INHIBITORY EFFECT OF ARECA CATECHU L. ON THE GROWTH OF STREPTOCOCCUS MUTANS JC-2 (Streptococcus mutans JC--2의 생육에 미치는 빈랑의 억제효과)

  • In its study, We investigated the effects of Areca catechu L on the growth and acid production of Streptococcus mutans JC-2 in broth system and the activity of glucosyltransferase. The results were summarized as follow; 1. The growth of Streptococcus mutans JC-2 was suppressed by adding Areca catechu L in broth system. Especially, its inhibitory effect was significant at 2,000ppm of concentration. 2. Areca catechu L decreased the acid production of Streptococcus mutans JC-2. Decrease of pH according to acid production was less in presence of Areca catechu L than in absence. 3. Areca catechu L exerted the inhibitory effect against glucosyltransferase activity form Streptococcus mutans JC-2.

Biological activities of Solvent Fractions Isolated from Areca catechu L

  • This study was conducted to investigate the biological activities of Areca catechu L. The antioxidative, fibrinolytic, thrombin inhibitory, and ${\alpha}$-glucosidase inhibitory activities of Areca catechu L extracted with hexane, $CHCl_3$, ethyl acetate, butanol, and water were measured. The water fraction showed the highest extraction yield at 3.65% (w/w). The butanol, $CHCl_3$, water, and ethyl acetate fractions showed strong antioxidative activities at 81.6%, 87.1%, 88.0%, and 89.5%, respectively. The fibrinolytic activity was strong only in the ethyl acetate fraction at 0.84 plasmin units/ml. The 100-fold dilution of the water fraction had the strongest thrombin inhibitory activity at 59.2%. The 100-fold dilution of butanol fraction displayed the strongest ${\alpha}$-glucosidase inhibitory activity at 88.6%. In conclusion, the extracts of Areca catechu L hold promise for use in the development of biofunctional foods to prevent cardiovascular diseases.

  • In order to develop natural anticariogenic agents, we investigated inhibitory effects of Areca catechu L. extracts on the acid production of Streptococcus mutans JC-2 and its cytotoxicity on NIH 3T3 fibroblasts were also examined. The results are as follows : 1. Major organic acid produced by Streptococcus mutans JC-2 were lactic acid and acetic acid, and their productions were decreased by additions of Areca catechu L. extracts. 2. Areca catechu L. extracts were showed noncytotoxicity on NIH 3T3 fibroblasts.

Desmutagenic Effect of Water Extract from Areca catechu L. on the Mutagenicity of N-Methyl-N-Nitro-N'-Nitrosoguanidinein in E. coli PQ37 (N-methyl-N-nitro-N'-nitrosoguanidine의 변이원성에 대한 빈랑 물 추출물의 돌연변이 억제효과)

  • The desmutagenic activity of the water extract of Areca catechu L. on the mutagenicity induced by aflatoxin $B_1$ ($AFB_1$), N-methyl-N-nitro-N'-nitrosoguani-dine (MNNG), mitomycin C (MMC) and 4-nitroquinoline 1-oxide (4-NQO) was studied by using the SOS Chromotest with Escherichia coli PQ37. The inhibition rates of water extract of Areca catechu L. at concentration of $100{\mu}g/assay$ were 41.0%, 47%, 46%, and 32% against $AFB_1$, MNNG, MMC and 4-NQO, respectively. The water extract of Areca catechu L. was separated into methanol soluble and methanol insoluble parts. The methanol insoluble part exhibited higher inhibition effect than the methanol soluble part against the mutagenic activities of MNNG. Step-wise fractionation of methanol insoluble part was done to obtain methanol, ethyl acetate and water fractions. Among these fractions, water fraction had the strongest inhibitory effect of 45.0% against mutagenicities of MNNG. The inhibition rates of aqueous fraction of methanol-insoluble from water extracted Areca catechu L. at concentrations of 1.61, 16.13, 161.29 and $322.58{\mu}g/mL$ were 12.0%, 24.0%, 47.5% and 62.0%, respectively. The water fraction showed the inhibitory effects with dose response against the mutagenic activity induced by MNNG.

  • The inhibitory activities against bacterial ${\alpha}-amylase$of oriental drugs from animals, plant and mineral origin were investigated. In final screening test, it was found that Areca catechu L., Cinnamomum cassia Pres 1. and Ephedra sinica Stapf had stronger inhibitory activities against ${\alpha}-amylase$ than other oriental drugs used in this experiment.

  • The emergence and spread of drug-resistant malaria parasites is a serious public health problem in the tropical world. Useful antimalarial drugs such as chloroquine have resistance in the world now. Moreover, other antimalarialdrugs such as mefloquine, halofantrine, atovaquone, proguanil, artemether and lumefantrine retain efficacy but have limitations, one of which is their high cost. New antimalarial drugs are clearly needed now. Cytotoxicity assay and susceptibility assay were performed for the selectivity of herb extracts in vitro. On the basis of high selectivity, 4-day suppressive test and survival test were progressed in Plasmodium berghei-infected mice. The selectivity of Areca catechu L. (ACL) and butanol extract of ACL (ACL-BuOH extract) were 3.4 and 3.0 in vitro, respectively. Moreover in vivo, 4-day suppressive test showed 39.1 % inhibition effect after treated with 150 mg/kg/day ACL-BuOH to P. berghei-infected mice. Survival test also showed 60% survival rate with ACL-BuOH-treated group while all other group mice died. In this study, ACL and ACL-BuOH were investigated for antimalarial activity in vitro and in vivo and they showed a potent antimalarial activity. In particular,ACL-BuOH could specifically lead higher survival rate of mice in vivo. Therefore ACL-BuOH would be a candidate of antimalarial drugs.

The Scavenging Effect of 207 Oriental Medicines and 19 Oriental Prescriptions on 1,1-Diphenyl-2-Picrylhydrazyl Radical (207종 한약과 활성 한약이 포함된 19종 한약 방제의 1,1-Diphenyl-2-Picrylhydrazyl 라디칼 소거작용)

  • The inhibitory effects of 207 oriental medicines and 19 oriental prescriptions on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical were examined. The most effective oriental medicines were the fruit of Terminalia chebula, followed in order by the seed of Areca catechu, the heart wood of Caesalpinia sappan, the pericarp of Castanea crenata, the seed of Nelumbo nucifera, Rhus galls, the root of Sanguisorba officinalis, and the bark of Betula platyphylla at the concentration of 10 ${\mu}g$/mL. Of the 19 oriental prescriptions, Buyeonsan and Samsinhwan showed the potent scavenging activities of 88% and 71% on DPPH radical at the concentration of 5 ${\mu}g$/mL, respectively.

Antioxidant activity of hot-water extracts and floral waters from natural plant pigments (천연색소 함유 식물추출물과 플로럴 워터의 항산화 활성)

  • The antioxidant potentials of ten kinds of medicinal plants used as natural dyeing agents were tested. Among the plant extracts and floral waters that were examined in this study, the antioxidant activities of the DPPH and ABTS radical scavengers increased with increasing amounts of the extracts. The hot-water extracts from Aphis chinensis, Caesalpinia sappan L., Rumex crispus L., and Areca catechu effectively inhibited the DPPH and ABTS radicals at concentration below 0.1 mg/mL. The floral water obtained through steam distillation from Areca catechu, Rubia akane Nakai, Coptis chinensis, and Phellodendron amurense showed relatively valid antioxidant activities. In particular, the natural colorants extracted from Rumex crispus L., Areca catechu, and Aphis chinensis effectively suppressed the photogenerated singlet oxygen induced by a photosensitizer in in-vitro assay systems. The concentrations ($IC_{50}$) of the hot-water Aphis chinensis extract required to exert a 50% reduction effect on DPPH, ABTS, and singlet oxygen were found to be 8.5, 8.0, and $210{\mu}g/mL$, respectively. The study results suggest that extracts derived from naturally occurring colorants as well as floral waters could be used as natural antioxidants in the food and health-care industries.

Phytochemicals and antioxidant capacity of some tropical edible plants

  • Objective: To find biological functions such as antibacterial and antioxidant activities in several tropical plants and to investigate the possibility of antibiotic substitute agents to prevent and treat diseases caused by pathogenic bacteria. Methods: Plants such as Poncirus trifoliata fruit (Makrut), Zingiber officinale Rosc (Khing), Areca catechu L. (Mak), Solanum melongena L. I (Makkhuayao), and Solanum melongena L. II (Makhurapro) were extracted by methanol, n-hexane, chloroform, ethyl acetate, butanol and water. The free radical scavenging activities were measured using 2-diphenyl-2-picryl hydrazyl photometric assay. Antibacterial activities with a minimum inhibitory concentration (MIC) were observed by agar diffusion assay against pathogenic strains of Escherichia coli, Burkholderia sp., Haemopilus somnus, Haemopilus parasuis, Clostridium perfringens, and Pantoea agglomerans. Results: Poncirus trifoliata fruit methanol extract showed antibacterial activities against gram-negative and gram-positive pathogens. Additionally, this showed the strongest antibacterial activity against Burkholderia sp. and Haemopilus somnus with MIC $131{\mu}g/mL$, respectively. Areca catechu L. water extract showed antibacterial activities against Burkholderia sp., Haemopilus somnus, and Haemopilus parasuis. The MIC value for Haemopilus parasuis was $105{\mu}g/mL$ in this. Antioxidant activity of Zingiber officinale Rosc n-hexane extract showed 2.23 mg/mL effective concentration 50% ($EC_{50}$) value was the highest activity among tropical plants extracts. Total polyphenol content in Zingiber officinale Rosc methanol extract was $48.4{\mu}g/mL$ and flavonoid content was $22.1{\mu}g/mL$ showed the highest values among tested plants extracts. Conclusion: Taken together, these results suggest that tropical plants used in this study may have a potential benefit as an alternative antibiotics agent through their antibacterial and antioxidant activities.

The Antiviral Effects of Areca catechu L. Extract (빈랑 추출물의 새로운 항바이러스 활성)

  • Trafficking of viral glycoproteins to the cell surface results in syncytium formation in baby hamster kidney (BHK) cells infected with Newcastle disease virus (NDV). An extract from the medicinal Areca catechu L plant inhibited not only syncytium formation, but also trafficking of the hemagglutinin-neuramidase (HN) glycoprotein to the cell-surface. The viral glycoprotein was processed within the endoplasmic reticulum during transit to the cell membrane. Fungal extracts showed inhibitory activities ($IC_{50}10{\mu}g/mL$) against ${\alpha}$-glucosidase. These results suggested that A. catechu L. extracts inhibited the cell-surface expression of NDV-HN glycoprotein without significantly affecting HN glycoprotein synthesis in NDV-infected BHK cells.
