Monitoring Pathogen Infection of Freshwater Cyprinid Fish and Crustacean in Soyang Lake in 2016 (2016년 소양호에서 채집한 자연산 잉어과 어류와 갑각류의 법정전염병 및 기생충성 질병 모니터링)

  • The presence of pathogens in wild fish is a potential threat to the fish being raised nearby fish farm. Surveillance of these pathogens in the wild is, thus, highly important to keep fish in the farm safe from serious communicable diseases. Fish and crustacean were sampled 4 times at Soyang Lake in 2016. Pathogens for five reportable communicable diseases including KHVD (koi herpesvirus disease), SVC (spring viraemia of carp), EUS (epizootic ulcerative syndrome) and WSD (white spot disease), and parasites were investigated. In Soyang Lake, pale chub Zacco platypus was the most abundant. Forty seven pale chub, 8 Korea piscivorous chub Opsarichthys uncirostris, 3 oily shiner Sarcocheilichthys variegatus wakiyae and 2 crucian carp Cyprinus carassius were investigated for detection of target diseases of fish. Seventeen Caridina denticulata denticulata, 12 Oriental river prawn Macrobrachium nipponense and 1 Chinese mitten crab Eriocheir sinensis were used for detection of WSD. As the result, reportable communicable diseases were not detected in all the freshwater animals. Dactylogyrus was detected in Korea piscivorous chub. Copepoda was also detected in pale chub and Korea piscivorous chub. Metacercaria was detected in the pale chub sampled in September and October. Those were not metacercaria of liver fluke Clonorchis sinensis.

Toxic effects of ammonia on the survival, growth, and oxidative and immune responses in the Pacific abalone, Haliotis discus hannai

  • The accumulation of ammonia, a consequence of intensive aquaculture activity, can damage cultured animals. We analyzed the survival rates, growth, antioxidant responses, and immune responses of abalones, Haliotis discus hannai (mean shell length 70.2 ± 4.9 mm; mean body weight 36.9 ± 3.6 g), biweekly for four weeks, to determine the accumulated concentrations of ammonia (0, 0.1, 0.2, 0.4, 0.8, and 1.6 mg/L). In our biweekly measurements, a survival rate of ≥ 95% was observed for all concentrations of ammonia. The specific growth rate (SGR) decreased at ≥ 0.8 mg/L (p < 0.05), but the daily increment in shell length (DISL) showed no significant change (p < 0.05). Regarding antioxidant responses, the superoxide dismutase (SOD) activity of the hepatopancreas was increased at ammonia concentrations over 0.8 and 0.2 mg/L (p < 0.05) at two and four weeks, respectively, and the SOD activity of the gills increased at concentrations over 0.4 and 1.6 mg/L (p < 0.05) at four weeks. The catalase (CAT) activity of the hepatopancreas and gills increased at ammonia concentrations > 0.8 mg/L (p < 0.05). Phenoloxidase (PO) activity increased at ammonia concentrations over 0.8 mg/L (p < 0.05), and Lysozyme (LZM) increased at concentrations over 0.8 and 0.4 mg/L (p < 0.05). Overall, our findings indicated that ammonia concentrations over 0.8 mg/L in seawater might damage the SGR, antioxidant responses, and immune responses in H. discus hannai. Our findings suggest the necessity of improvement or reinstallation for abalone aquaculture systems and can be used to assess the toxic effects of ammonia on H. hannai.

Toxicity of doxycycline, a broad-spectrum semi-synthetic antibiotic, in Pacific whiteleg shrimp (Litopenaeus vannamei) (흰다리새우(Litopenaeus vannamei)에서 광범위 반합성 항생제 doxycycline의 독성연구)

  • Doxycycline is a semi-synthetic broad-spectrum antibiotic, and it has been used to get rid of bacteria in animals and humans. The use of antibiotics has greatly contributed to the aquaculture production although its misuse sometimes presents public health problems. This study was performed to investigate the toxic effects of doxycycline on whiteleg shrimp (Litopenaeus vannamei) administered for possible infection treatments. The shrimp were allocated into four groups and doxycycline was fed three times a day for 7 days at 0, 20, 50 and 100 mg/kg to each group. After 24 hr following the 7-day treatment, hemolymph and hepatopancreas were used for blood and biochemical analysis: Total hemocyte counts, Total protein, Total cholesterol, Gluscose, Glutamic pyruvic transaminase, Glutamic oxaloacetic transaminase, Glutathione peroxidase, Superoxide dismutase, Glutathione-s-transferases, Total antioxidant capacity colorimetric and Acid phosphatase. In addition, histopathological examination was performed on the hepatopancreas and muscle. It was observed that body weight gain was significantly retarded in 100 mg/kg doxycycline group. Doxycycline was found to induce biochemical or functional disorders at 100 mg/kg as observed many of the blood and biochemical parameters were significantly reduced. In conclusion, it was judged that there will be no major toxicity problems with doxycycline when used for shrimp aquaculture at regular doses.

Changes in Fish Viral Disease Outbreaks in the Coastal Area of Korea Due to Increasing Water Temperature, an Impact of Climate Change (기후변화에 기인한 연안 수온상승에 따른 연안 어류의 바이러스성 질병 발생 예측)

  • The impact of global warming on aquatic systems has been a priority research area in the past decade. However, the possibility that increased temperatures will cause shifts in viral disease outbreaks has not been well addressed. In the present study, with increasing water temperature (WT) in the coastal area of Korea, we estimated the possibility of changes in fish viral diseases. From the present time, WT may rise between 0.62 and $1.7^{\circ}C$ by 2050, and the effect on aquaculture could be more adverse than benefitial. Red seabream iridovirus disease (RSIVD) and viral nervous necrosis (VNN) cause high mortality above 22 and $24^{\circ}C$, respectively, and outbreaks could commence earlier and persist for prolonged periods. Nevertheless, the period of occurrence of viral hemorrhagic septicemia (VHS), which outbreaks at a lower WT (< $18^{\circ}C$), could be shorter than the current infectious period. Thermal stress in fish causes reductions in growth and immunocompetence; thus, increases in summer WT can lead to the development of new viral diseases. WT has a strong influence on fish population dynamics; therefore, entry of new viruses and changes in the prevalence of infection can be expected if carrier fishes are introduced or migrate to Korean waters.

Applications and Prospects of Stable Isotope in Aquatic Ecology and Environmental Study (수생태 환경 연구에 있어 안정동위원소의 활용과 전망)

  • Stable isotope approach for aquatic ecology and environmental sciences has been introduced as very useful technique since 1980s and also has been applied to investigate various issues in aquatic ecology and environmental study last 10 years in Korea. Especially carbon and nitrogen isotope ratios have been mainly used to understand food web energy flow and ecosystem structure. In addition, nitrogen isotope ratio has been applied for nitrogen cycle and source identification as well as biomagnification studies. However, large temporal or spatial variations of nitrogen isotope ratio of primary producer have been found in many aquatic environments, and it is regarded as the critical problems to determine trophic level of aquatic animals. Recently, the compound specific isotope analysis of nitrogen within individual amino acids has been developed as an alternative method for trophic ecology. This article introduces the progress history of stable isotope application in aquatic ecology and environmental sciences, and also suggests new direction based on future prospects in stable isotope ecology and environmental study.

  • Amphibian population declines globally. Aquatic contamination by organic pollutants including endocrine disrupters has been suspected to the one of the reason for distinction of amphibia which has obligate aquatic life style during larval period. Amphibians have been widely accepted as animal model for the study of endocrine disruption in aquatic ecosystem at molecular as well as individual levels. There are increasing need for toxicological data in amphibians at multiple endpoints for management of contamination and development of safety guideline for important EDs in aquatic media. Alkylphenols have been widely used in agricultural, industrial, and housekeeping activities, contaminating the aquatic media and evoking endocrine disruption in aquatic animals. In this review, we summarized data concerning the endocrine disruption by alkylphenol organic pollutants on amphibians according to route, concentration, terms, and developmental stage of exposure together with mechanism of endocrine disruption.

Effect of the Gonadotropin-Releasing Hormone (GnRH) on Induction of Maturation in White-Spotted Bambooshark Chiloscyllium plagiosum (성 성숙 자극호르몬방출호르몬(GnRH) 투여를 이용한 백점얼룩상어 (Chiloscyllium plagiosum)의 성 성숙 유도에 관한 연구)

  • Shark populations are constantly decreasing owing to environmental destruction and overfishing; thus, sharks are now at risk of extinction, with 30.5% of species classified as endangered on the International Union for Conservation of Nature's Red List. Sharks are apex predators and keystone species in balancing the marine food chain; their extinction would create an imbalance in the entire marine ecosystem. Assisted reproductive technology is a last resort for protecting animals facing extinction. Here, as a proactive effort toward building a hormone-induced artificial insemination protocol for endangered wild sharks, we identified the possibility of germ cell maturation by administration of GnRH, a commercially produced synthetic salmon gonadotropin-releasing hormone, and calculated its optimum dosage and injection timing. The experiment was conducted on one shark species, Chiloscyllium plagiosum. Injections were administered in 24 h intervals to C. plagiosum females, and 0.2 mL/kg+0.2 mL/kg were the optimal doses. These doses effectively induced maturation and, and ovulation, and oocyte release. Our results confirm that GnRH is a suitable tool for shark hormone-induced artificial insemination and indicate that this method may facilitate the conservation of endangered shark species.

Effects of Biocontrol Agents on Weed Control, Abundance of Aquatic Animals and Insects, and Yield in Paddy Rice Fields (생물자원의 논 잡초 생물학적 방제 효과와 그에 따른 수서동물과 곤충 종수 및 벼 수량 변화)

  • Golden apple snails (Pomacea canaliculat) (GAS) are widely used for weed control in rice cultivation. However, concerns on the ecological risk of invasive GAS species are increasing. This study aimed to evaluate the overall impacts of GAS on weed control, abundance of aquatic animals and insects, and rice yield, in comparison with that from other biocontrol agents such as loach (Misgurnus mizolepis) and catfish (Silurus asotus), which are alternatives for biological weed control in rice paddy cultivation. Field experiments included five treatments; control, herbicide, GAS, loach, and catfish. During the rice growth, weed appearance and biological abundance were monitored, and at harvest, the rice yield was determined. Weed control efficiency was the highest for GAS treatment (100%), followed by that for herbicide (95.8%), loach (57.5%), and catfish treatments (31.7%). Insect abundance was considerably decreased in GAS treatment due to heavy weed removal, which affects the habitat of aquatic animals and insects. The amount of rice yield (unit: kg 10 a-1) was in the order of GAS (798.9) > loach (708.1) = herbicide (700.7) > catfish (629.4) > control (496.0). Therefore, considering the weed control efficiency, biological abudance, and rice yield, loach could be a potential alternative for biological weed control in organic rice farming. However, the activity of the biocontrol agents are susceptible to environmental conditions; therefore, further studies under different conditions are essential to confirm these findings.

  • On maturation of abalone KIKUCHI'S concept of accumulative temperature expressed ${\Sigma}(t_i-{\theta})$ have been apply importantly for an anniversary spawning of the animals. In case of Haliotis discus hannai he was formulated ${\Sigma}(t_i-7.6)>1300$. This expression is able to modify by UKI'S the relationship between the daily feeding rate ($\%$) (Y) ana temperature ($^{\circ}C$) (t) expressed Y= 0.4291 t-0.1501. The modified expression is as the formula ${\Sigma}(Y-3.05)>570$. And the concept its meaning involves is replaced with accumulative ingestion instead of accumulative temperature. This modified concept is understood that on maturation of abalone temperature decide on satiation level, food ingestion decide on gonad maturity rate.

  • The materials were obtained from Busan (=Pusan), Chilcheondo in Geoje, Chungmu, Yeosu, Beolgyo, Boseong, Jangheung, Seo-gwipo of the southern coast of Korea during the period from 1970 to 1971. As a result of the study on the animals of Nereidae, the following five species were identified, which are newly recorded to Korean fauna : Tylorrhynchus heterochaetus Perinereis nuntia var. vallata Perinereis nuntia var. brevicirris Neanthes japonica Nectoneanthes oxypoda.
