콩기름의 자동산화에 미치는 스쿠알렌과 토코페롤의 합산화 효과 (The effect of squalene and tocopherols on their autooxidation of bean oil)

  • In this study, the attempts were made to Investigate the effect of squalene (200 ppm) on the autooxidation of soybean oil The effect of mixed tocopherols (200 ppm) was also studied In the same way and the results of the study were compared with those of squalene. A put of a commercial soybean oil was stored at 45.0$\pm$0.5t for their autooxidation. The extent of the autooxidation was estimated from the changes of the peroxide value and % conjugated dienoic acid content of the ell substrates The results of the autooxidation, squalene In the soybean oil substrate showed antioxidant activity but the activity was weaker than that of mixed-tocopherols nth the same concentration. The induction periods of the control and substrates with the same concentration. The Induction periods of the control and substrates with squalene and mixed-tocopherols were 448.5 504.9 and 668.9 hours, respectively. Time required to Tech a POV of 30meq/kg oil.

Biological screening of 100 plant extracts for cosmetic use (II) Inhibitory activities of tyrosinase and DOPA autooxidation

  • In eastern Asian region, cosmetic preparations containing the molecues possessing the whitening activity are promising. In this study, 100 plant extracts were screened in order to elucidate the whitening effects using in vitro tyrosinase assay and DOPA autooxidation. Several plant extracts such as Morus alba, Glycyrrhiza glabra showed the inhibitory activity (>50%) against mushroom tyrosinase at the concentration 333$mu extrm{m}$/ml. And plant extracts including Morus alba and Bupleurum falcatum showed the inhibitory activity (>50%) at the concentration of 500($\mu\textrm{g}$/ml against DOPA autooxidation. The IC50 values of the several extracts were also found.

  • 추출방법과 용매가 조 채종유중의 미량성분과 그 자동산화성에 미치는 영향에 관해 검토하였다. 불검화물과 Sterol 함량은 추출방법(Twisselmaun과 Soxhlet)과 사용한 용매의 종류(Pentan, Hexan, Heptan) 종류에 따라 변화되지 않았으며 Chlorophyll 함량은 추출용매의 분자량의 증가에 따라 증가되고 사용된 추출법 상호간에는 Chlorophyll 수율에 유의한 영향을 미치지 않았다. 채종유가 자동산화되는 경향을 추출용매의 비점이 상승함에 따라 증가되는 것으로 관찰되었다.

  • 인삼(Panax ginseng C.A. Meyer) 엽소병에서 chorophyll의 bleaching 현상에 대한 생리적 반응기작을 조사, 분석한 결과 chorophyll의 bleaching 현상은 singlet oxygen ($^1O_2$)에 의한 photooxidation 과 catalase와 peroxidase의 inactivation에 의한 $H_2O_2$의 autooxidation이었다. Chorophyll의 bleaching 현상에 원인으로 추리된 $H_2O_2$를 농도별로 인삼잎 절편에 처리하였던 바, chorophyll의 함량과 chorophyll의 흡수스펙트럼은 자연광 처리시 유발된 bleaching 현상과 일치하였으며, saponin 첨가로 인하여 bleaching 현상은 현저히 촉진되었다.

  • Tetrahydropapaveroline (THP), a dopamine-derived 6, 7-dihydroxy-1-(3' ,4'-dihydroxybenzyl)-1,2,3,4-tetrahydroisoquinoline, has been suspected as a possible dopaminergic neurotoxin to elicit Parkinsonism. Autooxidation or enzymatic oxidation of THP and subsequent generation of reactive oxygen species (ROS) may contribute to the degeneration of dopaminergic neurons induced by this isoquinoline alkaloid.(omitted)

숙성된 3종 인삼추출물의 항산화 및 주름개선 효과 (Antioxidant and Anti-wrinkle Effects of Steamed Three Ginseng Extracts)

  • 숙성된 인삼은 인간의 건강을 회복시키고 향상시키는 강장제로서 알려져 있으며, 백삼보다 더 많은 약리적인 활성을 가진다고 알려져 왔다. 전이 가능한 종양, 림프구 증식과 쥐간의 지질과산화에 대한 숙성된 인삼의 효과도 연구되었다. 본 연구는 3가지 인삼 추출물의 항산화와 항노화 효과를 평가하기 위해서 실험하게 되었다. 시장에서 이미 만들어진 홍삼엑기스를 사용하였고, 그 외 수삼, 건삼을 구매하여 홍삼제조기로 만들어진 추출물로 실험에 사용하였다. 숙성된 인삼의 3가지 추출물들은 superoxide radical 소거 활성, hydroxyl radical 소거 활성, 리놀산 자동산화 억제, 섬유아세포에서 collagenase 억제와 collagen 합성의 효과를 연구했다. 숙성시켜 만든 수삼은 100 ug/mL의 농도에서 가장 높은 superoxide radical 소거 활성뿐만 아니라 vitamin C보다 더 높은 hydroxyl radical 소거 활성을 나타냈다. 또한 리놀산 자동산화 억제, collagen 합성과 collagenase 억제에서 가장 높은 활성을 보였다.

  • The mechanisms of benzene toxicity is not fully elucidated, although the metabolism of benzene is very well understood. In order to study the mechanism of benzene toxicity, we investigated DNA damage induced by benzene metabolite, 1,2,4-benzenetriol (BT) in HL-60 cells by alkaline comet assay. To investigate the mechanism of cellular DNA damage induced by BT, the cells were treated with antioxidant such as vitamin C, SOD, catalase, and chelating agent such as deferoxamine (DFO), bathocuproinedisulfonic acid (BCDS). BT induced DNA damage in dose-dependent manner at concentration between 10$\mu\textrm{m}$ and 100$\mu\textrm{m}$. The antioxidant vitamin C itself induced DNA damage at higher concentration. The DNA damage induced by BT in HL-60 cells was protected at low concentraiton of vitamin C whereas no protective effect was found at high concentration. In hibitory effect of SOD on DNA damage by BT was observed and this suggested that BT produce superoxide anion (O2-) causing DNA damage. Catalase protected BT-induced DNA damage suggesting that BT produce H2O2 during autooxidation of BT. Both Fe(II)-specific cheiating agent, deferoxamine (DFO) and Cu(I)-specific chelating agent, bathocuproinedisulfonic acid (BCDS) inhibited BT0induced DNA damage. This suggested that DNA damage was caused by active species which was produced DAN damage. This suggested that DNA damage was caused by active species which was produced by the autooxidation of BT in the presence of Cu(II) and Fe(III). These findings suggest that reactive oxygen species play an important role in the mechanism of toxicity induced by benzene metabolites.

감마선 조사에 의한 돈지의 산패에 산화방지제의 첨가효과 (Effects of Antioxidants on Oxidation of Lard Induced by Gamma Irradiation)

  • The effects of antioxidants such as ascorbyl palmitate(AP), tocopherol( Toc), BHA and ascorbyl palmitate+ tocopherol(AP+ Toc) on lipid peroxide formations in lard have been investigated during storage at 50oC after gamma irradiation with the dose of 1~10kGy. Immediately gamma irradiation greatly increased the initiative oxidation of lard as expected. But antioxidants were found to be greatly effective in minimizing the radiation induced peroxidation of lard. The decreasing order of effects on their antioxidative activities was AP>AP+ Toc>BHA> Toc. Oxidation of lard during storage at 50oC after irradiation induced the acceleration of autooxidation. But the additions of an tioxidants inhibited the formation of peroxides. Their antioxidative activities during storage were BHA> AP+ Toc> Toc>AP. Especially AP+ Toc mixture was not significantly different from BHA (p>0.05).
