자초 추출액을 이용한 염색직물의 항균성 및 소취성 (The Antibacterial Activity and Deodorization of Fabrics Dyed with Lithospermi Radix Extract)

  • This study shows the colorfastness, antibacterial activity, and deodorizing effect about cotton and silk fabrics dyed with Lithospermi Radix Extract. In the case of colorfastness, the light colorfastness showed the lowest grade, the other colorfastness was relatively good. However there are some differences according to fabrics and mordants. In the case of antibacterial activity to Staphylococcus aureus, the cotton fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted SnC1$_2$ㆍ2$H_2O$, and the silk fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted A1($CH_3$COO)$_3$. In the case of deodorizing effect, the cotton fabrics showed the most deodorizing effect at the dyed fabrics that had been mordanted SnC1$_2$ㆍ2$H_2O$, and the silk fabrics showed the comparatively high antibacterial effect at the dyed fabrics that had been mordanted both SnCl$_2$ㆍ2$H_2O$ and Al($CH_3$COO)$_3$.

Antibacterial and Pharmacological Evaluation of Fluoroquinolones: A Chemoinformatics Approach

  • Fluoroquinolone (FQ) antibiotics are an important class of synthetic antibacterial agents. These are the most extensively used drugs for treating bacterial infections in the field of both human and veterinary medicine. Herein, the antibacterial and pharmacological properties of four fluoroquinolones: lomefloxacin, norfloxacin, ciprofloxacin, and ofloxacin have been studied. The objective of this study was to analyze the antibacterial characteristics of the different fluoroquinolones. Also, the pharmacological properties of the compounds including the Lipinski rule of five, absorption, distribution, metabolism, and excretion, LD50, drug likeliness, and toxicity were evaluated. We found that among all four FQ molecules, ofloxacin showed the highest antibacterial activity through in silico assays with a strong interaction (-38.52 kJ/mol) with the antibacterial target protein (topoisomerase-II DNA gyrase enzyme). The pharmacological and pharmacokinetic analysis also showed that the compounds ciprofloxacin, ofloxacin, lomefloxacin and norfloxacin have good pharmacological properties. Notably, ofloxacin was found to possess an IGC50 (concentration needed to inhibit 50% growth) value of $0.286{\mu}g/L$ against the Tetrahymena pyriformis protozoa. It also tested negative for the Ames toxicity test, showing its non-carcinogenic character.

Influence of the N- and C-Terminal Regions of Antimicrobial Peptide Pleurocidin on Antibacterial Activity

  • Pleurocidin, a 25-mer antimicrobial peptide, has been known to exhibit potent antibacterial activity. To investigate the functional roles in N- and C-terminal regions of pleurocidin on the antibacterial activity, we designed four truncated analogs. The antibacterial susceptibility testing showed that pleurocidin and its analogs exerted antibacterial effect against various bacterial strains and further possessed specific activity patterns corresponding with their hydrophobic scale [pleurocidin > Anal 3 (1-22) > Anal 1 (4-25) > Anal 4 (1-19) > Anal 2 (7-25)]. Fluorescence experiments using 1,6-diphenyl-1,3,5-hexatriene (DPH) and 3,3'-dipropylthiadicarbocyanine iodide [$diSC_3(5)$] indicated that the differences in antibacterial activity of the peptides were caused by its membrane-active mechanisms including membrane disruption and depolarization. Blue shift in tryptophan fluorescence demonstrated that the decrease in net hydrophobicity attenuates the binding affinity of pleurocidin to interact with plasma membrane. Therefore, the present study suggests that hydrophobicity in the N- and C-terminal regions of pleurocidin plays a key role in its antibacterial activity.

Antioxidant and Antibacterial Activity of Caprylic Acid Vanillyl Ester Produced by Lipase-Mediated Transesterification

  • Vanillyl alcohol (VA), which is abundant in Vanilla bean, has strong antioxidant activity. However, the use of VA in the food and cosmetics industries is limited, due to its low solubility in emulsion or organic solvents. Meanwhile, medium chain fatty acids and medium chain monoglycerides have antibacterial activity. We synthesized butyric acid vanillyl ester (BAVE) or caprylic acid vanillyl ester (CAVE) from VA with tributyrin or tricaprylin through transesterification reaction using immobilized lipases. BAVE and CAVE scavenged 2,2-diphenyl-1-picrylhydrazyl radicals in organic solvents. In addition, BAVE and CAVE decreased the production rate of conjugated diene and triene in the menhaden oil-in-water emulsion system. While BAVE showed no antibacterial activity, CAVE showed antibacterial activity against food spoilage bacteria, including Bacillus coagulans. In this study, the antibacterial activity of vanillyl ester with medium chain fatty acid was first revealed. Zeta potential measurements confirmed that BAVE and CAVE were inserted into B. coagulans membrane. In addition, the propidium iodide uptake assay and fluorescent microscopy showed that CAVE increased B. coagulans membrane permeability. Therefore, CAVE is expected to play an important role in the food and cosmetics industries as a bi-functional material with both antioxidant and antibacterial activities.

Synergistic Killing Effect of Synthetic Peptide P20 and Cefotaxime on Methicillin-Resistant Nosocomial Isolates of Staphylococcus aureus

  • The salt resistance of antibacterial activity and synergistic effect with clinically used antibiotic agents are critical factors in developing effective peptide antibiotic drugs. For this reason, we investigated the resistance of antibacterial activity to antagonism induced by NaCl and $MgCl_2$ and the synergistic effect of P20 with cefotaxime. P20 is a 20-residue synthetic peptide derived from a cecropin A (CA)-melittin(ME) hybrid peptide. In this study, P20 was found to have potent antibacterial activity against clinically isolated methicillin-resistant Staphylococcus aureus (MRSA) strains without hemolytic activity against human erythrocytes. The combination study revealed that P20 in combination with cefotaxime showed synergistic antibacterial activity in an energy-dependent manner. We also confirmed the synergism between P20 and cefotaxime by fluorescence-activated flow cytometric analysis by staining bacterial cells with propidium iodide (PI) and bis-(1,3-dibutylbarbituric acid) trimethine oxonol (BOX). This study suggests that P20 may be useful as a therapeutic antibiotic peptide with synergistic effect in combination with conventional antibiotic agents.

Streptomyces sp. J46의 세균성구멍병원균 Xanthomonas arboricola pv. pruni에 대한 항균 활성 (Antibacterial Activity of Streptomyces sp. J46 against Bacterial Shot Hole Disease Pathogen Xanthomonas arboricola pv. pruni)

  • BACKGROUND: Bacterial shot hole of stone fruits is a seriuos plant disease caused by Xanthomonas arboricola pv. pruni (Xap). Techniques to control the disease are required. In this study, microorganisms with antibacterial activity were isolated to develop as a microbial agent against the bacterial shot hole. METHODS AND RESULTS: An isolate with the strongest activity among the isolates was identified as Streptomyces avidinii based on 16S rRNA gene sequence analysis and designated Streptomyces sp. J46. J46 showed suppression of bacterial leaf spot with a control value of 90% at 10 times-diluted cell free supernatant. To investigate antibacterial metabolites produced by J46, the supernatant of J46 was extracted with organic solvents, and the extracts were subjected to chromatography works. Antibacterial metabolites were not extractable with organic solvents. Both reverse and normal phase techniques were not successful because the metabolites were extremely water soluble. The antibacterial metabolites were not volatiles but protein compounds based on hydrolysis enzyme treatment. CONCLUSION: Our study suggests that Streptomyces sp. J46 may be a potential as an microbial agent against bacterial shot hole. Further study to identify the metabolites is required in more detail.

국화 추출액을 이용한 염색직물의 염색성 및 항균성 (The Dyeability and Antibacterial Activity of Fabrics Dyed with Chrysanthemum indicum L. Extract)

  • This study shows the dyeability and antibacterial activity about cotton and silk fabrics dyed with Chrysanthem indicum L. In the case of dyeability, in the first, K/S value was shown comparatively high numerical value. The value of all test sample was over 20. In the next, in the case of colorfastness, the light colorfastness showed the lowest grade, the other colorfastness was relatively good. However there are some differences according to fabrics and mordants. In the case of antibacterial activity to Klebsielia pneumoniae, the cotton fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted $CuSO_4$.$5H_2O$, and the silk fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted $CuSO_4$.$5H_2O$.$Al_2$ and $(SO_4)_3$.

Simultaneous Determination of Benzoic Acid, Caffeic Acid and Chlorogenic Acid in Seeds of Eriobotrya japonica and their Antibacterial Effect

  • We aim to develop a simple method for simultaneous and quantitative determination of benzoic acid, caffeic acid and chlorogenic acid in seeds of Eriobotrya japonica. In addition, antibacterial effect of these three phenolic acids was examined. A basic method is performed on the high performance liquid chromatography system coupled to an UV-detector (230 nm) and reverse phase C-18 column ($4.6{\times}150mm$, $5{\mu}m$). Each phenolic acid was confirmed via liquid chromatography-mass spectrometry (MS)/MS system under the multiple-reaction monitoring with negative-ion electrospray ionization (ESI(-)) mode. It is demonstrated that the method was could be applied to samples for an analytical study of the phenolic acids. On the other hand, three phenolic acids in seeds of E. japonica exhibited antibacterial effect against several pathogenic bacteria. Of these, benzoic acid was found to have stronger antibacterial effect.

A Study on the Antibacterial Activity of Chitosan

  • Water-soluble chitosan and water-insoluble chitosan with molecular weight of 2,000,000, 500,000, 80,000, and 40,000 and more than 90% degree of deacetylation were controlled to evaluate the antibacterial activity of chitosan against a pathogenic bacteria, methicillin resistant Staphylococcus aureus(MRSA), which is being issued in the world. The Shake Flask Method and Modified Shake Flask Method were used to find out the antibacterial activities of 5types of chitosan/acetic acid solution, and the other antibacterial activities test with the cotton filter treated with chitosan /acetic acid solution. Those test methods showed the great differences ,but the results of the antibacterial activites showed the same difference.

방아(Agastache rugosa)로부터 소폐렴균에 대한 항균성 물질의 생산 (Production of Antibacterial Substance against Bovine Pneumoniae Bacteria from Agastache rugosa)

  • The objective of this study was to develop a new potent antibacterial compound against bovine pneumoniae bacteria from medicinal plants or herbs. Among 65 kinds of medicinal plants and herbs, ethanol extracts of Citrus unishiu showed the highest solid yield of $54\%$. However, ethanal extracts from Agastache rugosa had the highest antibacterial activities against bovine pneumoniae bacteria, Mannheimia haemolytica A and Haemophilus somnus (size of clear zone: 16.0 mm and 10.0 mm, respectively). The antibacterial compound was also maximally extracted when the powder of A. rugosa was treated with $70\%$ ethanol at $45^{\circ}C$ for 12 hours.
