Nematode destroying-fungi from ginseng field were isolated and then identified on 2% wateragar and malt extract agar media. 1. Six strains of Arthrobotrys sp. and three strains of Harposporium sp. were isolated and identified. 2. Arthrobotrys sp. formed trapping apparatus when they were cultured with Nematode and appeared to be destroying fungi. 3. Harposporium sp. appeared to be endoparasitic fungi. 4. Both Arthrobotrys sp. and Harposporium sp. were grown well on nutrient agar and malt extract agar.

Nematophagous fungi can capture, kill, and digest nematodes using a specific capturing organ. Of the nematophagous fungi, while Arthrobotrys flagrans and A. superba have been described previously, certain characteristics have not been described. For a detailed description of the two nematophagous fungi, the fungi were isolated from soil samples and produced in a pure culture. Morphological characteristics, such as predatory ability (according to the nematode species), shape, and size of predatory organ, conidia, and chlamydospore were investigated and they were used for identification of the fungal isolates along with molecular phylogenetic analysis. Furthermore, this study provides the classification key for 21 nematophagous species.
Most commonly occurring predacious fungus Arthrobotrys oligospora showed great variation in size and shape of conidia on some media. The formation of larger conidia was recorded on beef extract and nutrient agar media. The length of conidia in Richard's YPSS, Sabouraud's, PDA and com meal agar media was of medium size while smaller conidia were produced on Czapek's, Jensen's, Martin's medium. Maximum width of conidia was recorded on YPSS medium followed by Sabouraud's medium. The average size of spores on nematode infested com meal agar medium was slightly increased than those on com meal agar medium.

Nematode-trapping fungi use various specialized traps to capture nematodes. A fungus that can capture nematodes in three dimensional adhesive networks was isolated from the soil around the root of Cucumis melo L. (Oriental melon) in Seongju, Korea. The conidiophores were found to be septate, hyaline, erect and $290-528(342.8){\mu}m$ high. It produces obovoid shape and 1-3 septate (commonly 2-septate) conidia with a size of $30.5{\times}20.3{\mu}m$. Molecular analysis of 5.8 S rDNA displayed 99% similarity to Arthrobotrys sinensis. On the basis of morphological, morphometric and molecular studies, the fungus was identified as A. sinensis. It is the first report in Korea which can be one of biological control resource of plant-parasitic nematode.
For the biological control of the root-lesion nematodes, Pratylenchus spp., which damage directly and indirectly to the leaf perilla, the nematical effect of three nematode-trapping fungi, Arthrobotrys oligospora, A. conoides and A. dactyloides was evaluated in the field. Three species of Arthrobotrys were isolated from the culture soil of leaf perilla in 1998 and were observed the capture of the root-lesion nematodes, Pratylenchus spp. by adhesive hyphal networks or constricting rings on agar. At 40 days after treatment, the plant-parasitic nematodes and root-lesion nematode populations were approximately increased 3.5 fold in untreated control plot, while the nematode population in fungi treatment plots was similar to initial population. In the A. dactyloides plot, however, the population of plant-parasitic nematodes and Pratylenchus spp. was approximately reduced 65% and 53%, respectively. Thus, the fungus A. dachyloides should provide as biological agent for the control of Pratylenchus spp.
Chitinase AO-801 is a hydrolase secreted by Arthrobotrys oligospora during nematode feeding, while its role remained elusive. This study analyzed the molecular characteristics of recombinant chitinase of Arthrobotrys oligospora (reAO-801). AO-801 belongs to the typical glycoside hydrolase 18 family with conserved chitinase sequence and tertiary structure of (α/β)8 triose-phosphate isomerase (TIM) barrel. The molecular weight of reAO-801 was 42 kDa. reAO-801 effectively degraded colloidal and powdered chitin, egg lysate, and stage I larval lysate of Caenorhabditis elegans. The activity of reAO-801 reached its peak at 40℃ and pH values between 4-7. Enzyme activity was inhibited by Zn2+, Ca2+, and Fe3+, whereas Mg2+ and K+ potentiated its activity. In addition, urea, sodium dodecyl sulfate, and 2-mercaptoethanol significantly inhibited enzyme activity. reAO-801 showed complete nematicidal activity against C. elegans stage I larvae. reAO-801 broke down the C. elegans egg shells, causing them to die or die prematurely by hatching the eggs. It also invoked degradation of Haemonchus contortus eggs, resulting in apparent changes in the morphological structure. This study demonstrated the cytotoxic effect of reAO-801, which laid the foundation for further dissecting the mechanism of nematode infestation by A. oligospora.
The nematode-trapping process by Arthrobotrys conoides was investigated with the aid of scanning and transmission electron microscopy. 1. A. conoides captures nematode by means of three-dimensional network. 2. The wall of trap cell was thicker than that of vegetative hypha and the trap cell was more rich in cell organelles such as endoplasmic reticulum, mitochondria and electrondense granule. 3. The electron-dense granule, which could be found only in trap organs, gradually disappeared during its penetration into nematode cuticle. 4. The osmiophilic area was found at adhering site between the trap organ and nematode cuticle. 5. In some cases, any appressorium was not found at the site of penetration. 6. When the fungal-nematode culture was conserved for 2~3 weeks, numerous young nematodes were found to be adhered to spores, resulting in death.
Application of fertilizers such as urea, diammonium phosphate (DAP) and muriate of potash in soil adversely affected the spore germination of Arthrobotrys dactyloides. Amendment of soil with urea at the concentrations of 1.0%, 0.5% and 0.1 % completely inhibited spore germination and direct trap formation on the conidium, whereas muriate of potash delayed and reduced the spore germination even at the lowest concentration. DAP also inhibited spore germination at 1.0% concentration, while at lower concentration the percentage of spore germination was reduced. Application of neem cake at the concentration of 0.5% also inhibited spore germination after 24 h of amendment. The inhibitory effect of neem cake was reduced after 15 days of amendment, while after 30 days after amendment the inhibitory effect was completely lost and the spore germinated by direct trap as in unamended soil. Nematodes were not attracted to ungerminated spores after 24 h of amendment. After 15 days of amendment nematodes were attracted to agar blocks containing fewer germinated spores after 24 h of incubation but after 48 h of incubation large number of nematodes were attracted and trapped by the germinated spores with direct traps. After 30 days of amendment, larger number of nematodes were attracted and trapped by direct traps.
Pinewood nematode (PWN) trapping by nematophagous fungi, Arthrobotrys conoides, A. dactyloides and A. oligospora and the fungal growth were characterized. The three Arthrobotrys species each was inoculated into the PWN cultured on Botrytis cinera fungal colony on potato dextrose agar (PDA). The effects of temperature, pH, PWN inoculation density and nutrients on the growth of the three Arthrobotrys spp were measured. A. conoides grew fast, 13.9 mm/day while A. dactyloides grew slow, 3 mm/day. PDA medium was the best for the fungal growth at $25^{\circ}C$ and pH 4.5. The Arthrobotrys spp growth was stimulated by 500 nematodes inoculation but not by 1000 inoculation. A. dactyloides did not grow below pH 4.5 and at high PWN density. A. conoides and A oligospora formed trapping organs with thick constricting hyphal network only when PWN present, while A. dactyloides formed the organ with circular hyphae constitutively. A. conoides formed trapping organs faster than A. oligospora did. The nematode trapping hyphae of the fungi penetrated into PNW inside to form many tiny infection bulbs and to digest the nematode. However, A. dactyloides formed a few trapping organs but no trapping was observed. Infection rate of PWN was 95% by A. conoides, 80% by A. oligospora and 92% by the combination inoculation of A. conoides and A. oligospora. In contrast A. dactyloides increased PWN density without infecton. There was no interaction effect in any combination inoculation of the three Arthrobotrys spp. A. conoides enhanced PWN infection rate by rapid hyphal growth and early trapping, while A. oligospora did it by increasing hyphal density. In conclusion A. conoides is the most effective in both hyphal growth and infection, and thus these characteristics can be utilized as a biological control of PWN.
Monacroporium ullum sp. nov. the captures nematodes on adhesive spherical knobs was isolated from soil around Codonopsis lanceolata at Ulleung island, Korea. The spindle-shaped conidia sized $17-44{\mu}m\;(26.5{\mu}m)\;long,\;7-10{\mu}m\;(8.1{\mu}m)$ wide, containing from 1 to 4 cross-walls but most often divided by 2 septa (47%). Resting bodies sized $57{\times}30{\mu}m$. Arthrobotrys amerospora has almost spherical non-septate conidia with a small truncate protuberance at the base and sized $20-27{\mu}m\;(23.3){\mu}m$ long and $11-17{\mu}m\;(14.1){\mu}m$ wide. Conidiophores are somewhat longer $362.8{\mu}m\;(311-418{\mu}m)$ than its original description ($75-250{\mu}m$).
