ANTIBIOTICS RESIDUES IN RAW MILK IN THAILAND

  • One thousand eight hundreds and twenty two samples of raw milk were detected for antibiotic residues using Bacillus subtilis ATTCC 6633, B. stearothermophilus var. calidolactis C 593 and Micrococcus luteus ATCC 9341 as test organisms, were carried out from July 1991 through June 1992. Apparent antibiotic residues were found through out the study period, except in January. The detection rate varied from 0.7% in March and May to 11% in April. One hundred and thirty six (72%) samples of the 187 screening positive samples were considered to contain only the indigenous antimicrobial agents. Of the total, 51 (2.8%) samples were positive for antibiotic residues. Among the tested organisms, B. stearothermophilus var. calidolactis was the most sensitive organism in detection of the antibiotic residues.

Prevalence of Antibiotic Residues and Antibiotic Resistance in Isolates of Chicken Meat in Korea

  • The aim of study was to investigate the correlation between the level of 17 antibiotic residues and 6 antibiotic resistances of Escherichia coli isolates in chicken meats. A total of 58 chicken meats were collected from retail grocery stores in five provinces in Korea. The total detection rate of antibiotic residues was 45% (26 out of 58). Ten out of 17 antibiotics were detected in chicken meats. None of the antibiotics exceeded the maximum residue level (MRLs) in chicken established by the Ministry of Food and Drug Safety (MFDS). The most detected antibiotics were amoxicillin (15.5%), followed by enrofloxacin (12.1%) and sulfamethoxazole (10.3%). In a total of 58 chicken meats, 51 E. coli strains were isolated. E. coli isolates showed the highest resistance to ampicillin (75%), followed by tetracycline (69%), ciprofloxacin (65%), trimethoprim/sulfamethoxazole (41%), ceftiofur (22%), and amoxicillin/clavulanic acid (12%). The results of study showed basic information on relationship between antibiotic residue and resistance for 6 compounds in 13 chicken samples. Further investigation on the antibiotic resistance patterns of various bacteria species is needed to improve food safety.

소 및 돼지의 정육과 내부장기중의 항생물질 잔류 조사 (Detection of Antibiotic Residues in Meats and Internal Organs of Cattle and Pigs)

  • The purpose of the present survey was to evaluate the antibiotic residues in meats and internal organs such as muscle, liver, heart, kidney and spleen of cattle (n=59) and pigs (n=115). The EEC-4-plate-method were employed. The results were obtained as follows ; 1. In BS 6.0, BS 7.2 and BS 8.0 used as media to detect antibiotic residues, the zone($M{\pm}SD,$ cm) of bacterial growth inhibition was narrow($1.40{\pm}0$) in meats, whereas the zone was wide($1.69{\pm}0.25-1.88{\pm}0.23$ and $1.58{\pm}0.18-1.86{\pm}0.15$ in cattle and pigs, respectively) in internal organs. But in SL 8.0, it was difficult to detect the zones ($0-1.40{\pm}0$) of both meats and internal organs. 2. Residues of antibiotic in beef and pork were rarely detected in BS 6.0, BS 7.2 and BS 8.0 (range 1.7-11.9% and 2.6-4.3%, respectively), whereas residual percentages of internal organs were relatively higher(range 69.5-96.6% and 43.5-84.3%, respectively). But in SL 8.0, it was not detected in both beef or pork, whereas they were 0-13.6% and 0-4.3% in interanal organs.

  • Antibiotic residues are undesirable in milk and milk products for a number of reasons. In particular, they can have harmful effects on public health and harm to the manufacturer of the cultured milk products, e.g. MRSA etc. Although government regulatory agencies and the dairy industry have been successful in decreasing the presence of high concentrations of antibiotic residues, violations still occur and lead to contaminated products. As a result, several rapid and reliable methods for the detection of antibiotic residues have been developed, including microbiological and instrumental analysis methods. The conventional methods are time consuming, but recent improvements have allowed for better detection time, sensitivity, and accuracy. An example of an advanced detection instrument is the biosensor, which has several applications in food and environmental science, e.g. food-born pathogen detection, antimicrobial residues etc. In the present review, the recent trends in the methods used to test for antibiotic residues in milk and dairy products, as well as their specific applications, have been discussed.

항생물질 발효 부산물이 수도 및 토마토 생육에 미치는 영향 (Effect of Antibiotic Fermentation Residues on Rice and Tomato Growth)

  • Erythromycin 및 Rifamycin 두 종의 항생물질 발효 부산물의 농업적 이용에 대한 검토를 위하여 발효 부산물 자체의 이화학적 성질의 조사와 함께 경기도 벽제 소재 일반 농가의 논과 밭토양을 이용하여 수도 및 토마토를 표식작물로 하여 두 종 시험물질은 시용효과를 조사한 결과는 다음과 같다. 1) 항생물질 발효균제 두 종은 모두 중성물짙($pH 6.8{\sim}7.4$)로서 다량의 유기물($21.6{\sim}24.2%$), 인산 ($2900{\sim}4600 ppm$) 및 치환성 염류($55.4{\sim}138.3meq/100g$)를 함유하였다. 2) 두 종의 항생물질 부산물은 일반토양 보다 높은 등전점($pH 7.0{\sim}8.0$)을 가짐과 동시에 양전하의 발달이 급격하여 음 ion의 흡착용량이 큰 것으로 나타났다. 3) 수도에 대한 두 종 항생물질 부산물의 시용효과는 삼요소 단용구 및 기타 유기질 비료의 시용효과와 대응하였으며 반당 40㎏ 시용에서 최대의 정조 수량을 나타내었다. 4) 토마토에 대한 두 종 항생물질 부산물의 시용효과도 수도에 대한 결과와 유사한 경향을 보여 평균적으로 기타 비료 시용효과와 대등하였다. 5) 생육상황 중 수도는 식물체 초장, 건물중 및 유효 분얼 주당 수수, 그리고 토마토에 있어서는 묘목수장과 생체중이 각기 수량과 유사한 결과를 나타내어 이들이 수량증가에 관계가 있음을 나타내었다.

원유내 약물잔류의 원인에 관한 연구 (The reasons of drug residues in bulk milk)

  • The 102 farms received a positive result of the bulk milk drug residue test were selected to investigate the reasons of drug residues in bulls milk. The most frequent causes of drug residues were milker or producer mistakes (28.4%), failure to observe withdrawal time (21.5%), and withholding milk from treated quarters only (19.6%). Milker or producer mistakes occurred high at the farms having a parlor system (4 cases out of 11 farms), and related to the inadequate records and marking of treated cows. The lack of knowledge on the absorption of antibiotic from treated quarters and its excretion from untreated quarters caused mainly withholding milk from treated quarters only. Among the 91 farms identified the cause of drug residues, most of the route of drug administration was intramammary infusion (81.3%), and mostly drug used for the treatment of cows was ${\beta}$-lactam antibiotic (57.1%)

Analysis of antibiotic residues in milk from healthy dairy cows treated with bovine mastitis ointment using ultra-performance liquid chromatography coupled with electrospray tandem mass spectrometry

  • This study was conducted to analyze penicillin G (PEG), streptomycin (STR) and neomycin (NEO) residues in milk of healthy lactating cows. Milk samples were collected from all four quarters of 12 dairy cows 2−7 days after intramammary infusions of an ointment containing PEG, STR and NEO once (n = 4; group I) or twice (n = 4, group II) daily. Ultra-performance liquid chromatography coupled with electrospray tandem mass spectrometry was used to determine the antibiotic residues in the samples. The correlation coefficient ($r^2$) of the calibration curves for all antibiotics was > 0.999 and the limits of detection and quantification were $0.002-0.005{\mu}g/mL$ and $0.007-0.02{\mu}g/mL$, respectively. Recovery rates were ranged from 75.5 to 92.3%. In group I, PEG, STR and NEO residues were detected in milk at 2, 3 and 2 days post-treatment, respectively, which were below the maximum residue limit (MRL). In group II, PEG, STR and NEO residues were detected in milk at 2, 3 and 3 days post-treatment, respectively, which were bellow the MRL. These results suggest that a 3-day for milk withdrawal period after the ointment treatment might be sufficient for reduction of the antibiotic residues below the MRL.

Lab-on-a-Chip for Monitoring the Quality of Raw Milk

  • A lab-on-a-chip (LoC) was designed for simultaneous monitoring of microorganisms, antibiotic residues, somatic cells, and pH in raw milk. The LoC was fabricated from polydimethylsiloxane (PDMS) using microelectromechanical system (MEMS) technology, which consisted of two parts; a protein array and microchannel. The protein array was fabricated by immobilizing five types of antibodies corresponding to two microorganisms, two antibiotic residues, and somatic cells. A sol-gel film was deposited on a glass substrate to immobilize the antibodies. The target analytes in raw milk could be bound with the corresponding antibody by an immunoreaction, and the antigen-antibody complex was detected using fluorescence microscopy. SNARF-dextran was used as a pH indicator, and the SNARF-entrapped hydrogel was attached to the microchannel in the chip. After injecting the milk sample into the channel, the pH was measured by monitoring the change in fluorescence intensity by fluorescence microscopy. The on-chip simultaneous assay of two microorganisms (E. coli O157:H7 and Streptococcus agalactiae), two antibiotic residues (penicillin G and dihydrostreptomycin), and neutrophils was successfully accomplished using the proposed LoC system.

시료 중 잔류 항생제 분석 방법: II. 액상 시료 전처리 방법 (Determination of Antibiotic Residues: II. Extraction and Clean-up Methods for Liquid Samples_A Review)

  • Increased attention has been paid to the presence of veterinary antibiotics in various environmental matrices due to their toxicological behavior in the ecosystem and development of antibiotic-resistant strains of pathogenic bacteria. In the this review, 37 target antimicrobials were selected based on annual sales of antibiotics for livestock in South Korea 2014. Also, extraction and clean-up methods for the determination of the antibiotic residues in liquid samples including water, milk, and honey were comprehensively reviewed in the literature. Solid-phase extraction (SPE) was commonly used as a pre-treatment method for the samples. Most of the analytes were extracted in acidic conditions (2.5~4.0) except for aminoglycosides, which were extracted in neutral conditions (7.0~8.0). ${\beta}-Lactams$ showed the highest recoveries in neutral pH due to their degradation characteristics in acidic media. Starta-X, Oasis HLB, and Oasis MCX were frequently applied as an SPE cartridge and Oasis HLB showed the highest recoveries for the majority of antibiotic classes. The homogenized honey and milk were extracted by mixing with acids for deproteinization. Solids and other interfering substances in the extract were eliminated by centrifugation followed by membrane filtration or SPE before injection into HPLC.

Application of a Lateral Flow Immunoassay to Determine Ampicillin Residues in Muscle Tissue of Olive Flounder (Paralichthys olivaceus)

  • Antibiotic Detection Kit (Combination I), a lateral flow immunoassay (LFIA) developed for the detection of antibiotic residues in milk, was utilized for the analysis of antibiotic residues in the muscle tissue of olive flounder. After 60-min treatment by dipping in water dosed with ampicillin (200-g/ton water), the residue depletion of ampicillin was investigated in 25 cultured olive flounder (Paralichthys olivaceus). Muscles of fish were sampled on the 1st, 2nd, 3rd, 4th and 5th day after drug treatment. The concentration of ampicillin in the muscle was determined by LFIA. The absorbance ratio of the sample to the control blank (Bs/Bo) was employed as an index to determine the muscle residues in olive flounder. To investigate the recovery rate, standard solutions were added to muscle samples to give final concentrations in the muscle of 4 and 8 ng/ml. The recovery rates of all spiked samples were > 96% of the spiked value. Ampicillin was detected in the muscle of fish treated with the drug until the 2nd day of the withdrawal period. The present study showed that the LFIA can be easily adopted to predict ampicillin residues in tissue of farmed fishes.
