Immunosuppressive Effects of Bryoria sp. (Lichen-Forming Fungus) Extracts via Inhibition of CD8+ T-Cell Proliferation and IL-2 Production in CD4+ T Cells

  • Lichen-forming fungi are known to have various biological activities, such as antioxidant, antimicrobial, antitumor, antiviral, anti-inflammation, and anti proliferative effects. However, the immunosuppressive effects of Bryoria sp. extract (BSE) have not previously been investigated. In this study, the inhibitory activity of BSE on the proliferation of $CD8^+$ T cells and the mixed lymphocytes reaction (MLR) was evaluated in vitro. BSE was non-toxic in spleen cells and suppressed the growth of splenocytes induced by anti-CD3. The suppressed cell population in spleen cells consisted of $CD8^+$ T cells and their proliferation was inhibited by the treatment with BSE. This extract significantly suppressed the IL-2 associated with T cell growth and $IFN-{\gamma}$ as the $CD8^+$ T cell marker. Furthermore, BSE reduced the expression of the IL-2 receptor alpha chain ($IL-2R{\alpha}$) on $CD8^+$ T cells and CD86 on dendritic cells by acting as antigen-presenting cells. Finally, the MLR produced by the co-culture of C57BL/6 and MMC-treated BALB/c was suppressed by BSE. IL-2, $IFN-{\gamma}$, and CD69 on $CD8^+$ T cells in MLR condition were inhibited by BSE. These results indicate that BSE inhibits the MLR via the suppression of $IL-2R{\alpha}$ expression in $CD8^+$ T cells. BSE has the potential to be developed as an anti-immunosuppression agent for organ transplants.

Immunostimulatory Effects of Cordyceps militaris on Macrophages through the Enhanced Production of Cytokines via the Activation of NF-${\kappa}B$

  • Background: Cordyceps militaris has been used in traditional medicine to treat numerous diseases and has been reported to possess both antitumor and immunomodulatory activities in vitro and in vivo. However, the pharmacological and biochemical mechanisms of Cordyceps militaris extract (CME) on macrophages have not been clearly elucidated. In the present study, we examined how CME induces the production of proinflammatory cytokines, transcription factor, and the expression of co-stimulatory molecules. Methods: We confirmed the mRNA and protein levels of proinflammatory cytokines through RT-PCR and western blot analysis, followed by a FACS analysis for surface molecules. Results: CME dose dependently increased the production of NO and proinflammatory cytokines such as IL-$1{\beta}$, IL-6, TNF-${\alpha}$, and $PGE_2$, and it induced the protein levels of iNOS, COX-2, and proinflammatory cytokines in a concentrationdependent manner, as determined by western blot and RT-PCR analysis, respectively. The expression of co-stimulatory molecules such as ICAM-1, B7-1, and B7-2 was also enhanced by CME. Furthermore, the activation of the nuclear transcription factor, NF-${\kappa}B$ in macrophages was stimulated by CME. Conclusion: Based on these observations, CME increased proinflammatory cytokines through the activation of NF-${\kappa}B$, further suggesting that CME may prove useful as an immune-enhancing agent in the treatment of immunological disease.

Cordyceps militaris Enhances MHC-restricted Antigen Presentation via the Induced Expression of MHC Molecules and Production of Cytokines

  • Background: Cordyceps militarys water extract (CME) has been reported to exert antitumor and immunomodulatory activities in vivo and in vitro. However, the therapeutic mechanism has not yet been elucidated. In this study, we examined the effects of CME on the antigen presenting function of antigen presenting cells (APCs). Methods: Dendritic cells (DCs) were cultured in the presence of CME, and then allowed to phagocytose microspheres containing ovalbumin (OVA). After washing and fixing the efficacy of OVA, peptide presentation by DCs were evaluated using CD8 and CD4 T cells. Also, we confirmed the protein levels of proinflammatory cytokines through western blot analysis. Results: CME enhanced both MHC class I and class II-restricted presentation of OVA in DCs. In addition, the expression of both MHC class I and II molecules was enhanced, but there was no changes in the phagocytic activity of exogenous OVA. Furthermore, CME induced the protein levels of iNOS, COX-2, proinflammatory cytokines, and nuclear p65 in a concentration-dependent manner, as determined by western blot. Conclusion: These results provide an understanding of the mechanism of the immuno-enhancing activity of CME on the induction of MHC-restricted antigen presentation in relation to their actions on APCs.

목련과 수목의 추출성분에 대한 세포독성평가 (Evaluation of Cell Cytotoxicity on the extractives of Magnoliaceae)

  • 8가지 목련과 수목의 수피 및 잔가지를 채집하고 건조하여 MeOH로 열추출하고, 사람의 암세포에 대한 독성을 측정하였다. 이중 일본 목련이 강한 세포독성이 관찰되어 세포독성을 갖는 순수한 물질 즉, magnonol, honokiol, dihydroxybiphenyl ether, linodenine, anonaine, asimilobine에 대한 세포독성을 측정하였다. 그리고 암세포 살해능 및 apoptosis 유도능을 측정한 결과 magnonol, honokiol, dihydroxybiphenyl ether가 초기단계의 apoptosis 유도에 기인함을 밝혔다. 따라서 세포독성이 강한 물질로 판명된 magnolol, honokiol, dihydroxybiphenyl ether에 대한 정량분석을 위해서 MeOH 추출물을 클로로포름, 에틸아세테이트, 수용성 분획으로 나누고, 에틸 아세테이트 분획을 n-Hexane : Acetone(4:1, v/v)용매로 silica gel chromatography를 한 후, 이 들 화합물이 함유된 분획을 만들었다. 이 분획을 HPLC system으로 이들의 함량을 측정한 결과 MeOH 추출물에 대하여 각각 0.9%, 0.3%, 0.24%가 있음을 밝혔다

동충하초 균사체를 이용한 법제 유황의 항산화활성 및 항염증 효과 (Study on the Anti-oxidative Activity and Anti-inflammatory Effects of Processed Sulfur with Cordyceps Militaris Mycelium)

  • The purpose of this study was evaluated physiological activity of processed sulfur with Cordyceps militaris mycelium that antioxidative and antiinflammatory effects. Proliferation of processed sulfur (PS) with Cordyceps militaris mycelium was increased in dose-dependent manner. In organic sulfur contents of Cordyceps militalis mycelium fortified processed sulfur, CM+PSH (CM+3000 ppm of PS) was significantly higher than other groups. However, CM+PSL (CM+1500 ppm of PS) was almost changed organic sulfur. Content of total polyphenol compounds was similarity to CM, CM+PSL and CM+PSH. The EDA (electron donating ability) and SOD-like activity was increased in dose-dependent manner and the activity of CM were significantly higher than CM+PSL and CM+PSH. We examined cytotoxicity, nitric oxide production of Raw 264.7 cell and inhibition of HT 1080 cell by MTT assay. CM, CM+PSL and CM+PSH do not have any toxic effects in macrophages (Raw 264.7). And CM+PSL and CM+PSH inhibited the production of nitrite in Raw 264.7 cells activated with LPS. The antitumor effects of processed sulfur with Cordyceps militaris mycelium on HT 1080 cell was indicated a significantly inhibition activity. These results suggested that processed sulfur with Cordyceps militaris mycelium have activities of antioxidant, antiinflammatory effects.

Obatoclax Regulates the Proliferation and Fusion of Osteoclast Precursors through the Inhibition of ERK Activation by RANKL

  • Obatoclax, a pan-Bcl2 inhibitor, shows antitumor activities in various solid malignancies. Bcl2-deficient mice have shown the importance of Bcl2 in osteoclasts, as the bone mass of the mice was increased by the induced apoptosis of osteoclasts. Despite the importance of Bcl2, the effects of obatoclax on the proliferation and differentiation of osteoclast precursors have not been studied extensively. Here, we describe the anti-proliferative effects of obatoclax on osteoclast precursors and its negative role on fusion of the cells. Stimulation with low doses of obatoclax significantly suppressed the proliferation of osteoclast precursors in a dose-dependent manner while the apoptosis was markedly increased. Its stimulation was sufficient to block the activation of ERK MAP kinase by RANKL. The same was true when PD98059, an ERK inhibitor, was administered to osteoclast precursors. The activation of JNK1/2 and p38 MAP kinase, necessary for osteoclast differentiation, by RANKL was not affected by obatoclax. Interestingly, whereas the number of TRAP-positive mononuclear cells was increased by both obatoclax and PD98059, fused, multinucleated cells larger than $100{\pm}m$ in diameter containing more than 20 nuclei were completely reduced. Consistently, obatoclax failed to regulate the expression of osteoclast marker genes, including c-Fos, TRAP, RANK and CtsK. Instead, the expression of DC-STAMP and Atp6v0d2, genes that regulate osteoclast fusion, by RANKL was significantly abrogated by both obatoclax and PD98059. Taken together, these results suggest that obatoclax down-regulates the proliferation and fusion of osteoclast precursors through the inhibition of the ERK1/2 MAP kinase pathway.

연교를 함유한 처방단 추출물들의 항염증 효과 (Anti-inflammatory Effects of Prescription Extracts Containing Forsythia viridissima L.)

  • 본 연구는 연교를 함유한 한방처방단을 이용하여 항염 소재를 개발하여 피부 진정 및 여드름, 아토피용 한방화장품 소재를 개발하고자 하였다. 한방에서 염증 질환 치료에 이용되고 있는 연교를 함유한 처방단을 기성한의서를 바탕으로 조사하였다. 그 중 아토피 증상과 가장 연관이 있을 것으로 판단되는 연교승마탕(連翹升麻湯), 연교음(連翹飮), 청열소독음(淸熱消毒飮), 회춘양격산(回春凉膈散)의 처방단을 추출하여 항산화 및 항염 관련 효능을 알아보았다. 연교 함유처방단들의 자유라디칼 소거활성과 superoxide dismutase 유사활성을 알아보았으며, lipoxygenase 활성 저해 효과와 대식세포주에서의 LPS 처리에 의해 유도되어지는 nitric oxide (NO)와 prostaglandin $E_2$ ($PGE_2$)의 생성을 저해하는 효능을확인하였다. 그 결과 연교 함유 처방단들의 우수한 항산화효과와 lipoxygenase 활성 저해 효과를 확인하였다. 특히, 연교승 마탕과 회춘양격산의 NO와 $PGE_2$의 생성 억제 효과를 확인하였다. 또한, 인체 피부 일차자극시험 결과 무자극으로 나타났다. 이로써, 연교를 함유한 처방단들은 항염 효능을 이용한 피부 자극 완화제로서의 개발 가능성을 확인하였다.

Acer okamotoanum Nakai Leaf Extract Inhibits Adipogenesis Via Suppressing Expression of PPAR γ and C/EBP α in 3T3-L1 Cells

  • The genus Acer contains several species with various bioactivities including antioxidant, antitumor and anti-inflammatory properties. However, Acer okamotoanum Nakai, one species within this genus has not been fully studied yet. Therefore, in this study, we investigated the anti-adipogenic activities of leaf extract from A. okamotoanum Nakai (LEAO) on 3T3-L1 preadipocytes. Adipogenesis is one of the cell differentiation processes, which converts preadipocytes into mature adipocytes. Nowadays, inhibition of adipogenesis is considered as an effective strategy in the field of anti-obesity research. In this study, we observed that LEAO decreased the accumulation of lipid droplets during adipogenesis and down-regulated the expression of key adipogenic transcription factors such as peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) and CCAAT/enhancer binding protein ${\alpha}$ (C/EBP ${\alpha}$). In addition, LEAO inactivated PI3K/Akt signaling and its downstream factors that promote adipogenesis by inducing the expression of PPAR ${\gamma}$. LEAO also activated ${\beta}$-catenin signaling, which prevents the adipogenic program by suppressing the expression of PPAR ${\gamma}$. Therefore, we found that treatment with LEAO is effective for attenuating adipogenesis in 3T3-L1 cells. Consequently, these findings suggest that LEAO has the potential to be used as a therapeutic agent for preventing obesity.

Multiple Effects of a Novel Epothilone Analog on Cellular Processes and Signaling Pathways Regulated by Rac1 GTPase in the Human Breast Cancer Cells

  • The epothilones are a class of microtubule inhibitors that exhibit a strong antitumor activity. UTD2 is a novel epothilone analog generated by genetic manipulation of the polyketide biosynthetic gene cluster. This study investigated the effects of UTD2 on the actin cytoskeleton and its critical regulators, and the signaling pathways which are essential for cell motility, growth and survival in MCF-7 breast cancer cells. Results showed that UTD2 inhibited the cellular functions of actin cytoskeleton, such as wound-closure, migration and invasion, as well as adhesion. Our study further demonstrated that UTD2 suppressed Rac1 GTPase activation and reduced the activity of PAK1, which is a downstream effector of Rac1, while the activity of Cdc42 was not affected. Additionally, the phosphorylation of p38 and ERK were significantly inhibited, but the phosphorylation of JNK remained the same after UTD2 treatment. Moreover, UTD2 inhibited the activity and mRNA expression of MMP-2, which plays a key role in cell motility. UTD2 also reduced the phosphorylation of Akt, which is an important signaling kinase regulating the cell survival through Rac1. Furthermore, UTD2 interrupted the synergy between Rac1 and Raf in focus formation assays. Taken together, these results indicated that UTD2 exerted multiple effects on the actin cytoskeleton and signaling pathways associated with Rac1. This study provided novel insights into the molecular mechanism of the antineoplastic and antimetastatic activities of epothilones. Our findings also suggest that the signaling pathways regulated by Rac1 may be evaluated as biomarkers for the response to therapy in clinical trials of epothilones.

추출조건에 따른 마늘 추출물의 항산화 및 항암활성 효과 (Effect of Garlic Extracts with Extraction Conditions on Antioxidant and Anticancer Activity)

  • The purpose of this study was to investigate the effect of garlic(Allium sativum L.) extracts with extraction conditions on antioxidant and anticancer activity. The extracts prepared for garlic by hot temperature extraction (HG), low temperature extraction (LG), UMPM extraction (UG), fermentation (FG) and black garlic hot temperature (BG) method. Content of total polyphenol compound was the BG higher than other extracts. The EDA (electron donating ability) and SOD-like activity was increased in dose-dependent manners, and the activity of BG and UM was significantly higher than LG and FG. We examined cytotoxicity, nitric oxide production of Raw 264.7 cell and inhibition of HT 1080 cell by MTT assay. All extracts does not have any toxic effects in macrophages(Raw 264.7). And UG inhibited the production of nitrite in Raw 264.7 cells activated with LPS. The antitumor effects of LG and UG on HT 1080 cell was indicated a significantly inhibition activity. These results suggested that UG (UMPM extraction of garlic) have activities of antioxidant, anticancer effects.
