Effect of Samhwangsasim-tang, Samigangap-tang and Bangtan-tang on Blood Pressure in Stroke Prone Spontaneously Hypertensive Rats (뇌졸중성(腦卒中性) 본태성(本態性) 고혈압(高血壓) 백서(白鼠) 모델에서 삼황사심탕(三黃瀉心湯), 사미강압탕(四味降壓湯), 방탄탕(防癱湯)의 혈압(血壓)에 미치는 영향(影響))

  • Objectives : The aim of the present study is to investigate the hypotensive effect of Samhwangsasim-tang (SHSST), Samigangap-tang (SMGAT) and Bangtan-tang (BTT) in stroke-prone spontaneously hypertensive rats (SHR-SP). Methods : SHR-SP rats were treated with SHSST, SMGAT and BTT at dose of 200 mg/kg/day orally for 5 weeks, respectively. Results : Treatment SHR-SP rats with SMGAT significantly lowered blood pressure but not in the SHSST or BTT treat groups. On the other hand, SHSST, SMGAT and BTT ameliorated endothelium-dependent and independent vascular relaxation in the phenylephrine-precontracted aorta and carotid artery, respectively. Conclusions : These results indicated that SMGAT has an antihypertensive effect and SHSST, SMGAT and BTT improve vascular function in stroke-prone hypertensive rat model, respectively.

Analysis of Nutritional Characteristics and Physiological Functionality of Hypsizygus marmoreus (Brown cultivar) (갈색 느티만가닥버섯의 영양특성 및 생리기능성 분석)

  • In order to apply into functional food or medicinal industry, nutritional characteristics and physiological functionality of Hypsizygus marmoreus (brown cultivar) were investigated. Fruiting body of H. marmoreus contained 27.3% of crude protein, 55.8% of total sugar and 11,109.3 mg/100 g dry weight of malic acid. Furthermore, 66.7% of antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity and 37.3% of antigout xanthin oxidase inhibitory activity showed in the water extract from H. marmoreus. The economically ACE inhibitory activity (81.4%) was obtained when the fruiting body of Hysizygus marmoreus was extracted with distilled water of $50^{\circ}C$ for 12 h, even though maximal ACE inhibitory activity (84.4%) was showed the extracts from $70^{\circ}C$ for 12 h.

The depressive effect of S'NC nerve control treatment in hypertension patients (고혈압 환자에서 뇌신경조절의학 S'NC의 혈압강하 효과)

  • Objectives : The aim of this study was to investigate the antihypertensive effect of S'NC nerve control treatment in hypertensive patients. Methods: We measured the blood pressure of 5 patients who were admitted in the Oriental Medical Clinic of S'NC Medicine Research Institute from 19th April 2007 to 29th June 2007. Eligible participants had systolic blood pressure ${\ge}$140mmHg or diastolic blood pressure ${\ge}$90mmHg. Blood pressure measurements were after the patient had been in bed rest for at least 10 min. Ten sessions of S'NC nerve control treatment over 2 weeks were performed in the patients. Blood pressure were measured twice before and after each treatment. Results : After 2 weeks, blood pressure reduction was observed in the treatment patients, with an average decline of systolic blood pressure up to 21mmHg and diastolic blood pressure up to 20mmHg. But, there were not statiscally significant The effects of S'NC nerve control treatment by measuring time on blood pressure were as follows: In a systolic blood pressure(p=0.087) and diastolic blood pressure(p=0.609) was gradually deceased not significantly from 1st to 10th. Conclusion: These results suggest that S'NC nerve control treatment may be efficacious in decreasing arterial blood pressure in hypertensive patients. Controlled trials investigating the efficacy of S'NC nerve control treatment for lowering blood pressure are warranted.

Preparation of Fermented Sauces with Increased ACE Inhibitory and Antioxidative Activities (ACE 저해능 및 항산화능이 향상된 발효소스의 제조)

  • For development of premium sauce, fermented sauces containing ferment (with wheat flour and soybean) and extract(s) of Styela clava (Korean name: miduduk) tunic and/or mulberry leaves and/or onion were prepared, and their angiotensin converting enzyme (ACE) inhibitory and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were evaluated. All sauces containing extract(s) showed increased ACE inhibitory and DPPH radical scavenging activities. Especially, sauces containing extracts of mulberry leaves and onion showed superior activities, with 26.92% and 40.66% relatively increased ACE inhibitory and DPPH radical scavenging activities, respectively, compared to control (no extract was added). These results suggest that extracts of mulberry leaves and onion could improve antihypertensive and antioxidant activities of fermented sauce.

Scientific Study of Magnetic Acu-point Therapy on Eight Extraordinary Meridian for Lowering Blood Pressure (기경팔맥 자석침의 혈압 강하 효과에 대한 과학적 검증 연구)

  • This study assessed effects of magnetic acu-point therapy as an add-on to conventional antihypertensive managements such as medication or lifestyle modification for hypertensive or pre-hypertensive subjects. This study was conducted with the purpose to evaluate the decreasing effect of the blood pressure on the hypertensive patients who were given to magnetic acu-point therapy. A single-blind, placebo-controlled trial by cross matching was conducted at Daejeon University Oriental Hospital. The washout periods of cross matching are 10 days. 22 hypertensive (systolic BP>140 mmHg or diastolic BP>90 mmHg) volunteers were recruited and treatments were donemagnetic acu-points on Oegwan(TE 5), Jogimeup(GB 41), Naegwan(PC 6), Gongson(SP 4), Hugye(SI 3), Sinmaek(BL 62). The treated group was statistically significant decrease of the blood pressure, while control group show no statistically significant difference of BP. From the results, magnetic acu-point therapy seems to offer a benefit to the treatment of hypertensive patients.

Anti-obesity effects of onion juice in high fat diet-induced obese rats (비만 유도 흰쥐에 대한 양파의 항비만 효과)

  • This study investigated the effects of onion juice on the serum lipid components and blood pressure in obese rats fed high fat diets. Thirty-nine of Sprague-Dawley rats were divided into six groups, and were treated for 8 weeks: (1) normal diet (ND); (2) high fat diet (HFD); (3) HFD for first 4 weeks and high fat diet with 40% onion juice for the last 4weeks (H-H+O); (4) HFD with 40% onion juice for 8 weeks (H+O); (5) HFD for first 4 weeks and ND for the last 4 weeks (H-N); (6) HFD for first 4 weeks and ND with 40% onion juice for the last 4 weeks (H-N+O). The rates of increasing body weight were reduced in H+O and H-H+O groups compared with HFD group. The levels of triglyceride, low density lipoprotein-cholesterol and total cholesterol in blood serum were significantly decreased in the H+O and H-H+O groups compared with the HFD group. Administration of onion reduced the size of adipocyte, steatosis, and serum hyperlipidemia in obese rats fed HFD. Moreover, the antihypertensive effects of onion were observed in obesity rat fed HFD. Overall results suggest that onion reduces the serum lipid components and improves hypertension in obese rat fed HFD.

The inhibition of Hypertension-related Response by $17\beta$-estradiol and the Increase of $17\beta$-estradiol Activity by Electrical Stimulation ($17\beta$-estradiol의 고혈압 유도반응 억제와 인체적용 전기자극의 $17\beta$-estradiol 활성 증가)

  • Purpose: $17\beta$-estradiol is the most active endogenous estrogen, which is related to favorable changes in the plasma lipid profile, to relaxation of the coronary vessels, and to a decrease in platelet aggregation and vascular smooth muscle cell migration. However, although the beneficial effect of estrogens on plasma lipoproteins (ie, lowering low-density lipoprotein and increasing high-density lipoprotein cholesterol) contributes to cardiovascular protection, it does not fully account for the protective effect, particularly in the application of physical therapy, including low frequency electrical stimulation. Methods: The aim of this study was to demonstrate the inhibition of stressors, such as endothelin-1 (ET-1), serotonin (5-hydroxytryptamine, 5-HT), prostaglandin $F2\alpha$ ($PGF2\alpha$), and a protein kinase C (PKC) activator 12-deoxyphorbol 13-isobutyrate (DPB), induced isometric tension by $17\beta$-estradiol in vascular smooth muscle strips, respectively. In addition, the effects of low frequency electrical stimulation at the meridian points (CV-3, -4, Ki-12, SP-6, LR-3, BL-25, -28, -32, -52) on the indirect antihypertensive effect were examined by monitoring the changes in the serum $17\beta$-estradiol concentration in healthy volunteers. Results: Isometric tension analysis showed that the responses of inhibited tension by $17\beta$-estradiol were similar to the same stressors in rat aortic smooth muscle strips. Furthermore, although the continued amplitude modulation (AM) type of electrical stimulation was not increased significantly by electrical stimulation, the current of the frequency modulation (FM) type of low frequency electrical stimulation increased the serum $17\beta$-estradiol concentration in normal volunteers. Conclusion: These results, in part, suggest that $17\beta$-estradiol has the capacity to supress stressor-induced muscle tension, and electrical stimulation, particularly current of the FM type, has a modulatory effect on the sex steroid hormones, particularly $17\beta$-estradiol, in healthy volunteers.

Effects of DaeSiHo-Tang extract on Hypertension and Arterial Contraction (대시호탕(大柴胡湯)이 고혈압과 수축혈관에 미치는 영향)

  • This study was undertaken to define the effect of DaeSiHo-Tang extract on the hypertension in spontaneous hypertensive rat and norepinephrine-induced arterial contraction in rabbit. Systolic blood pressure and blood velocity were significantly attenuated by administration of DaeSiHo-Tang extract. but blood flow and renin-angiotensin-aldosterone system unaffected by DaeSiHo-Tang extract. The relaxation effect of DaeSiHo-Tang extract was dependent on the presence of endothelium, showing that DaeSiHo-Tang extract-induced relaxation was not observed in the strips without endothelium. The endothelium-dependent relaxation induced by DaeSiHo-Tang extract was decreased by the pretreatment of $N{\omega}$-nitro-L-arginine or methylene blue, but it was not observed in the strips pretreated with indomethacin or tetraethylammonium chloride. When $Ca^{2+}$ was applied, the strips which were contracted by norepinephrine in a $Ca^{2+}$-free solution, arterial contraction was increased. But pre-treatment of DaeSiHo-Tang extract inhibited contractile response to $Ca^{2+}$. These results indicate that antihypertensive effect of DaeSiHo-Tang extract is due to descend arterial resistance by the arterial relaxation through the formation of nitric oxide in the vascular endothelial cells.

INFLUENCE OF PINACIDIL ON CATECHOLAMINE SECRETION EVOKED BY CHOLINERGIC STIMULATION AND MEMBRANE DEPOLARIZATION FROM THE RAT ADRENAL GLAND

  • It has been known that potassium channel openers are a new class of molecules that have attracted general interest because of their potent antihypertensive activity in vivo and vasorelaxant activity in vitro (Hamilton and Weston, 1989). In the present study, it was attempted to examine the effect of the potassium channel opener on catecholamine (CA) secretion evoked by cholinergic stimulation, membrane depolarization and calcium mobilization from the isolated perfused rat adrenal gland. The perfusion of pinacidil (30-300 uM) into an adrenal vein for 20 min produced relatively dose-dependent inhibition in CA secretion evoked by ACh (5.32 mM), high $K^{+}$ (56 mM), DMPP (100 uM for 2 min), McN-A-343 (100 uM for 2 min), cyclopiazonic acid (10 uM for 4 min) and Bay-K-8644 (10 uM for 4 min). Also, under the presence of minoxidil (100 uM), which is also known to be a potassium channel activator, CA secretory responses evoked by ACh, high potassium, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were also significantly depressed. However, in adrenal glands preloaded with pinacidil (100 uM) under the presence of glibenclamide (1 uM), an antidiabetic sulfonylurea that has been shown to be a specific blocker of ATP-regulated potassium channels (for 20 min), CA secretory responses evoked by ACh, high potassium, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were considerably recovered to a considerable extent of the normal release as compared to that of pinacidil only. These results, taken together, suggest that pinacidil cause the marked inhibition of CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as by membrane depolarization, indicating strongly that this effect may be mediated by inhibiting influx of extracellular calcium and release in intracellular calcium in the rat adrenomedullary chromaffin cells. Furthermore, these findings suggest strongly that these potassium channel openers-sensitive membrane potassium channels also play an important role in regulating CA secretion.

The Study on Association of Calcium Channel SNPs with Adverse Drug Reaction of Calcium Channel Blocker in Korean

  • Rapid advances in pharmacogenomic research have provided important information to improve drug selection, to maximize drug efficacy, and to minimize drug adverse reaction. The SNPs that are the most abundant type of genetic variants have been proven as valid biomarkers to give information on the prediction of pharmacokinetic/pharmacodynamic properties of drugs based on genotype. In order to elucidate a correlation between SNPs of calcium channel encoding gene and adverse reactions of calcium channel blockers, we investigated SNPs in CACNA1C gene known as a binding site of calcium channel blocker. 96 patients with hypertension who had taken or are taking an antihypertensive drug, 1,4-dihydropyridine (DHP) were included for analysis. These patients were composed of 47 patients with adverse drug reactions (ADR) such as edema from calcium channel blockers and 49 patients without ADR as a control group. The exons encoding the drug binding sites were amplified by PCR using specific primers, and SNPs were analyzed by direct sequencing. We found that there was no SNP in the exons encoding DHP binding site, but four novel SNPs in the exon-intron junction region. However, four novel SNPs were not associated with the ADR of calcium channel blockers. In conclusion, this study showed that ADR from calcium channel blockers may not be caused by SNPs of the binding sites of calcium channel blockers in CACNA1C gene.
