An efficient method for the rapid micropropagation of Camptotheca acuminata from axillary buds was established by application of various plant growth regulators. Among various cytokinins, $0.5mg\;L^{-1}$ BA showed the best performance on shoot multiplication, number average multiple shoots up to 10.8. The propagated shoot cuttings in vitro were elongated on NN basal medium without plant growth regulators. The secondary multiple shoots were induced at the site of initially induced buds. Rooting was induced directly near the base of the shoot on half-strength NN medium containing $0.5mg\;L^{-1}$ of IBA, whereas high concentration of $1.0mgL^{-1}$ IBA could induce callus at the base of the shoot. The camptothecin content, anticancer compound of the micropropagated plants was contained in various tissues. Camptothecin contents were 1.8 and $2.5mg\;g^{-1}$ dry weight in stems from propagated in vitro and mother plant, respectively. This result may be used to develop strategies for large-scale propagation of elite C. acuminata trees.
The Cassava (Manihot esculenta Crantz) is one of the major food crops in the tropical or subtropical regions. Recently, clean planting materials of improved cassava cultivars are in high demand. Problems in the propagation of cassava are virus vulnerable and low rates of seed germination. Thus, the study was undertaken to develop an efficient in vitro mass propagation protocol of Manihot esculenta Crantz. So we tried to optimize protocols for mass production from axillary buds of Cassava. Young and actively growing stem segments were excised from adult plants of cassava. Samples were cut into a 3~4 cm nodal segments with axillary buds, and cultivated in the different medium supplemented with various plant growth regulators for 4 weeks. For shoot multiplication, axillary buds approximately 1 cm in length were taken from in vitro derived shoots and subcultured. After 4~6 weeks, the shoot generation rate showed 55.6%. The shoot number and its length was 1.0/explant and 2.3 cm in the most favorable medium composition. The auxin β-indolebutyric acid(IBA) 0~2.0 mg/L was proved to be effective on root development. Plantlets with fibrous roots easily generated tuberous roots in vitro. The tuberous roots were induced only when both kinetin and IBA were used in combination. after 8 weeks, the root generation rate showed 100%. The root number and its length was 17.2/explant and 2.2 cm in the most promising medium composition. Our experiments confirmed that in vitro growth and multiplication of plantlets could depend on its reaction to the different medium composition, and this micropropagation techniques could be a useful system for healthy and vigorous plant production.

A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.
아배양을 통한 물박달나무 성숙목의 기내증식은 DKW 배지에서 양호하였다. 이 배지는 다른 두 배지보다 줄기증식 및 줄기생장이 양호하여 물박달나무의 적정 배지로 생각되었다. DKW 배지에서 액아배양과 정아배양에는 큰 차이는 없었으나 증식에는 액아배양이, 생장에는 정아배양이 다소 양호하였다. 한편 1/2 MS 배지에서는 절편기부에 callus가 직경 1.0cm이상 지나치게 형성되어 줄기신장을 억제하였고, WPM에서는 줄기형태는 정상이나 생장이 저조하여 물박달나무의 아배양 배지로는 부적당하였다. 발근은 NAA 처리가 IBA 보다 효과적이었다. l/2 DKW 배지에 1.0 mg/L NAA 처리로 80%의 기내발근 되었으며 유도된 뿌리수도 많았다 얻어진 줄기는 직접 기외삽목도 가능하였는데, 이 같은 결과는 물박달나무 성숙목의 효율적인 기내번식이 가능함을 시사한다.
한반도에 태풍내습이 빈번한 7, 8, 9월에 참다래(키위)가 조기낙엽되었을 때 당년 액아의 재발아, 익년발아, 개화 등 수체생장에 미치는 영향을 알아보고자 하였다. 비가림조건에서 포트재배(13L)된 골드키위 신품종 '골드러쉬' 착과유목 1-2년생을 대상으로 하고, 적엽처리는 7, 8, 9월에 각각 0(무처리), 50, 100% 잎을 제거하는 것으로 하였다. 적엽처리 30 일뒤 액아 재발아율은 적엽수준에 비례하여 증가하였으며, 시기적으로는 7월이 가장 높았고 8월과 9월로 갈수록 낮았다. 전년도 조기낙엽에 따른 이듬해 발아율은 처리간에 차이가 없었다. 무처리구의 주당 평균 결과지율과 결과지당 꽃수는 각각 27.4%와 2-3개였다. 그러나 전년도 8월과 9월의 100% 적엽구의 결과지율과 결과지당 꽃수는 무처리에 비해 크게 감소하였다. 특히, 8월 16일 100% 적엽구에서는 전혀 꽃이 형성되지 않아 태풍에 의해 조기낙엽이 이 시기에 발생할 경우 개화량 감소로 인한 생산량 감소는 불가피할 것으로 판단된다. 참다래 생산농가에서는 7월 이후에 낙엽피해가 발생하지 않도록 반드시 파풍망, 비가림 등 바람피해 예방시설을 갖추어야 할 것으로 판단된다.
The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.
Ginseng zygotic embryos, seedlings, and exised cotyledonary nodes were cultured on Murashinge and Skoog's(MS) medium, supplemented with 6-benzyladenine(BA) and gibberellic acid(GA3) to induce flower buds. As the concenteration of nitrogen compounds in MS medium was reduced to half of its strength, the flowering frequency of zygotic embryos increased up to 90%. The optimum concentration of sucrose in the medium for flowering of seedlings was 30-60 g/1. In all cases flower buds were formed on elongated axillary branches from the cotyledonary nodes, while the apices remained vegetative. When zygotic embryos and excised cotyledonary nodes were cultured on the medium, supplemented with all possible combinations of BA, GA3, and abscisic acid(ABA) of 5 $\mu$M indole-3-acetic acid(IAA) in the above combinations did not affect flowering. These results suggest that cytokinins, gibberellins, and inhibitors play primary, permissive, and preventive roles, respectively, in the induction of flowering of ginseng.
작약절편을 접종하면 배지와 절편이 흑갈색으로 변하고 수 이로부터 배양이 죽기 시작하는데 배지성분 중에 이를 조장하는 물질이 있음을 발견하였다. 즉 변색의 주원인은 철분과 염화칼슘(Cacl$_2$)이었고, 절편을 괴사시키는데 가장 큰 영양을 미친 것은 NO$_3$었다. 이 결과를 토대로 피해가 비교적 적은 1/4 MS를 번식용 배지로 이용하여 식물체의 모든 부분을 배양하였을 때 조직으로부터 재생은 전혀 일어나지 않았다. 다만 줄기절편에서 이미 형성된 액아가 유식물체로 성장하기도 하였으나 건전한 것으로 계속 성장하지 못하였으며 이것을 다시 번식체로 이용할 때는 초기 배양의 절편에서 나타나는 현상과 같은 원인으로 인하여 생존하지 못하였다.
