A STUDY OF IONIC DISSOCIATION ON VARIOUS CALCIUM HYDROXIDE PASTES USING MOLECULAR SIEVING MODEL (분자체 모델을 이용한 수종의 수산화칼슘 제재의 이온 용출에 관한 연구)

  • The purpose of this study was two-fold. First was to evaluate whether the molecular sieving model was appropriate for ionic dissociation experiment. Second was to compare the dissociation of calcium and hydroxyl ions from five types of calcium hydroxide pastes (Pure calcium hydroxide paste, DT temporary $dressing^{\circledR},{\;}Metapaste^{\circledR},{\;}Chidopex^{\circledR},{\;}Metapex^{\circledR}$) in three vehicles (aqueous, viscous and oily) and the antibacterial effect. Each calcium hydroxide pastes was placed into 0.65ml tube with cap and then 15% polyacrylamide gel was placed onto calcium hydroxide pastes. After the gel was hardened, the tubes were filled with tridistilled water (pH 7.14) and closed with cap. The tubes were stored in $37^{\circ}C$ 100% incubator The pH reading and the concentration of calcium ions were taken at 1, 4, 7. 10, and 14 days. The brain heart infusion agar plates with S. mutans and A. actinomycetemcomitans were used far antibacterial activity test. Middle of agar plate was filled with the calcium hydroxide pastes. The plates were incubated at $37^{\circ}C$ and observations were made to detect the zones of inhibition. These data were evaluated statistically by use of the analysis of variance and duncan test. The results were as follows. 1. In fresh mixing state, the pH of five types of calcium hydroxide pastes were measured between 12.5 and 12.8. 2. The pH was increased in all five types of calcium hydroxide pastes compared with control group. In 14 days, Pure calcium hydroxide paste (11.45) and DT temporary $dressing^{\circledR}$ (11.33) showed highest pH, followed by $Metapaste^{\circledR}$ (9.49), $Chidopex^{\circledR}$ (8.37) and $Metapex^{\circledR}$ (7.59) 3. Calcium was higher in all five types of calcium hydroxide pastes compared with control group. In 14 days, Pure calcium hydroxide paste (137.29 mg%) and DT temporary $dressing^{\circledR}$ (124.6 mg%) showed highest value, followed by $Metapaste^{\circledR}$ (116.74 mg%), $Chidopex^{\circledR}$ (111.84 mg%) and $Metapex^{\circledR}$ (60.22 mg%). 4. The zones of bacterial inhibition were seen around all five types of calcium hydroxide pastes. $Chidopex^{\circledR}{\;}and{\;}Metapex^{\circledR}$ groups which include iodoform were observed significantly larger zone of inhibition in A. actinomycetemcomitans compared with the other calcium hydroxide groups (p<0.05) However, $Metapex^{\circledR}$ showed the least antibacterial effect on S. mutans compared with other groups (p<0.05). The molecular sieving model was found to be acceptable in dissociation experiment of hydroxyl and calcium ions when compared with the previous tooth model study. But this model was not appropriate for the antibacterial test.

  • For this study copper ions-containing bioactive glass nanoparticles commonly used in mineral trioxide aggregate (MTA) was developed to improve the mechanical properties and promote antibacterial effect of MTA with the original material. The mechanical properties and antibacterial activity of Cu-BGn incorporating varying amounts cooper incorporated bioactive glass nano particles(BGn) 0.5,1.0,2.0 and 4.0 wt% in MTA were characterized composition of the resulting were investigated. The compressive strength was calculated by weighing specimens with a diameter of 4 mm and a thickness of 6 mm according to ISO 6876 (2012). The antimicrobial effect was evaluated using two strains of S. mutans and E. faecalis. The mechanical properties of the test results was Cu-BGn increased no statistically significant difference was observed (p>0.05). Adhesion experiment results S. mutans in contrast to the control group Ortho MTA, 4.0 wt% of Cu-BGn added experimental group showed a significant difference was observed (p<0.05). Also, E. faecalis statistical analysis indicated a significant difference for antibacterial agents between control and Cu-BGn containing(p<0.05). It seems that this Cu-BGn proved that even a antibacterial effect was demonstrated. Therefore, it was suggest that it is necessary for in-depth research into various environments that can reproduce the oral environment.

Dyeability and Antibacterial Activity against Food Poisoning Bacteria Dyed with Geranium nepalense Extracts (이질풀 추출물로 염색한 면직물의 염색성과 항식중독균성)

  • This study was conducted to develop antibacterial hand towel and dishcloth against food poisoning bacteria, and to examine the dyeability and the antibacterial activity of cotton fabrics against food poisoning bacteria, which was dyed with 1 % extracts of Geranium nepalense at $50\;to\;60^{\circ}C$ minutes. Surface colors of cotton fabrics treated nothing, alum $[AIK(SO_4)_2{\cdot}nH_2O]$, and calcium hydroxide $[Ca(OH)_2]$ with mordants showed grayed-yellow group, that treated copper sulfate $(CUSO_4)$ with mordant was grey-brown group, and that treated sulfate of iron $(FeSO_4{\cdot}7H_2O)$ made grayed-green group. Hue value of cotton fabrics dyed with the extracts of Geranium nepalense and dried until twenty times was an order of descent Y, that dried thirty to forty an order of descent YR. Cotton fabrics dyed with the extracts of Geranium nepalense had the antibacterial activity against food poisoning bacteria such as Staphylococcus aureus, Escherichia coil and Salmonella typhimurium by over 99.99%. And that dried over thirty times showed the antibacterial activities against food poisoning bacteria by 52.91%.

Improvement of Hygienic Characteristics of Material for Patients Clothing through Treatment with Chitosan/Nanosilver Mixed Solution (키토산/은나노 혼합용액 처리에 의한 환자복 소재 위생 성능 향상)

  • In order to enhance the functionality of a cotton fabric actually used as the clothing material for patients, the fabrics are treated with chitosan/nanosilver mixed solution. The nanosilver has excellent biocompatibility, not causing an environmental pollution as a natural polymer, provides expectation of an additional performance, does not harm human beings, and shows a strong antibacterial activity even in a small amount, and supplements chitosan, which is disadvantaged if used alone for fabric treatment. This study evaluates functional improvement of the clothing material for patients and observes through hygienic characteristics which are the most important function. In antibacterial activity of the fabrics treated with chitosan/nanosilver mixed solution against Staphylococcus aureus, higher ratio of chitosan was observed to achieve better antibacterial activity. In antibacterial activity against Klebsiella pneumoniae, higher ratio of nanosilver was observed to achieve better activity. Regarding laundry durability of antibacterial activity after repeated laundering, activity against Staphylococcus aureus was little reduced, but was greatly lowered against Klebsiella pneumoniae. Deodorization activity was excellent as the mixed ratio of chitosan was increased, and air permeability, moisture permeability and moisture regain were reduced as the mixed ratio of nanosilver was increased.

Antibacterial Activities of Flower Tea Extracts against Oral Bacteria (꽃차용 꽃 추출물의 구강위생균에 대한 항균특성)

  • In this study, we analyzed flower tea activity against oral bacteria. Lagerstroemia indica, Paeonia suffruticosa and Hemerocallis fulva showed high extract yields. Bellis perennis, Punica granatum and Cercis chinensis showed the high rates of yield by ethanol extraction. Extract yield seemed to be related to the characteristics of the specimens rather than to the solvent. Streptococcus mutans, Streptococcus obrinus, Porphyromonas gingivalis and Prevotella intermedia were used to investigate extracts activity against bacteria; the former two cause dental caries and the latter two cause halitosis. Cornus officinalis, L. indica, P. granatum and P.s uffruticosa showed high antibacterial activities against S. mutans. In specimens extracted with ethanol, P. suffruticosa, Camellia sinensis, Camellia japonica L. and Rosa hybrida showed high antibacterial activities. L. indica, P. granatum and C. officinalis showed high antibacterial activities against S. sobrinus. C. officinalis, P. granatum, L. indica and P. suffruticosa showed high activities for specimens extracted with ethanol. The results show that the warm extracts of C. officinalis, L. indica and P. granatum may be effective to prevent dental caries. In particular, the ethanol-based extracts of C. officinalis, P. suffruticosa and C. sinensis were effective to prevent dental caries and thus may be highly marketable. Chrysanthemum zawadskii, R. hybrida, P. granatum, C. japonica L. and Zinnia elegans showed high antibacterial activity against P.gingivalis. R. hybrida showed the highest ethanol extract activity, followed by P. suffruticosa, P. granatum, C. japonica L. and L. indica. R. hybrida, P. granatum, C. morifolium and C. japonica showed high activity against Pr.intermedia in the order named. C. zawadskii, P. granatum, L. indica, C. japonica and A. princeps showed high ethanol extract activity. Thus, the warm extracts of R. hybrida, P. granatum and C. japonica may be helpful to reduce halitosis. In addition, the ethanol-based extracts of P. granatum, C. japonica and L. indica are expected to be highly marketable as mouthwashes.

Antibacterial and Antibiofilm Activities of Alnus japonica Stem Extract against Porphyromonas gingivalis (Porphyromonas gingivalis에 대한 오리나무 줄기 추출물의 항균활성 및 생물막 형성 억제 효과)

  • This study investigated the potential of dye plants as natural oral health products. The antibacterial activity of ethanol stem extracts of A. japonica, R. verniciflua Stokes, G. jasminoides, D. morbifera, P. amurense Rupr., and S. japonica against P. gingivalis KCTC 5352, S. mutans KCTC3065, S. downei KCTC3634, S. sanguinis KCTC3284, and S. gordonii KCTC 3286 was confirmed. Among the stem extracts from 6 dye plants grown in Korea, ethanol extract from A. japonica stem (1 mg/disc) showed the highest antibacterial activity against P. gingivalis KCTC5352. The A. japonica stem extracts showed antibacterial activity similar to chlorhexidine, which was used as a positive control. The MIC and MBC of P. gingivalis KCTC5352 were 0.4 mg/ml and 0.6 mg/ml, respectively. The biofilm production rate and cell growth of P. gingivalis KCTC5352 in the cultures treated with 0.2-2.0 mg/ml of A. japonica extract were significantly decreased in a concentration-dependent manner. In addition, the mRNA expression of the superoxide dismutase and fimA associated with fimbriae formation in these cultures was suppressed, also in a concentration-dependent manner. Based on these results, it is concluded that A. japonica stem extracts can be used as an oral health product derived from natural materials, as demonstrated by its antibacterial action against and inhibition of biofilm formation of P. gingivalis KCTC5352.

Comparison of Fatty Acids and Antibacterial Activity against Pathogen of Acne in Different Parts of Ripened Black Raspberry (Rubus coreanus Miquel) (완숙 복분자 부위별 지방산 조성과 여드름 원인균에 대한 항균 활성)

  • The purpose of this study was to obtain the fatty acid composition and the antibacterial activity against pathogen of acne in various parts including seed, flesh and fruit from ripened black raspberry. In fatty acid analysis of lyophilized black raspberry, total fat of dried seed (11.047%) was much higher than in other parts. Moreover, linoleic acid (5.577%) and linolenic acid (2.317%) identified as the major fatty acids in the seed. By contrast, highest fatty acid of dried flesh was palmitic acid (0.323%) but it was remarkably low level compared with fatty acid composition of dried seed. Total polyphenol content in seed extract and flesh extract were 123.64 mg/g and 55.43 mg/g, respectively. DPPH radical scavenging ability of seed extract was higher than flesh and fruit extract. In antibacterial activity experiment against strains related acne such as Propionibacterium acne and Staphylococcus epidermidis, antibacterial activity of flesh extract was stronger than seed extract. However, seed extract showed antibacterial activity of 76.96~89.08% against each strain compared to activity of flesh extract. Based on these results, we concluded that seed of black raspberry can be useful in related field.

Invitro and Virtual Screening of Bioactive Molecule from Mycelium of Trichoderma atroviride Inhibit the UDP-3-O-(R-3-hydroxymyristoyl)-N-acetylglucosamine Deacetylases (LpxC) for Treatment of Bacterial Infection

  • Trichoderma species are a rich source of metabolites, but less known for biomedical potential. This work deals with antibacterial and antioxidant potentials of intracellular non-cytotoxic metabolites, extracted from Trichoderma atroviride (KNUP001). A total of 53 fractions was collected by column chromatography and tested for cytotoxicity by MTT assay. Only one fraction (F41) was found to be non-toxic to Vero cells with $95.4{\pm}0.61%$ of survival. The F41 was then subjected to chemical analysis, antibacterial and antioxidant assays. The F41 at $500{\mu}g.ml^{-1}$ showed the total antioxidant of $48.70{\pm}2.90%$, DPPH radical scavenging activity of $37.25{\pm}2.25$, nitric oxide (NO) radical scavenging activity of $54.55{\pm}1.95$ and $H_2O_2$ radical scavenging activity of $43.75{\pm}3.21$. The F41 at $25{\mu}g.ml^{-1}$ displayed antibacterial activity against E. coli ($14.25{\pm}0.2mm$), P. mirabilis ($10.4{\pm}0.6mm$), S. dysenteriae ($18.6{\pm}03mm$), S. paratyphi A ($14.1{\pm}1.1mm$), E. aerogenes ($5.6{\pm}0.4mm$) and S. marcescens ($14.25{\pm}0.2mm$). GC-MS analysis revealed the dominant presence of oleic acid C 18.1 (63.18%), n-hexadecanoic acid (6.17%), and ethyl oleate (4.93%) and potent molecules such as 8-[(2E)-2-(3-hydroxybenzylidene)hydrazinyl]-1,3,7-trimethyl-3,7-dihydro-1H-purine-2,6-dione, 2-(Dimethylamino)ethyl (1Z)-N-hydroxy-2-(4-morpholinyl)-2-oxoethanimidothioate, Fluorene in the F41, and virtual study revealed that these molecules are likely responsible for the antibacterial activities of F41. Hence, further investigation deserves on purification and characterization of the active metabolites from T. atroviride strain KNUP001 towards developing molecular leads to effective antibacterial drugs, and non-toxic to host cells.

Effect of antibacterial substances produced by probiotic lactic acid bacteria on histamine formation in rennet curd (렌넷 커드 내 히스타민 생성에 관한 프로바이오틱 유산균이 생산한 항균 물질의 영향)

  • Purpose of the present study was to investigate the factors affecting the production of antibacterial substances and histamine in rennet curd prepared by inoculation of histamine-producing lactic acid bacteria (LAB) and probiotic LAB. Probiotic Lactobacillus sakei PIL52 and Lactobacillus plantarum FIL20 produced strong antimicrobial agents against histamine-producing bacteria Lactobacillus brevis LAS129, Enterococcus faecium SBP12, and Enterococcus faecalis SBP58. The lactic acid and crude bacteriocin produced from the probiotic LAB inhibited histamine-producing bacteria in a concentration-dependent manner. As the number of probiotic LAB inoculated for the production of rennet curd increased, the antibacterial activity against histamine-producing bacteria was elevated due to the increased amount of lactic acid and crude bacteriocin in the sample. The growth of probiotic LAB as well as histamine-producing bacteria was inhibited by addition of 10% NaCl, thus the antibacterial substances and histamine contents in rennet curd were significantly lower than those of the control (P < 0.05). Meanwhile, the histamine content was not significantly increased when the rennet curd prepared by mixing probiotic LAB and histamine-producing bacteria was stored at $25^{\circ}C$ for 5 days. However, the amount of histamine detected in the rennet curd was significantly (P < 0.05) increased because the antibacterial activity of the bacteriocin produced by the probiotic LAB was decreased at $20^{\circ}C$ for 20 days.

Antioxidant and Antibacterial Activity of Commercially Available Herbs in Korean Markets (국내 시판되는 허브류의 항산화 및 항균효과 검색)

  • In this study, we investigated the antioxidant and antibacterial activity of methanol extracts from 6 plants which were Chrysanthemum zawadskii Herb. var. latilobum (Maxim.) Kitamura (Gu-jeol-cho), Lavandula spica L. (Lavender), Rosmainus offcinals L. (Rosemary), Cymbopogon citrates (Lemongrass), Saussureae radix (Mok-hyang), Calendular officinalis L. (Calendular). Antioxidative effects of herbal extracts were measured by polyphenols, flavonoids contents and DPPH radical scavenging activity assay. We also evaluated the antibacterial activity against Staphylococcus aureus and Escherichia coli O157:H7. Methanol extracts from Gu-jeol-cho, lavender, rosemary and lemongrass showed high polyphenols contents as well as strong DPPH scavenging activity. In particular, rosemary extract contained highest polyphenol levels as $126.69{\pm}2.62{\mu}g/mg$ compared to other herbs. As for DPPH radical scavenging activities, $IC_{50}$ values of rosemary extracts were $6.23{\pm}0.58{\mu}g/mL$. The rosemary extracts also showed higher antibacterial effects against S. aureus and E. coli O157:H7. These results indicate that rosemary could be used as natural antioxidant and antibacterial agents.
