Comparision of antioxidant and anti-inflammatory activities of enzyme assisted hydrolysate from Ecklonia maxima blades and stipe

  • Marine brown seaweeds are a source of functional ingredients with various biological properties. They have been used in the food and functional food industries. Brown seaweeds are divided into three parts of blades, stipe, and root. Normally seaweed blades were used as raw materials for biological research. However, there are limited uses on stipes of Ecklonia maxima (E. maxima) depending on the physicochemical, nutritional, and biological properties. Besides, the comparative studies of two structures of E. maxima, blades and stipe didn't discover previously. This study aimed to compare the potent antioxidant and anti-inflammatory activities of the two structures of E. maxima, blades and stipe in vitro studies to increase the utilization of the two structures of E. maxima. The enzyme-assisted hydrolysate from E. maxima showed significant antioxidant and anti-inflammatory activities. Among them, celluclast-assisted hydrolysate from E. maxima blades (EMBC) and viscozyme-assisted hydrolysate from E. maxima stipe (EMSV) expressed significant protection on hydrogen peroxide-induced oxidative stress. Moreover, EMBC and EMSV treatment remarkably reduced nitric oxide production by downregulation of pro-inflammatory cytokine expressions in lipopolysaccharide-stimulated Raw 264.7 cells. Especially EMBC showed strong inhibition on pro-inflammatory cytokine production compared to EMSV. Taken together research findings suggest that EMBC and EMSV possessed potent antioxidant and anti-inflammatory properties and may be utilized as functional ingredients in the food and functional food sectors.

Evaluation of Antioxidant and Anti-diabetic Effects of Sappan Lignum by Extraction Method (추출방법에 따른 소목 심재의 항산화 및 항당뇨 활성 평가)

  • Objectives : The heartwood of Sappan Lignum has been used since ancient times as an ingredient in folk medicines against anti-bacterial and anti-anemia purposes. Many bioactive constituents have been derived from this biomass such as chalcones and homoisoflavonoids. In the current investigation, the antioxidant and anti-diabetic properties using DPPH and $ABTS^+$ radicals scavenging, ${\alpha}-glucosidase$, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Sappan Lignum. Methods : In our continuing investigation for bioactive natural ingredients, the antioxidant and ${\alpha}-glucosidase$ inhibitory properties of Sappan Lignum extracts were prepared from different extraction methods and the biological efficacies were investigated in vitro. The antioxidant properties were evaluated employing radical scavenging assays using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radicals. In addition, the anti-diabetic effects of Sappan Lignum extracts were tested via ${\alpha}-glucosidase$ and AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested samples showed dose-dependent radical scavenging and ${\alpha}-glucosidase$ inhibitory activities. Among the tested extracts, the 80% methanolic extract of Sappan Lignum was showed the most potent activity with an $IC_{50}$ value of $82.3{\pm}1.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. While, $ABTS^+$ radical scavenging activity of 80% methanolic extract was higher than those of other extracts. Also, ${\alpha}-glucosidase$ inhibitory and AGEs formation effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Sappan Lignum can be considered as a new effective source of natural antioxidant and anti-diabetic materials.

Volatile Compound, Physicochemical, and Antioxidant Properties of Beany Flavor-Removed Soy Protein Isolate Hydrolyzates Obtained from Combined High Temperature Pre-Treatment and Enzymatic Hydrolysis

  • The present study investigated the volatile compound, physicochemical, and antioxidant properties of beany flavor-removed soy protein isolate (SPI) hydrolyzates produced by combined high temperature pre-treatment and enzymatic hydrolysis. Without remarkable changes in amino acid composition, reductions of residual lipoxygenase activity and beany flavor-causing volatile compounds such as hexanol, hexanal, and pentanol in SPI were observed after combined heating and enzymatic treatments. The degree of hydrolysis, emulsion capacity and stability, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity, and superoxide radical scavenging activity of SPI were significantly increased, but the magnitudes of apparent viscosity, consistency index, and dynamic moduli (G', G") of SPI were significantly decreased after the combined heating and enzymatic treatments. Based on these results, it was suggested that the enzymatic hydrolysis in combination with high temperature pre-treatment may allow for the production of beany flavor-removed SPI hydrolyzates with superior emulsifying and antioxidant functionalities.

Antioxidant Activity of Prunus mume Extract in Cooked Chicken Breast Meat (매실씨 부산폐기물의 항산화 기능)

  • The antioxidant properties of methanolic extracts from the fruit of Prunus mume were determined in chicken breast meat systems. When P. mume extract (PM) was added to chicken breast meat, 2-thiobarbituric acid-reactive substances (TBARS) value at Day 3 was decreased by about $45\%$ of the control. PM did not affect color of chicken meat compared to the control. The amounts of volatile aldehydes and hydrocarbons were significantly decreased by the addition of PH. Especially, hexanal was the most predominant volatile compound in the control taking up almost more than $50\%$ of the total volatiles, and PM reduced the amount into $26\%$ of the control meat at 3 days.

Quality and Antioxidant Properties of White Breads Enhanced with Broccoli (Brassica oleracea L.) Powder (브로콜리 분말을 첨가한 식빵의 품질 및 항산화 특성)

  • This study was conducted to investigate the quality and antioxidant properties of white breads enhanced with broccoli powder (BP) (0%, 2.5%, 5.0%, 7.5% and 10.0%). L-value significantly decreased with increasing levels of BP, but a- and b-values increased (p<0.05). Baking loss rate, bread volume, and specific volume were reduced with the addition of BP, whereas the pH of the dough and bread weight increased. The hardness, gumminess and brittleness of breads with BP were higher than those of control breads (p<0.05). Upon sensory evaluation, the 2.5% and 5.0% BP breads showed no significant differences in color, flavor, taste, texture, or overall acceptability compared with controls (p<0.05). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of bread significantly increased with increasing BP content (p<0.05). Accordingly, bread quality can be improved by the substitution of 5% BP in place of flour.

Antioxidant Enzyme Activities and Soil Properties of Healthy and Declining Abies koreana (Wils.) in Mt. Halla (한라산 구상나무 건전개체와 쇠약개체의 항산화효소활성 및 토양특성)

  • In order to examine the differences in antioxidant enzyme activities which represent defence mechanism to stressful environments, and soil properties between healthy and declining (or unhealthy) trees, we selected three sites, Witseorum, Youngsil and Sungpanak (Jindallebat). Antioxidant enzymes including Ascorbate peroxidase (APX) and Glutathione Reductase (GR), forest soil properties including soil texture, soil pH, organic matter, total nitrogen, available phosphate, cation exchange capacity, exchangeable cation content and nutrient contents in leaves of Abies koreana (Korean fir) trees were analyzed. There were no significant differences between healthy and declining trees in GR activity. However, seasonal difference in antioxidant enzyme activity was observed. GR activity was lower in June and August than that of September. Soil chemical and physical properties of each site showed a tendency that organic content, total nitrogen content, available phosphorus, cation exchange capacity and cation content were lower at the site of declining trees than the site of healthy trees.

Perfluorosulfonic Acid Composite Membranes Containing Antioxidant Grafted Graphene Oxide for Polymer Electrolyte Membrane Fuel Cell Applications (산화방지 작용기를 함유한 산화 그래핀이 도입된 과불소화계 복합 막의 고분자 전해질 막 연료전지로의 응용)

  • In this study, hindered amine-grafted graphene oxide (HA-GO) with antioxidant properties was prepared and incorporated into Nafion-based composite membranes as an effective filler material for polymer electrolyte membrane fuel cell applications. HA-GO was synthesized via a ring-opening reaction between amine groups in 4-amino-2, 2, 6, 6-tetramethyl piperidine and epoxy groups on the surface of GO. Nafion-based composite membranes containing different weight contents of HA-GO were fabricated to compare the polymer electrolyte membrane properties with those of the pure Nafion membrane. The composite membranes with HA-GO were found to have better mechanical properties, chemical stability, and proton conductivity than the pure Nafion membrane. In particular, the conductivity retention behavior confirmed by the decrease in proton conductivity after Fenton's test of the composite membranes was better than that of the pure Nafion membrane due to the incorporation of HA-GO with effective antioxidant properties.

Changes in the Components of Astragalus membranaceus Fermented by Korean Traditional Nuruk (전통 누룩을 이용한 발효황기의 성분 변화)

  • The major active components of Astragalus membranaceus (AM) are isoflavones, which exist in the form of various glycosides. Nuruk is a traditional fermentation starter in Korea, and is suitable for the biotransformation of isoflavone glycosides because it contains various microorganisms and enzymes. This study was performed to determine changes in the isoflavones and antioxidant properties of AM fermented (AF) with nuruk over 24 hours. AF was sampled after 0, 2, 4, 6, 12, 18, and 24 h of fermentation, and calycosin 7-glucoside, ononin, calycosin, and formononetin content, and the antioxidant properties of AF were analyzed. The total phenolic content increased with fermentation time, and the ABTS radical scavenging activity increased until 6 h of fermentation and then decreased. During fermentation, the isoflavone glycosides decreased significantly as fermentation time increased. The contents of calycosin and formononetin, which are aglycons of calycosin-7-glucoside and ononin, increased from 100.54 ㎍/g to 276.84 ㎍/g and from 56.29 ㎍/g to 123.04 ㎍/g, respectively, at 18 h of fermentation. Significant correlations were observed between fermentation time, isoflavone content, and antioxidant properties. The results of this study showed that fermentation with nuruk is suitable for the biotransformation of isoflavones in AM.

Antioxidant Compounds and Antioxidant Activities of the Methanolic Extracts from Milling Fractions of Black Rice (흑미의 도정분획 메탄올 추출물의 항산화 성분 및 항산화 효과)

  • The colored rice, particularly black rice, has been well characterized to possess antioxidant properties. Rice bran, a by-product of the rice milling process, contains high levels of several phytochemicals which have antioxidant activities as well as health-beneficial properties. The purposes of this study were to evaluate the antioxidant compounds and antioxidant activities of the methanolic extracts from milling fractions and whole grain of two black rice cultivars. Whole black rice of the two cultivars were milled into rice bran and endosperm using the gradual milling system. These were evaluated for antioxidative activities by DPPH and ABTS radical scavenging activities. To determine the antioxidant compounds in the methanolic extract from the milling fractions, the content of polyphenolics, flavonoids, anthocycanins and ${\gamma}$-oryzanol were measured by spectrophotometric methods and vitamin E analysis was carried out by HPLC. The yield of whole black rice, rice bran, and endosperm were 3.1, 15.3, and 0.9% for Heugjinjubyeo and 2.7, 15.5, and 1.1% for Heugkwangbyeo, respectively. The methanolic extracts from rice bran showed generally higher antioxidant activities than the extracts from whole grain and endosperm. In addition, antioxidant compounds distributed much higher contents in rice bran extract than in the extracts from whole grain and endosperm. A significant correlation was also noted between free radical scavenging activity and polyphenolic compounds. The results of this study show that notable antioxidant activity in black rice bran are considered to have significant health benefits.

Effects of Antioxidant and Quality Characteristics of Herb Soy Sauce (허브간장의 항산화성 및 품질 특성에 미치는 영향)

  • Herb soy sauce was prepared by extracting rosemary, lemon balm, spearmint and peppermint at a low temperature in fresh soy sauce. The antioxidant and physicochemical properties of herb soy sauces were examined. Herb soy sauces were prepared by adding 2.5% (w/v) of herbs to fresh soy sauce at 60℃. The total polyphenol, DPPH%, ABTS% increased with extracting time but five minutes extraction with sufficient antioxidant herb soy sauce could be prepared. The antioxidant powers of herb soy sauces were higher in lemon balm extract, and the rosemary extract showed the lowest. Soy sauces added with herbs had lower pH and salinity (p<0.05) and higher sugar content than without herbs. The colorness according to the type of herbs showed significant difference.
