Synthesis and Evaluation of Coumaroyl Dipeptide Amide as Potential Whitening Agents

  • Coumaroyl dipeptide amide, Coumaric acid-LG-$NH_2$, was prepared successfully using the solid-phase method, and its efficacy as a skin whitening agent was studied. Coumaric acid-LG-$NH_2$ was prepared with Rink-amide resin, and 96.354% of purity was obtained. Using MTT assay and LDH release assay, we found that it exhibited very low cytotoxicity. And, we found that Coumaric acid-LG-$NH_2$ inhibited tyrosinase activity dose-dependently and showed superior tyrosinase inhibitory activity to well-known whitening agent, arbutin. $IC_{50}$ value of Coumaric acid-LG-$NH_2$ was 182.4 ${\mu}M$, and $IC_{50}$ value of arbutin was 384.6 ${\mu}M$. Also, in measurement of melanin contents using B16F1 melanoma cell lines, Coumaric acid-LG-$NH_2$ reduced melanin production induced by ${\alpha}$-MSH statistically significant, and showed superior melanin inhibitory activity to p-coumaric acid or arbutin. In addition, Coumaric acid-LG-$NH_2$ reduced MC1R mRNA expression level. Thus, we concluded that MC1R pathway is the significant pathway of Coumaric acid-LG-$NH_2$, and Coumaric acid-LG-$NH_2$ has great potential to be used as novel whitening agents.

The Anticarcinogenic and Antioxidative Activity of Hemicentrotus pulacherrimus Fractions in Various Cancer Cells. (말똥성게 분획물에 의한 항 발암 및 항산화 효과)

  • This study was carried out to investigate the anticarcinogenic and antioxidative activities of Hemicentrotus pulacherrimus (HP). HP was extracted with methanol (HPM), which was then further fractionated into four sub-fractions by using the solvent partition method, affording methanol (HPMM), hexane (HPMH), butanol (HPMB) and aqueous (HPMA) soluble fractions. We determined the anticarcinogenic activities of these four fractions in four kinds of cancer cell lines, such as HepG2, HT29, MCF-7 and B16-F10, by MTT assay. Among various fractions from HPM, the HPMH showed the strongest growth inhibition effect. We also determined the inductive effect on quinone reductase (QR) of HP fractions. HPMB fraction exhibited strong inductive effects in HepG2 cells at a level of 90 ${\mu}g/ml$, showing inductive indexes of 2.26 compared to the control value of 1.0. The antioxidant activities of fractions from HP were also investigated by measuring the scavenging activities of HP against reactive oxygen speicies (ROS), peroxynitrite (ONOO-) and NO. Among the various solvent fractions, HPMH fractions displayed marked antioxidative activities.

Antitumor and Immuno-modulatory Effect of Crude Polysaccharides from Fruiting Body of Tremella aurantialba Against Mouse Sarcoma 180 (생쥐의 Sarcoma 180에 대한 금목이(Tremella aurantialba) 자실체 추출 조다당류의 항암 및 면역조절 효과)

  • Tremella aurantialba, one of edible and medicinal mushrooms belonging to Tremellaceae of Basidiomycota, has been known to have outstanding curing effects on coughing, tracheitis and hypertension of humans and antitumor activity on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests showed that all crude polysaccharides extracted from the fruiting body were not cytotoxic against cancer cell lines such as RAW 309 CR.1 and Sarcoma 180 at the concentration of $2000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $11.1{\sim}66.7%$ in mice inoculated with Sarcoma 180. Fr. MeOH improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.16 folds at the concentration of $200\;{\mu}g/ml$. In case of Fr. HW, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 1.42 and 2.87 folds, respectively.

Studies on the Antioxidative Effect of the Buckwheat (Fagopyrum esculentum Moench) Extract and its Protective Role against Cadmiun-mediated Stress (메밀의 항산화 및 카드뮴 방어 효능에 관한 연구)

  • In the present study, the Chuncheon buckwheat extracts prepared from its seed coats, seeds and stems were used to determine anti-oxidative effects, the content of rutin and phytic acid, and the protective role against cadmium at the cellular level. futhermore, it was evaluated whether the buckwheat, mainly known as a healthy food source, might be applicable to functional cosmetics. Up to $100 {\mu}g/mL$ of the extract was not toxic in HaCaT and B16F10 cell lines using MTT assay. The anti-oxidative capacity of superoxide radicals was shown in seed coats extracts > stem extracts=seed extracts. Although its content of rutin, known as one of effective anti-oxidants, mainly exists in the stem, any extract did not eliminate hydroxyl radicals. Phytic acid, known as a heavy metal-chelate agent, was highly concentrated in the stem. The Chuncheon buckwheat extract had $10\%$ protective effect against the treatment of $50{\mu}M$ cadmium at which $50\%$ of HaCaT cells survived. Confocal laser scanning microscope revealed the intracellular generation of reactive oxygen species (ROS) by cadmium treatment. Finally, we identified that the stem extract had the most protective effect on the elimination of ROS.

An In Vitro Study on the Activity of Abelmoschus manihot L. Flower Extract on Skin Anti-wrinkle and Skin Whitening (금화규(Abelmoschus manihot ) 꽃 추출물의 In Vitro 피부 미백 및 주름 개선 효능 연구)

  • In this study, the skin whitening and anti-wrinkle activity of Abelmoschus manihot (A. manihot) flowers were evaluated as a cosmetic material. A. manihot flowers were extracted using water and 70% ethanol. Tyrosinase inhibitory activity was evaluated to increase concentration-dependent inhibitory activity in both water (AMW) and 70% ethanol extracts (AME), and was found to inhibit melanin biosynthesis at concentrations that did not affect survival in B16F10 cell lines at 10, 25, 50, 75, and 100 ㎍/mL. As a result of measuring the anti-wrinkle effectiveness of A. manihot flowers, 70% ethanol extract (AME) showed higher anti-wrinkle activity than water extract (AMW). Through the results of this study, it is considered that the A. manihot flower extract can be used as a functional cosmetic material with whitening and anti-wrinkle activity.

Diversification of Rice Quality for Processing. Physicochemical Characteristics and Inheritance of Floury Endosperm Mutants (특수 가공용 미질개발 : 분상질배유 돌연변이 계통의 이화학적특성과 유전)

  • This study was carried out to assess the agronomic characters and physicochemical properties of floury and chalky-endosperm mutant lines induced by chemical mutagen treatment to rice varieties, Hwacheongbyeo and IR24. Linkage analysis of a floury-endosperm gene was carried out using linkage testers. The grain size of brown rice of the mutants was smaller than that of the original varieties. The l, 000-grain and 1$\ell$ weight were lighter in the mutants compared with those in the original varieties. The compound starch granules in the endosperm cell of the mutants showed a loosely-packed crystalline structure. Amylose contents in mutants ranged from 16.9 to 28.5%. Crude protein contents of the mutants were not significantly different from the original rice variety, Hwacheongbyeo, but white core mutant(line 47106) derived from IR24 showed higher protein(l1.32%) compared with IR24(8.30%). The mutants showed slightly harder gel characteristics, and much lower viscosity in Amylograph than original varieties. Steamed rice-cakes from mutant lines showed greater volume than those from original varieties. During the process of alcohol fermentation, Brix in the mutants(especially floury mutants) decreased faster and the alcohol production after 10-day fermentation was much greater in the mutants than in the original varieties. Three different gene loci for floury endosperm characteristics were identified from the allelism test among mutant lines, and the genes were tentatively symbolized as flo-a, flo-b and flo-c, respectively. A floury gene, flo-a, was linked with lg(liguleless) gene in the linkage group N, with R.V. 5.76$\pm$1.72%.

Ampicillin treated German cockroach extract leads to reduced inflammation in human lung cells and a mouse model of Asthma

  • Cockroaches can cause allergic sensitization in humans via contact with their feces or frass. Antibiotics can affect concentration of major allergen and total bacteria production in German cockroaches (Blattella germanica). This study examined the ability of antibiotic-treated German cockroaches to induce allergic airway inflammation and the effect of antibiotics on their lipopolysaccharide and Bla g1, 2, and 5 expression levels. Specifically, we measured the ability of German cockroach extract (with or without prior antibiotic exposure) to induce allergic inflammation in human bronchial epithelial cells and a mouse model of asthma. Bacterial 16S rRNA and lipopolysaccharide levels were lower in ampicillin-treated cockroaches than in the control group. The Bla g1, Bla g2, and Bla g5 expression in ampicillin-treated cockroaches decreased at both the protein and RNA levels. In human bronchial epithelial cell lines BEAS-2B exposed to the ampicillin-treated extract, expression levels of interleukin-6 and interleukin-8 were lower than that in the control group. The total cell count and eosinophil count in bronchoalveolar lavage fluid was also lower in mice exposed to the ampicillin-treated extract than in those exposed to normal cockroach extract. Mouse lung histopathology showed reduced immune cell infiltration and mucus production in the ampicillin group. Our results showed that ampicillin treatment reduced the symbiont bacterial population and major allergen levels in German cockroaches, leading to reduced airway inflammation in mice. These results can facilitate the preparation of protein extracts for immunotherapy or diagnostics applications.

Characterization and Antiviral Effects of Mx Proteins from Various MHC Haplotype Chickens Showing Different Susceptible to Marek's Disease Virus

  • Chicken Mx protein (cMx) induced interferon (IFN) is an antiviral protein to inhibit replication of RNA virus, particularly negative stranded RNA virus, through blockage of transfortation of viral RNA and proteins. In order to determine antiviral effects of cMx from different MHC haplotype chicken, we characterized cMx gene by studying on nucleotide sequencing, antiviral effects to Newcastle disease virus, VSV and MDV, and transcription activities. Three types of eMx genes (2,118 bp) were detected from the different MHC haplotype chickens [B19 (N), B15(F) and B21 (GSP)] chickens, which have showed different susceptible to Marek's disease (MD). Several amino acid substitutions were showed in the cMx. The amino acid 548 and 631 in the cMxs from N and F, chickens susceptible to MD, was Val and Asn which was important on antiviral effects, and showed in resistant cMx. Those in the cMx from GSP, chicken resistant to MD, were same that showed in susceptible cMx. Though every cMx transactivated the expression of the reporter gene, the transcription activation by resistant cMx from N and F was lower compared to that by susceptible cMx from GSP. The decease of the cell growth in the resistant cMx cloned cells was seen in comparison with another cMx clone cells. Replication of NDV and VSV was suppressed in the clones with resistant cMx from N and F. NMx258-transducted cells lack of antiviral effects, and NMx437 or NMx646-transducted cells was showed 60% of antiviral effects compared to NMx705. Mean death time (MDT) and hemaggutination (HA) titer to NDV was long and low in the eggs of N and F lines, but short and high in the egg of GSP line. Interestingly, strong suppression to NDV was observed in the clone with N-Mx and in the eggs of N line. However, the effects of Mx for replication of vvMDV1 have not been. Thus, resistant types of cMx, N- and F-Mx, have showed the anti-viral effects to only RNA virus including NDV and VSV, but not to DNA virus. Antiviral effects of cMx were required whole length of amino acid including Val and Asn in amino acid 548 and 631.

Antitumor and Immuno-potentiating Activities of Crude Polysaccharides from Fruiting Body of Agaricus brasiliensis (신령버섯(Agaricus brasiliensis) 자실체 추출 조다당류의 항암 및 면역증강 작용)

  • Agaricus brasiliensis, one of edible mushroom belonging to Basidiomycota, has been used for curing gastric ulcer and stomach cancer of human beings and also known to have good inhibitory effects on sarcoma 180 and Ehrlich carcinoma of mice. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were prepared from fruiting body of the mushroom. ${\beta}$-glucan and total protein contents were identify from fractions of edible mushrooms extract. The ${\beta}$-glucan and protein contents of all fractions of the mushrooms ranged from 21.54~32.31% and 0.16~9.34%, respectively. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180, HT-29, NIH3T3 and RAW 264.7 at the concentration of 10~2000 ${\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of 18.8~50.6% in mice previously inoculated with Sarcoma 180. Fr. HW increased the numbers of spleen cell by 1.2 fold at the concentration of 200 ${\mu}g/ml$ compared with control. Fr. MeOH and Na improved the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.6 fold compared with control at the concentration of 50~500 ${\mu}g/ml$. Fr. Na generated 15.9 ${\mu}M$ of nitric oxide (NO) when cultured with RAW 264.7 at the concentration of 200 ${\mu}g/ml$, while lipopolysaccharide, a positive control, produced 3.7 ${\mu}M$. The Fr. NaCl, Fr. HW and Fr. MeOH increased the secretion of TNF-${\alpha}$, IL-$1{\beta}$, Il-2 and IL-6 by 2.2 times compared with the control group. Fr. Na increased the numbers of peritoneal exudate cells by 4 folds at the concentration of 50mg/kg compared with control. Circulating leukocytes increased by 2.7 folds when Fr. HW from A. brasiliensis was inoculated at the concentration of 50 mg/kg body weight. The hematological and blood chemical analysis of the 3 fractions did not show any difference in blood compositions and enzyme activities compared with the control group (p<0.05). Therefore, the experimental results suggested that crude polysaccharides extracted from A. brasiliensis contain antitumor and immuno-potentiating activities against Sarcoma 180 in ICR mice.

Anticarcinogenic Effects of Extracts from Gloiopeltis tenax (참가사리 분획물의 암 예방효과)

  • In this study, we investigated anticarcinogenic effects of extracts from Gloiopeltis tenax (GT). GT was extracted with methanol (GTM), which was then further fractionated into four fractions by using solvent fractionation method, affording methanol (GTMM), hexane (GTMH), butanol (GTMB) and aqueous (GTMA) soluble fractions. We determined the cytotoxic effects of these fractions on cancer cells by MTT assay. Among various fractions of GT, the GTMM showed the strongest cytotoxic effect at concentration of $150{\mu}g/mL$, displaying 95.97% on HepG2 cell lines and 93.64% on HT-29 cell lines, respectively. And, the anti-proliferative effect of GT was accompanied by a marked in increase of levels of Bad, Bax, Bok and Bak protein and activation of caspase-3, caspase-7 and PARP protein. Also, we observed quinone reductase (QR) induced effects in all fraction layers of GT on HepG2 cells. The QR induced effects of the GTMM and GTMB on HepG2 cells at concentration of $60{\mu}g/mL$ showing inductive indexes of 2.86 and 2.04 compared to the control value of 1.0.
