Two hundred 10-day-lid, male Arbor Acres broiler chicks divided randomly into 4 groups of 50 chicks each were used. Different feeding treatment was carried out for each group. Chicks in treatment 1 were fed a basal diet(Starter feed)(control); treatment 2, a basal diet + 0.1% B. subtilis culture; treatment 3, a basal diet + 0.2% lactobacilli culture in the feed; and treatment 4, a basal diet + 5 g lactobacilli in the drinking water. The viable bacterial counts for each treatment were approximately $10^9cells/kg$ feed. The weight gain in chickens given feeds incorporated with B. subtilis and lactobacilli was significantly(p < 0.05) higher than those of the control. With regard to feed efficiency, there was a definite tendency towards a higher feed : gain lower(p < 0.05) feed : gain ratio. A significantly(p < 0.05) larger population of Lactobacillus was found in the small intestine of chickens fed with feed incorporated with B. subtilis at 21 and 28 days and with lactobacilli at 14, 21 and 28 days. Populations of intestinal E. coli in broilers given feed added with B. subtilis were not significantly(p < 0.05) different from those of the control, but in chickens fed lactobacilli-added feed, their populations wee significantly lower(p < 0.05) at 14 and 21 days. No significant differences were found among the treatments and the control in the occurrence of Salmonella and Campylobacter during the whole experimental period.
A five wk feeding experiment was conducted with day-old one thousand broiler chicks (Arbor Acres) to determine the effects of microbial phytase (Natuphos$^{(R)}$) supplemented to low nonphytate P (NPP) corn-soy diets. Five pens of 50 mixed sex birds each were randomly assigned to each of the four dietary treatments: T1, control diet containing normal NPP level; T2, T1-0.1% NPP+600 U of phytase/kg diet; T3, T1 - 0.2% NPP+600 U of phytase/kg diet; and T4, T1 - 0.3% NPP+600 U of phytase/kg diet. T1, T2, and T3 showed similar growth rate, feed intake, and feed efficiency, indicating that NPP level in broiler diets could be reduced by approximately 0.2% by the microbial phytase supplementation. But T4 showed significantly (p<0.05) lower weight gain than others. The phytase supplementation improved P availability resulting in low P excretion. Weight and girth of metatarsal bone were increased by phytase supplementation at low NPP diet treatments but ash contents were not significantly different. It can be concluded that NPP level of corn-soy broiler diets can be safely lowered by approximately 0.2 % by supplementing 600 U of microbial phytase/kg diet. With the adjusted level of NPP and phytase supplementation, P excretion could be reduced by 50%.
The 5'-adenosine monophosphate-activated protein kinase (AMPK) is a key part of a kinase-signaling cascade that acts to maintain energy homeostasis. The objective of this experiment was to investigate the possible effects of fasting and refeeding on the gene expression of hypothalamic AMPK, some appetitive regulating peptides and lipid metabolism related enzymes. Seven-day-old male broiler (Arbor Acres) chicks were allocated into three equal treatments: fed ad libitum (control); fasted for 24 h; fasted for 24 h and then refed for 24 h. Compared with the control, the hypothalamic gene expression of $AMPK{\alpha}2$, $AMPK{\beta}1$, $AMPK{\beta}2$, $AMPK{\gamma}1$, Ste20-related adaptor protein ${\beta}$ ($STRAD{\beta}$), mouse protein $25{\alpha}$ ($MO25{\alpha}$) and agouti-related peptide (AgRP) were increased after fasting for 24 h. No significant difference among treatments was observed in mRNA levels of $AMPK{\alpha}1$, $AMPK{\gamma}2$, LKB1 and neuropeptide Y (NPY). However, the expression of $MO25{\beta}$, pro-opiomelanocortin (POMC), corticotropin-releasing hormone (CRH), ghrelin, fatty acid synthase (FAS), acetyl-CoA carboxylase ${\alpha}$ ($ACC{\alpha}$), carnitine palmitoyltransferase 1 (CPT-1) and sterol regulatory element binding protein-1 (SREBP-1) were significantly decreased. The present results indicated that 24 h fasting altered gene expression of AMPK subunits, appetite regulation peptides and lipometabolism related factors in chick's hypothalamus; the hypothalamic FAS signaling pathway might be involved in the AMPK regulated energy homeostasis and/or appetite regulation in poultry.
This study was conducted to explore the effects of squid ink on growth performance, immune functions and antioxidant ability of broiler chickens during a period of six weeks. Either sex Arbor Acres broilers were equally allotted to 4 groups with 3 replicates of 20 chickens each. Broilers diets for the 4 test groups were prepared separately with starter and finisher phases. Control chickens were fed with basal diet and birds of group Exp 2, Exp 4 and Exp 6 were fed with the basal diet supplemented with 2%, 4% and 6% of squid ink, respectively. Broilers were sacrificed to investigate antioxidant parameters of sera, indices of thymus, spleen and bursa of fabricius and spleen lymphocyte proliferation, as well as growth performance on the $21^{th}$ and $42^{th}$ day. The results revealed that, i) squid ink promoted growth performance of broilers during days 22 to 42 and the whole trial period (p<0.05 or p<0.01); ii) squid ink elevated relative weight of the three immune organs during the starter phase and spleen lymphocyte proliferation throughout the experiment (p<0.05); iii) squid ink increased SOD activity and decreased MDA level in sera from broilers during the whole period (p<0.05). The above results suggest that squid ink could improve growth performance, antioxidant ability and immune functions of growing broiler chickens and be employed in the development of feed additives for animals.
This study was conducted to investigate the effects of dietary coconut oil as a medium-chain fatty acid (MCFA) source on performance, carcass composition and serum lipids in male broilers. A total of 540, one-day-old, male Arbor Acres broilers were randomly allotted to 1 of 5 treatments with each treatment being applied to 6 replicates of 18 chicks. The basal diet (i.e., R0) was based on corn and soybean meal and was supplemented with 1.5% soybean oil during the starter phase (d 0 to 21) and 3.0% soybean oil during the grower phase (d 22 to 42). Four experimental diets were formulated by replacing 25%, 50%, 75%, or 100% of the soybean oil with coconut oil (i.e., R25, R50, R75, and R100). Soybean oil and coconut oil were used as sources of long-chain fatty acid and MCFA, respectively. The feeding trial showed that dietary coconut oil had no effect on weight gain, feed intake or feed conversion. On d 42, serum levels of total cholesterol, low-density lipoprotein cholesterol, and low-density lipoprotein/high-density lipoprotein cholesterol were linearly decreased as the coconut oil level increased (p<0.01). Lipoprotein lipase, hepatic lipase, and total lipase activities were linearly increased as the coconut oil level increased (p<0.01). Abdominal fat weight/eviscerated weight (p = 0.05), intermuscular fat width (p<0.01) and subcutaneous fat thickness (p<0.01) showed a significant quadratic relationship, with the lowest value at R75. These results indicated that replacement of 75% of the soybean oil in diets with coconut oil is the optimum level to reduce fat deposition and favorably affect lipid profiles without impairing performance in broilers.
This trial was performed to study the effects of replacing soybean meal (SBM) with fermented rapeseed meal (RSM) on growth performance, serum biochemistry variable and intestinal morphology of broilers. A total of 640 d-old Arbor Acres broiler chicks were randomly allocated to 4 dietary treatments, 4 pens per treatment and 40 birds per pen for a 6-wk feeding trial. In the four treatment groups, fermented RSM replaced soybean meal at 0, 5, 10, and 15%, respectively. On 21 d and 42 d, two birds from each pen were randomly selected and slaughtered. Blood samples and sections of duodenum, jejunum, and ileum were collected for measurement of serum biochemical variables and intestinal morphology, respectively. Results showed that body weight gain (BWG) and feed conversion (FC) were significantly (p<0.01) poorer for birds fed the 15% fermented RSM diet than those fed with 0, 5 and 10% fermented RSM diets during all periods. Compared with 0 and 5% fermented RSM groups, IgG content in the serum of birds in 10 and 15% fermented RSM groups was improved (p<0.01) urea nitrogen content of serum was reduced (p<0.01) during both growing and finishing periods. However, IgM, phosphorus and calcium levels increased (p<0.05) only during the growing period. Increased (p<0.05) villus height was observed in the duodenum and jejunum of broilers fed the diet with 10% fermented RSM. In addition, villus height to crypt depth ratio in the jejunum was significantly higher (p<0.01) for birds fed the diet with 10% fermented RSM than for those fed diets with 0, 5 and 15% fermented RSM. The present results suggest that RSM fermented with Lactobacillus fermentum and Bacillus subtilis is a promising alternative protein source and that it could be safely used replace up to 10% SBM in broiler diets.
Objective: The objective of this experiment was to characterize the mRNA expression profile of type IIb sodium-inorganic phosphate cotransporter (NaPi-IIb) and the biochemical values of serum alkaline phosphatase (AKP), calcium, inorganic phosphorus, tibial ash and minerals of broiler chickens with aging. Methods: A total of 56 one-day-old Arbor Acres male broiler chickens were used. Broiler chickens were weighed and samples were collected weekly from day 1. Results: The result showed that before the growth inflection point, ash, calcium, and phosphorus content in the tibia of broiler chickens increased with growth (before 3 weeks of age), although there were no significant differences in chicks at different ages in the later period of the experiment and weight gain rate was relatively slow at this stage (4 to 6 weeks). NaPi-IIb gene expression in the small intestine in the early growth stage was higher than that in the later growth stage. Expression of calbindin and the vitamin D receptor protein in the intestinal mucosa increased with age in the duodenum and jejunum. Serum AKP activity first increased and subsequently decreased after peaking at 1 week of age, but there was no significant difference after 3 weeks of age. Conclusion: These results show that compared with the early growth stage, the weight-gain rate of broiler chickens in the late growth stage gradually decreased with gradual tibia maturation, along with weaker positive transport of phosphorus in the intestine and reinforced re-absorption of phosphorus in the kidney, which might be the reason that phosphorus requirement in the late growth stage was decreased.
In order to study performance enhancing effects of supplementary antibiotics (avoparcin, nosiheptide and enramycin), two feeding trials were conducted. In Experiment 1, 1, 040 male Arbor Acres were reared in floor pens for 6 wk. Chicks were assigned to one of the following four treatments: Basal diet(B), B+avoparcin 10 ppm, B+nosiheptide 2.5 ppm and B+enramycin 5 ppm. Each treatment had five replications of 52 chicks each. In Experiment 2, two antibiotics (avoparcin and enramycin) were compared in $2{\times}2$ (antibiotics$\times$sex) factorial design. One thousand broiler chicks were reared in floor pens for 6 wk. Each of the 4 treatments had five replications of 50 chicks each. The results of Experiment 1 showed that antibiotic treatments (enramycin, avoparcin and nosiheptide) significantly(P <0.05) improved weight gain. Feed/gain ratio of avoparcin treatment and enramycin treatment tended to he lower than the control but they were not statistically significant. Dressing percentages were high in avoparcin and enramycin treatments compared to the control and nosiheptide treatment. The number of E. coli and Cl. perfringens in ileum and cecal contents were decreased by antibiotic treatments. Moisture contents in excreta of the birds were not significantly affected by the treatments. Leg abnormality and mortality were not significaniy different among treatments. In Experiment 2, weight gain was significantly (P<0.01) different between sexes, but not between antibiotic treatments. Significant effects of antibiotics (P<0.01), sex (P<0.01) and interaction (P<0.05) were shown in feed in take. Feed/gain ratio of avoparcin treatment was significantly (P<0.01) lower than that of enramycin treatment. leg abnormality and mortality were not significantly different among treatments but those in male broiler tended to be higher than in female broilers. It was concluded that nonsystemic antibiotics supplemented to the broiler diets suppress undesirable microorganisms and improve broiler performace in general and avoparcin was most effective in improving feed/gain ratio.
An experiment was conducted with day-old 300 commercial male broiler chicks (Arbor Acres$^{(R)}$) to evaluate the efficacy of crude phytase preparerations produced from a culture of Aspergillus ficcum. The experiment consisted of five dietary treatments; T1, com-soy control diet with 0.45% non-phytate phosphorus (NPP) for starter period and 0.35% NPP for grower period; T2, control - 0.1% NPP; T3, control 0.2% NPP; T4, T3+600 U of crude phytase (broth+cell); and T5, T3+600 U of crude phytase (broth). The body weight gain, feed intake, and feed/gain of chickens fed T1 diet was highest (p<0.01) among treatments. BW gain and feed intake of T4 and T5 were greater than those of T3 but were less than those of T1 and T2. T3 was highest in mortality among treatments. Decreasing the NPP level lowered availability of DM, crude ash, ether extract, crude fiber, Zn, and Fe but supplementation of crude phytase preparations improved the availability of these nutrients as well as those of Ca, P and Cu. Excretion of P and Cu significantly decreased as the NPP level in the diet decreased. Further reduction of P and Cu excretion and reduction of Ca, Mg and Fe excretion were achieved by supplementation of crude phytase preparations. The serum concentrations of Ca, P, Mg, Zn, Fe, and Cu were significantly increased by crude phytase supplementation. The weight and length of tibia, and contents of crude ash, Ca, P, Mg, and Zn were adversely affected by lowering NPP level but partially recovered by supplementation of crude phytase preparations. In conclusion, lowering NPP level in the broiler diet significantly depressed the performance. Supplementation of crude phytase preparations produced from Aspergillus ficuum could partially recover the depression.
This study was conducted to examine the effects of different soy-oil and soy-lecithin levels on growth performance, nutrient utilization and serum parameters in broiler chickens. Two hundred and forty 1-day-old Arbor Acres chicks were randomly divided into 4 groups and treated as follows: basal diet with 2% soy-oil (SO); soy-oil and soy-lecithin mixture in proportion of 75/25 (SOL1), 50/50 (SOL2) and 2% lecithin (SL). At the end of the trial (42 d), birds in SOL1 group grew faster (p<0.05) and had better feed conversion efficiency (p<0.05) than other groups while SL group had the lowest performance (p<0.05). The utilization of ether extract was improved in SOL1 group (p<0.05) but apparent metabolizable energy (AME) and utilization of other nutrients decreased in SOL2 and SL group from 19 to 21 d. No significant effects were observed in apparent metabolizable energy, dry matter, crude protein and ether extract but the utilization of calcium and phosphorus was significantly improved in SL group (p<0.05) during 39 to 42 d. The birds fed with lecithin had lower serum total cholesterol and triglyceride than the control group (SO). Broilers fed with 2% lecithin (SL) had the highest insulin level (p<0.05). The results implied that soy-lecithin and soy-oil in a proportion of 25:75 had the highest growth performance and that soy-lecithin had cholesterol lowering capacity.
