A Study on the Antibacterial and Antifungal Properties of Zeolite/Zinc-polypeptide Coated Polypropylene Film (Zeolite/Zinc-polypeptide를 코팅한 폴리프로필렌필름의 항균 및 항진균 특성에 관한 연구)

  • This study is for the application of functional antibacterial packaging to fresh food. Zeolite/Zinc-polypeptide was coated on PP film at concentrations of 5%, 10%, and 15%, degree of dispersion was verified through FESEM and FT-IR analysis. In addition, the antibacterial and antifungal properties of the films were analyzed according to the control group and the concentration of coating materials. As a result, the degree of dispersion of coating material was irregular but wide, depending on the concentration of Zeolite/Zinc-polypeptide on the surface of PP film. The antibacterial effect against E. coli was over 99.9%, and the growth of R. oryzae was inhibited about 70%. Therefore, it was confirmed that Zeolite/Zinc-polypeptide had antibacterial and antifungal properties against E. coli and R. oryzae even after coated on PP film. In conclusion, Zeolite/Zinc-polypeptide coating film is expected to be effective in preventing corruption and improving the shelf life of fresh food as a functional packaging material. In order to be applied to various fresh foods in the future, storage experiments are additionally required with temperature and humidity conditions according to fresh foods.

A Study on the Use of Human Antibacterial Peptide LL-37-derived FK-13 as a Cosmetic Preservative (인간 항균펩타이드인 LL-37 유래의 FK-13의 화장품보존제로 활용에 대한 연구)

  • Here, we conducted the study on the possibility of using FK-13, a short analog of human-derived antibacterial peptide LL-37, as a cosmetic preservative to discover a natural cosmetic preservative that is safe for human body. For the purpose, FK-13 composed of 13 amino acids was synthesized by solid-phase peptide synthesis, and purified using reversed phase-high performance liquid chromatography (RP-HPLC). The purity and molecular weight were confirmed by liquid chromatography-mass spectrometry (LC-MS) analysis. FK-13 showed high antimicrobial activity on the three gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, and Staphylococcus epidermidis), the three gram-negative bacteria (Escherichia coli, Salmonella typhimurium, and Pseudomonas aeruginosa), and also even the fungus Candida glabrata. FK-13 had a broad spectrum of antibacterial activity, showing a suitability as a cosmetic preservative. In addition, FK-13 showed high thermostability and higher antibacterial activity in a comparative test with existing natural herbal cosmetic and chemical preservatives. Therefore, as FK-13 is a safe material and has high antibacterial activity at a low concentration, it is likely to be applied as a peptide natural cosmetic preservative that can replace existing chemical preservatives.

  • The number of households with pets has increased in recent years. Consequently, interest in mites and their odors has increased. In this study, we investigated whether pyrethrum extract and water-soluble phytoncides are effective tick repellents, deodorizing agents, and antibacterial agents. In the tick-climbing test, the pyrethrum extract (23.33 points) and water-soluble phytoncide (22.00 points) showed a high repelling effect. To confirm the tick avoidance effect of the composite extract, a tick climbing test was conducted with a pest control extract, a composite extract (referred to as a sample) containing a water-soluble phytoncide, and a third-party product (control). The sample showed a high avoidance effect (13.00 points and Control 26.67 points. An indoor repellent efficacy test for ticks confirmed that the sample showed an excellent tick-repellent effect (88%) compared to the control (12%). As a result of a deodorization test, ammonia was shown to have a deodorizing effect of 98.3% and trimethylamine of 99.5%, whereas, in the antibacterial test, the E. coli, Staphylococcus aureus, and pneumonia groups showed an antibacterial effect of 99.9%. Combining the results of this study, it is judged that the complex extract including the above material has high utility value as a material that is effective in mite repellents, deodorization, and antibacterial activity.

Effect of Chitosan Treatment for Improving Physical Properties of Taegwondo Uniform (시판 태권도복의 기능성 증진을 위한 키토산 처리 효과)

  • Taegwondo uniforms need powerful durability in fabrics against friction and shock because of higher exercising power. Therefore, in this study we developed more comfortable Taegwondo uniform for functional fabrics(deodorization rate, antibacterial activity and static charge). Chitosan is a deacetylated product of chitin, has widely been used in the biomedical sector, food industry, and textile industry. For the purpose of this study, we used the chitosan, which was congenial to the human body. Then, we compared the differences between the chitosan treated fabrics and non treated fabrics of Taegwondo uniform(100% polyester, 65% polyester/35% cotton). Chitosan was dissolved in 1% acetic acid. Also, the fabrics were washed using distilled water. Afterward, its treatment with chitosan was completed by padding the sample to its wet pick-up at $80{\pm}5%$ and by heating $150^{\circ}$ for 3 minutes. The chitosan treated Taegwondo uniform was improved on deodorization rate than non treated uniform. And the chitosan treated Taegwondo uniform was improved remarkably the antibacterial activity in all samples. In the further researches, the static charge was reduced in chitosan treatment uniform.

Anti-Bacterial Effects of Basil Oil on Streptococcus mutans and Porphyromonas gingivalis (Streptococcus mutans와 Porphyromonas gingivalis에 대한 Basil Oil의 항균효과)

  • Purpose : The study objective was to assess the antibacterial activity of essential oil of basil against S. mutans and P. gingivalis and to explore its potential to prevent dental caries and peridontal disease. Method : Essential oil of basil, extracted using steam distillation, was diluted with triple distilled water and Tween 20 to generate samples at various concentration, that is 30%, 50%, and 70% (v/v). Strains of S. mutans and P. gingivalis were incubated in the medium under anaerobic condition. Broth microdilution susceptibility testing and plate incubation diffusion were utilized to determine the minimum inhibitory concentration (MIC) and to measure antibacterial activity, respectively. Result : An upsurge in antibacterial activity was seen to correlate with and increase in the concentration used for both bacterial strains, but was more significant with S. mutans. A statistically significant growth inhibition effect and reduction in the number of colonies was also observed with both strains dependent on the concentration used following 24 hours of incubation. Conclusion : Thus, the current study finding was that essential oil of basil was effective against dental caries and periodontal disease and could be used in dentifrice to help prevent oral disease.

Structure-Activity Relationships of Peptide Antibiotics with Improved Bacterial Cell Selectivity of Pseudin

  • Pseudin is a naturally occurring 24 amino-acid-residue antimicrobial peptide derived from the skin of paradoxical frog Pseud's paradoxa. It shows potency against the bacteria and antibiotic-resistant bacteria strain, but has high cytotoxicity against mammalian cell. In our previous study, substitution of $Pro^{11}$ for Gly (Ps-P) increased bacterial cell selectivity but decreased the antibacterial activity of pseudin. In this study, we designed pseudin analogue, Ps-4K-P with increased cationicity up to +7 in Ps-P by substituting Glu14, Gln10, Gln24, and Leu18 with Lys. Ps-4K-P showed improved potent antibacterial activity with high bacterial cell selectivity. We determined the tertiary structure of Ps-4K-P in the presence of DPC micelles by NMR spectroscopy and it has a hinge structure at $Pro^{11}$ followed by three turn helices from $Pro^{11}$ to $Val^{23}$ at the C-terminus. Amphipathicity with increased cationicity as well as helix-hinge-helix structural motif provided by introduction of a Pro at position $Gly^{11}$ are the crucial factors which confer antibacterial activity with bacterial cell selectivity to Ps-4K-P.

Synthesis, Characterization, and Antibacterial Applications of Novel Copolymeric Silver Nanocomposite Hydrogels

  • Copolymeric silver nanocomposite hydrogels were synthesized by using acryloyl phenylalanine (APA), N'-isopropylacrylamide (NIPAM) and crosslinked by N,N-methylene bisacrylamide (MBA) via radical redox polymerization. Present study allows entrapping silver nanoparticles into hydrogel networks. UV-visible spectroscopy and X-ray diffraction (XRD) studies confirmed the formation of silver nanoparticles in hydrogel matrix. 11% of weight loss difference between hydrogel and silver nanocomposite hydrogel is clearly indicates the formation and silver nanoparticles by thermo-gravimetrical analysis. The order of swelling capacity values of hydrogels and silver nanocmposite hydrogels were found to be in the order of placebo copolymeric hydrogel >Ag-copolymeric silver nanocomposite hydrogels. The particle size of silver nanoparticles was analysed and are in the range of 5 - 10 nm which has been confirmed by transmission electron microscopy (TEM) as well as particle size analysis. The silver nanocomposite hydrogel has shown very good antibacterial activity on gram-positive and gram-negative bacteriocides.

A Study on efficacy and safety of antibacterial(pefloxacin methanesulfonate) to cultured fish, Cyprinus caprio and Paralichthys olivaceus (양식어류(이스라엘 잉어, 넙치)에 대한 항균물질 pefloxacin의 효능 및 안전성에 관한 연구)

  • A study on quinolone antibacterial (pefloxacin methanesulfonate) was performed to use for the drug of fisheries. Petloxacin was proved excellent in antibacterial activity and resistance against fish pathogens when compared with the existing antibacterials. And any side effect was not observed during the period of indicated use. An outline of MIC(Minimal Inhibitory Concentration) was $1.6{\sim}6.4{\mu}g/ml$, $TLm_{48h}$ value were 380~420 ppm in Israeli carp(Cyprinus caprio) and 2100~2300 ppm in flounder(Paralichthys olivaceus). The residual time of fish body was less than 15 days. So we can treat some bacterial disease of fish by the dosage of 100 g/day/ton of fish body weight for 3 days and pefloxacin is thought to be used effectively and widely against most bacterial fish pathogens.

  • We have found out the relationship of nanoemulsion containing nano vitamin C, E and propolis and gingival disease. We've confirmed effect of nanoemulsion through the experiment of in vivo and in vitro. We tested cell viability of gingival fibroblast cells by MTT assay and mRNA appearance of interleukin-$1{\beta}$, using mouse that was guided inflammation. Anti-microbacterial activity for Antibacterial effect's experiment was carried out by using S.aureus and E.coli. In addition, inflammation tissue has been observed with scanning electrical microscopy. In this study, expression of interleukin-$1{\beta}$ was decreased after adding nanoemulsion containing nanovitamin C, E and propolis. We've also obtained good results from the test of Antibacterial effect against S.aureus and E.coli. Also, swelling of inflammation tissues observed by scanning electrical microscopy has gone down. In conclusion, we have gained confidence that nanoemulsion containing nano vitamin C, E and propolis has very high Antibacterial effect against bacteria in oral. And it made us guess that inflammation of gingival reduces after decreasing interleukin-$1{\beta}$. Thus, we expect that nanoemulsion containing nano vitamin C, E and propolis gives good effects to patient having gingival disease.

A STUDY ON THE ANTIBACTERIAL EFFECT OF RESTORATIVE MATERIALS (수복재의 항균효과에 관한 연구)

  • The purpose of this study was to investigate the antibacterial effect of zinc oxide eugenol(ZOE), zinc phosphate cement(ZPC), glass ionomer cement, resin, and Vitapex to S. muntans, S. sanguis, S. fecalis and E. coli by agar diffusion method. Four wells were punctured in mitis-salivarius agar plate per each group and each wells were filled with restorative matetials. The width of inhibition zones produced in mitis - salivarius agar were measured as the parameter of the antibacterial effect after 16 hours and 40 hours. In S. mutans and S. sanguis, the largest inhibition zone was produced on ZOE, followed by glass ionomer cement, and ZPC. Inhibition zones was not observed in resin and Vitapex. In S. fecalis, ZOE and glass ionomer cement showed wider inhibition zone than ZPC. In E. coli, ZOE showed wider inhibition zone than ZPC, but no inhibition zone was observed on glass ionomer cement.
