AN EXPERIMENTAL STUDY ON THE LOW-DOSE RADIOSENSITIVITY OF TUMOR CELL LINES (수종의 암세포주의 저선량 방사선감수성에 관한 실험적 연구)

  • The purpose of this study was to aid in the radiation therapy of head and neck cancer patients. For this study, radiation survival curves were generated for Bl6, MG-63 and YAC-l cell lines using semiautomated MTT assay and Dye Exclusion Assay. Irradiation of 2, 4, 6, 8, 10Gy were delivered at room temperature at a dose rate of 210.2cGy/min using / sup 60/Co γ-ray Irradiator ALOORAOO 8. The viable cells were determined for each radiation dose and compared to control values. The obtained results were as follows: 1. There was significantly different absorbance at 10Gy on B16 cell line in MTT assay(P<0.05). 2. There was significantly different absorbance at 4, 6, 8, 10Gy on MG-63 cell line in MTT assay(P<0.05). 3. YAC-l cell line was more sensitive than B16 or MG-63 cell line to all doses of radiation(P<0.05). 4. There was significantly different absorbance among all tumor cell lines except between B16 and MG-63 cell line at ZGy in MTT assay(P<0.05). 5. Good correlation was obtained between MTT assay and DEA(P<0.05). The efficient of correlation of B16, MG-63 and YAC-l cell line was 0.845, 0.824 and 0.906, respectively.

  • In order to investigate the antitumor effect by Chungsangbohahwan after B-16 cells were transplanted in C57BL/6 mice, and the immune responses in mice induced by methotrexate, the extract of Chungsangbohahwan was orally administered to the mice for 21 days. Experimental studies were performed for measurance of metastasis, cell cytotoxicity in vitro, natural killer cell activity, productivity of interleukin-2. The results were summarized as follows: 1. Inhibition of metastasis in Chungsangbohahwan-treated group was higher than control group with significance on 7th day and 14th day. 2. On the MTT assay, cell viability was significantly inhibited by $5{\mu}g/well$, $2.5{\mu}g/well$, $1.25{\mu}g/well$, and $0.625{\mu}g/well$ of Chungsangbohahwan concentration inhibited cell viability significantly(P<0.05). $IC_{50}$ for cell viability was $2.17{\mu}g/well$. 3. Natural killer cell activity in Chungsangbohahwan-treated group was significantly increased on 100:1, 50:1 E/T(effect cell/target cell) ratio(P<0.05). 4. Production of interleukin-2 in Chungsangbohahwan-treated group was significantly increased(P<0.05).

  • Objective : The purpose of this experiment is to study on the anti-cancer, anti-metastasis and immune response improvement effects of Herbal-acupuncture with Sinomenii acuti lignun infusion solution(SAL-HAS). Methods : We injected Sinomenii acuti Lignum infusion solution into Chung-wan(CV12) of C57BL/6 mouse which is corresponding to human Chung-wan(CV12). We observed its effect on the nunter of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK^+/CD3e^+$ cells in mouse PBMCs, the number of the pulmonary colony, and the effect on MST and ILs of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results & Conclusions : 1. The spleen cells proliferation of the sample groups treated with SAL-HAS extract has increased significantly compared with that of the control group. 2. The percentage of the $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK^+/CD3e^+$ cells in C57BL/6 mouse PBMCs of the sample groups treated with SAL herbal-acupuncture has increased compared with that of the control group. 3. The lung colony number of the sample groups SAL Herbal-acupuncture has decreased significantly compared with that of the control group. 4. MST and ILS of the sample groups SAL herbal-acupuncture have increased significantly compared with those of the control group.

Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of Taraxinic Acid from Taraxacum coreanum (흰민들레(Taraxacum coreanum)에서 분리한 taraxinic acid의 tyrosinase 활성저해 및 melanin 생성저해 효과)

  • This study was to investigate the Taraxinic acid from Taraxacum coreanum on tyrosinase activity and melanogenesis. In B16BL6 cell, Taraxinic acid did not show cytotoxicity even at concentrations of up to 100 ㎍/mL. In addition, tyrosinase inhibitory activity and melanogenesis inhibitory activity were confirmed by stimulation with α-melanocyte stimulating hormone (α-MSH) in the presence of taraxinic acid. Taraxinic acid was added to cells at concentrations of 10, 50 and 100 ㎍/mL and treated for 48 hours to confirm tyrosinase inhibitory activity and melanin production. The tyrosinase inhibitory activity increased in proportion to the amount of the sample, and showed an inhibitory activity of about 54.5% at a concentration of 50 ㎍/mL. Melanin production decreased in proportion to the sample amount, and it was about 62.2% at the concentration of 10 ㎍/mL. From the above results, it was found that Taraxinic acid had higher tyrosinase activity and melanin synthesis inhibitory activity in melanocyte than arbutin. The results suggest that Taraxinic acid can be utilized in natural whitening cosmetics.

Inhibitory Effect of Methanolic Extract of Cnidii Rhizoma on the Melanogenesis (천궁 메탄올 추출액의 멜라닌 형성 억제 효과)

  • Down-regulation of melanin synthesis is required for recovery of pigmentary disorders and it is known that direct inhibitors of tyrosinase suppress melanin synthesis. We screened several oriental medicinal plants using B16/Fl0 cells and found that the methanolic extract of Cnidii Rhizoma down-regulated melanin synthesis effectively. Although the proliferation of Bl6/Fl0 cells was decresed by the methanolic extract of Cnidii Rhizoma, it did not appear necrosis. Bl6/F10 cells incubated with the methanolic extract of Cnidii Rhizoma showed reduced pigmentation and tyrosinase activity. Western blotting revealed that the amount of tyrosinase was decreased by the methanolic extract of Cnidii Rhizoma. These results suggest that the inhibitary effect of the methanolic extract of Cnidii Rhizoma on melanogenesis is due to the suppression of tyrosinase in Bl6/F10 cells and Cnidii Rhizoma is a candidate for an efficient whitening agent.

Isolation and Identification of the Antilisterial Bifidobacterium Isolates from the Infants Fecal Samples (유아의 분변으로부터 항리스테리아 활성의 Bifidobacterium 속 균주의 분리 및 동정)

  • This study was conducted to isolate antilisterial strains of the Bifidobacterium isolates from the infant feces. The bifidobacteria were isolated anaerobically on BL agar and screened for their inhibitory activity on the MRS-cysteine medium against three foodborne pathogens: Listeria monocytogenes, Bacillus cereus, and Staphylococcus aureus. Among the 52 bifidobacterial isolates, 5 strains(A24, Bl, B6, B10, and Bl2) were finally selected based on their stronger antilisterial activity against Listeria monocytogenes than other isolates tested. Morphologically, all the isolates were typically shown Y-and V-shaped under electron microscopic examination. Each isolate was primarily subjected to identification by a polymerase chain reaction(PCR) using a genus-specific primer designed for targeting the 16S rRNA gene sequence, and confirmed the primary identification data using an API-kit(Biomeriuex, France), commercially available product for identification based on biochemical and physiological traits. Of the isolates with antilisterial activity, strain A24 was finally confirmed as the Bifidobacterium longum A24.

The percentage of the $CD25^{+}/CD4^{+}$, $CD8^{+}/CD3e^{+}$, $CD69^{+}/B220^{+}$, $NK^{+}/CD3e^{+}$ cells in C57BL/6 mouse PBMCs of the sample groups treated with CTT herbal-acupuncture has increased compared with that of the control group. 3. The lung colony number of the sample groups CTT Herbal-acupuncture has decreased significantly compared with that of the control group. 4. MST and ILS of the sample groups CTT herbal-acupuncture have increased significantly compared with those of the control group.

MST and ILS of the sample groups treated with EDR herbal-acupuncture have increased significantly compared with those of the control group.
