=3,509), Eptesicus serotinus (n=6), Hypsugo alaschanicus (n=349), Miniopterus fuliginosus (n=5,919), Murina hilgendorfi (n=417), Myotis aurascens (n=294), Myotis bombinus (n=2), Myotis formosus (n=401), Myotis macrodactylus (M.mac, n=151), Myotis petax (n=2,131) and Plecotus ognevi (n=109). We studied the thermal preference and selection of hibernacula of seven dominant bat species. Four species (Myotis petax, Hypsugo alaschanicus, Plecotus ognevi and Murina hilgendorfi) hibernated mainly at the cold site below than $7^{\circ}C$, while three species (Myotis formosus, Rhinolophus ferrumequinum and Miniopterus fuliginosus) hibernated at warm site above than $7^{\circ}C$. Rhinolophus ferrumequinum had broad-ranged temperature zone for their hibernating site. The mean body temperature of each species was $2.64{\pm}0.98^{\circ}C$ for Murina hilgendorfi, $2.76{\pm}1.68^{\circ}C$ for Hypsugo alaschanicus, $2.78{\pm}0.98^{\circ}C$ for Plecotus ognevi, $4.52{\pm}1.02^{\circ}C$ for Myotis petax, $7.83{\pm}1.94^{\circ}C$ for Miniopterus fuliginosus, $9.19{\pm}2.35^{\circ}C$ for Rhinolophus ferrumequinum and $13.64{\pm}0.76^{\circ}C$ for Myotis formosus, respectively. The body temperatures of hibernating bats were closely related to the rock surface temperatures rather than the ambient temperatures. In conclusion, the diversity of bats community in hibernacula were closely related to the range of inner ambient temperature of hibernacula, and more species of bats were occupied at sites presenting a broad range of ambient temperatures.

Taxonomical Classification of Jangho Series (장호통의 분류)

  • This study was conducted to reclassify Jangho series based on the second edition of Soil Taxonomy : A Basic System of Soil Classification for Making and Interpreting Soil Surveys. Morphological properties of typifying pedon of Jangho series were investigated and physico-chemical properties were analyzed according to Soil Survey Laboratory Methods Manual. The typifying pedon of Jangho series has very dark grayish brown (10YR 3/2) gravelly silt loam A horizon (0~14 cm), very dark brown (7.5YR 3/2) silty clay BAt horizon (14~31 cm), dark brown (7.5YR 3/4) silty clay Bt1 horizon (31~58 cm), brown (7.5YR 4/4) silty clay Bt2 horizon (58~90 cm), and brown (7.5YR 4/4) clay Bt3 (90~120 cm) horizon. That is developed on river terraces. The typifying pedon has an argillic horizon from a depth of 14 to more than 120 cm and a base saturation (sum of cations) of less than 35% at 125 cm below the upper boundary of the argillic horizon. That can be classified as Ultisol. The typifying pedon has 0.9 % or more organic carbon in the upper 15 cm of the argillic horizon, and can be classified as Humult, which is reported for the first time in Korea. That does not have fragipan, kandic horizon, sombric horizon, plinthite, etc. in the given depths, and keys out as Haplohumult. Also that meets the requirements of Typic Haplohumult. That has 35 % or more clay at the particle-size control section, and has mesic soil temperature regime. Jangho series can be classified as fine, mixed, mesic family of Typic Haplohumults, not as fine silty over clayey, mixed, mesic family of Mollic Hapludalfs.

Home-range Analysis of Pipistrelle Bat (Pipistrellus abramus) in Non-Reproductive Season by Using Radio-tracking (원격무선추적을 이용한 집박쥐의 비번식기 행동권 분석)

  • This study was conducted to analyze the home range size of Pipistrellus abramus in non-reproductive season. The survey was conducted in the day-roosting site of Pipistrellus abramus located in Gyeongju City(North Gyeongsang Province). We radio-tracked six Pipistrellus abramus(male 3, female 3) and LTM single stage radio transmitter(0.38g), R2000 ATS receiver, three element yagi antenna, roof mounted antenna and ArcGIS 3.3(ESRI, Animal Movement Extension 2.0) were used to locate bats and home range analyze. The home range sizes of the 6 radio-tracked Pipistrellus abramus ranged from 8.97 to 19.07ha(Maen $14.46{\pm}3.44ha$). Mean home range size of female($16.83{\pm}1.96ha$) was larger than that of male($12.08{\pm}2.96ha$) but there were no significant differences in home range sizes between male and female(t=2.32, p>0.05). Also, mean maximum distances from the roost was $468.73{\pm}94.38m$ but there were no significant differences between sexes(female, $422.73{\pm}10.38m$; male, $514.74{\pm}125.74m$; t=-1.26, p>0.05).

A Study on Integrated Approaching Factors of Environmentally-Friendly Companies Certification Scheme and Environmental Impact Assessment of Korea (환경친화기업지정제도와 환경영향평가제도에서의 통합적 환경관리 요소에 대한 기초적 연구)

  • A worldwide trend of permitting system for industrial installation to achieve a high level of protection of the environment has been moved from single media to multimedia in approach. The Council of the European Community issued the Directive 96/61/EC, the IPPC Directive, concerning integrated pollution prevention and control in 1996. The IPPC Directive is one of the most ambitious legal measures that the European Union (EU) has initiated with a view to applying the prevention principle for industrial activities. The IPPC aims to achieve the integrated prevention and reduction of environmental pollution emitted by those industrial installations with a higher potential of emissions to the environment. Organization for Economic Cooperation and Development (OECD) recommended on Environmental Performance Reviews of Korea in 2006 that IPPC permitting concept should be considered for large stationary sources at the national and regional levels. Any Korean law doesn't provide for integrated pollution control with a single process covering all pollution from economic activities. However, one exception might be the "environmentally-friendly companies" certification scheme, introduced in 1995, in which participants agree to meet targets beyond the legal emission limit values in exchange for government technical and financial support to operate environmental management systems. The other exception might be Environmental impact assessment (EIA) of projects, in 1977, which has been strengthened and reinforced to be more preventive through development of the prior environmental review system (PERS) in 1999. The aim of this work is to introduce the contents of IPPC Directive at the viewpoint of Korea policy and to survey the integrated approaching concept of Environmentally-Friendly Companies (EFC) Certification Scheme and EIA policy of Korea. The study will be helpful in the future to prepare the infrastructure of integrated permitting system and to enforce the integrated permit which the authorities of local government issues on industrial activities. It can be said that the data calculated through both EFC Certification Scheme and EIA will be discussed as worthful information to determine Korean BAT reference notes for integrated permitting process.

The Study on Ancient Embroidered Norigae and Modern Embroidered Norigae. -Focus on the Current Embroiered Norigae in 1990- (유물 수노리개와 현대 수노리개의 비교 연구 (1990년 시중 수노리개를 중심으로))

  • Woman in Choson era had various kind of accdssories like rings, ear ring, ornamentak hairpins and so on. Among them their favorite was the Norigae, the pendent trinket. With the embroidered ornament at its center, beads and knots were attached to the upper and lower part, and it was hung with a tassel. It was carried on the string of the Chogori. norigae oreginated in waist string of Three Countries era(Sills, Paekje and Koguryo) ; had been changed with the development of fashion, embroidery and the influence of Buddhistic culture. The differences between Embroidered Norigae and Norigae are following. If the ornament at its center is made of such jewels as gold, silver, jade and the like, it is called Norigae. But if the ornament at its center is made of the embroidered Norigae. Such a Embroidered Norigae in which perfume was contained, was as a perfume box or a perfume bag and was carried on the strings of Chogori by Danjak(single work). The Differences between ancient Embroidered Norigae of late Choson era and modern Embroidered Norigae are Following; 1) The type of main body-the embroidered ornament at its center was mainly embroidered with the shape of animals in both of ancient Embroidered Norigae was usually formes like a bat and that of modern Embroidered Norigae like butterfly. 2) plants were adapted as the main design of ancient Embroidered Norigae, but plants and animals as that of modern Embroidered Norigae. 3) The colors of the background and the design are usually red in both ancient and modern Embroidered norigae. 4) The tassels of ancient Embroidered Norigae were mode in the shape of srick and a strawberry. but the tassels of modern Embroidered Norigae were mainly made in the shape of a stick. 5) Nowadays the beauty of external form is considered more important while the symbol of main body was considered more important in Choson era. 6) The role of Thidon in the composition of Embroidered Norigae is declined and a lot of main bodies are used in modern Embroidered Norigae. 7) Modern Korean people are apt to choose Embroidered Norigae depending upon the design of main body and the traditional Korean beauty. 8) Modern people who desire to get new thing, try to make the design of main body various. Analysis of Liquiritigenin, an Aglycone of Liquiritin in Licorice by High Performance Liquid Chromatography (감초 중 리퀴리티게닌 분석법 개발 및 함량분석)

  • Licorice(Glycyrrhizae Radix et Rhizoma) is recorded as the root of Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne or Glycyrrhiza inflata Bat.(Leguminosae) in Korean Pharmacopoeia $9^{th}$ edition (KP9) and Chinese Pharmacopoeia 2005(CP2005), Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne in Japanese Pharmacopoeia 2005(JP2005). It is established the content standard of Glycyrrhizin 2.5 % and liquiritin 1% in KP9 and CP2005. But, according to the reports, all Licorice species were not sufficient for content standard of liquiritin 1.0% for licorice in KP9 and CP2005. It shows different content of liquiritin among G. uralensis, G. glabra and G. inflata. Also, it was reported liquiritin, liquiritin apioside are transformed into liquiritigenin by human internal flora. Therefore, we have studied for the pre-treatment condition and analytical method of liquiritigenin; It was good efficinet in 2M HCl reflux(1 hr) for hydrolysis and in methylene chloride for solvent fractionation. And 1% acetic acid in DW(A) and acetonitrile(B) with gradient condition as a mobile phase was most effective in HPLC analytical condition. According to these experimental methods, we have anlayzed content of liquiritigenin about 77 Licorice sample. In this research, it was also examined the content of liquiritin and liquiritigenin for Glycyrrhizae Radix related growing area. According to the results, we suggested the content standard of glycyrrhizin more than 2.5%, liquiritigenin more than 0.7%(after hydrolysis) of licorice.

Analysis on the Textile and Dye Used for the Book Cover and Slipcase Housed in the Oryundae Korean Martyrs Museum (오륜대 한국순교자박물관 소장 필첩 및 첩갑에 사용된 직물 및 염료분석)

  • The purpose of this study is to analyze on the textile used for book covers of "Gukgiboksiksoseon" and "Boepboksajeolboksaek", and slipcase of these books kept in the Oryundae Korean Martyrs Museum in Busan. These records are estimated to be written by Gyeongbin Kim(1831-1907), who was a royal concubine of 24th King Heonjong (reign 1834~1849) of the Joseon Dynasty. The cover textile of slipcase and two books are investigated to be silks by the FT-IR. The cover textile of slipcase is flower patterned satin with silver thread and the cover textile of two books are green and red with Su characters and bat patterned satin. The blackish part of pattern of slipcase is investigated by silver thread by FE-SEM-EDAX. Moreover, by the dye analysis, berberine, brazilin, and carthamin are detected from the cover textile of "Boepboksajeolboksaek". It is indicated that it was dyed with an amur cork-tree, a sappanwood, and a safflower. And rutin which is the main dyestuff of the sophora flower of the pagoda tree was detected from the yellow thread of the cover textile of slipcase.
