글루코실세라마이드 함유 파인애플과실추출물의 경구 투여가 아토피 피부염 동물모델의 피부 장벽기능 개선에 미치는 효과 (Effect of Oral Administration of Pineapple Fruit Extract Containing Glucosylceramide on Skin Barrier Function Improvement in Animal Model of Atopic Dermatitis)

  • 본 연구는 PFEG(글루코실세라마이드 4.0% 함유)가 저 미네랄 특수사료(HR-AD)를 섭취시킨 헤어리스 마우스에서 발생하는 피부장벽 붕괴에 대한 억제작용이 있는지를 확인하고자 하였다. 4주간의 투여를 통해 정상군(Rabo MR Stock)과 비교하여 대조군(HR-AD)에서는 경피 수분 손실량(TEWL)이 서서히 상승하여 피부장벽기능의 붕괴가 발생함과 동시에 표피 수분량 및 각층 수분량도 서서히 저하되는 것이 확인되었다. 육안적으로는 깊은 주름 형성이 확인되었으며 조직학적 소견에서는 표피의 비후와 각화 항진이 확인되었다. 그러나 PFEG를 섭취시킨 시험군에서는 대조군에서 유발되는 피부장벽기능 붕괴에 대한 억제 효과가 확인되었다. 즉, 글루코실세라마이드 0.01% 배합군 및 0.1% 배합군에서는 섭취 시작 2주일째 이후 야기되는 TEWL 상승을 완전히 억제하였다. 또한, 저하되는 표피 수분량, 각층 수분량에 대해서도 유의하게 억제하여 피부 보습 기능을 유지하고 있었다. 육안적으로도 깊은 주름 형성은 억제되었으며, 조직학적인 검토에서도 표피의 비후나 각화 항진은 확인되지 않았다. 이러한 결과는 HR-1 마우스의 피부 세라마이드 생산 개선에 기인한 결과라고 예상된다. 즉, HR-AD 섭취에 의해 HR-1 마우스는 세라마이드를 포함한 피부 지질의 함량 및 구성에 이상이 생기고, 피부 수분량이 급격히 줄어들어 피부 보습 능력이 저하된다. 그런데 PFEG의 혼합 투여는 손상된 피부의 세라마이드 함량 및 구성을 정상과 가깝게 만들어, 대조군에서 나타나는 증상의 발현을 억제하는 것으로 판단된다. 이상의 결과로부터 PFEG의 섭취는 피부장벽기능의 손상으로 발생하는 다양한 피부 수분 손실을 억제함으로써 피부 보습 개선에 우수한 효능이 있음을 알 수 있었다. 따라서 PFEG가 피부 보습에 도움을 주는 건강기능성 원료로서 이용 가능성이 높음을 확인할 수 있다.

고삼 추출물의 피부장벽 강화와 염증완화 효과 (Effect of Sophora flavescens Extract on Reinforcing Skin Barrier and Alleviating Inflammation)

  • 아토피성 피부염은 피부장벽 기능장애, 염증 및 만성 소양증을 특징으로 하는 다인성의 염증성 피부질환이다. 아토피성 피부염은 유전적, 면역학적, 환경적 요인 등의 복합적인 요인으로 피부장벽 기능과 면역기능의 장애를 유발한다고 알려져 있다. 고삼 추출물은 중국전통의학에서 사용되고 있으나, 이의 항아토피 효능에 대한 연구는 거의 진행되지 않았다. 본 연구에서는 아토피성 피부염의 주요 증상인 피부장벽 기능과 면역이상 개선에 대한 고삼추출물의 효과를 평가하였다. 고삼추출물은 피부장벽 기능에서 중요한 역할을 하는 각질세포막의 형성을 강화하는 결과를 나타내었다. 또한 피부의 보습작용에 있어서 중요한 히알루론산의 발현을 증가시키는 결과를 나타내었다. 아토피성 피부염 병변에서 특이적으로 증가하는 황색포도상구균에 대한 고삼추출물의 효능도 확인하였으며, 고삼추출물이 황색포도상구균으로부터 유도된 전염증성사이토카인의 생성을 억제함을 확인하였다. 또한 피부 스트레스 등으로 부터 생성되는 신경전달 물질인 substance P에 의해 유도된 전염증성사이토카인의 발현도 억제하는 것을 확인하였다. 이러한 결과들은 고삼추출물이 피부장벽기능과 면역반응 개선을 통해 아토피 피부염 치료에 사용될 수 있는 잠재적 후보물질임을 제시한다.

Zanthoxylum Piperitum Attenuates the Allergic Inflammation in vivo and in vitro

  • Natural products have received revived interest via traditional remedies or alternative medicine used for the treatment of various diseases. Zanthoxylum piperitum (ZP) has been utilized in traditional medicine for various medicinal purposes. The present study was conducted to evaluate whether ZP modulates allergic inflammation both in vivo and in vitro. We examined the pharmacological effects of ZP on 2, 4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) symptoms in mice. Additionally, in order to clarify the anti-inflammatory mechanisms of ZP, we elucidate the effect of ZP on the expression levels of inflammatory cytokines and nuclear factor-κB (NF-κB) in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). The results demonstrated that ZP attenuated AD clinical symptoms such as erythema, edema and dryness as well as histamine and IgE serum levels in DNCB-induced AD model mice. Additionally, ZP suppressed the expression of inflammatory cytokines and activation of NF-κB in AD-like skin lesions and stimulated HMC-1. These results provide experimental evidence that ZP may be useful candidate for treating allergic inflammation including AD.

실크 단백질의 식이 공급이 아토피 피부염 동물 모델 NC/Nga Mice 표피의 스핑고이드 베이스 및 인산화물 함량 변화에 미치는 영향 (Dietary effect of silk protein on epidermal levels of free sphingoid bases and phosphate metabolites in NC/Nga mice)

  • In our previous studies, dietary supplements of silk protein, sericin, and fibroin, were beneficial for improving epidermal levels of ceramides, which are the major lipids for maintaining the epidermal barrier. In this study, we investigated the dietary effects of silk protein on epidermal levels of free sphingoid bases and their phosphates such as $C_{18}$ sphingosine (So), $C_{18}$ sphinganine (Sa), $C_{18}$ sphingosine-1-phosphate (S1P), and $C_{18}$ sphinganine-1-phosphate (Sa1P), which are either synthetic substrate or degradative metabolites of ceramides. Forty-five male NC/Nga mice, an animal model of atopic dermatitis (AD), were divided into three groups: group CA was an atopic control and fed a control diet, group S was fed a 1% sericin diet, and group F was fed a 1% fibroin diet. Fifteen male BALB/c mice served as group C (control group) and were fed the control diet. All mice were fed with diets and water $ad$ $libitum$ for 10 weeks. Sa in group CA was lower than that in group C, but So in group CA was similar to that in group C. So and Sa were higher in groups S and F than those in group CA; So level was even higher than that in group C, and Sa level was similar to that of group C. The So/Sa ratio in group CA, which is reported to increase in AD, was significantly higher than that of group C. The So/Sa ratio was lower in groups S and F than that in group CA, and decreased further in group F. However, S1P and Sa1P in groups S and F were similar to those in group CA. Taken together, we demonstrated that silk protein, sericin and fibroin dietary supplements, increased So and Sa levels, and decreased the So/Sa ratio.

청화탕(淸華湯)의 항산화 및 항염증 효능 (Effect of CHT in anti-oxidative and anti-inflammatory related factors)

  • To investigate the clinical aspects of CHT in atopic dermatitis (AD) treatments, the effect of CHT in anti-oxidative and anti-inflammatory cytokines were tested. 100% or higher cell viability was observed in all tested groups from 25 to 200 ${\mu}g/m{\ell}$ using Raw 264.7 cells. CHT showed dose-dependent DPPH scavenging activity, with more than 90% scavenging activities at 800 ${\mu}g/m{\ell}$ concentrations. CHT showed dose-dependent suppression activity of ROS production, especially at 200 ${\mu}g/m{\ell}$ of 37.5%. CHT decreased NO production activity, with significant decrease of 33.2% at 200 ${\mu}g/m{\ell}$. IL-6, MCP-1, TNF-${\alpha}$ production rate were decreased by approximately 25% when Raw 264.7 cells were treated with LPS and with CHT of 200 ${\mu}g/m{\ell}$. Also, IL-$1{\beta}$ production rate was decreased by 25% at 100 ${\mu}g/m{\ell}$. The results above indicate that CHT significantly reduces the effect of oxidative and inflammatory cytokines. The use of CHT in dermatitis can be widely suggested.

  • KCSYJT medicines controlled $CD4^+/IFN-\gamma$, and $CD4^+/CD25^+/foxp3^+$ revelation that an experiment that motive allergy immune reponse because an in vitro experiment stimulates T cells of a NC/Nga mouse same time by anti-CD40/rmIL-4, and interleukin-$1{\beta}$, IL-6, TNF-$\alpha$, and TGF-$\beta$ mRNA outturn that bear in T and B cells decreased remarkably by KCSYJT medicines. Intracellular staining of splenocytes anti-CD40/rmIL-4 plus rmIL-4 stimulated as described in a, assessed after 24 h, KCSYJT exerts a mainly immunosuppressive effect that acts at least partially through suppression of the transcription factor GATA3 expression in $CD4^+$ T cells. We found that skin lesions, which were clinically and histologically very similar to human AD, mite antigen-induced dermatitis on the face, neck, ears and dorsal skin of inbred NC/Nga mice. Result that Th1 cell and Th2 cell observe to be shifted by cytokine expression with GATA3 regulation by KCSYJT medicines could know that KCSYJT medicines can use usefully in allergy autoimmnune diease.

아토피성 피부질환 동물 모델 NC/Nga 생쥐에서 내재면역 T와 B 세포의 변형 (Alteration of Innate Immune T and B Cells in the NC/Nga Mouse)

  • Background: Millions of people in the world are suffering from atopic dermatitis (AD), which is a chronic inflammatory skin disease triggered by Th2 immune responses. The NC/Nga mouse is the most extensively studied animal model of AD. Like human AD, NC/Nga mice demonstrate increased levels of IgE, a hallmark of Th2 immune responses. Adaptive immunity cannot be generated without help of innate immunity. Especially natural killer T (NKT) cells and marginal zone B (MZB) cells have been known to play important roles in linking innate immunity to adaptive immunity. Methods: Through flow cytometric analysis and ELISA assay, we investigated whether these lymphocytes might be altered in number in NC/Nga mice. Results: Our data demonstrated that the number of NKT cells was reduced in NC/Nga mice and IFN${\gamma}$ production by NKT cells upon ${\alpha}-GalCer$ stimulation decreased to the levels of CD1d KO mice lacking in NKT cells. However, reduction of NKT cells in NC/Nga mice was not due to CD1d expression, which was normal in the thymus. Interestingly, there was a significant increase of $CD1d^{high}B220^+$ cells in the spleen of NC/Nga mice. Further, we confirmed that $CD1d^{high}B220^+$ cells are B cells, not dendritic cells. These $CD1d^{high}B220^+$ B cells show $IgM^{high}CD21^{high}CD23^{low}$, a characteristic phenotype of MZB cells. Conclusion: We provide the evidence that there are decreased activities of NKT cells and increased number of MZB cells in the NC/Nga mice. Our findings may thus explain why NC/Nga mice are susceptible to AD.

울산지역 초등학생 알레르기 질환 유병률: 지역적 차이와 환경위험인자 (Prevalence of Children's Allergic Diseases in Ulsan: Local Differences and Environmental Risk Factors)

  • Objectives: This study aims to investigate the AD (allergic diseases: asthma, allergic rhinitis and atopic dermatitis) prevalence among elementary schoolchildren in an industrial city, Ulsan, and identify major environmental risk factors associated with AD prevalence. Methods: Data on the physician-diagnosed prevalence over the past 12 months and potential risk factors of AD were collected through a questionnaire from a 2009-2010 survey of 4,067 schoolchildren living in different urban environments. The logistic regression analysis was performed to assess differences in AD prevalence among the areas and to determine which environmental factors impacted AD. Results: Our survey results showed that the AD prevalence rate ranged between 26.2% and 35.9%. Children living in polluted areas (near industrial and central urban areas) had about a 10% higher prevalence of AD than did those living in coastal or suburban residences. The Chi-Square test demonstrated that this local difference was statistically significant before and after adjustment of major confounders such as parental AD history and parental education. The results of the logistic regression analysis showed a statistically significant association between several environmental factors (ventilation in winter, odor conditions and exposure to traffic smoke, and outdoor $PM_{10}/O_3$ pollution) and the prevalence of AD found by multivariate model after adjusting confounders. Conclusion: These results suggest that local differences in AD prevalence are significantly associated with outdoor environmental factors. Although there are likely to be other risk factors for AD, living in a polluted area and exposure to high levels of air pollutants can contribute to an increased risk of childhood AD.

Transduced PEP-1-Grb7 Fusion Protein Suppressed LPS-induced COX-2 Expression

  • Although the incidence and severity of atopic dermatitis (AD) is steadily increasing at an alarming rate, its pathogenic mechanisms remain poorly understood yet. Recently, we found that the expression of Grb7 protein was markedly decreased in AD patients using proteomic analysis. In the present study, human Grb7 gene was fused with PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame PEP-1-Grb7 fusion protein. The expressed and purified PEP-1-Grb7 fusion proteins transduced efficiently into skin cells in a time- and dose-dependent manner when added exogenously in culture media. Once inside the cells, the transduced PEP-1-Grb7 protein was stable for 48 h. In addition, transduced PEP-1-Grb7 fusion protein markedly increased cell viability in macrophage RAW 264.7 cells treated with LPS by inhibition of the COX-2 expression level. These results suggest that the PEP-1-Grb7 fusion protein can be used in protein therapy for inflammatory skin disorders, including AD.

  • Atopic dermatitis (AD) were caused by localized hypersensitivity reaction to an allergen. Therefore, to reduce inflammatory response of AD had been developed that natural extracts and oils with anti-inflammatory activities. This study were investigated that anti-inflammatory effects of Malva verticillata L. oil induced by LPS with RAW 264.7 cells. We measured to production of NO and expression of COX-2, iNOS, TNF-${\alpha}$ by RT-PCR. The Malva verticillata L. oil had decreased the production of NO (p<0.05) and the mRNA level of iNOS in concentration dose dependent manner. In conclusion, this study have shown here may be of help to understand the action mechanism of the anti-inflammatory and we hope that Malva verticillata L. oil used in skin diseases such as AD.
